Overexpression of epidermal growth factor receptor (EGFR) in cancer is a key cause of recurrence of cervical cancer (CC). Although the EGF-EGFR pathway has been studied for decades, preventing tumor growth and recurrence caused by peripheral EGF remains a great challenge. In this work, a strategy is proposed to reduce the stimulation of high concentration EGF on tumor growth by using a thermo-sensitive hydrogel. The hydrogel is a triblock copolymer composed of polyethylene glycol (PEG) and poly (lactide glycolide) (PLGA). Based on the excellent temperature sensitivity, carrier capacity, swelling property and biocompatibility, the hydrogel can absorb the liquid around the tumor by injection and release EGF continuously at low concentration. The inhibitory effect of hydrogel on tumor growth is fully confirmed by an implanted tumor mouse model with human cervical cancer cell lines (HeLa) using triple-immunodeficient NCG mice. Compared with free EGF, the EGF-loaded hydrogel can hardly induce surface plasmon resonance (SPR) response, which proves that hydrogel can effectively weaken cytoskeleton rearrangement and inhibit cell migration by continuously releasing low concentration EGF. In addition, the EGF-loaded hydrogel can reduce cell proliferation by delaying the progress of cell cycle progression. Taken together, the hydrogel can effectively protect tumor microenvironment from the stimulation of high concentration EGF, delay cancer cellular processes and tumor growth, and thus providing an approach for inhibiting tumor recurrence of CC.

Chronic neuropathic pain severely impairs quality of life, with current therapies often causing adverse effects. Our research group identified αO-conotoxin GeXIVA[1,2] as a potent analgesic candidate derived from marine cone snails. However, its clinical application is limited by rapid clearance and complex administration. This study developed a sustained-release hydrogel microneedle patch encapsulating GeXIVA[1,2] to address these challenges. Optimized 4:3 (w/w) polyvinyl alcohol (PVA)–sucrose hydrogel formulation achieved 98.6% structural integrity and controlled swelling (ratio = 1.9 at 48 h). The microneedles demonstrated uniform conical morphology (height: 889 ± 49 µm, base: 381 ± 26 µm) enabling epidermal penetration. In spared nerve injury (SNI) models, a single microneedle patch application increased mechanical paw withdrawal thresholds from 0.056 g to 0.7269 g, maintaining efficacy for 3 days. Chronic constriction injury (CCI) models showed comparable pain relief. Notably, microneedle patch treatment improved locomotor function in SNI mice (total movement: 1518 cm vs. 1126 cm untreated). This hydrogel microneedle patch platform extends GeXIVA[1,2]’s analgesic duration from hours to days through sustained release, while resolving administration challenges through transdermal delivery, expanding the potential applications of GeXIVA[1,2], and demonstrating a promising strategy for the chronic neuropathic pain management.

The regeneration of critical-sized bone defects is severely hampered by a dual challenge: insufficient vascularization and limited osteogenic activity at the injury site. Histatin-1 (Hst1), a salivary peptide with known pro-angiogenic and regenerative activities, is a hopeful candidate for bone regeneration, yet its dual function in osteogenesis and angiogenesis remains underexplored. Here, we systematically elucidated this dual function. In vitro , Hst1 was demonstrated to stimulate the osteogenic differentiation of rat bone marrow mesenchymal stem cells (BMSCs) through the activation of the Wnt/β-catenin signaling pathway. Concurrently, Hst1 promoted tube formation through paracrine signaling. In vivo , the hydrogel formulated from oxidized sodium alginate (OSA), gelatin (GE), and adipic acid dihydrazide (ADH) was employed to ensure the sustained delivery of Hst1. This Hst1-loaded hydrogel was applied to a 5-mm critical-sized cranial defect in rats. Compared to the Control group, Hst1-loaded hydrogel significantly enhanced bone regeneration, with the bone volume fraction (BV/TV) increasing to 20.0 % ( vs. 8.2 % for Control). This was accompanied by a marked increase in vascularization, as evidenced by a 3.0-fold rise in vessel density. In conclusion, this study reveals the dual pro-regenerative function of Hst1 in simultaneously promoting osteogenesis and angiogenesis, demonstrating that harnessing Hst1 within a sustained-release hydrogel is a powerful therapeutic strategy for achieving vascularized bone regeneration.

Chronic osteomyelitis remains a major challenge in orthopedic therapy. Developing a biodegradable, non-toxic material capable of providing sustained antibiotic release has emerged as a promising approach for localized antibiotic delivery in managing this condition. In this study, copper-tannic acid (CuTA) nanosheets were synthesized and employed as a coating material for vancomycin, resulting in the formation of vancomycin@CuTA (Van@CuTA) nanocomposites. The morphological and structural characterization of CuTA and Van@CuTA was performed using various techniques. The sustained release behavior of vancomycin, and in vitro effects of Van@CuTA on methicillin-resistant Staphylococcus aureus (MRSA) growth, bone marrow mesenchymal stem cells (BMSCs) viability, osteogenesis, and human umbilical vein endothelial cells (HUVECs) angiogenesis were systematically investigated. A rabbit model of chronic osteomyelitis was established to assess the therapeutic effect of Van@CuTA, in combination with fibrin gel, in controlling infection, preventing bone destruction, and inhibiting the progression of chronic osteomyelitis. The characterization results confirmed the formation of Van@CuTA nanocomposites. In vitro experiments revealed that Van@CuTA enabled gradual vancomycin release, effectively suppressed MRSA growth, and demonstrated no toxicity to BMSCs. Furthermore, Van@CuTA significantly promoted osteogenic differentiation of BMSCs and improved angiogenesis in HUVEC. The in vivo studies demonstrated that Van@CuTA coated with fibrin gel ameliorated the appearance of local infection, reduced bone structural damage, and diminished inflammatory infiltration within the bone marrow in the rabbit model of chronic osteomyelitis. Current findings indicated that CuTA nanosheets served as a promising in-situ antibiotic carrier for sustained release in chronic osteomyelitis treatment. Van@CuTA demonstrated improved antibacterial properties, enabled sustained vancomycin release, and promoted both osteogenesis and angiogenesis, leading to its preliminary therapeutic efficacy in rabbit models of chronic osteomyelitis and strong potential for clinical application in osteomyelitis treatment.

The purpose of this study is to provide a new strategy for constructing a temperature-controlled hydrogel as a promising agent for wound healing using natural products through physical co-assembly. Herein, the temperature-controlled physically assembled hydrogel consisting of gallic acid and lysozyme (GL) could be co-assembled into a regular fibrous structure accompanied by strong blue fluorescence with three-dimensional networks at micron levels through hydrophobic interactions, π–π interactions and hydrogen bonding. This GL hydrogel has excellent temperature sensitivity and self-healing properties, as proved by cycle high-low temperature tests. In addition, it possesses stable rheological properties, great sustained release ability, and could realize the spatiotemporal delivery of gallic acid and lysozyme. Biocompatibility and antibacterial tests proved that this well-assembled GL hydrogel has no cytotoxicity but excellent antibacterial activity. Both in vitro and in vivo experiments demonstrated that the GL hydrogel has excellent anti-inflammation efficiency and promotes the healing of chronic wounds by suppressing the expression of pro-inflammatory related genes. Tests using an E. coli-infected wound model confirmed that the GL hydrogel could terminate the inflammatory phase early and ultimately promote the healing of wounds infected by E. coli. This study provides a promising strategy for the effective treatment of wounds through a physical self-assembled hydrogel.

Soybean products rich in high-quality proteins have a short shelf life owing to their high water activity characteristics. In this study, sustained-release antibacterial composite films of cinnamaldehyde microcapsules (CAMs) were constructed for the preservation of dry tofu using soy hull nanocellulose (SHNC), polyvinyl alcohol (PVA), and pullulan (PUL) as the composite matrix. CAMs with an encapsulation efficiency of 70.53% were prepared via complex coacervation using gelatin–Arabic gum as the wall material. Different contents of CAMs (0.5%, 1%, 2%, and 3% w /w) were added to the SHNC–film using the solution casting method. Fourier transform infrared spectroscopy and X-ray diffraction analysis confirmed that cinnamaldehyde was successfully embedded in the CAMs and crosslinked with the composite film matrix via hydrogen bonding to form a dense network structure. Scanning electron microscopy showed that the CAMs were well integrated into the composite film matrix without forming pores or cracks. Physical performance tests showed that after adding 2% ( w /w) CAMs, the mechanical strength of the composite film was significantly improved (the tensile strength increased from 13.79 ± 1.67 MPa to 26.94 ± 2.02 MPa; P < 0.05), as were its hydrophobic properties (the contact angle increased from 25.7° ± 1.6° to 68.3° ± 1.6°), thermal stability, and ultraviolet barrier properties. The CAMs composite film (CAMs-film) significantly inhibited the proliferation of Pseudomonas fragi by inhibiting energy metabolism, degrading cell components and metabolic intermediates, and disrupting secondary metabolism (inhibition rate of 96.03%, P < 0.05). The composite film with 2% CAMs could extend the shelf life of dried tofu to 20 days at 4 °C. As a new environmentally friendly packaging material, the CAMs-film developed in this study can effectively extend the shelf life of perishable foods, providing a solution for the development of sustainable food packaging.

Peimine (P) and curcumin (CM) possess significant bioactivities but suffer from poor water solubility, low bioavailability, and limited stability. To address these challenges, P-CM self-assembled microparticles (SMPs) were prepared via non-covalent interactions and systematically characterized. The results demonstrated that optimal self-assembly reaction conditions occurred at pH 9, 25 °C, and a P:CM molar ratio of 1:1, yielding uniform nanoparticles (284 nm, PDI = 0.202) with a zeta potential of −19.77 mV. P-CM SMPs exhibited superior antioxidant activities, with ABTS, DPPH radical scavenging activity and FRAP values of 62.69%, 34.18%, and 81.02%, respectively, as well as enhanced anti-inflammatory activity compared to their individual components. To further improve stability and control release, the P-CM SMPs were encapsulated in a hollow zein-chitosan (HZ-CH) composite, achieving high encapsulation efficiency (96.0%). HZ-CH-SMPs exhibited storage stability over time and sustained-release behavior under simulated gastrointestinal conditions. This study provides a theoretical basis for designing delivery strategies integrating molecular self-assembly and protein-polysaccharide encapsulation to overcome solubility and bioavailability limitations of bioactive compounds in functional foods.

The triblock copolymer F127 (molecular formula (EO)106(PO)70(EO)106) was adopted as template with TMOS as the silica source. They were hydrolyzed by acidic aqueous solution in which chlorpyrifos (CH) and metal ions (Mn+) were introduced to the products to construct "F127-Mn+-CH" coordination structure in one step. Different sustained release systems were prepared through changing the composition of acidic aqueous solution added during the preparation. The characterization results confirmed the coordination interaction existing among metal ions, mesoporous silica and chlorpyrifos. It was also proved that the chlorpyrifos loaded in the sustained release system existed as amorphous state. According to SEM and TEM observation, the pore of the material presented as face-centered cage-type cubic structure with IM-3M symmetry similar to SBA-16 which is in accordance with N2 adsorption isotherms. The as-synthesized system showed significant pH sensitivity in the sustained release process indicating that the sustained release system can be well stored in neutral environment and activated while sprayed into earth in acid or basic environment. Their sustained release curves can be described by Korsmeyer-Peppas equation in neutral while accords with Higuchi under acid or basic environment.

Novel amphiphilic chitosan derivative of N , N -dimethylhexadecyl carboxymethyl chitosan (DCMCs) was synthesised. The physical properties of DCMCs were characterised by Fourier transform-infrared spectroscopy, nuclear magnetic resonance spectroscopy, X-ray diffraction and differential scanning calorimetry. DCMCs can form core–shell micelles with low critical micelle concentration (31 mg L −1 ). The core–shell micelles exhibited spherical shape with positive charge (+50.7 mV) and narrow size distribution (polydispersity index <0.5). Vitamin D 3 (VD 3 ) was initially used to load into the core–shell micelles, and the solubility of VD 3 was improved with higher encapsulation efficiency (53.2%). The in vitro release processes of VD 3 from the core–shell micelles were initially rapid and then followed by a sustained release. The Higuchi model was the most suitable for describing the entire release procedure. DCMCs core–shell micelles are promising carriers for VD 3 and other hydrophobic bioactive food factors.

Glaucoma filtration surgery (GFS) is the definitive treatment for reducing intraocular pressure in patients with refractory glaucoma. However, postoperative scarring of the filtration tract is a major cause of surgical failure. Although artesunate (ART) exhibits significant antifibrotic efficacy, its clinical application is severely limited by poor solubility and a short half-life. This study designed and developed a new injectable hydrogel cross-linked by collagen and four-armed polyethylene glycol N-hydroxysuccinimide encapsulating ART (C-P@ART) to overcome the limitations of ART in ocular antifibrotic therapy and extensively examined its underlying mechanisms. Stable three-dimensional network structure, excellent biocompatibility, and sustained drug release characteristics of C-P@ART were validated through FTIR, rheological analysis, endotoxin-testing, scanning electron microscope (SEM), and in vitro and in vivo release assays. In a TGF-β1-induced fibroblast fibrosis model, C-P@ART significantly inhibited α-SMA expression concentration-dependently and exerted antifibrotic effects through dual regulation of TGF-β1/SMAD and PI3K/Akt pathways. Subsequently, we evaluated its antifibrotic efficacy in the rabbit GFS model, where the C-P@ART treatment group exhibited a significant reduction in postoperative collagen deposition and α-SMA-positive expression compared to the control group. C-P@ART inhibited NF-κB pathway activation and reduced inflammatory cell infiltration. In conclusion, these findings demonstrated that C-P@ART synergistically mitigates fibrosis and inflammation through sustained ART release, providing an innovative therapeutic strategy to enhance GFS outcomes.

While poly (vinyl alcohol) (PVA)-based aerogels are promising porous carriers for controlled drug delivery, drying-induced pore collapse generally limits their internal pore accessibility, drug-loading capacity, and release regulation. In this study, duration-controlled tert-butanol (TBA) solvent exchange was developed as a pore-engineering strategy to regulate the structure-performance relationship of PVA/cellulose nanocrystal/Ca2+composite aerogels. This strategy focuses on balancing pore preservation, liquid-accessible pore connectivity, and network densification, instead of simply increasing the total surface area. With an optimized exchange duration, the TBA6 aerogel exhibited a well-preserved porous network. Moreover, its specific surface area and porosity increased from 33.11 to 96.02 m2·g−1and from 93.93% to 96.03%, respectively. This optimized structure enhanced salicylic acid (SA) loading, elevating the equilibrium adsorption capacity from 40.91 to 61.18 mg·g−1. Release kinetic analysis revealed that the Higuchi constant and the Korsmeyer-Peppas release constant decreased from 18.148 to 12.751 and from 25.070 to 17.283%·h−1/2, respectively, indicating a slower diffusion-regulated release process. Confocal Raman mapping further confirmed that TBA6 promoted a more homogeneous SA distribution within the aerogel matrix and enabled gradual release from the internal porous network. To sum up, controlling solvent-exchange duration is an effective strategy for tuning pore accessibility and diffusion resistance. This study provides new insight into the design of PVA-based composite aerogels for drug-loading and sustained-release applications.

Chronic wound infections, particularly those complicated by antibiotic-resistant bacteria and biofilm formation, present persistent clinical challenges due to limited drug penetration and unresolved inflammation. Here, we report a lipid-droplet-engineered, gallium-loaded sonodynamic therapeutic platform (LDTG) incorporated into a bilayer microneedle (MN) patch co-delivering all-trans retinoic acid (ATRA), termed LDTG A-MNs, for synergistic antibacterial therapy and accelerated wound repair. The engineered LDTG exhibit strong bacteria-targeting capability by mimicking host lipid droplets and efficiently generate reactive oxygen species (ROS) under ultrasound irradiation to achieve potent sonodynamic bactericidal activity. The bilayer MN design enables rapid release of LDTG for targeted bacterial capture, followed by sustained ATRA release to modulate macrophage polarization and promote tissue regeneration. In vitro, LDTG A-MNs demonstrate robust antibacterial and biofilm-disrupting effects, excellent biocompatibility, and enhanced M2-type macrophage polarization. In vivo, LDTG A-MNs combined with ultrasound effectively eradicate bacterial infection, suppress excessive inflammation, improve collagen deposition, enhance angiogenesis, and significantly accelerate wound closure in infected murine skin wounds. This work presents a minimally invasive and infection-responsive MN system that integrates pathogen targeting, sonodynamic antibacterial therapy, and immune-regulated tissue repair, offering a promising therapeutic strategy for the management of complex and refractory infected wounds.
