Switchable temperature-responsive deep eutectic solvents: A sustainable strategy for polyphenol extraction from sorghum bran

Jing Su,Xue Mi,Jianming Wang,Huihui Lu,Jieru Li,Yizhen Wu,Meimei Pan,Xu Jing,Jun Wang

Innovative TRDES-UAE for green polyphenol extraction from sorghum bran • Higher phenolic yields than conventional solvents & control DES • Enhanced bioactivity linked to SEM/molecular docking findings • Excellent recyclability (>85% efficiency over multiple cycles) This study developed a green extraction method using a switchable temperature-responsive deep eutectic solvent (TRDES) combined with ultrasound-assisted extraction (UAE) for polyphenol recovery from sorghum bran. TRDES formation was confirmed by FT-IR. Under optimized conditions, TRDES-UAE significantly enhanced the total polyphenol yields (22.14 mg/g DW), total flavonoid yields (24.71 mg/g DW), and total proanthocyanidins yields (29.47 mg/g DW), representing 1.15- to 6.02-fold higher yields than conventional solvents and a conventional DES. SEM revealed pronounced plant matrix disruption by TRDES, and molecular docking confirmed strong binding affinities between TRDES and target polyphenols. The TRDES extract exhibited potent antioxidant activity—DPPH IC 50 : 90.39 μg/mL; ABTS + IC 50 : 101.55 μg/mL; superoxide anion IC 50 : 117.30 μg/mL; hydroxyl radical IC 50 : 124.24 μg/mL; FRAP value: 1081.29 μmol Fe 2+ /L—as well as strong antibacterial activity, with MIC values of 0.15 mg/mL against Staphylococcus aureus and Escherichia coli , and 0.10 mg/mL against Salmonella enterica . Six greenness metrics validated the environmental benefits, and TRDES maintained >85% extraction efficiency over five consecutive reuse cycles. This work establishes TRDES-UAE as a sustainable strategy for sorghum bran upcycling with potential for functional food applications.

Orchestrated Engineered Polyphenol-Peptide Condensates Coupled With β-GP Reverses Diabetic Osteoporosis by Remodeling Mitophagy

Xiuyun Xu,Meiqin Zhang,Ting Wu,Erfan Wei,Andrei Y. Hancharou,Zeying Wang,Zijuan Wang,Letian Lv,Likun Wu,Xingtong Pan,Qiyue Zhu,Xinyi Dong,Hao Liu,Yongsheng Zhou

Diabetic osteoporosis (DOP) is a chronic complication of diabetes mellitus characterized by reduced bone mass, disrupted microarchitecture, and an elevated fracture risk. Persistent oxidative stress and inflammation further inhibit osteogenesis and angiogenesis, accelerating bone degeneration. In this study, we used a peptide-polyphenol conjugation strategy to develop a multifunctional colloidal nanoplatform (β-GP@EGCG-E7). The bone-targeting peptide E7 was covalently conjugated with epigallocatechin-3- gallate (EGCG) and subsequently loaded with β-glycerophosphate (β-GP). Sequentially, this nanoplatform integrates bone-targeted delivery, improving mitochondrial quality by anti-inflammation and antioxidation, as well as osteogenesis and angiogenesis within a single system. In detail, in a high glucose microenvironment, the nanoplatform selectively accumulates in bone lesions, scavenges excess intracellular reactive oxygen species (ROS) triggered by high glucose, clears damaged mitochondria by activating PINK1-Parkin mediated mitophagy, and exerts anti-inflammatory effects to rebalance the microenvironment of bone regeneration. Concurrently, sustained phosphate release supports mineralization, promotes osteogenic differentiation, enhances angiogenesis, and improves local microcirculation. In diabetic osteoporotic models, β-GP@EGCG-E7 significantly reduced oxidative stress, restored mitochondrial homeostasis, and promoted bone and vascular regeneration. This study provides a promising therapeutic strategy for DOP and highlights the potential of peptide-polyphenol hybrid nanomaterials for regenerative applications.

Metal-polyphenol network-encapsulated Prussian blue nanoparticles for enhanced photothermal/chemodynamic combined therapy of cancer

Yuqi Feng,Jinyu Wang,Tao Liao,Bojie Li,Qi Yu,Ying Kuang,Cao Li

Cancer remains a significant global health concern. Traditional treatments, such as surgery, chemotherapy, and radiation therapy, often cause severe side effects due to their lack of specificity. In recent years, nanomedicine-based approaches, particularly photothermal therapy (PTT) and chemodynamic therapy (CDT), have emerged as promising strategies for the precise treatment of tumors. However, the efficacy of PTT is often limited by the overexpression of heat shock proteins (HSPs) in cancer cells, which confer thermoresistance. To address this issue, we developed a multifunctional nanosystem called PB@MPN. This nanosystem encapsulates Prussian blue nanoparticles (PB NPs) within a metal-polyphenol network (MPN) formed from Fe 3 + and (-)-epigallocatechin gallate (EGCG). This system enhances photothermal conversion and enables CDT via Fenton reactions catalyzed by Fe 2+ , which is reduced from Fe 3+ by EGCG. Furthermore, EGCG effectively suppresses HSP expression, overcoming thermoresistance and improving therapeutic outcomes. In vitro and in vivo studies have demonstrated that PB@MPN exhibits excellent biocompatibility and significantly promotes tumor ablation under near-infrared (NIR) laser irradiation. These results suggest that PB@MPN has great potential as a versatile nanoplatform for combined photothermal and chemodynamic cancer therapy.

Quantification of polyphenol composition and multiple statistical analyses of biological activity in Portuguese red wines

Bai Shuang,Cui Can,Liu Jing,Li Pei,Li Qing,Bi Kaishun

The objective of the research was to evaluate potential biological activities of Portuguese red wines with different grape types on the level of composition characterization. An ultrafast liquid chromatography coupled with triple quadrupole mass spectrometry method was validated and used for qualitative analysis of commercial red wine samples through simultaneously determining 31 major active components. Meanwhile, the antioxidant activities of red wines were measured by DPPH (1,1-diphenyl-2-picrylhydrazyl), ABTS [2,2′-azino-bis-(3-ethylbenzothiazoline-6-sulphonic acid)] and CUPRAC (cupric ion reducing antioxidant capacity) assay. The relations between polyphenols and antioxidant values were further considered by multivariate statistical analysis. Results of correlation analysis and stepwise multiple regression analysis showed that antioxidant activities were highly correlated to polyphenolic profiles, with significant contributions of procyanidins and phenolic acids. Consequently, the assessment method we established appeared to be an effective tool for appraising the nutritional value of polyphenols in wines, and the research strategy could be applied to the evaluation of other beverages.

Antidiabetic and Antioxidant Effects and Phytochemicals of Mulberry Fruit (Morus alba L.) Polyphenol Enhanced Extract

Yihai Wang,Limin Xiang,Chunhua Wang,Chao Tang,Xiangjiu He

The antidiabetic and antioxidant activities of the ethyl acetate-soluble extract (MFE) of mulberry fruit (Morus alba L.) were investigated. In vitro , MFE showed potent α-glucosidase inhibitory activity and radical-scavenging activities against DPPH and superoxide anion radicals. In vivo , MFE could significantly decrease fasting blood glucose (FBG) and glycosylated serum protein (GSP), and increase antioxidant enzymatic activities (SOD, CAT, GSH-Px) in streptozotocin (STZ)-induced diabetic mice. Bioactivity-guided fractionation of the MFE led to the isolation of 25 phenolic compounds, and their structures were identified on the basis of MS and NMR data. All the 25 compounds were isolated from mulberry fruit for the first time. Also, the α-glucosidase inhibitory activity and antioxidant activity of the phenolics were evaluated. Potent α-glucosidase inhibitory and radical-scavenging activities of these phenolics suggested that they may be partially responsible for the antidiabetic and antioxidant activities of mulberry fruit.

Molecular basis of sediment formation in cereal vinegar driven by polysaccharide–protein–polyphenol ternary interactions

Dantong Liu,Zhenhua Su,Mengjia Zhang,Jintong Duan,Jia Song,Fanfan Lang,Yanbing Shen,Min Wang,Yu Zheng

Polysaccharide-protein-polyphenol interactions drive cereal vinegar sedimentation. • Polyphenols induce protein rigidification via stable hydrogen bond networks. • Competitive displacement of polysaccharides triggers loss of steric stability. • Displaced polysaccharides induce depletion flocculation for irreversible aging. Abiotic sediment formation during aging compromises visual stability and quality consistency of cereal vinegar, yet its molecular basis in acidic matrices remains unclear. Here, Shanxi aged vinegar was investigated as a polyphenol-rich system comprising polysaccharides, proteins, and phenolics. Sedimented polysaccharides were glucose-rich heteropolysaccharides containing arabinose, xylose, and mannose. In reconstructed systems, mannan–wheat bran protein–vanillic acid showed the greatest destabilization, with an instability index exceeding 0.60 under elevated-temperature conditions. Quantitative excitation–emission matrix fluorescence analysis showed that mannan decreased the vanillic acid-associated maximum fluorescence intensity by 42.8%, indicating a pronounced change in its fluorescence environment. In a gliadin-based molecular model, vanillic acid preferentially associated with hydrophobic protein regions and restricted conformational mobility, while the gliadin–mannan distance increased from 21.49 to 38.5 Å, consistent with redistributed protein–polysaccharide contacts. These findings support a polyphenol-associated interaction-reorganization model and provide a multiscale framework for ternary colloidal destabilization under the tested acid–salt conditions. Metal-polyphenol “prison” attenuated bacterial outer membrane vesicle for chemodynamics promoted in situ tumor vaccines

Weidong Nie,Anqi Jiang,Xu Ou,Jiaxin Zhou,Zijin Li,Chao Liang,Li-Li Huang,Guanghao Wu,Hai-Yan Xie

As natural adjuvants, the bacterial outer membrane vesicles (OMV) hold great potential in cancer vaccines. However, the inherent immunotoxicity of OMV and the rarity of tumor-specific antigens seriously hamper the clinical translation of OMV-based cancer vaccines. Herein, metal-phenolic networks (MPNs) are used to attenuate the toxicity of OMV, meanwhile, provide tumor antigens via the chemodynamic effect induced immunogenic cell death (ICD). Specifically, MPNs are assembled on the OMV surface through the coordination reaction between ferric ions and tannic acid . The iron-based “prison” is locally collapsed in the tumor microenvironment (TME) with both low pH and high ATP features, and thus the systemic toxicity of OMV is significantly attenuated. The released ferric ions in TME promote the ICD of cancer cells through Fenton reaction and then the generation of abundant tumor antigens , which can be used to fabricate in-situ vaccines by converging with OMV. Together with the immunomodulatory effect of OMV, potent tumor repression on a bilateral tumor model is achieved with good biosafety.

Adsorption mechanism of a biodegradable plant polyphenol depressant for selective separation of chalcopyrite from talc

Jiaozhong Cai,Shimei Li,Jiushuai Deng,Bozeng Wu,Hongxiang Xu,Le Yang,Mingzhen Wu,Hongxin Qiu,Naikai Chen,Mingzhen Hu

In this paper, the effect of Tannic acid (TA), a green, biodegradable plant polyphenol, as a talc depressant on the separation behavior of talc and chalcopyrite was investigated. The results of micro-flotation studies revealed that at a TA dosage of 20 mg/L at pH 7, talc recovery was barely 10 % whereas chalcopyrite recovery remained above 90 %, indicating that TA can selectively depress talc flotation. The results of the artificial mixed minerals test further confirmed the depressive effect of TA. The adsorption results showed that the adsorption density of TA on talc was greater than that on chalcopyrite surface. Similar conclusions were observed in zeta potential tests. It is noteworthy that the zeta potential of the chalcopyrite treated with TA and butyl xanthate sodium continued to shift negatively in comparison to the zeta potential of chalcopyrite treated with TA alone, suggesting that butyl xanthate sodium can continue to adsorb on the surface of chalcopyrite following TA treatment. Besides, X-ray photoelectron spectroscopy and Fourier-transform infrared spectroscopy tests concluded that TA adsorbs on talc surface by physical (hydrophobic) interaction. Differences in the crystal structures of chalcopyrite and talc, as well as the large number of phenolic hydroxyl structures contained in TA, are the main driving forces for the selective adsorption of TA on the talc surface. This work provides an insight to the selection of inhibitors for the separation of chalcopyrite and talc, particularly from the perspective of the hydrophobic effect of the talc layered structure.

Effect of High-pressure Homogenization on Structure and Properties of Soy Protein Isolate/polyphenol Complexes

Ren Cong,Xiong Wenfei,Li Bin

Understanding the effects of typical unit operations on the structure and properties of protein/polyphenol complexes has important guiding value for the design and development of two-component functional foods. Therefore, the effects of high-pressure homogenization (HPH) on the structure, physicochemical properties, and in vitro digestive characteristics of soy protein isolate (SPI)-black soybean coat extract (BE) complexes were investigated. UV-Vis and intrinsic fluorescence spectra manifested that the tertiary conformation of protein was partially unfolded after HPH treatment. In addition, with the enhancement of HPH pressure, the zeta-potential magnitude and average particle size of SPI-BE complexes increased and decreased, respectively. On the other hand, HPH had no significant effect on in vitro digestion, anthocyanin content, and free radical scavenging ability of SPI-BE complexes. These findings can provide new insights and guidance for the optimization and regulation of protein-polyphenol co-existing food systems, and provide reference for their application in food industry.

Interfacial deposition of Ag nanozyme on metal-polyphenol nanosphere for SERS detection of cellular glutathione

Yuxin Li,Ping Li,Yiqing Chen,Yue Wu,Jing Wei

The low polarization and low Raman cross section characteristics of glutathione (GSH) make it challenging to directly detect GSH molecules through surface enhanced Raman scattering (SERS) technology. Development of nanostructures for indirect detection of GSH applied to the SERS platform is of great interest. Herein, silver nanoparticles (Ag NPs)/copper-polyphenol colloidal spheres (denoted as CuTA@Ag) with adjustable Ag NPs coverage are prepared by deposition of Ag NPs on the metal-polyphenol colloidal spheres via an interfacial polyphenol reduction method. The size and density of the Ag NPs deposited on the out layer can be readily adjusted by tailoring the concentrations of silver precursor. It leads to activity difference for the nanozyme and SERS characteristics. The SERS properties of the obtained CuTA@Ag are studied using oxTMB , catalytic products of nanozyme, as the probing molecules. They provide satisfactory SERS performance with a low detection limit of 10 −7  M (S/N = 3) and linear determination in the 1–100 μM range for GSH. Moreover, it is further able to detect the glutathione content in cancer cells with well accurate and reproducible capability, catching the signs of rising cancer marker levels. This work proposes structurally tunable nanomaterials platform for a catalytic-based SERS assay, which is expected to utilize the high sensitivity of SERS tool for GSH detection in the cellular environment.

Polydopamine nanoparticles coated with a metal-polyphenol network for enhanced photothermal/chemodynamic cancer combination therapy

Jianbo Su,Tao Liao,Zhe Ren,Ying Kuang,Wenqian Yu,Qianqian Qiao,Bingbing Jiang,Xueqin Chen,Ziqiang Xu,Cao Li

Polydopamine nanoparticles (PDA NPs) are commonly used for photothermal therapy (PTT) of cancer because of their good biocompatibility and photothermal conversion capability. However, it is difficult to achieve a good tumor inhibition effect with a single PTT of PDA. Therefore, in this work, we prepared a combined anticancer nanosystem for enhanced chemodynamic therapy (CDT)/PTT by coating PDAs with an (−)-epigallocatechin gallate (EGCG)/iron (Fe) metal-polyphenol network (MPN). The MPN shell of this nanosystem named EGCG@PDA is degraded by the weakly acidic environment intracellular, releasing EGCG and Fe 3+ . EGCG inhibits the expression of heat shock proteins (HSPs) in cancer cells, thus eliminating their thermal protection against cancer cells for enhanced PTT. Meanwhile, the reductive EGCG can also reduce Fe 3+ to Fe 2+ , to catalyze the decomposition of overexpressed hydrogen peroxide (H 2 O 2 ) in cancer cells to generate strong oxidative hydroxyl radicals ( OH), i.e. , catalyzing the Fenton reaction, for CDT. After the Fenton reaction, the re-oxidized Fe ions can be reduced again by EGCG and reused to catalyze the Fenton reaction, which can achieve enhanced CDT. Both in vitro and in vivo studies have shown that EGCG@PDA has low dark toxicity and good anticancer effects. It is expected to be used for precision cancer therapy.

Construction of multifunctional hydrogel with metal-polyphenol capsules for infected full-thickness skin wound healing

Nanbo Liu,Shuoji Zhu,Yuzhi Deng,Ming Xie,Mingyi Zhao,Tucheng Sun,Changjiang Yu,Ying Zhong,Rui Guo,Keluo Cheng,Dehua Chang,Ping Zhu

Damaged skin cannot prevent harmful bacteria from invading tissues, causing infected wounds or even severe tissue damage. In this study, we developed a controlled-release antibacterial composite hydrogel system that can promote wound angiogenesis and inhibit inflammation by sustained releasing Cu-Epigallocatechin-3-gallate (Cu-EGCG) nano-capsules. The prepared SilMA/HAMA/Cu-EGCG hydrogel showed an obvious inhibitory effect on Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) . It could also promote the proliferation and migration of L929 fibroblasts. In vivo full-thickness infected wound healing experiments confirmed the angiogenesis and inflammation regulating effect. Accelerate collagen deposition and wound healing speed were also observed in the SilMA/HAMA/Cu-EGCG hydrogel treated group. The findings of this study show the great potential of this controlled-release antibacterial composite hydrogel in the application of chronic wound healing.
