Identification of a Novel Ibuprofen Biotransformation Pathway in Streptomyces sp. D218 and Detoxification as Indicated by the Green Algae Scenedesmus obliquus

Junwei Huang,Zilin Zhu,Ruomu Chen,Dandan Pan,Qing X. Li,Xiangwei Wu

Ibuprofen, a widely used nonsteroidal anti-inflammatory drug, contaminates agricultural products and potentially threatens human health due to its frequent detection and poor biodegradability. Microbial metabolism dominates the elimination of residual ibuprofen in the environment. In mineral salt medium at pH 6 with 5 mM glucose, Streptomyces sp. D218 transformed ibuprofen concentrations ranging from 0.05 to 0.40 mM in 24 h. The optimal temperature, pH, and initial OD600 nm for ibuprofen transformation by strain D218 were 25–37 °C, 5.0–6.0, and 1.0–1.5, respectively. Strain D218 could simultaneously transform ibuprofen into the intermediates 2-hydroxyibuprofen and ibuprofen amide (IBUA). The two intermediates were further metabolized to 2-hydroxyibuprofen amide (2HIBUA), thus relieving the growth inhibition of ibuprofen in Scenedesmus obliquus. This is the first complete pathway reported for the detoxification of ibuprofen transformation by a Gram-positive strain. These findings further our understanding of the microbial catabolism of the IBU.

Differential effects of oxytetracycline on detoxification and antioxidant defense in the hepatopancreas and intestine of Chinese mitten crab under cadmium stress

Lin Zeng,Yong-Hong Wang,Chun-Xiang Ai,Bin Zhang,Hui Zhang,Zi-Ming Liu,Min-Hui Yu,Bing Hu

This study aims to evaluate the effects of oxytetracycline (OTC) on detoxification and oxidative defense in the hepatopancreas and intestine of Chinese mitten crab ( Eriocheir sinensis ) under cadmium (Cd) stress. The crab was exposed to 0.6 μM Cd, 0.6 μM OTC, and 0.6 μM Cd plus 0.6 μM OTC for 42 days. Our results showed that in the intestine, OTC alone enhanced protein carboxylation (PC) and malondialdehyde (MDA) contents, which was associated with the increased OTC accumulation. Compared to Cd alone, Cd plus OTC increased Cd and OTC contents, and reduced detoxification (i.e., glutathione (GSH) content, gene expressions of cytochrome P450 ( CYP ) isoforms, 7-ethoxyresorufin O-deethylase (EROD) activity, mRNA levels and activities of glutathione peroxidase (GPx), glutathione reductase (GR) and glutathione-S-transferase (GST)), and antioxidant defense (i.e., gene expressions and activities of catalase (CAT) and superoxide dismutase (SOD)) in the intestine, leading to the increased in PC and MDA contents, suggesting that OTC had a synergistic effect on Cd-induced oxidative damage. In the hepatopancreas, although OTC alone increased OTC accumulation, it did not affect PC and MDA contents. Compared to Cd alone, Cd plus OTC reduced MDA content, which was closely related to the improvement of detoxification (i.e., GSH content, mRNA levels of CYP isoforms, EROD activity, gene expressions and activities of GPx, GR and GST), and antioxidant defense (gene expressions and activities of CAT and SOD, metallothionein content). Aryl hydrocarbon receptor ( AhR ) and nuclear factor E2-related factor 2 ( Nrf2 ) transcriptional expressions were positively correlated with most detoxification- and antioxidant-related gene expressions, respectively, indicating that AhR and Nrf2 were involved in the regulation of these gene expressions. Our results unambiguously demonstrated that OTC had tissue-specific effects on Cd-induced toxicological effect in E. sinensis , which contributed to accurately evaluating Cd toxicity modulated by TCs in crab.

Geopolymer sound-absorbing materials from municipal solid waste incineration fly ash and foundry ash: Dioxin detoxification, heavy metal stabilization and acoustic performance

Meiling Guan,Defu Xu,Zanmei Chen,Ying Zhuang,Jinhai Li,Mingyang Sun,Alan Howard

Sound-absorbing materials are widely used to mitigate noise pollution. In this study, a geopolymer sound-absorbing material was prepared using municipal solid waste incineration fly ash (MSWIFA) and foundry ash (FA), and its dioxin and heavy metal characteristics were investigated. The toxicity equivalent quantity (TEQ) of dioxins in raw MSWIFA was 78 ng TEQ kg−1, with 2,3,4,7,8-PeCDF contributing 34 ng TEQ kg−1. Detoxification using calcium hydroxide combined with pyrolysis at 400°C reduced the TEQ in MSWIFA to 0.62 ng TEQ kg−1, achieving a removal rate of 99.21%. A geopolymer was prepared by mixing low-temperature detoxified fly ash (LTDFA, pyrolysed at 400°C) with activated foundry ash (AFA, calcined at 800°C) at a 1:1 mass ratio, resulting in LTDFA-FABG with a TEQ of 12 ng TEQ kg−1 – well below the 50 ng TEQ kg−1 limit specified in the Technical Specification for Pollution Control of Fly Ash from Municipal Solid Waste Incineration (HJ 1134-2020). Compared to LTDFA-FABG, the geopolymer prepared using LTDFA, AFA and lime (LA-LTDFA-FABG) showed significantly reduced leachate concentrations of Cd and Mn, from 0.115 and 0.206 mg L−1 to below detection limits. LA-LTDFA-FABG exhibited a sound absorption bandwidth of 400–1000 Hz, a noise reduction coefficient of 0.49, and a compressive strength of 0.34 MPa, representing increases of 28.9% and 13.3%, respectively, over a commercial sound-absorbing panel. With an average pore size of 13.71 nm and a closed-pore structure, LA-LTDFA-FABG enhances contact and friction between sound waves and air, improving low-frequency sound absorption. These findings demonstrate that MSWIFA and FA can be effectively combined to produce geopolymer sound-absorbing materials, offering a promising route for the resource utilization of MSWIFA.

The voltage-gated sodium channel mutation and detoxification enzymes associated with the resistance of Grapholita molesta (Busck) (Lepidoptera: Tortricidae) to lambda-cyhalothrin

Xiaohe Zhang,Sha Su,Caiyun Qiu,Bufei Ma,Zheng Zhao,Xiong Peng,Jaime C. Piñero,Maohua Chen

The oriental fruit moth , Grapholita molesta (Busck) (Lepidoptera: Tortricidae), a major fruit pest, is primarily controlled with pyrethroid insecticides such as lambda-cyhalothrin and resistance has emerged as a serious threat to management. In this study, we compared a lambda-cyhalothrin-resistant strain (RS) with a susceptible strain (SS) of G. molesta . Sequencing of the voltage-gated sodium channel (VGSC) gene revealed the L1014F mutation exclusively in RS individuals, strongly associated with knockdown resistance. Bioassays showed that the RS exhibited a 13.1-fold cross-resistance to deltamethrin and a 6.0-fold cross-resistance to bifenthrin, as well as moderate cross-resistance to chlorantraniliprole, abamectin, and emamectin benzoate. While showing only low or negligible cross-resistance to other insecticides tested. Synergist assays indicated that piperonyl butoxide (PBO) increased lambda-cyhalothrin toxicity by 1.72-fold in RS, implicating cytochrome P450 as key contributors. Biochemical assays confirmed significantly elevated activities of glutathione S-transferase (GST) and P450 in RS relative to SS. Transcriptomic analysis identified 758 differentially expressed genes, including 13 P450s, 5 GSTs, and 2 carboxylesterases (CarEs), with qRT-PCR validating strong up-regulation of CYP333B121 (14-fold), CYP6B230 (15-fold), CYP6AB117 (13-fold), and GSTs2 (5-fold). These findings suggest that the target-site resistance conferred by the L1014F kdr mutation, together with metabolic resistance mediated by overexpressed P450 and GST genes, jointly contribute to the resistance of G. molesta to lambda-cyhalothrin.

Genome-wide identification, characterization, and expression profiling of ATP-binding cassette (ABC) transporter genes potentially associated with abamectin detoxification in Cydia pomonella

Di Ju,Youssef Dewer,Shipan Zhang,Chao Hu,Peirong Li,Xueqing Yang

The codling moth Cydia pomonella L. (Lepidoptera: Tortricidae) is one of the most notorious pests of pome fruits and walnuts worldwide, which has developed resistance to almost all classes of insecticides, including abamectin (ABM). ATP-binding cassette (ABC) transporters are thought to play a vital roles in insecticide detoxification by reducing the toxic concentrations of insecticides in an organism tissues. Despite the tremendous progress in understanding the detoxification mechanisms at the molecular level, the physiological functions of ABC transporters in insects have been poorly investigated. In this study, we found that the ABC inhibitor verapamil synergized significantly the toxicity of ABM, suggesting a potential role of ABC in detoxification. A total of 54 ABC genes were identified in the third-instar larvae of C. pomonella after treatment with sublethal doses (LD 10 and LD 30 ) of ABM. The expression profile of these genes in ABM-treated larvae at different time points (24, 48, 72 hr) using transcriptomic analysis (RNA-seq) was also investigated. The results showed that the expression of about 30 ABC genes was significantly co-upregulated after treatment. Several specific genes were up-regulated at 48 hr after treatment of larvae with LD 10 ABM. Among these up-regulated genes, we found that the relative expression level of the CPOM19553 was 29.7-fold and 16.0-fold higher when larvae were exposed to ABM at the LD 10 and LD 30 doses compared to control, respectively. Unlike other ABC genes, only CPOM08323 exhibited significant expression levels in the head and cuticle of the third-instar larvae of C. pomonella exposed to the two sublethal doses of ABM, with no expression was observed in the detoxification tissues such as midgut and Malpighian tubule. This study suggests that these up-regulated genes may be involved in ABM resistance in C. pomonella . Our findings will provide an additional information required for further analysis of ABC transporter genes associated with xenobiotic metabolism in C. pomonella .

A MOF-doped molecularly imprinted polymer/MOF hybrid gel incorporating with pH-buffering sodium acrylate for practical detoxification of organophosphorus nerve agents

Peng Jiang,Yalin Niu,Junpeng Cao,Dong Xie,Jiaqi Li,Tianying Guo

The fatal toxicity of organophosphorus nerve agents (ONAs) and the realistic condition of chemical attack occurring place trigger the urgent need to develop catalyst materials with degrading ability under the natural environment without bulk water and bases. Here, we strategically design a molecularly imprinted polymer (MIP)/MOF hybrid catalyst material by integrating enzyme-mimic MIP with Zr-MOF UiO-66-NH 2 which demonstrate promising catalytic performance for degradation of ONAs. Furthermore, sodium acrylate functions as a hydrophilic and alkaline functional unit together with pyridine-amidoxime based functional monomers (PAAO) to copolymerize into molecularly imprinted polymeric network. The basic sodium carboxylate groups were installed near the active sites to provide appropriate alkaline microenvironment, non-bulk water and acid buffer capacity for the catalysis in pure water. The resultant MIP/MOF composite catalyst demonstrate outstanding catalytic activities in both liquid and solid phase hydrolysis of ONAs. Specifically, the half-life of liquid phase hydrolysis catalyzed by MIP/UiO-66-NH 2 -0.5 achieves 9.4 min for diethyl-4-nitrophenyl phosphate (DENP) and 10 min for dimethyl 4-nitrophenyl phosphate (DMNP), and solid-state hydrolysis present rapid degradation efficiency with a half-life of 28.6 min. In addition, composite catalyst MIP-Ag/UiO-66-NH 2 -0.5 that retains substrate selectivity of MIP, enables the simultaneous degradation of two types of organophosphorus substrates (phosphate and thiophosphate) by MIP and MOF in a synergistic way. Summarily, this work displays promising potential of MIP/MOF hybrid enzyme-like catalyst as effective and practical material for the practical protection and detoxification of organophosphorus nerve agents.

Leveraging oxygen-vacancy-rich Yb-MnO2/N-carbon quantum dots heterostructure for solar light-driven tetracycline detoxification and antibiotic-resistant bacteria inactivation

Zhiquan Chen,Didi Yang,Zhenda Liang,Yongtao Xu,Yijun Pang,Gang Xu,Chaoxiang Chen,Kunyu Ma,Li Zhou,Bing Yan

Microbial pollution and antibiotic contamination can pose a serious threat to human health. The rational development of efficient and low-cost methods is critical to solving these problems. In this study, novel Z-scheme heterojunctions consisting of Yb-MnO 2 and nitrogen-doped carbon quantum dots (N-CQDs) were fabricated to achieve efficient photocatalytic degradation of tetracycline (TC) and the destruction of bacteria ( Escherichia coli used as a model). Results showed that the Yb-doping strategy improves the characteristics of host MnO 2 by weakening the lattice interface and increasing its surface area. Compared with Yb-MnO 2 , Yb-MnO 2 /N-CQDs heterostructures exhibited superior photocatalytic activity in TC degradation under simulated solar light. The remarkable photocatalytic and mild oxidation abilities of Yb-MnO 2 /N-CQDs led to an impressive 97.88% removal rate of TC. Additionally, the structure displayed robust resistance to various ions and broad pH levels. Notably, the Yb-MnO 2 /N-CQDs heterostructures exhibited excellent bactericidal activity, with no observed bacterial resistance even after 20 consecutive trials. Furthermore, quenching experiments demonstrated that 1 O 2 and O 2 − were the primary radicals involved in the Yb-MnO 2 /N-CQDs-based photocatalysis process. The remarkable photocatalytic activity of Yb-MnO 2 /N-CQDs can be attributed to two key factors. Firstly, the large surface area and abundant oxygen defects provide rich active sites for reactant degradation. Secondly, the Z-scheme heterojunctions formed between Yb-MnO 2 and N-CQDs effectively facilitate the separation of photogenerated electron-hole pairs and increase the reduction/oxidation potentials for reactive oxygen species generation. This study presents a promising novel photocatalyst for the facile and energy-saving treatment of antibiotic and microbial pollution, and it may provide new insight into the design of highly efficient heterogeneous photocatalysts.

Carbon nitride-doped melamine-silver adsorbents with peroxidase-like catalysis and visible-light photocatalysis: Colorimetric detection and detoxification removal of total mercury

Mengyuan Yin,Yuqi Wan,Shuai Li,Xiaoting Zhao,Wenwen Zhang,Yun Zhang,Hua Wang

Mesoporous melamine-silver (MA-Ag) nanocomposites doped with carbon nitride quantum dots (CN QDs ) were fabricated simply by the controlled supramolecular self-assembly. It was discovered that the resulting nanoflower-like CN QDs @MA-Ag nanocomposites could exhibit the peroxidase-like catalysis, which could be specifically enhanced by Hg 2+ by forming [email protected] alloys. A double catalysis-based colorimetric method was thereby developed for the fast detecting of Hg 2+ and Hg 0 in wastewater samples, with the levels down to 0.050 nM and 18.3 nM, respectively. Moreover, strong visible-light-driven photocatalysis of the nanocomposites was demonstrated for oxidizing Hg 0 into Hg 2+ through photocatalytic H 2 O 2 production so as to realize the detoxification of Hg 0 in the environmental wastewater. Besides, the fabricated mesoporous CN QDs @MA-Ag nanocomposites with large specific surface areas might facilitate the high Hg adsorption through the powerful MA-Hg chelate interaction, showing the efficient adsorption and/or removal of total Hg. The catalysis-selective colorimetric analysis and photocatalysis-based detoxification removal of total mercury may promise for wide applications in the environmental monitoring and wastewater treatment of toxic heavy metals of mercury.

Sublethal Effects of Abamectin on the Development, Reproduction, Detoxification Enzyme Activity, and Related Gene Expression of the Oriental Fruit Moth (Lepidoptera: Tortricidae)

Su Sha,Jian Chengzhi,Zhang Xiaohe,Fang Sensen,Peng Xiong,Piñero Jaime C,Chen Maohua

Grapholita molesta is one of the most important fruit pests worldwide. Abamectin is a biological pesticide frequently used to control fruit borers like G. molesta in part owing to its translaminar properties. In this study, we characterized the toxicity of abamectin to G. molesta larvae using the diet incorporation method. The sublethal effects of abamectin on the development, reproduction, detoxification enzyme activity, and related gene expression of G. molesta were assessed. The results showed that the LC20 and LC50 values of the insecticide against G. molesta 72 h post-treatment were 1.17 mg L–1 and 5.85 mg L–1, whereas the LC20 and LC50 values 96 h post-treatment were 0.34 mg L–1 and 3.63 mg L–1. When compared to the control, sublethal concentrations of abamectin 1) significantly increased the mortality of the larvae, prepupae, and pupae of G. molesta, 2) prolonged the duration of 3rd to 5th instar larva, prepupal and pupal periods, 3) shortened the longevity of adults, and 4) reduced female fecundity. The enzymatic activity of glutathione S-transferase (GST) varied significantly after exposure to sublethal concentrations of abamectin, but the cytochrome P450 monooxygenases and carboxylesterase activity were not significantly affected. Thirteen of the 25 GST genes were significantly upregulated under different sublethal concentrations of abamectin. The combined findings increase our understanding of the effects of abamectin on G. molesta and the potential role of GSTs in the metabolic interactions of abamectin in this pest, and have applications for more rational and effective use of abamectin to control G. molesta.

Facile preparation of UiO-66-NH2-stabilized high internal phase molecularly imprinted polymers for synergistic catalytic hydrolysis detoxification of organophosphate nerve agents

Yalin Niu,Peng Jiang,Tianying Guo

Developing high-performance artificial enzyme materials with practical applications in efficient catalytic hydrolysis detoxification of organophosphate (OP) nerve agents remains a formidable challenge. Herein, UiO-66-NH2-stabilized Pickering high internal phase molecularly imprinted polymer systems (MOFs@PHIPMIPs) were developed using OPs as template molecules. Thus, UiO-66-NH2-doped high-internal-phase MIP composites were obtained, which integrate MIPs and MOFs successfully, showing remarkable catalytic hydrolysis activity towards OPs. In addition, by introducing methyl methacrylate (MMA) during copolymerization, the MOFs@PHIPMIPs would effectively acquire pH buffering properties. The multi-porous composite catalysis system exhibits an exceptionally high detoxification efficiency towards dimethyl-4-nitrophenyl phosphate (DMNP) in pure water (kobs = 0.9162 min−1, t1/2 = 0.76 min). This work presents a new strategy for the preparation of artificial phosphatase materials with high-performance catalytic detoxification of OPs with great application prospects.

Synergistic detoxification efficiency and mechanism of triclocarban degradation by a bacterial consortium in the liver-gut-microbiota axis of zebrafish (Danio rerio)

Mingyu Sang,Shuyu Liu,Haohao Yan,Bing Zhang,Siyuan Chen,Bowen Wu,Tian Ma,Hanyi Jiang,Peichao Zhao,Guanjun Sun,Xinyan Gao,Hailian Zang,Yi Cheng,Chunyan Li

Triclocarban (TCC), an emerging organic contaminant, poses a potential threat to human health with long-term exposure. Here, Rhodococcus rhodochrous BX2 and Pseudomonas sp. LY-1 were utilized to degrade TCC at environmental related concentrations for enhancing TCC biodegradation and investigating whether the toxicity of intermediate metabolites is lower than that of the parent compound. The results demonstrated that the bacterial consortium could degrade TCC by 82.0% within 7 days. The calculated 96 h LC 50 for TCC, as well as its main degradation product 3,4-Dichloroaniline (DCA) were 0.134 mg/L and 1.318 mg/L respectively. Biodegradation also alleviated histopathological lesions induced by TCC in zebrafish liver and gut tissues. Liver transcriptome analysis revealed that biodegradation weakened differential expression of genes involved in disrupted immune regulation and lipid metabolism caused by TCC, verified through RT-qPCR analysis and measurement of related enzyme activities and protein contents. 16 S rRNA sequencing indicated that exposure to TCC led to gut microbial dysbiosis , which was efficiently improved through TCC biodegradation, resulting in decreased relative abundances of major pathogens. Overall, this study evaluated potential environmental risks associated with biodegradation of TCC and explored possible biodetoxification mechanisms, providing a theoretical foundation for efficient and harmless bioremediation of environmental pollutants.

Mechanistic Insights into Aflatoxin B1 and M1 Degradation by Bacillus subtilis HNGD-Mq02 from Wheat Koji and Its Application in Food Detoxification

Ranran Ma,Weiwei Peng,Yanli Xie,Nazish Muzaffar,Weibin Ma,Yuhui Yang,Qian Li,Hang Jia

Aflatoxin B1 (AFB1), the most toxic and prevalent mycotoxin in food and feed, and its hydroxylated metabolite aflatoxin M1 (AFM1), common in dairy products, pose serious health risks. Effective degradation of these toxins is thus a key goal in food safety. In this study, Bacillus subtilis HNGD-Mq02 was isolated and assessed for its ability to degrade AFB1 and AFM1. Under optimal conditions (50 °C, pH 7), the fermentation broth achieved 94.06 and 98.26% degradation, respectively. Ultrahigh-performance liquid chromatography–mass spectrometry (UHPLC-MS) analysis confirmed that both mycotoxins were converted into less-toxic compounds. Zebrafish assays and Ames tests further verified the reduced toxicity of the degradation products. In practical application, the strain reduced AFB1 by 72.90% in corn flour and AFM1 by 42.25% in milk powder. Fermentation also led to partial improvements in nutritional quality. These results highlight the potential of B. subtilis HNGD-Mq02 for enhancing the safety and value of food and feed products.
