Prebiotic Absolute Asymmetric Synthesis of d‐Ribose on Wollastonite

Wendi Zhang,Yang Gao,Jiao Wang,Quanzheng Deng,Lu Han,Wanning Zhang,Yuxi Fang

ATP is the core substance of energy metabolism in all forms of life. The chirality of d ‐ribose is essential for ATP's function, ensuring efficient molecular recognition, metabolic stability, and co‐evolution with RNA in biological processes. However, origin of chirality of d ‐ribose is still unknown. Here, we demonstrate that wollastonite, an abundant Earth mineral, catalyzes the formose reaction between formaldehyde and glycolaldehyde and achieves high enantioselectivity (63.3%–91.3% ee ) toward d ‐ribose. The lower energy barrier for the transition state of d ‐glyceraldehyde compared to that of l ‐glyceraldehyde, which possesses a similar configuration to glyceraldehyde, results in the enantioselective synthesis of ribose. Experimental and computational studies reveal that the surface chirality of wollastonite, which is characterized by its wavy atomic arrangement, selectively reduces the adsorption energy of the d ‐glyceraldehyde intermediate compared to its l ‐enantiomer. This preferential adsorption kinetically promotes the d ‐glyceraldehyde pathway and steers the reaction toward d ‐ribose. Our work establishes wollastonite as an intrinsically enantioselective mineral catalyst, provides a mechanistic foundation for prebiotic chiral selection toward d ‐ribose, and highlights surface design principles for asymmetric synthesis. Wollastonite's inherent surface chirality lowers the energy barrier for d ‐glyceraldehyde, kinetically favoring its pathway over l ‐forms. This preferential adsorption steers the formose reaction toward d ‐ribose, offering a prebiotic mechanism for chiral selection.

D-Amino acids inhibit initial bacterial Adhesion: Thermodynamic evidence

Su-Fang Xing,Xue-Fei Sun,Alicia A. Taylor,Sharon L. Walker,Yi-Fu Wang,Shu-Guang Wang

Bacterial biofilms are structured communities of cells enclosed in a self-produced hydrated polymeric matrix that can adhere to inert or living surfaces. D-Amino acids were previously identified as self-produced compounds that mediate biofilm disassembly by causing the release of the protein component of the polymeric matrix. However, whether exogenous D-amino acids could inhibit initial bacterial adhesion is still unknown. Here, the effect of the exogenous amino acid D-tyrosine on initial bacterial adhesion was determined by combined use of chemical analysis, force spectroscopic measurement, and theoretical predictions. The surface thermodynamic theory demonstrated that the total interaction energy increased with more D-tyrosine, and the contribution of Lewis acid–base interactions relative to the change in the total interaction energy was much greater than the overall nonspecific interactions. Finally, atomic force microscopy analysis implied that the hydrogen bond numbers and adhesion forces decreased with the increase in D-tyrosine concentrations. D-Tyrosine contributed to the repulsive nature of the cell and ultimately led to the inhibition of bacterial adhesion. This study provides a new way to regulate biofilm formation by manipulating the contents of D-amino acids in natural or engineered systems. Biotechnol. Copper nanocluster-based fluorescence enhanced determination of d-penicillamine

Chunlei Ma,Wensheng Ren,Jianke Tang,Xiaoli Wang,Dongxin Ji,Rongqian Meng,Cuihong Zhang,Qi Wang

Taking advantage of the compelling properties of d -penicillamine ( d -PA) combined with copper, a method for the sensitive and selective determination of d -PA was established using copper nanocluster (Cu NC)-based fluorescence enhancement. d -PA molecules containing a thiol compound showed a strong tendency to combine with the surface of Cu NCs, causing the re-dispersion of nanoclusters and therefore fluorescence intensity was enhanced. Fluorescence enhancement efficiency of Cu NCs induced by d -PA was linear, with the d -PA concentration varying from 0.6–30 μg ml −1 (R 2 = 0.9952) and with a detection limit of 0.54 μg ml −1 . d -PA content in human urine samples was detected with recoveries of 104.8–112.99%. Fluorescence-enhanced determination of d -PA using Cu NCs was established for the first time and this rapid, easy and sensitive method should attract much attention for this application.

Improving Powder Performances of d-Ribose through Spherulitic Growth

Dongbo Wang,Xuan Zhou,Yanbo Liu,Mingyang Chen,Dandan Han,Junbo Gong

With the rise of research on functional sugar, we found that preparing crystalline powders into spherical crystals will significantly improve quality, stability, appearance, and texture. In this work, d-ribose (D-RIB) spherical crystal aggregates were prepared for the first time. And D-RIB spherical crystal aggregates with superior filterability, flowability, and tap density can be prepared in ethanol, 1-propanol, and isopropanol. Specifically, we used ethanol as an example to reveal the formation mechanism of D-RIB spherical crystal aggregates: solvent-induced crystal defects, which in turn trigger noncrystallographic branching and further spherulite growth. Finally, a comparison of the properties of the D-RIB spherical crystal aggregates with those of commercially available products is given, demonstrating the fabrication of D-RIB spherulites with better tap density and flowability by solvent-induced noncrystallographic branching. This will provide a new insight into the preparation of high-quality D-RIB crystals.

Congcong Jia,Min Zhu,Yingtung Lo,Meixiang Yu,Xinyi Xia,Weiwei Ji,Xiang Cao,Yakun Wan,Jianzhong Lu,Hai Zhang

Background Early screening for venous thromboembolism and the precise use of anticoagulant drugs are crucial, and further basic innovations are urgently needed to advance related diagnostic and therapeutic practices. This study aimed to develop an innovative antibody for detecting D-dimer and to verify its performance. The core aim was to screen a pair of variable domains of the heavy chain of heavy-chain antibodies (VHH) that can specifically recognize unique epitopes on the D-dimer molecule. This study is expected to promote technological advancements in the diagnosis, risk assessment, and efficacy monitoring of thrombotic diseases. Results D-dimer-positive clones underwent positive and negative screening across seven rounds of phage biopanning, yielding a pair of VHHs. The superior thermal stability of the sandwich VHH pair was demonstrated after continuous heating at 80 °C for up to 1 h by comparison with a commercial monoclonal antibody. Subsequently, by integrating the reliable and convenient Luminex platform with these novel VHHs, a new detection method for D-dimer was established under optimal conditions, with a wide quantitative range of 0.5 to 1000 ng/mL and detection limit as low as 0.25 ng/mL. The recovery rate of spiked samples ranged from 99.3% to 106.0%, with a relative standard deviation of 1.1% to 3.2%. Furthermore, this newly developed VHH-based Luminex assay was utilized to examine clinical D-dimer samples, and its performance was evaluated by correlation analysis with the clinical INNOVANCE® D-Dimer Assay, resulting in a Pearson correlation coefficient of 0.99086 (P < 0.0001). Significance This study identified a novel VHH pair targeting D-dimer, demonstrating good binding activity and exceptional thermal stability, making it highly effective for bioanalysis experiments. Utilizing the reliable and efficient Luminex platform, a detection method for D-dimer was developed, exhibiting substantial potential for applications in venous thrombus screening, anticoagulant efficacy assessment, and related fundamental research.

D-π-D type fluorescent compounds based on dehydroabietic acid triarylamine: Synthesis, photophysical properties and application as hole-transporting materials in perovskite solar cells

Yanan Gao,Liwei Dou,Hong Gao,Jie Song,Shibin Shang,Zhanqian Song

D-π-D hole-transporting materials have the potential to be excellent dopant-free hole-transporting materials (HTMs) for perovskite solar cells (PSCs) because of their tunable energy levels, high charge mobility, good solubility, and excellent film-forming properties. In this paper, two series of D-π-D type compounds, Series 1 (2TPA-CZ1~2TPA-CZ3) and Series 2 (2DTPA-CZ1~2DTPA-CZ3), were synthesized by linking a carbazole derivative (as a π-bridge) to triaryamine or dehydroabietic acid triarylamine groups (the donor groups) via C–C coupling reactions. The optical, electrochemical, thermal, and photovoltaic properties in PSCs of these compounds were fully investigated. The two series of compounds exhibit good ultraviolet absorption, fluorescence emission , and electrochemical and thermal properties. In addition, their highest occupied molecular orbital levels and energy gaps make them suitable for use as hole-transporting materials in PSCs. Among the PSCs developed, Device 1, based on 2TPA-CZ1, showed the best performance, with a power conversion efficiency (PCE) of 12.27%, a open circuit voltage (V OC ) of 1.13 V, a short-circuit photocurrent density (J SC ) of 15.10 mA cm −2 , and a fill factor (FF) of 71.61%, which is comparable to that of a device based on the conventional hole-transporting material Spiro-OMeTAD. While the PCE of Devices 4–6 was lower than the PCE of Devices 1–3, Devices 4–6 exhibit better stability. This may be due to the higher T g and better film-forming properties of compounds formed using the dehydroabietic acid skeleton. In additional, the introduction of alkyl groups on the carbazole moiety decreased the efficiency of the PSC devices. These results demonstrate that the 2TPA-CZ1 compounds are promising HTMs for PSC applications.

Novel potential and current type chiral amino acids biosensor based on L/D-handed double helix carbon nanotubes@polypyrrole@Au nanoparticles@L/D-cysteine

Qi Zhang,Mingxuan Fu,Haijun Lu,Xinyu Fan,Haiyang Wang,Yufan Zhang,Huan Wang

Novel chiral amino acids biosensor based on both potential difference and current ratio simultaneously was first established by employing left/right-handed double helix carbon nanotubes (L/D-DHCNT) with chiral structure as substrate material, and by self-assembly anchoring L/D-cysteine (L/D-Cys) on the surface of the left/right-handed double helix carbon nanotubes@Polypyrrole@Au nanoparticles nanocomposites (L/D-DHCNT@PPy@AuNPs). In this work, L/D-DHCNT@PPy@AuNPs@L/D-Cys was successfully constructed via a facile and environment-friendly process. Interestingly, in order to obtain highly dispersed AuNPs deposited on L/D-DHCNT, polypyrrole polymerized from pyrrole acts as a locating agent. Owing to the combination of unique performance of the L/D-Cys and L/D-DHCNT with chiral structure, and PPy and AuNPs with outstanding conductivity, an extremely efficient electrochemical chiral biosensing system was constructed that exhibited enhanced conductivity, sensitivity and chiral recognition capacity for tyrosine (Tyr), tryptophan (Trp) and glutamic acids (Glu) enantiomers. Utilizing both the potential difference and current ratio signal to commendably realize the electrochemical recognition and quantitative determination of amino acid enantiomers in their racemic solution. In addition, achieving the detection of Tyr in tablet samples by fabricated ultra-efficient electrochemical chiral biosensing system. Therefore, L/D-DHCNT@PPy@AuNPs@L/D-Cys as novel and ultrasensitive chiral biosensing system possesses the capability of chiral recognition of amino acid enantiomers, and it opens up the potential application of L/D-DHCNT@PPy@AuNPs@L/D-Cys in the field of chiral biosensors.

Reshaping the binding pocket of D-tagaturonate epimerase UxaE to improve the epimerization activity of C4-OH for enabling green synthesis of d-tagatose

Wenhao Xia,Shimeng Liu,Lihui Huang,Hao Wu,Tao Bai,Shanshan Wang,Fei Hao,Xiao Wang,Wen Wang

d -tagatose, a rare sugar with broad applications in the food and pharmaceutical industries, can be synthesized from d -fructose by epimerases, an approach with significant biosynthesis potential. However, so far there has been only one attempt to engineer tagatose 4‑epimerase through random mutagenesis. The lack of exploration has severely restricted the application of epimerases in the industrial production of d -tagatose. The previous research primarily focused on enhancing the catalytic activity of epimerases through random mutagenesis, with limited understanding of how substrate-adjacent residues affect enzyme efficiency, which was hence the subject of this study. First, using a semi-rational design approach, after three rounds of mutagenesis and screening, the mutant (S125D/S268A/Q124E) of D-tagaturonate epimerase (UxaE) from Thermotoga neapolitana was obtained, which increased d -tagatose production efficiency by 10.8-fold compared to the wild type, with a specific activity of 1505.6 U/g, representing the highest specific activity reported for a UxaE mutant to date. Molecular dynamics simulations showed that the substitution of Ser to Asp-125 enhanced substrate-enzyme interactions, while the substitution of Glu to Gln-124 reinforced the hydrogen bond network around the catalytic residue Glu-128, further reshaping the substrate-binding pocket and enhancing catalytic activity. This study for the first time explored the enhancement of the C4 epimerization function of epimerases by reshaping their substrate-binding pockets, thereby providing a feasible route for the industrial biosynthesis of d -tagatose.

Breaking the linear scaling relation through synergistic modulation of d-band center and spin state via p-d hybridization for enhanced bifunctional oxygen electrocatalysis

Guanhua Zhang,Mingwang Lu,Qitong Sheng,Guangjian Xing,Xiaojing Yang,Xiaofei Yu,Xinghua Zhang,Zunming Lu,Lanlan Li

The development of highly efficient and stable bifunctional electrocatalysts capable of simultaneously catalyzing the oxygen reduction reaction (ORR) and the oxygen evolution reaction (OER) is critical for advancing zinc-air batteries (ZABs). However, the linear scaling relationship (LSR) governing the adsorption energies of oxygen species imposes constraints on the concurrent enhancement of ORR and OER performance. In this study, we report the synthesis of a novel FeSn/NC catalyst that exhibits strong p-d orbital hybridization by incorporating p-block Sn metal into d-block Fe metal. This strategy facilitates the manipulation of the d-band center and enables the effective formation of low-spin (LS) Fe 3+ simultaneously. The regulation of the d-band center is identified as an effective strategy to enhance the adsorption energy of OOH* and OH* intermediates, while the LS Fe 3+ configuration can effectively weaken the adsorption strength of the O* intermediate. Consequently, this p-d hybridized Fe-based catalyst successfully overcomes the constraints imposed by LSR, thereby boosting bifunctional activity for both ORR and OER. The synthesized FeSn/NC catalyst demonstrates exceptional bifunctional performance with a potential difference (ΔE) of 0.65 V between ORR and OER activities. Furthermore, ZAB based on FeSn/NC achieves an impressive peak power density of 186.0 mW cm −2 , while maintaining stable operation over 700 h during cycling tests. This work offers new insights into designing high-performance bifunctional electrocatalysts through strategies involving synergistic modulation of d-band center and spin state.

Manipulating d-Band Center and d–p Orbital Coupling to Mitigate Adsorption–Desorption Trade-Off for Efficient CO2-to-CO Photoreduction over MOF Nanofibers

Yong Yang,Jianfeng Jiang,Ya-Nan Feng,Zanyong Zhuang,Fei-Fei Chen,Dantong Zhang,Yan Yu

The strong CO 2 adsorption and facile CO desorption are a trade-off issue for CO 2 -to-CO photoreduction pathways. Synchronous optimization of these two steps remains a substantial hurdle and has been less explored. Herein, an O/N mixed coordinated Co-NTA (NTA: nitrilotriacetic acid), featuring an ultralong nanofiber architecture, is reported for efficient CO 2 photoreduction. The mixed coordination can regulate the positions of d -band center of Co sites and also d – p orbital coupling with coordination atoms, which are recognized as main activity descriptors. Consequently, free energies of both steps of CO 2 →*CO 2 and *CO→CO are well optimized, making the CO 2 -to-CO photoreduction pathways more favorable thermodynamically. By contrast, the classic N-coordinated Co-MeIm (MeIm: 2-methylimidazole) and O-coordinated Co-BDC (BDC: 1,4-benzenedicarboxylic acid) suffer from unfavorable CO 2 adsorption and intractable CO desorption, respectively. In addition, achiral NTA molecules can direct the growth of Co-NTA into one-dimensional (1D) nanofibers, while Co-BDC and Co-MeIm possess typical 2D nanosheet and 3D polyhedron architectures. The fast electron transport along the axis directions makes charge-transfer kinetics most efficient over 1D Co-NTA. The thermodynamic and kinetic benefits jointly contribute to the 1.8-fold increase in CO production over 1D Co-NTA.

Puerarin alleviates memory deficits through the modulation of D‑serine-mediated N-methyl-D-aspartate receptor activity in presenilin 1/2 conditional double knockout mice

Waimao Gao,Kaifan Hu,Guang Yang,Jie Pan,Xiao Xu,Pingping You,Kun Li,Yan Zhao,Xingyu Wang,Ying Xu

Background Alzheimer’s disease (AD) is a neurodegenerative disorder characterized by progressive cognitive decline, the formation of amyloid-β plaques and neurofibrillary tangles, and synaptic dysfunction. Puerarin (PUE), an isoflavone C-glycoside extracted from Pueraria lobata , has demonstrated potential in attenuating AD-related cognitive decline; however, the mechanisms governing the effects of PUE remain incompletely understood. Purpose To investigate the mechanisms underlying PUE’s anti-AD effects. Methods Presenilin 1/2 conditional double knockout mice served as the AD model. Following one month of PUE administration, memory function was evaluated through behavioral testing, and hippocampal synaptic plasticity was assessed using electrophysiological recordings. D‑serine levels were quantified via ELISA. Additionally, Western blotting and qRT-PCR were employed to analyze the expression of glutamate receptor subunits and key enzymes involved in D‑serine metabolism. The mechanistic role of D‑serine was further validated through intrahippocampal D‑serine injections in mice and the application of a serine racemase (SR) inhibitor in N2a cells. Results PUE administration reversed memory impairment and enhanced both hippocampal long-term potentiation and synaptic transmission by increasing the open probability of N-methyl-D-aspartate receptor (NMDAR) channels. Furthermore, PUE elevated D‑serine levels and upregulated the expression of SR. Notably, these therapeutic effects were replicated by exogenous D‑serine and abolished by treatment with an SR inhibitor. Conclusion These findings indicate that PUE alleviates cognitive deficits by promoting SR-mediated D‑serine biosynthesis and subsequently restoring NMDAR activity. Accordingly, PUE represents a viable therapeutic candidate for the prevention and management of AD.
