For the rapid and sensitive detection of vascular endothelial growth factor 165 (VEGF165) in clinical blood samples, a microfluidic sensing chip that integrates a centrifugal separation pretreatment unit and a composite nanosensing film was proposed in this paper. An efficient sensing strategy and method was established. The blood sample was first separated and extracted by centrifugal force on the centrifugal microfluidic chip within 5 min after injection. The separated plasma can be automatically transferred through the designed microchannels to the detection area integrated electrodes for subsequent differential pulse voltammetric detection. The Au NPs/MCH/Apt2 sensing film was constructed on the surface of the Au working electrode. A sandwich sensing strategy based on “double aptamers” and “nanoprobe” for VEGF165 detection was established, by which the synthetic Apt1/PThi/Au NP nanoprobe was applied to capture VEGF165 in plasma and bind to the sensing film. By this method, the detection limit of VEGF165 in whole blood was 0.67 pg/mL and the linear range was between 1 pg and 10 ng, which met the needs of clinical VEGF165 detection. It was illustrated that the proposed methodology based on the centrifugal microfluidic chip had potential application prospects in the development of the point-of-care testing fields.

Exosomes are lipid bilayer extracellular vesicles secreted by various types of cells and inherit abundant molecular information from parental cells. Tumor-derived exosomes have been widely recognized as noninvasive biomarkers for early cancer diagnosis and surveillance, but the separation of intact exosomes and detection of exosomal proteins remain challenging. Herein, we proposed a microfluidic chip for specific exosome isolation, integrated with sensitive quantification by a novel PTCDI–aptamer signal switch strategy. To enhance the capture efficiency, an alternating drop-shaped micropillar array was designed to assist the capture of tumor-derived exosomes by Tim4-modified magnetic beads (Tim4 beads) on the chip. Following capture, a chelating agent can easily elute intact exosomes which were further used for profiling exosomal surface proteins by the multiplexed fluorescence turn-on approach. Profiting from the efficient on-chip enrichment of the Tim4 beads and superior fluorescence signal transduction strategy, the detection limit of the analysis platform for HepG2 exosomes is as low as 8.69 × 103 particles/mL with a wide linear range spanning 6 orders of magnitude. Meanwhile, the proposed platform could recognize subtle changes in protein levels on the exosomal surface from various cell lines. More importantly, this strategy is successfully applied to analyze exosomes in human serum to distinguish liver cancer patients from healthy individuals. Combined analysis of different types of biomarkers on the exosomal membrane surface can greatly improve the accuracy of cancer type identification and disease monitoring. We hope that this convenient, rapid, and sensitive platform may become a powerful tool in the field of exosome analysis and early cancer screening.

Gas sensors based on metal oxide semiconductor adduce a low cost and reliable solution for the leakage detection of methane, which is highly explosive. However, it is a fact that MOS gas sensors suffer from a heavy cross sensitive response to volatile organic compounds such as ethanol and acetic acid. With the help of filters, metal oxide semiconductor gas sensors could become quite selective to methane. Here, a novel on-chip microfilter was designed for metal oxide semiconductor gas sensors. The on-chip microfilter is fabricated from porous alumina ceramics loaded with platinum on the surface. The active filter was continuously heated by sensor chips. Whereas, it was kept intact with sensing films and reactive gas molecules have to diffuse through the porous ceramics before reaching the surface of metal oxide semiconductor. It was found that both selective response and sensitive response to methane were enhanced by the on-chip microfilter. This is similar to the reported active filter, which is prepared from catalytic films and deposited directly on the top of sensor layer. Additionally, it was indicated that the on-chip microfilter not only limited to tin dioxide but also valid for indium oxide based on sensors.

To meet the evolving requirements of material designability, sustainability, and eco-friendliness, the development of syntactic foams puts great emphasis on filler optimization and matrix selection. Here, we present a novel microfluidic expansion coupled with the thermal contraction method to improve the fabrication of syntactic foam fillers (SFFs), highlighted by the independently regulated parameters in contrast to the hydrodynamic regulation approach. The eccentricity of droplets can be reduced via osmotic swelling under a hypotonic circumstance and further preserved through real-time UV polymerization. The eccentricity of shrunken SFFs after heat treatment is found to inherit the regulated configurations of droplets and this homogeneity difference of shell thickness can result in the mechanical performance deviation. Then, we choose greener material, geopolymer, as the binder matrix and prepare geopolymer syntactic foams (GSFs) by mold casting. The inclusion of SFFs has increased the atomic volume ratio of Si : Al in the matrix, thereby forming a strong interfacial bonding. The compressive yield strength of GSFs is inversely proportional to the volume fraction of SFFs, and the specimens after thermal exposure can still maintain their shape integrity and exhibit similar mechanical performance even with the formation of cracks. Moreover, the fire-resistant performance of GSFs has been visually validated by the combustion comparison experiment, characterized as flame retardancy, smoke-free, and basically intact morphology, which should give insights into the design and fabrication of functional composites.

Continuous-flow PCR (CF-PCR) can realize rapid DNA amplification because of the high temperature variation rate. However, off-line detection methods for PCR may induce cross contamination. To overcome this problem, we herein fabricated an integrated CF-PCR and electrophoresis microfluidic chip. The optimal voltage applied in the electrophoresis part of the microfluidic chip was achieved by simulation in COMSOL. Coating the inside wall of the microchannel can inhibit electroosmotic flow and improve the resolution for DNA fragments. The temperature distribution of the serpentine part can meet the PCR and has no obvious suppressive effect on sample separation. Finally, we have performed the amplification of target genes forPorphyromonas gingivalis,Tannerella forsythia, andTreponema denticolaand detected the corresponding PCR products in the microfluidic chip within 11 min. Such work provides a new method for the rapid detection of bacteria. Keywords:capillary electrophoresis;periodontal pathogens;continuous flow PCR;microfluidic chip;fluorescence detection

Uniform and stable droplet generation is critical for accurate and efficient digital nucleic acid analysis (dNAA). In this study, an integrated microfluidic step emulsification device with wide-range droplet generation capability, small device dimensions, convenient fabrication strategy, low contamination and high robustness was developed. A tree-shaped droplet generation nozzle distribution design was proposed to increase the uniformity of droplet generation by equating flow rates, and the flow field in the design was numerically simulated. Theoretical analysis and comparative experiments on droplet size were performed regarding the influences of nozzle dimensions and surface properties. With incubation and hydrophobic reagent treatment, droplets as small as 73.1 μm were generated with multiplex nozzles of 18 μm (h) × 80 μm (w). The droplets were then collected into a standard PCR tube and an on-chip monolayer droplet collection chamber, without manual transfer and sample contamination. The oil-to-sample volume ratio in the PCR tube was recorded during collection. In the end, the droplets generated and collected using the microfluidic device proved to be stable and uniform for nucleic acid amplification and detection. This study provides reliable characteristic information for the design and fabrication of a micro-droplet generation device, and represents a promising approach for the realization of a three-in-one dNAA device under a step emulsification method. Keywords:step emulsification;micro-droplet;integrated microfluidic chip;nucleic acid detection

We present the development and application of a multilayer microfluidic lung chip designed to accurately replicate the human respiratory bronchi, providing an innovative platform for controlled particle deposition in the lung. By employing a quantitative control method of fluid velocity through the deformation of an elastic PDMS membrane, this platform mimics the passive breathing process in humans and allows for precise simulation of the respiration cycle. We utilized time-lapse photography of fluorescent particles in a water/glycerol solution to qualitatively observe fluid morphology in the channel, while a chip-aerosol exposure device combined with microscopy imaging was employed to visualise aerosol deposition. Both experimental and numerical simulation results showed that particle concentration decreased towards the distal generations of the lung, and that changes in breathing pattern significantly affected particle deposition trends. Furthermore, we found that increasing the residence time of particles in the channel facilitated deeper particle deposition, achievable by adjusting parameters such as breath-hold time, exhalation time, respiration cycle length, and tidal volume. The proposed microfluidic lung chip device has significant potential for future research in respiratory health and inhaled drug delivery, providing an efficient, cost-effective, and ethical alternative to traditional in vivo studies.

Syntactic foams with fly ash cenospheres or commercial microballoons as fillers have been widely used in various applications ranging from aerospace to marine fields and the automotive industry. However, these two extensively adopted fillers possess multiple shortcomings, such as variations in the composition, material degeneration and distinct structural heterogeneity, which will inevitably hamper accurate prediction of the structure–property relationship and the corresponding design of the syntactic foams, reducing material utilization. Here, we present a microfluidic-based approach integrated with a subsequent heat treatment process to engineer syntactic foam fillers with a predefined composition, specified dimensional scope and reduced structural heterogeneity. These fillers are fully guaranteed by the synergy of the flexible and controllable generation of droplet templates with hydrodynamic regulation and rational selection of the nanoparticle dynamic response with respect to the heating temperature. In addition, two distinct surface morphologies have been observed with a temperature demarcation point of 1473 K, further endowing the fillers with multiplicity and optionality, simultaneously laying the foundation to regulate the properties of the syntactic foams through the diversity of the filler selection. Then, we fabricated a syntactic foam specimen by mold casting, and the integrity of the fillers inside was verified using an elaborate buoyancy comparison experiment, exhibiting its potential value in lightweight related applications. As the fillers derived from our approach show significant advantages over conventional ones, they will provide considerable benefits for the regulation and improvement of syntactic foam fillers in many practical applications.

Induced phase separation extraction (IPSE) is an efficient sample clean-up technique that can replace liquid-liquid extraction (LLE). The purpose of this study was to miniaturize IPSE by carrying it out in a microfluidic chip. An IPSE chip was designed and evaluated for its ability to separate and purify samples on a microscale. The 5 × 2 cm chip was fed with a solution of polar to non-polar model compounds in acetonitrile-water (1:1). In the 100 µm wide and 40 µm deep microchannels, the sample solution was efficiently separated into two immiscible phases by adding a hydrophobic solvent as inducer. Analytes present in the sample solution each migrated to their own favorable phase upon phase separation. After optimization, extraction and fractionation were easily and efficiently achieved. The behavior of analytes with a pH-dependent partitioning could be influenced by adjusting the pH of the sample solution.Scutellaria baicalensisextract, used in Traditional Chinese Medicine (TCM), was successfully separated in aglycones and glycosides. In this microscale system, the sample and solvent consumption is reduced to microliters, while the time needed for the sample pretreatment is less than one minute. Additionally, the extraction efficiency can reach up to 98.8%, and emulsion formation is avoided. Keywords:fast sample clean-up;IPSE;miniaturization;green analytical chemistry;μLPME;low solvent consumption;microfluidics;on-chip separation;TCM;Scutellaria baicalensis

Aim: To explore the hepatotoxicity of copper sulfide nanoparticles (CuSNPs) toward hepatocyte spheroids. Materials & methods: Other than the traditional agarose method to generate hepatocyte spheroids, we developed a multi-concave agarose chip (MCAC) method to investigate changes in hepatocyte viability, morphology, mitochondrial membrane potential, reactive oxygen species and hepatobiliary transporter by CuSNPs. Results: The MCAC method allowed a large number of spheroids to be obtained per sample. CuSNPs showed hepatotoxicity in vitro through a decrease in spheroid viability, albumin/urea production and glycogen deposition. CuSNPs also introduced hepatocyte spheroid injury through alteration of mitochondrial membrane potential and reactive oxygen species, that could be reversed by N-acetyl-l-cysteine. CuSNPs significantly decreased the activity of BSEP transporter by downregulating its mRNA and protein levels. Activity of the MRP2 transporter remained unchanged. Conclusion: We observed the hepatotoxicity of CuSNPs in vitro with associated mechanisms in an advanced 3D culture system.

The authors describe an integrated microfluidic chip for immunodetection of the prostate specific antigen (PSA) by using giant magnetoimpedance (GMI) sensor. This chip contains an immunoreaction platform and a biomarker detection system. The immunoreaction platform contains an incubation chamber and a reactive chamber to implement immunological reaction in microfluidics. The system can detect PSA rapidly with ultra-high sensitivity. Both are fabricated by MEMS technology. Immunomagnetic beads (If PSA binds to its antibody (that is labeled with immunomagnetic beads; IMBs) it will be trapped on the surface of self-assembled film. Trapped IMBs generate a stray magnetic field under the magnetization of the external applied magnetic field and can be detected by the GMI sensor. The chip can detect PSA with a detection limit as low as 0.1 ng ∙ mL −1 and works in the 0.1 ng ∙ mL −1 to 20 ng ∙ mL −1 concentration range. Compared to established GMI biosensors, the magnetic microfluidic chip reduces assay time, and lends itself to fast detection. It also avoids complex handling steps, enhances reaction efficiency and decreases experimental errors.
An integrated magnetic microfluidic chip which contains immunoreaction platform and biomarker detection system was designed and microfabricated by micro-electromechanical systems (MEMS) technology to detect prostate specific antigen (PSA) rapidly, and has promise in Point-of-care (PoC) diagnostic applications.

The high sensitivity and long-term storage stability of a plasmonic substrate are vital for practical applications of the surface-enhanced Raman scattering (SERS) technique in real-world analysis. In this study, a rationally designed, ternary film-packaged, silver-coated gold-nanoparticle (Au@Ag NP) plasmonic array was fabricated and applied as a stable and high-performance SERS chip for highly sensitive sensing of thiabendazole (TBZ) residues in fruit juices. The ternary films played different roles in the plasmonic chip: a newborn poly(methyl methacrylate) (PMMA) film serving as a template for fixing the self-assembled closely packed monolayer Au@Ag NP array that provided an intensive hot spot, a fluorescent quantitative polymerase chain reaction adhesive film (qPCR film) acting as a carrier to retrieve the Au@Ag/PMMA film that was used to improve the robustness of the plasmonic array, and a polyethylene terephthalate (PET) film covered over the Au@Ag/PMMA/qPCR film performing as a barrier to improve the stability of the chip. The Au@Ag/PMMA/qPCR-PET film chip showed high sensitivity with an enhancement factor of 3.14 × 106, long-term storage stability without changing SERS signals for more than 2 months at room temperatures, and a low limit of detection for sensing TBZ in pear juice (21 ppb), orange juice (43 ppb), and grape juice (69 ppb). In addition, the procedure for fabricating the Au@Ag/PMMA/qPCR-PET film SERS chip was easy to handle, offering a new strategy to develop flexible and wearable sensors for on-site monitoring of chemical contaminants with a portable Raman spectrometer in the future.
