Oral administration of polysaccharide-iron complexes (UCP-Fe(III)) is limited by their degradation in the gastric environment, which reduces bioavailability. We developed core–shell sodium alginate/chitosan hydrogel microsphere for pH-responsive UCP-Fe(III) delivery. High sodium alginate (SA) concentrations increased viscosity and storage modulus, enhancing mechanical stability. The encapsulation efficiency of UCP-Fe(III) reached 94.41% using 2.0% SA. Noncovalent interactions between UCP-Fe(III) and SA contributed to enhanced hardness and chewiness of the microspheres, while higher SA concentrations promoted a denser cross-linked network. The hydrogel microspheres showed low swelling in simulated gastric fluid (SGF, pH 2.0; swelling ratio: 467.33–588.50%) but rapid swelling in simulated intestinal fluid (SIF, pH 7.4; swelling ratio: 800–962.17%), indicating significant pH responsiveness. They effectively prevented premature release (only 12.14–23.48% in SGF after 2 h) while enabling sustained release (87.60–95.16% in SIF after 6 h). This study provides a new strategy for developing oral iron supplements with pH-responsive sustained-release properties.

Menthol and thymol, as natural antiseptics, have the advantages of nontoxicity and broad-spectrum antimicrobial activity, which solves the problems of antibiotic resistance and toxicity of metal antiseptics. However, their practical applications face the challenges of chemical instability and difficult solubility in water. In this study, menthol and thymol were tactfully physically combined, forming hydrophobic deep eutectic solvent (HDES), which were then encapsulated via an emulsion system to address their instability and hydrophobicity. Then, 2-hydroxyethyl acrylate was added and polymerized into a covalent network to fabricate a HDES-loaded heterogeneous gel, featuring sustained-release characteristics for the preservation of perishable fruits. The combination of emulsion encapsulation and gelation improves the stabilization of menthol and thymol, allowing for delayed release and providing materials with a 34-day antibacterial cycle. Additionally, the emulsion gel presents tensile strain as high as 2250%, making it appropriate as practical packaging materials. Through cherry, tomato, and broccoli preservation tests, it is found that compared with the monomer such as thymol, menthol, and the deep eutectic mixture, the emulsion gel presented the best antibacterial activity, enabling the preservation days to significantly increase. We calculated that it was probably due to the HDES and cross-linking network displaying a synergistic effect. This design will provide a perspective for the preservation of perishable fruits and vegetables.

Using butyl methacrylate (BMA) and styrene (St) as monomers and divinylbenzene (DVB) as a crosslinking agent, P(St-BMA) microspheres were prepared by suspension polymerization. Tea tree oil (TTO) microspheres were prepared by adsorbing TTO on P(St-BMA) microspheres. The structure and surface morphology of P(St-BMA) microspheres and TTO microspheres were characterized by Fourier transformed infrared spectroscopy (FTIR), optical microscopy, and Thermogravimetric analysis (TGA). In doing so, the structural effect of P(St-BMA) microspheres on oil absorption and sustained release properties could be investigated. The results show that the surface of the P(St-BMA) microspheres in the process of TTO microsphere formation changed from initially concave to convex. The TTO microspheres significantly improved the stability of TTO, which was found to completely decompose as the temperature of the TTO increased from about 110 °C to 150 °C. The oil absorption behavior, which was up to 3.85 g/g, could be controlled by adjusting the monomer ratio and the amount of crosslinking agent. Based on Fickian diffusion, the sustained release behavior of TTO microspheres was consistent with the Korsmeyer-Pappas kinetic model. After 13 h of natural release, the anti-bacterial effect of the TTO microspheres was found to be significantly improved compared to TTO.

Novel amphiphilic chitosan derivative of N , N -dimethylhexadecyl carboxymethyl chitosan (DCMCs) was synthesised. The physical properties of DCMCs were characterised by Fourier transform-infrared spectroscopy, nuclear magnetic resonance spectroscopy, X-ray diffraction and differential scanning calorimetry. DCMCs can form core–shell micelles with low critical micelle concentration (31 mg L −1 ). The core–shell micelles exhibited spherical shape with positive charge (+50.7 mV) and narrow size distribution (polydispersity index <0.5). Vitamin D 3 (VD 3 ) was initially used to load into the core–shell micelles, and the solubility of VD 3 was improved with higher encapsulation efficiency (53.2%). The in vitro release processes of VD 3 from the core–shell micelles were initially rapid and then followed by a sustained release. The Higuchi model was the most suitable for describing the entire release procedure. DCMCs core–shell micelles are promising carriers for VD 3 and other hydrophobic bioactive food factors.

Background: Linagliptin (LNG) exhibits poor bioavailability and numerous side effects,
significantly limiting its use. Transdermal drug delivery systems (TDDS) offer a potential solution to
overcome the first-pass effect and gastrointestinal reactions associated with oral formulations.
Objective: The aim of this study was to develop LNG microparticle gels to enhance drug bioavailability
and mitigate side effects.
Methods: Linagliptin hyaluronic acid (LNG-HA) microparticles were prepared by spray drying
method and their formulation was optimized via a one-factor method. The solubility and release were
investigated using the slurry method. LNG-HA microparticle gels were prepared and optimised using
in vitro transdermal permeation assay. The hypoglycaemic effect of the LNG-HA microparticle gel
was examined on diabetic mice.
Results: The results indicated that the LNG-HA microparticle encapsulation rate was 84.46%. Carbomer
was selected as the gel matrix for the microparticle gels. Compared to the oral API, the microparticle
gel formulation demonstrated a distinct biphasic release pattern. In the first 30 minutes,
only 43.56% of the drug was released, followed by a gradual release. This indicates that the formulation
achieved a slow-release effect from a dual reservoir system. Furthermore, pharmacodynamic
studies revealed a sustained hypoglycemic effect lasting for 48 hours with the LNG microparticle gel
formulation.
Conclusion: These findings signify that the LNG microparticle gel holds significant clinical value
for providing sustained release and justifies its practical application.

Emamectin benzoate (EB), a widely used pesticide, is prone to decomposition by ultraviolet light and suffers from the corresponding loss of efficacy. The timed release of EB based on microspheres is one of the effective methods to solve this issue. As a non-toxic cellulose ester, cellulose acetate butyrate (CAB) is regarded as one of the best wall-forming materials for microcapsules with a good controlled release performance. Herein, two methods—mechanical activation (MA) technology and a conventional liquid phase (LP) method—were employed to synthesize different CABs, namely CAB-MA and CAB-LP, respectively. The molecular structure, rheological property, and thermal stability of these CABs were investigated. The two CABs were used to prepare microspheres for the loading and release of EB via an o/w (oil-in-water) solvent evaporation method. Moreover, the performances such as drug loading, drug entrapment, and anti-photolysis of the drug for these microspheres were studied. The results showed that both CABs were available as wall materials for loading and releasing EB. Compared with CAB-LP, CAB-MA presented a lower molecular weight and a narrower molecular weight distribution. Moreover, the MA method endowed the CAB with more ester substituent groups and less crystalline structure in comparison to the LP method, which had benefits including pelletizing and drug loading. Keywords:cellulose acetate butyrate;solid-phase synthesis;mechanical activation;microspheres;emamectin benzoate;release

Peimine (P) and curcumin (CM) possess significant bioactivities but suffer from poor water solubility, low bioavailability, and limited stability. To address these challenges, P-CM self-assembled microparticles (SMPs) were prepared via non-covalent interactions and systematically characterized. The results demonstrated that optimal self-assembly reaction conditions occurred at pH 9, 25 °C, and a P:CM molar ratio of 1:1, yielding uniform nanoparticles (284 nm, PDI = 0.202) with a zeta potential of −19.77 mV. P-CM SMPs exhibited superior antioxidant activities, with ABTS, DPPH radical scavenging activity and FRAP values of 62.69%, 34.18%, and 81.02%, respectively, as well as enhanced anti-inflammatory activity compared to their individual components. To further improve stability and control release, the P-CM SMPs were encapsulated in a hollow zein-chitosan (HZ-CH) composite, achieving high encapsulation efficiency (96.0%). HZ-CH-SMPs exhibited storage stability over time and sustained-release behavior under simulated gastrointestinal conditions. This study provides a theoretical basis for designing delivery strategies integrating molecular self-assembly and protein-polysaccharide encapsulation to overcome solubility and bioavailability limitations of bioactive compounds in functional foods.

Chronic wound infections, particularly those complicated by antibiotic-resistant bacteria and biofilm formation, present persistent clinical challenges due to limited drug penetration and unresolved inflammation. Here, we report a lipid-droplet-engineered, gallium-loaded sonodynamic therapeutic platform (LDTG) incorporated into a bilayer microneedle (MN) patch co-delivering all-trans retinoic acid (ATRA), termed LDTG A-MNs, for synergistic antibacterial therapy and accelerated wound repair. The engineered LDTG exhibit strong bacteria-targeting capability by mimicking host lipid droplets and efficiently generate reactive oxygen species (ROS) under ultrasound irradiation to achieve potent sonodynamic bactericidal activity. The bilayer MN design enables rapid release of LDTG for targeted bacterial capture, followed by sustained ATRA release to modulate macrophage polarization and promote tissue regeneration. In vitro, LDTG A-MNs demonstrate robust antibacterial and biofilm-disrupting effects, excellent biocompatibility, and enhanced M2-type macrophage polarization. In vivo, LDTG A-MNs combined with ultrasound effectively eradicate bacterial infection, suppress excessive inflammation, improve collagen deposition, enhance angiogenesis, and significantly accelerate wound closure in infected murine skin wounds. This work presents a minimally invasive and infection-responsive MN system that integrates pathogen targeting, sonodynamic antibacterial therapy, and immune-regulated tissue repair, offering a promising therapeutic strategy for the management of complex and refractory infected wounds.

Semi-solid extrusion (SSE) 3D printing offers significant potential for fabricating personalized oral dosage forms, thanks to its mild processing conditions. However, post-printing drying often induces structural collapse and deformation due to solvent evaporation, severely compromising geometric fidelity. While freeze-drying can mitigate deformation, it is time-intensive and yields highly porous structures with inferior mechanical strength. This study introduces a dual strategy employing a phase-change sacrificial support and thermal curing to overcome these limitations. A printable matrix was formulated using microcrystalline cellulose and hydroxypropyl methylcellulose (HPMC) as rheological modifiers, with Eudragit® RL 30D aqueous dispersion serving as the sustained-release polymer and beeswax as a sacrificial phase. The optimized tablets demonstrated superior mechanical integrity, with Young's modulus increasing by 67%. During thermal curing at 80°C, the dispersed Eudragit® RL pseudo-latex underwent coalescence and formed an insoluble matrix, creating an insoluble polymeric network that reinforced the structure and modulated drug release. Crucially, beeswax (melting range 62–67°C) provided mechanical support during the critical drying phase at 50°C, preventing structural collapse, before being sacrificially removed during the 80°C curing step. This approach successfully produced high-fidelity, diclofenac sodium-loaded tablets with tunable release profiles governed by the Eudragit® RL-to-HPMC ratio. These findings present a robust strategy for producing dimensionally stable, mechanically strong, and personalized sustained-release formulations via SSE 3D printing.

Soybean products rich in high-quality proteins have a short shelf life owing to their high water activity characteristics. In this study, sustained-release antibacterial composite films of cinnamaldehyde microcapsules (CAMs) were constructed for the preservation of dry tofu using soy hull nanocellulose (SHNC), polyvinyl alcohol (PVA), and pullulan (PUL) as the composite matrix. CAMs with an encapsulation efficiency of 70.53% were prepared via complex coacervation using gelatin–Arabic gum as the wall material. Different contents of CAMs (0.5%, 1%, 2%, and 3% w /w) were added to the SHNC–film using the solution casting method. Fourier transform infrared spectroscopy and X-ray diffraction analysis confirmed that cinnamaldehyde was successfully embedded in the CAMs and crosslinked with the composite film matrix via hydrogen bonding to form a dense network structure. Scanning electron microscopy showed that the CAMs were well integrated into the composite film matrix without forming pores or cracks. Physical performance tests showed that after adding 2% ( w /w) CAMs, the mechanical strength of the composite film was significantly improved (the tensile strength increased from 13.79 ± 1.67 MPa to 26.94 ± 2.02 MPa; P < 0.05), as were its hydrophobic properties (the contact angle increased from 25.7° ± 1.6° to 68.3° ± 1.6°), thermal stability, and ultraviolet barrier properties. The CAMs composite film (CAMs-film) significantly inhibited the proliferation of Pseudomonas fragi by inhibiting energy metabolism, degrading cell components and metabolic intermediates, and disrupting secondary metabolism (inhibition rate of 96.03%, P < 0.05). The composite film with 2% CAMs could extend the shelf life of dried tofu to 20 days at 4 °C. As a new environmentally friendly packaging material, the CAMs-film developed in this study can effectively extend the shelf life of perishable foods, providing a solution for the development of sustainable food packaging.

Chronic osteomyelitis remains a major challenge in orthopedic therapy. Developing a biodegradable, non-toxic material capable of providing sustained antibiotic release has emerged as a promising approach for localized antibiotic delivery in managing this condition. In this study, copper-tannic acid (CuTA) nanosheets were synthesized and employed as a coating material for vancomycin, resulting in the formation of vancomycin@CuTA (Van@CuTA) nanocomposites. The morphological and structural characterization of CuTA and Van@CuTA was performed using various techniques. The sustained release behavior of vancomycin, and in vitro effects of Van@CuTA on methicillin-resistant Staphylococcus aureus (MRSA) growth, bone marrow mesenchymal stem cells (BMSCs) viability, osteogenesis, and human umbilical vein endothelial cells (HUVECs) angiogenesis were systematically investigated. A rabbit model of chronic osteomyelitis was established to assess the therapeutic effect of Van@CuTA, in combination with fibrin gel, in controlling infection, preventing bone destruction, and inhibiting the progression of chronic osteomyelitis. The characterization results confirmed the formation of Van@CuTA nanocomposites. In vitro experiments revealed that Van@CuTA enabled gradual vancomycin release, effectively suppressed MRSA growth, and demonstrated no toxicity to BMSCs. Furthermore, Van@CuTA significantly promoted osteogenic differentiation of BMSCs and improved angiogenesis in HUVEC. The in vivo studies demonstrated that Van@CuTA coated with fibrin gel ameliorated the appearance of local infection, reduced bone structural damage, and diminished inflammatory infiltration within the bone marrow in the rabbit model of chronic osteomyelitis. Current findings indicated that CuTA nanosheets served as a promising in-situ antibiotic carrier for sustained release in chronic osteomyelitis treatment. Van@CuTA demonstrated improved antibacterial properties, enabled sustained vancomycin release, and promoted both osteogenesis and angiogenesis, leading to its preliminary therapeutic efficacy in rabbit models of chronic osteomyelitis and strong potential for clinical application in osteomyelitis treatment.

In order to improve the loading capacity of the drug delivery system based on magadiite for procainamide (PA), the cationic PA was inserted into the interlayer of magadiite by the intercalation of tetra-n-butylammonium ions and multi-step ultrasonic ion exchange. X-ray diffraction, compositional analyses, and IR results confirmed that the cationic PA was successfully loaded into the interlayer of magadiite, and the loading capacity of the drug delivery system based on magadiite for PA was about 25.0%. Thermogravimetric analysis indicated that the thermal stability of PA was obviously improved after it was loaded into the interlayer of magadiite. Drug release revealed the sustained and controlled release of PA from drug delivery system either in phosphate buffer solution (PBS, pH = 7.4, simulating intestinal fluid) or in HCl solution (pH = 1.35, simulating gastric fluid). This sustained release of drugs will be beneficial to improve the availability and efficacy of drugs. Moreover, the kinetic behavior of drug release either in PBS or in HCl can be fitted to ExpAssoc model. Nuclear magnetic resonance (NMR) results indicated that there is an interaction between the magadiite and PA in the drug delivery system, and this interaction may determine the kinetic behavior of drug release. The present work may provide a refer for improving the loading capacity of positively charged drugs in some layered silicates, which would be important in medical field.
