Aging is a natural process accompanied by inflammation and oxidative stress and is closely associated with age-related diseases. As a direct precursor of glutathione, γ-glutamylcysteine (γ-GC) possesses antioxidant and anti-inflammatory properties; however, whether γ-GC plays an important role in anti-aging remains unknown. Here, we investigated the protective effects and mechanisms of γ-GC in D-galactose (D-gal)-induced senescence in PC12 cells and aging mice. Our results showed that γ-GC treatment significantly reduced the percentage of senescence-associated-β-galactosidase (SA-β-Gal)-positive cells and inhibited D-gal-induced cell cycle arrest in PC12 cells. The results of Nissl and hematoxylin and eosin (H&E) staining in mouse brain showed that γ-GC treatment markedly reversed the damage in the hippocampus of D-gal-induced aging mice. Moreover, γ-GC increased the phosphorylation of AMP-activated protein kinase (AMPK) to promote the nuclear translocation of glyceraldehyde-3-phosphate dehydrogenase (GAPDH) while inhibiting the nuclear translocation of deleted in breast cancer 1 (DBC1), which leads to the activation of sirtuin 1 (SIRT1) and deacetylation of p53 in the nucleus. Therefore, γ-GC may be a potential therapeutic candidate compound for the prevention and treatment of age-related diseases.

In this study, the synthesis of 3-O-β-D-galactopyranosyl-sn-glycerol (GG) was performed by the reverse hydrolysis of D-galactose and glycerol using β-galactosidase from Kluyveromyces lactis. Four process variables, reaction temperature (30.0–45.0 °C), reaction time (24–48 h), enzyme concentration (150.00–350.00 U/mL), and substrate molar ratio (glycerol:D-galactose, 7.5:12.5 mmol/mmol) were investigated and optimized via response surface methodology (RSM) for optimal GG synthesis. Both quadratic equations and the optimal reaction conditions were established. Results showed that the four variables, i.e., reaction temperature, reaction time, enzyme concentration, and substrate molar ratio had significant (p < 0.0001) effects on the synthesis of GG. The optimal reaction conditions were identified as 350.00 U/mL of β-galactosidase concentration and 8.65:1.00 of substrate molar concentration ratio (glycerol: D-galactose) at 39.8 °C and 48 h of reaction. Under these conditions, the GG concentration was 140.03 g/L and GG yield was 55.71%, which both were close to the predicted values (143.26 g/L and 56.73%). This finding proves the RSM to be a useful tool in optimizing process conditions for GG synthesis.

Acteoside, an important phenylethanol glycoside, is the main active component in Osmanthus fragrans flower. Our previous study found that acteoside showed high antiaging effect but its absorption rate was low. We speculated acteoside palliated aging-related cognitive impairment before being absorbed, that was intestinal homeostasis underlie the antiaging effect of acteoside. In this study, acteoside was confirmed to palliate cognitive impairment in d -galactose induced aging mice. Acteoside treatment dramatically reduced oxidative stress, alleviated intestinal inflammation, restored intestinal mucosal barrier, rebuilt gut microbiome structure and upregulated gut microbiome metabolites short-chain fatty acids (SCFAs) and amino acids (AAs). Furthermore, antibiotic treatment revealed that the antiaging ability of acteoside was abolished in microbiota depleted mice, which offered direct evidence for the essential role of gut microbiota in the attenuation of cognitive impairment of acteoside. Together, our study indicated that acteoside palliated cognitive impairment by regulating intestinal homeostasisand acteoside intake might be a promising nutritional intervention in prevention of neurodegenerative diseases.

Oxyresveratrol (Oxy) is a natural polyhydroxystilbene abundant in mulberry that has anti-inflammation and anti-oxidant activities. We evaluated the protective effect of Oxy in the context of the lipopolysaccharide and d -galactosamine (LPS/ d -GalN) induced acute liver injury. Oxy restricted the development of histopathological changes, markedly reduced the activity of alanine transaminase (ALT) and aspartate transaminase (AST), which are indicators of impaired liver function. Oxy significantly regulated the contents of oxidative stress related enzymes and products, and inhibited expressions of inflammatory mediators and cytokines. Oxy treatment diminished the Toll-like receptor 4/nuclear factor-kappa B (TLR4/NF-κB) signaling pathway in liver, activated the Kelch-like ECH-associated protein 1(Keap1)-nuclear factor erythroid 2-related factor 2 (Nrf2) pathway, and increased expressions of heme oxygenase 1 (HO-1) and quinine oxidoreductase 1(NQO1). Pretreatment with Oxy decreased LPS/ d -GalN stimulated hepatocyte apoptosis by efficaciously raising the B-cell lymphoma 2 (Bcl-2)/Bcl-2 associated X (Bax) ratio, inhibiting the expression and activation of caspases , and activating the phosphoinoside-3-kinase (PI3K)-Akt pathway. Our results demonstrate the hepatoprotective efficacy of Oxy. The protection is mainly due to the prevention of TLR4/NF-κB pathway activation, induced activation of Keap1-Nrf2 signaling pathway, and decreased hepatocyte apoptosis. Oxy warrants further study as a potential therapeutic agent candidate for the management of acute liver injury.

Background Bupleurum is one of the most important traditional Chinese medicines and an ingredient in many compound preparations. It is widely used together with other drugs in clinical practice, and thus there is great potential for drug–drug interactions. Saikosaponin D (SsD) is a major bioactive triterpenoid saponin extracted from Bupleurum with anti-inflammatory, anticancer, antioxidative, and antihepatic fibrosis effects. Effects of the main components of Bupleurum on cytochromes P450 (CYPs) need to be clarified in the clinical application of combination therapies of formulations containing SsD or Bupleurum . Purpose This study aimed to investigate the effects of SsD on the CYP1A2 and CYP2D6 mRNAs, protein expression, and relative enzyme activities in HepaRG cells. Methods HepaRG cells were cultured with SsD at concentrations of 0.5, 1, 5 and 10 μM for 72 hours. mRNA and protein expression of CYP1A2 and CYP2D6 were analyzed with real-time PCR and Western blot analysis. Relative enzyme activities were analyzed with HPLC based on consumption of the specific probe substrate. Results SsD significantly induced expression of mRNA and increased relative activity of CYP1A2 in HepaRG cells after the cells had been treated with SsD at concentrations of 1, 5 and 10 μM. SsD also induced protein expression of CYP1A2 at concentrations of 5 and 10 μM. SsD exhibited an inductive effect on CYP2D6 mRNA and protein expression, while increasing the relative activity of CYP2D6 at concentrations of 5 and 10 μM. Conclusion This study is the first to investigate the effect of SsD on CYP1A2 and CYP2D6 in HepaRG cells, and the results may provide some useful information on potential drug–drug interactions related to clinical preparations containing SsD or Bupleurum .

A novel 2-D photonic crystal (PC) sensing material for the visual detection of glucose with high selectivity at a physiological ionic strength (150 mM) is developed. A monolayer polystyrene crystalline colloidal array (CCA) is embedded in a phenylboronic acid (PBA) functionalized hydrogel film to diffract light to sensitively report on the hydrogel surface area. This 2-D PC sensor is superior to the previously reported 3-D PC sensors due to its fast preparation and simple detection. The binding of glucose would increase the cross-linking of the hydrogel that could shrink the hydrogel to increase the Debye diffraction ring diameter. At a physiological ionic strength of 150 mM, the 2-D PC sensor exhibits significant sensitivity for glucose across the entire human physiological glucose range. Additionally, the 2-D PC sensor shows high selectivity for glucose rather than other sugars (fructose and galactose). The diffraction color change of the 2-D PC sensor can be observed. For validation, the prepared 2-D PC hydrogel sensor is applied for the sensing of glucose in the artificial tear fluid.

The solubility of d -sorbitol was measured in methanol, ethanol, n-propanol, n-butanol, THF, acetone, ethyl acetate and acetonitrile at temperatures ranging from 278.15 to 333.15 K under atmospheric pressure by gravimetric method. The solubility data were correlated with modified Apelblat equation and λh equation which showed that the solubility of d -sorbitol in these organic solvents increased with increasing temperature. The thermodynamic properties of the dissolution process, including enthalpy, entropy and Gibbs energy , were calculated by means of van't Hoff analysis and Gibbs equation. According to the experimental results, ethanol showed to be a relatively better solvent in crystallization process of d -sorbitol.

Porous organic cages (POCs) are discrete, three-dimensional molecular species constructed from organic building blocks via covalent bonds. In this work, we found that RCC3, a tetrahedral [4 + 6] porous organic cage possessing 6 vicinal diamine pairs, can selectively separate d -glucose and d -fructose. RCC3 exhibited high binding capacity for d -glucose (0.4 mg/mg of RCC3). This binding process can be reversibly controlled. After a capture-dissociation cycle, the recovery yields of d -glucose and RCC3 were 75% and 83%, respectively. In addition, three binding/dissociation experiments on the binary system of d -glucose and RCC3 demonstrated its reusability. The binding mechanism of RCC3 to d -glucose was investigated via nuclear magnetic resonance spectroscopy (NMR), matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS), and control experiments with various POCs and monosaccharides, revealing the aminal formation between diamines on RCC3 and the carbonyl group of d -glucose. This work demonstrated a novel strategy for the molecular recognition and isolation of d -glucose utilizing porous organic cages.

In this work, RLWPF (Arg-Leu-Trp-Pro-Phe) and VLRLF (Val-Leu-Arg-Leu-Phe) were investigated for the effects against D-galactose (D-gal) induced cognitive impairment by modulating the gut microbiota composition. The effects on serum metabolite production were further investigated. The two novel peptides derived from walnut protein alkaline protease hydrolysates were predicted by docking to acetylcholinesterase (AChE) with the highest binding affinities, −10.3 and −9.9 kcal mol−1, considered as the potential neuroprotective peptides. In behavioral experiments, RLWPF and VLRLF treatment significantly restored spatial learning and memory impairment induced by D-gal. The results showed that RLWPF and VLRLF could alleviate cholinergic dysfunction, oxidative stress, and inflammation to varying degrees caused by D-gal-induced aging. Furthermore, 16S rRNA analysis revealed that RLWPF and VLRLF treatment improved cognitive impairment by regulating the composition of the gut microbiota and the abundance of harmful bacteria, including the ratio of Firmicutes to Bacteroidetes, Helicobacter, Allobaculum, Alistipes, Mucispirillum, and Odoribacter. In addition to the same regulation, RLWPF and VLRLF had their marker and regulatory flora. Studies based on the gut microbiota would allow a better understanding of the neuroprotective effects of walnut-derived peptides, supporting that walnut-derived peptides could be potential functional ingredients in foods and nutraceuticals or drug candidates in the treatment of cognitive dysfunction.

As one of the emerging inorganic graphene analogues, two-dimensional titanium carbide (Ti3C2) nanosheets have attracted extensive attention in recent years because of their remarkable structural and electronic properties. Herein, a sensitive and selective nanoprobe to fluorescently probe phospholipase D activity was developed on the basis of an ultrathin Ti3C2 nanosheets-mediated fluorescence quenching effect. Ultrathin Ti3C2 nanosheets with ∼1.3 nm in thickness were synthesized from bulk Ti3AlC2 powder by a two-step exfoliation procedure and further modified by a natural phospholipid that is doped with rhodamine B-labeled phospholipid (RhB-PL-Ti3C2). The close proximity between RhB and Ti3C2 leads to efficient fluorescence quenching (>95%) of RhB by energy transfer. Phospholipase D-catalyzed lipolysis of the phosphodiester bond in RhB-PL results in RhB moving away from the surface of Ti3C2 nanosheets and subsequent fluorescence recovery of RhB, providing a fluorescent “switch-on” assay for the phospholipase D activity. The proposed nanoprobe was successfully applied to quantitatively determine phospholipase D activity with a low limit of detection (0.10 U L–1) and to measure its inhibition. Moreover, in situ monitoring and imaging the activity of phospholipase D in living cells were achieved using this biocompatible nanoprobe. These results reveal that Ti3C2 nanosheets-based probes exhibit great potential in fluorometric assay and clinical diagnostic applications.

Targeting imaging to the desired site of action can increase the accuracy and effectiveness of diagnostic and treatment. In this work, a series of fluorescent carbon dots (CDs) were prepared by varying molar ratios of d -glucose (Glu) to l -aspartic acid (Asp). Their photophysical properties, morphologies and structures were investigated in detail. More important, the targeting ability was screened by confocal laser scanning microscopy and flow cytometry. The results indicate that CDs prepared from the optimal molar ratio of Glu/Asp (7:3) exhibit the highest targeting ability on C6 glioma cells. This work highlights the interplay of molecular design and corresponding functions, and open new possibility of developing state-of-art nanoparticles for biomedical applications.

Graphical A new ratio fluorescent probe (RF-probe) based on the mixing of red-emitting D-penicillamine (DPA) stabilized copper nanoclusters (DPA-CuNCs) with pyridoxal 5’-phosphate (PLP) was designed for sensitive and selective determination of Al 3+ . The excess accumulation of nonessential Al 3+ in the human body can cause serious health problems, thus there is significant interest in the detection of Al 3+ . A new ratio fluorescent probe (RF-probe) based on the mixing of red-emitting D-penicillamine (DPA) stabilized copper nanoclusters (DPA-CuNCs) with pyridoxal 5’-phosphate (PLP) was designed for sensitive and selective determination of Al 3+ . The −CHO group of PLP is expected to form an imine linkage with the free −NH 2 group available of the DPA on the surface of DPA-CuNCs. The content of Al 3+ could be mensurated by the as-prepared RF-probe with a linear response in the range of 1.33–46.67 μmol ⋅ L −1 , and corresponding limit of detection (LOD) was calculated to be 0.33 μmol ⋅ L −1 . Moreover, the RF-probe was successfully extended to the assay of Al 3+ in aluminum carbonate chewing tablets, baking powder, pond water and drink water with satisfactory results compared to the standard atomic absorption spectroscopy (AAS).
