Living materials that combine living cells and synthetic matrix materials have become promising research fields in recent years. While multicellular systems present exclusive benefits in developing living materials over single-cell systems, creating artificial multicellular systems can be challenging due to the difficulty in controlling the multicellular assemblies and the complexity of cell-to-cell interactions. Here, we propose a coculture platform capable of isolating and controlling the spatial distribution of algal-bacterial consortia, which can be utilized to construct photosynthetic living fibers. Through coaxial extrusion-based 3D printing, hydrogel fibers containing bacteria or algae can be deposited into designated structures and further processed into materials with precise geometries. In addition, the photosynthetic living fibers demonstrate a significant synergistic catalytic effect resulting from the immobilization of both bacteria and algae, which effectively optimizes sewage treatment for bioremediation purposes. The integration of microbial consortia and 3D printing yields functional living materials with promising applications in biocatalysis, biosensing, and biomedicine. Our approach provides an optimized solution for constructing efficient multicellular systems and opens a new avenue for the development of advanced materials.

Prosthesis implantation and fat transplantation are common breast reconstruction methods. In general, prosthesis implantation alone does not achieve a realistic enough appearance, and fat transplantation alone is difficult to achieve in the correct capacity. To date, no reports have focused on methods of combining fat with implanted prostheses for breast reconstruction. Using a newly designed bionic ink (i.e., polyether F127 diacrylate (F127DA) & poly(ethylene glycol) diacrylate (PEGDA)) and projection-based three-dimensional bioprinting (PBP), we report the development of a new method for printing porous prostheses. PEGDA was used to improve the printing precision of the prosthesis by increasing the gel point of F127DA and reducing the impact of external temperature. The compression modulus of the printed prosthesis was very close to that of prostheses currently used in clinical practice and to that of natural breasts. Finally, stromal vascular fraction gel (SVF-gel), a human fat extract, was injected into the pores of the synthesized prostheses to prepare a prosthesis mixed with adipose tissue. These were implanted subcutaneously in nude mice to observe their biological performance. After 14 and 28 days of observation, the prosthesis showed good biocompatibility, and adipose tissues grew well in and around the prosthesis. This result shows that a porous prosthesis containing pre-placed adipose tissues is a promising breast reconstruction material. Graphic abstract

Chronic wound infection often leads to irregular tissue closure and accompanies delayed healing and economy issues. Developing an ideal wound dressing that can control the occurrence of antibacterial infections and biological responses is highly desirable. In this study, a multifunctional hybrid hydrogel (GS@EG–Cu–CA NPs) containing synthesized thiolated gelatin, methacrylated silk fibroin, and (−)-epigallocatechin gallate–copper ionic–carrageenan nanoparticles (EG–Cu–CA NPs) was engineered by a thio–ene click reaction. The metal–polyphenol EG–Cu–CA NPs were encapsulated with kappa-carrageenan to enhance its aqueous-soluble, mechanical, and bioactive properties and endowed the hydrogel dressing with fascinating antibacterial, antioxidation, and nitric oxide (NO) generation by catalyzing. The hybrid hydrogels also illustrated a favorable cytocompatibility. Benefiting from the thio–ene click reaction, the hybrid hydrogels were injected and photocured rapidly in situ to cover an irregular wound. In an SD rat full-thickness skin-wound-infected model, the methicillin-resistant Staphylococcus aureus-infected wound covered with GS@EG–Cu–CA NPs was almost completely healed after 10 days. This study presents a facile design of hydrogel dressing incorporating metal–polyphenol nanoparticles, which demonstrates a promising potential way for dealing with effective wound infection management and other complicated wound healings.

Background Hydrogel dressings have been used as a crucial method to keep the wound wet and hasten the healing process. Due to safety concerns regarding the gel components, low mechanical adhesiveness, and unsatisfactory anti-inflammatory capacity qualities for practical uses in vivo, leading to the clinical translation of wound dressings is still difficult. Methods A type of composite hydrogel (acrylamide/polyethylene glycol diacrylate/tannic acid, ie, AM/PEGDA/TA) by double bond crosslinking, Schiff base, and hydrogen bond interaction is proposed. The mechanical characteristics, adhesiveness, and biocompatibility of the hydrogel system were all thoroughly examined. Additionally, a full-thickness cutaneous wound model was employed to assess the in vivo wound healing capacity of resulting hydrogel dressings. Results Benefiting the mechanism of multiple crosslinking, the designed composite hydrogels showed significant mechanical strength, outstanding adhesive capability, and good cytocompatibility. Moreover, the hydrogel system also had excellent shape adaptability, and they can be perfectly integrated into the irregularly shaped wounds through a fast in situ forming approach. Additional in vivo tests supported the findings that the full-thickness wound treated with the composite hydrogels showed quicker epithelial tissue regeneration, fewer inflammatory cells, more collagen deposition, and greater levels of platelet endothelial cell adhesion molecule (CD31) expression. Conclusion These above results might offer a practical and affordable product or method of skin wound therapy in a medical context.

Recently, hydrogels have gained enormous interest in three-dimensional (3D) bioprinting toward developing functional substitutes for tissue remolding. However, it is highly challenging to transmit electrical signals to cells due to the limited electrical conductivity of the bioprinted hydrogels. Herein, we demonstrate the 3D bioprinting-assisted fabrication of a conductive hydrogel scaffold based on poly-3,4-ethylene dioxythiophene (PEDOT) nanoparticles (NPs) deposited in gelatin methacryloyl (GelMA) for enhanced myogenic differentiation of mouse myoblasts (C2C12 cells). Initially, PEDOT NPs are dispersed in the hydrogel uniformly to enhance the conductive property of the hydrogel scaffold. Notably, the incorporated PEDOT NPs showed minimal influence on the printing ability of GelMA. Then, C2C12 cells are successfully encapsulated within GelMA/PEDOT conductive hydrogels using 3D extrusion bioprinting. Furthermore, the proliferation, migration and differentiation efficacies of C2C12 cells in the highly conductive GelMA/PEDOT composite scaffolds are demonstrated using various in vitro investigations of live/dead staining, F-actin staining, desmin and myogenin immunofluorescence staining. Finally, the effects of electrical signals on the stimulation of the scaffolds are investigated toward the myogenic differentiation of C2C12 cells and the formation of myotubes in vitro. Collectively, our findings demonstrate that the fabrication of the conductive hydrogels provides a feasible approach for the encapsulation of cells and the regeneration of the muscle tissue.

Osteosarcoma is a malignant bone tumor, which often occurs in adolescents. However, surgical resection usually fails to completely remove the tumor clinically, which has been the main cause of postoperative recurrence and metastasis, resulting in the high death rate of patients. At the same time, osteosarcoma invades a large area of the bone defect, which cannot be self-repaired and seriously affects the life quality of the patients. Herein, a bifunctional methacrylated gelatin/methacrylated chondroitin sulfate hydrogel hybrid gold nanorods (GNRs) and nanohydroxyapatite (nHA), which possessed excellent photothermal effect, was constructed to eradicate residual tumor after surgery and bone regeneration. In vitro , K7M2wt cells (a mouse bone tumor cell line) can be efficiently eradicated by photothermal therapy of the hybrid hydrogel. Meanwhile, the hydrogel mimics the extracellular matrix to promote proliferation and osteogenic differentiation of mesenchymal stem cells. The GNRs/nHA hybrid hydrogel was capable of photothermal treatment of postoperative tumors and bone defect repair in a mice model of tibia osteosarcoma. Therefore, the hybrid hydrogel possesses dual functions of tumor therapy and bone regeneration, which shows great potential in curing bone tumors and provides a new hope for tumor-related bone complex disease.

Recent advances in integrating elastomers and hydrogels have enabled numerous emerging and promising applications in ionotronics, soft machines, and soft robotics. A challenge has emerged to integrate elastomers and hydrogels of intricate configurations in various manufacturing processes. Here, a biomimetic strategy is reported to enable the integration of elastomers and hydrogels with regulable curved profiles. To do so, hydrophilic islands are imparted on the hydrophobic surface of an elastomer to allow the regulation of the profiles of the precursor, which can be cured into a hydrogel with high degree of fidelity of the profile and robust adhesion between the hydrogel and the elastomer. Parametric investigations pertaining to the diameter of the hydrophilic island and the volume of the liquid are first carried out to construct the designing space, which is then applied to a hydrogel precursor. An ionotronic luminescent array that can selectively light up in response to various extents of compression is fabricated. A soft tunable lens whose focal length can be mediated by the profile and mechanical stretch is also demonstrated. The proposed approach paves avenues for integrated elastomer–hydrogel hybrid systems.

Janus hydrogels have promising applications in tendon healing and anti-peritendinous adhesions. However, their complicated preparation methods, weak mechanical properties, and unstable adhesion interfaces have severely limited their application in suture-free and high-quality tendon healing. In this work, by controlling the interfacial distribution of free -COOH groups and cationic-π structures on both sides of the hydrogels, a series of PZBA-EGCG-ALC Janus hydrogels with varying degrees of asymmetric properties are successfully prepared using a simple and efficient one-step synthesis method. The tensile strength and elongation at the break of the Janus hydrogel are as high as 0.51 ± 0.04 MPa and 922.89 ± 28.59%. In addition, the Janus hydrogel can achieve a high difference in adhesion strength (nearly 20-fold) while maintaining a strong adhesion strength on their bottom sides (up to 524.8 ± 33.1 J m −2 ). In the spatial dimension, its excellent mechanical compliance and one-sided adhesion behavior can provide effective mechanical support and physical barriers for the injured Achilles tendons. More importantly, the Janus hydrogel can also minimize early inflammation generation in the time dimension via its ROS-responsive PZBA-EGCG prodrug macromolecules. This study provided a more effective and convenient suture-free strategy for constructing Janus hydrogels to promote high-quality tendon healing.

The field of cardiac tissue engineering has made significant strides in therapeutic and pharmaceutical applications, highlighted by the development of smart biomaterials. Scaffolds with appropriate properties mimicking the nature of a heart matrix will be highly beneficial for cardiac tissue engineering. In this study, high-aspect-ratio water-dispersed gold nanowires (AuNWs) were synthesized and incorporated into gelatin methacrylate (GelMA) hydrogels, demonstrating enhanced electrical conductivity and mechanical properties of the biomaterial scaffolds. Cardiac cells cultured on GelMA–AuNW hybrid hydrogels exhibited better biological activities such as cell viability and maturation state compared to those cultured on GelMA hydrogels. Moreover, cardiomyocytes showed synchronous beating activity and a faster spontaneous beating rate on GelMA–AuNW hybrid hydrogels. Our strategy of integrating high-aspect-ratio water-dispersed gold nanowires within gelatin methacrylate hydrogels provides a favorable biomaterial scaffold to construct functional cardiac tissue for further applications in cardiac tissue engineering and drug screening.

Simulating the structure and function of blood capillaries is very important for an in-depth insight into their role in the human body and treatment of capillary-related diseases. Due to the similar composition and structure, hollow hydrogel microfibers are well-recognized as potential biomimetic blood capillaries. In this paper, we report a novel, facile, and reproducible method to fabricate coaxial microfluidic chips via 3D printing-assisted soft lithography and then hollow hydrogel microfibers using the as-prepared coaxial microfluidic chips. Instead of traditional photoresist-based lithography, 3D printing of gelatin hydrogel under various extrusion pressures is used to construct sacrificial templates of coaxial microfluidic chips. Various solid and hollow hydrogel microfibers with complicated and hierarchical structures can be obtained via multitype coaxial microfluidic chips or a combination of coaxial microfluidic fabrication and post-treatment. The as-formed hollow hydrogel microfibers are evaluated in detail as biomimetic blood capillaries, including physicochemical and cytological properties. Our results prove that the hollow hydrogel microfibers exhibit excellent mass transport capacity, hemocompatibility, semipermeability, and mechanical strength, and their barrier function can be further enhanced in the presence of endothelial cells. Overall, our 3D printing-assisted fabrication strategy provides a new technique to construct microfluidic chips with complicated 3D microchannels, and the resulting hollow hydrogel microfibers are promising candidates for blood capillaries.

Tissue-engineered scaffolds have been extensively explored for treating bone defects; however, slow and insufficient vascularization throughout the scaffolds remains a key challenge for further application. Herein, a versatile microfluidic 3D printing strategy to fabricate black phosphorus (BP) incorporated fibrous scaffolds with photothermal responsive channels for improving vascularization and bone regeneration is proposed. The thermal channeled scaffolds display reversible shrinkage and swelling behavior controlled by near-infrared irradiation, which facilitates the penetration of suspended cells into the scaffold channels and promotes the prevascularization. Furthermore, the embedded BP nanosheets exhibit intrinsic properties for in situ biomineralization and improve in vitro cell proliferation and osteogenic differentiation. Following transplantation in vivo, these channels also promote host vessel infiltration deep into the scaffolds and effectively accelerate the healing process of bone defects. Thus, it is believed that these near-infrared responsive channeled scaffolds are promising candidates for tissue/vascular ingrowth in diverse tissue engineering applications.
