Protective Effects of Lactobacillus gasseri against High-Cholesterol Diet-Induced Fatty Liver and Regulation of Host Gene Expression Profiles

Tianhua He,Nikita Lykov,Xu Luo,Huiling Wang,Zhanxiang Du,Ziyi Chen,Shitian Chen,Lin Zhu,Ye Zhao,Chimeng Tzeng

Fatty liver is one of the most pervasive liver diseases worldwide. Probiotics play an important role in the progression of liver disease, but their effects on host regulation are poorly understood. This study investigated the protective effects of lactobacillus gasseri (L. gasseri) against high-cholesterol diet (HCD)-induced fatty liver injury using a zebrafish larvae model. Liver pathology, lipid accumulation, oxidative stress and hepatic inflammation were evaluated to demonstrate the changes in a spectrum of hepatic injury. Moreover, multiple indexes on host gene expression profiles were comprehensively characterized by RNA screening. The results showed that treatment withL. gasseriameliorated HCD-induced morphological and histological alterations, lipid regulations, oxidative stress and macrophage aggregation in the liver of zebrafish larvae. Furthermore, the enrichment of the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway revealed that the core pathways ofL. gasseriregulation were interleukin-17 (IL-17) signaling, phosphoinositide 3-kinase (PI3K)-AKT signaling pathway, the regulation of lipolysis and adipocytes and fatty acid elongation and estrogen signaling. The genes at key junction nodes, hsp90aa1.1, kyat3, hsd17b7, irs2a, myl9b, ptgs2b, cdk21 and papss2a were significantly regulated byL. gasseriadministration. To conclude, the current research extends our understanding of the protective effects ofL. gasseriagainst fatty liver and provides potential therapeutic options for fatty liver treatment. Keywords:fatty liver;Lactobacillus gasseri;high cholesterol diet;zebrafish;IL-17

Effect of Lactobacillus acidophilus fermentation on the composition of chlorogenic acids and anti-hyperuricemia activity of Artemisia selengensis Turcz

Yin Wan,Dengxiao Wang,Yuefeng Shen,Yanru Chen,Jin Qian,Guiming Fu

Background: Lactobacillus fermentation has become a typical food processing method with the development of the modern food industry. Aims: This study aimed to assess the effects of Lactobacillus acidophilus NCUF202.2 fermentation on Artemisia selengensis Turcz (AST) in relieving hyperuricemia (HUA) and the mechanisms involved. Results: The fermented Artemisia selengensis Turcz extracts (FASTE) could significantly increase the content of free polyphenol and enhance the inhibitory effect on xanthine oxidase (XOD) in vitro. The result of ultrafiltration coupled with UPLC-MS/MS screened out the components that might be combined with XOD in the caffeoylquinic acids (CQAs) of Artemisia selengensis Turcz extracts (ASTE) as 5-CQA, 3-CQA, 4-CQA, 1-CQA, 3,4-diCQA, 3,5-diCQA, 1,5-diCQA and 4,5-diCQA as well as caffeic acid (CA) of FASTE except the above. Our results also showed that ASTE and FASTE could alleviate HUA in rats. In contrast, FASTE had a better ability to reduce serum adenosine deaminase (ADA) activity than ASTE. Furthermore, FASTE could restore catalase (CAT) in HUA model rats in vivo, and superoxide dismutase (SOD) activity to a better degree, thereby inhibiting the production of excess malondialdehyde (MDA). Under the intervention of ASTE and FASTE, the levels of serum pro-inflammatory and anti-inflammatory factors in rats tended to be normal. However, FASTE could increase the expression of the uric acid secretion protein OAT1 and decrease the expression of reabsorption proteins URAT1 and GLUT9 in model rats, thereby reducing the serum uric acid (SUA) level in model rats. ASTE and FASTE can increase the abundance of beneficial bacteria, such as Bifidobacterium and Akkermansiaceae. Conclusion: Overall, part of diCQAs and CQAs was decomposed after fermentation. FASTE had a more substantial HUA-relieving effect than ASTE. It is expected to be applied to functional foods and contribute to the research and development of uric acid lowering functional foods.

Correlation between microorganisms and flavor of Chinese fermented sour bamboo shoot: Roles of Lactococcus and Lactobacillus in flavor formation

Caixia Chen,Guiguang Cheng,Yaping Liu,Yongxin Yi,Dongjie Chen,Lu Zhang,Xuejiao Wang,Jianxin Cao

Sour bamboo shoot is a popular traditional fermented vegetable in southern China. The current study aims to clarify the relationship among physicochemical properties, flavor, and microbial compositions of sour bamboo shoots during fermentation. Four organic acids (OAs) were considered to be the main acids in sour bamboo shoots. At least 7 volatile compounds (OAV >1) were identified as the characteristic aroma substances, especially p -cresol, which accounted for the sour odor of sour bamboo shoots. The microbial community showed that Firmicutes and Proteobacteria were the dominant phyla, while Lactococcus and Lactobacillus were the dominant genera in sour bamboo shoots. Correlation analysis indicated that Firmicutes positively correlated with p -cresol, phenol , 2-methoxy-phenol, malic acid , total titratable acidity (TTA), and some OAs. Lactococcus was positively correlated with phenol, 2-methoxy-phenol, benzyl alcohol, TTA, some OAs, and amino acids (AAs). This study provided a theoretical basis for screening indigenous aroma-producing microorganisms as the starter of sour bamboo shoots.

Different oligosaccharides and sodium caseinate glycosylated conjugates for the microencapsulation of Lactobacillus fermentum to improve stability and cholesterol-lowering activity

Chenglong Lv,Xiangfei Li,Jie Liu,Jiayan Gu,Fei Gao,Jing Sun,Xinyi Pang,Yingjian Lu

In this study, we aimed to develop microcapsules using sodium caseinate (NaCas)-oligosaccharides (OS) Maillard reaction conjugates (MRCs) to enhance the stability of Lactobacillus fermentum Q11 (LfQ11) in gastrointestinal fluids and food processing environments. Xylooligosaccharide (XOS), fructooligosaccharide (FOS), and inulin (INU) were conjugated with NaCas via the Maillard reaction to form MRCs for the microencapsulation of LfQ11. The characterization revealed that NaCas-XOS3h exhibited the highest grafting degree (24.97 %) and superior emulsifying activity, while NaCas-INU4h demonstrated enhanced prebiotic activity. Characterization of the microcapsules indicated that LfQ11@NaCas-XOS, LfQ11@NaCas-FOS, and LfQ11@NaCas-INU achieved high encapsulation efficiency (above 90 %) and were stabilized by hydrophobic interactions. Moreover, LfQ11@NaCas-XOS maintained higher viability than free LfQ11 after exposure to gastrointestinal fluids (82.46 %), heat stress at 70 °C (3.95 Log CFU/mL), and storage at 25 °C for 28 days (7.42 Log CFU/mL) than other microcapsules. All three MRCs-based microcapsules provided enhanced protection against ionic stress and acidic stress. Furthermore, all microcapsules exhibited low cytotoxicity and excellent biocompatibility. Notably, LfQ11@NaCas-XOS significantly reduced cholesterol levels in RAW264.7 foam cells more effectively than other treatments. These findings offer valuable insights into the application of different OS-protein conjugates as microencapsulated wall materials and the development of probiotic agents with cholesterol-lowering functions.

Restraining the TiO2 nanoparticles-induced intestinal inflammation mediated by gut microbiota in juvenile rats via ingestion of Lactobacillus rhamnosus GG

Yu Zhao,Yizhou Tang,Ling Chen,Sidi Lv,Shanji Liu,Penghui Nie,Zoraida P. Aguilar,Hengyi Xu

Human were given a lot of opportunities to ingest TiO 2 NPs in the environment. Children have low, sensitive intestinal tolerance, and they could be exposed to higher levels of TiO 2 NPs than adults. Few studies have been conducted on the interaction between TiO 2 NPs and juvenile intestine phase models. Thus, in this work, weaning rats were orally exposed to TiO 2 NPs for 7 and 14 days. Results indicate that Ti accumulated in the intestine, liver, and feces. Inflammatory infiltration damage was observed in the colonic epithelial tissue, and gut microbiota fluctuated with a decreased abundance of Lactobacilli in feces. Oral supplementation with Lactobacillus rhamnosus GG (LGG) lessened TiO 2 NPs-induced colonic inflammatory injury, which might due to downregulation of nuclear factor kappa-B (NF-κB). Meanwhile, LGG maintained normal intestinal microbiome homeostasis, thereby improving TiO 2 NPs-induced colon injury in juvenile rats. Moreover, fecal microbiota transplant (FMT) experiment indicated possible TiO 2 NPs-induced intestinal microbiota disorder led to colonic inflammation. Our works suggested the urgent need for additional studies on the risk safety assessment, mechanism, and prevention of juvenile health damage from exposure to TiO 2 NPs.

Complex pectin metabolism by Lactobacillus and Streptococcus suggests an effective control approach for Maillard harmful products in brown fermented milk

Zibian Fan,Wei Jia,An Du,Lin Shi

Harmful Maillard reaction products (HMRPs) derived from brown fermented milk pose a potential threat to human health, but the conversion mechanism during the manufacturing process remains elusive and urgently needs to be controlled. Acrylamide (FC 2.14, adjusted p -value = 0.041), 5-hydroxymethylfurfural (FC 2.61, adjusted p -value = 0.026) and methylglyoxal (FC 2.07, adjusted p -value = 0.019) were identified as the significantly increased HMRPs after browning in this study and the analysis of proteomics integrated with untargeted metabolomics demonstrated that the degradation of HMRPs was jointly accomplished by Streptococcus thermophilus and Lactobacillus bulgaricus . The galactose oligosaccharide metabolism in Streptococcus thermophilus was identified as a key biochemical reaction for HMRPs degradation, and the hydrolysates of pectin could be utilized as prebiotics for Streptococcus thermophilus . Eighteen classes of enzymes of L. bulgaricus and Streptococcus thermophilus related to energy metabolism were upregulated in the pectin-added group, indicating that the entry of acrylamide and methylglyoxal into the tricarboxylic acid cycle was accelerated. NAD-aldehyde dehydrogenase and alanine dehydrogenase are enzymes belonging to Streptococcus thermophilus, and their downregulation accelerated the efflux of acetate, which was beneficial for the proliferation of L. bulgaricus and prevented the conversion of pyruvate to l -alanine, thus facilitating the energy metabolism. The recoveries and relative standard deviations of the intraday and interday precision experiments were 89.1%-112.5%, 1.3%-8.4% and 2.1%-9.4%, respectively, indicating that the developed approach was credible. Sensory evaluation results revealed that the brown fermented milk added with pectin had a better flavor, which was due to the fact that the supplement of polysaccharide promoted the fatty acid metabolism of lactic acid bacteria and increased the aroma substances including octoic acid and valeric acid. This study provided an insight into the formation and degradation mechanism of HMRPs in brown fermented milk, aiming to reduce the intake of advanced glycation end products in the diet.

Cotton Fibers with A Lactic Acid-like Surface for Re-establishment of Protective Lactobacillus Microbiota by Selectively Inhibiting Vaginal Pathogens

Yuzheng Wu,Pei Liu,Qing Liao,Tao Jin,Zhengwei Wu,Guomin Wang,Huaiyu Wang,Paul K. Chu

Failure to reconstruct the Lactobacillus microbiota is the major reason for the recurrence of vaginal infection. However, most empiric therapies focus on the efficacy of pathogen elimination but do not sufficiently consider the viability of Lactobacillus . Herein, cotton fibers with a lactic acid-like surface (LC) are fabricated by NaIO 4 oxidation and L-isoserine grafting. The lactic acid analog chain ends and imine structure of LC can penetrate cell walls to cause protein cleavage in Escherichia coli and Candida albicans and inhibit vaginal pathogens. Meanwhile, the viability of Lactobacillus acidophilus is unaffected by the LC treatment, thus revealing a selective way to suppress pathogens as well as provide a positive route to re-establish protective microbiota in the vaginal tract. Moreover, LC has excellent properties such as good biosafety, anti-adhesion, water absorption, and weight retention. The strategy proposed here not only is practical, but also provides insights into the treatment of vaginal infections. This article is protected by copyright. All rights reserved

Preparation of alginate-whey protein isolate and alginate-pectin-whey protein isolate composites for protection and delivery of Lactobacillus plantarum

Xin Luo,Siqi Fan,Zhuosui He,Fangfang Ni,Chengzhi Liu,Min Huang,Lei Cai,Gerui Ren,Xuan Zhu,Qunfang Lei,Wenjun Fang,Hujun Xie

Probiotics are sensitive to external conditions, resulting in low survival rates after being ingested or during food production, transportation and storage. In order to improve the survival rate of Lactobacillus plantarum (LP) during gastrointestinal digestion, storage, and freeze-drying, alginate-whey protein isolate (ALG-WPI) and alginate-pectin-whey protein isolate (ALG-PEC-WPI) composites were employed to encapsulate LP. The encapsulation efficiency of ALG-WPI-LP and ALG-PEC-WPI-LP beads both reached more than 99 % . Scanning electron microscopy (SEM) indicated that dense and rough aggregates were formed on the surface of both composites, and attached LP cells could be observed inside the beads. The ALG-WPI and ALG-PEC-WPI composites can protect the viability of LP in simulated gastric fluid (SGF) and release the probiotics in simulated intestinal fluid (SIF). The storage stability of LP at 4 °C was improved by about 15 % in comparison with bare LP and the survival rates of LP in ALG-WPI-LP and ALG-PEC-WPI-LP powders after freeze-drying were increased by 65.37 % and 72.06 %, respectively. The formation mechanism of ALG-WPI and ALG-PEC-WPI composites was further explored by fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD) and thermogravimetric analysis (TGA). The ALG-WPI and ALG-PEC-WPI composites have great potential to protect and deliver probiotics in food systems.

Synergistic Effect of Rice Bran and Lactobacillus Ferulate Esterase on Dextran Sodium Sulfate (DSS)-Induced Ulcerative Colitis (UC) in Mice

Yingjian Lu,Le Li,Zhuolun Xin,Xiangfei Li

UC is a globally escalating intestinal disorder with limited safe and effective interventions. This study investigated the synergistic therapeutic effects and mechanisms of Lactobacillus johnsonii 46 ( L. johnsonii 46) (a feruloyl esterase-producing strain) and rice bran (LJRB) on DSS-induced UC in C57BL/6J mice. The animal groups were divided into control group (CON), DSS group (MOD), sulfasalazine group (SUL), rice bran group (RBN), L. johnsonii 46 group (LAJ), and LJRB treatment group (RBL). The organ index, disease index, and biochemical indicators of mice were measured, the histopathology of the colon was analyzed. The expression of inflammatory factor, tight junction protein and TLRs/TRAF6/NF-κB signaling pathway genes were detected by Real-Time Quantitative Polymerase Chain Reaction (RT-qPCR). LJRB supplementation exerted superior protective effects compared to single rice bran (RBN) or L. johnsonii 46 (LAJ) intervention: it significantly alleviated DSS-induced weight loss (RBL group: 1.46 ± 0.21 g vs. MOD group: ∼3 g, p < 0.05), restored colon length, and reduced the disease activity index (DAI) by ∼50%. Biochemical and molecular analyses showed LJRB remarkably suppressed serum pro-inflammatory cytokines (IL-1β, TNF-α, IL-6; 40%–60% reduction vs. MOD group) while elevating anti-inflammatory IL-10 (∼35% increase), enhanced colonic antioxidant capacity (reduced MDA by ∼30% and increased SOD, GSH, GSH-Px by 25%–40%), and enriched cecal short-chain fatty acids (SCFAs) with acetic acid (0.99 µmol/g), propionic acid (0.31 µmol/g), and butyric acid (0.21 µmol/g) significantly higher than the MOD group. Mechanistically, LJRB synergistically inhibited the TLRs/TRAF6/NF-κB signaling pathway, as indicated by downregulated mRNA expression of TLR4, MyD88, IKKβ, and NF-κB p65 (30%–50% reduction vs. MOD group) and decreased phosphorylation of IKKβ, IκBα, and NF-κB p65 proteins. Histopathological observations confirmed LJRB preserved colon tissue integrity, reduced goblet cell loss, and alleviated mucosal erosion and edema. Collectively, these findings demonstrate LJRB alleviates UC through multi-targeted mechanisms (anti-inflammation, antioxidation, SCFA production promotion, and nuclear factor kappa-B (NF-κB) pathway inhibition), highlighting its potential as a novel natural dietary supplement for UC prevention and management. Graphical Synergistic protective effects of rice bran combined with Lactobacillus johnsonii 46 on DSS-induced ulcerative colitis in mice: Evaluated by clinical, biochemical, histopathological and molecular biological assays.

Targeted inactivation of protein disulfide-isomerase 1 by Lactobacillus reuteri-derived reuterin promotes intestinal stem cell proliferation via ROS signaling

Jiaqi Chen,Jiayue Zhao,Yibin Xu,Xianglin Fei,Shujie Xu,Binke Chen,Wenzi Wu,Jiachen Zhuang,Xiuan Zhan,Aikun Fu

Enteric Lactobacillus plays a crucial role in maintaining the intestinal barrier integrity. Lactobacillus reuteri , one of the most described probiotic species of Lactobacillus , has been shown to alleviate enteritis-associated intestinal mucosal damage by stimulating the expansion of intestinal stem cells (ISCs). However, the underlying ligand-protein interactions remain unexplored. Therefore, this study aimed to elucidate the specific molecular mechanisms by which L. reuteri and its metabolites govern ISC stemness. We employed broiler enteritis model and organoid culture to compare the probiotic effect of a wild-type (WT) L. reuteri strain and a reuterin-deficient isogenic mutant. Compared to the WT strain, the deficient strain exhibited an impaired ability to alleviate inflammation, restore the intestinal barrier, and stimulate ISC proliferation both in vivo and ex vivo, indicating that reuterin is the essential effector. Furthermore, bioorthogonal click chemistry combined with in silico molecular docking were utilized to map the direct host protein targets. We identified protein disulfide-isomerase 1 (PDIA1), an endoplasmic reticulum (ER)-resident redox-active protein, as the direct binding partner of reuterin, characterized by a strong binding affinity. The inactivation of PDIA1 by reuterin binding increased ER-derived reactive oxygen species (ROS) level, leading to the upregulation of Wnt/β-catenin pathway and the maintenance of ISCs. In conclusion, by inactivating the ER-resident PDIA1, L. reuteri -derived reuterin promoted ISC proliferation and ameliorated intestinal mucosal damage.

PUL/HPMC water-in-water Pickering emulsions: A food-grade microculture platform for efficient encapsulation and cultivation of Lactobacillus plantarum

Pengrui Wu,Yefan Niu,Zejie Chen,Chunling Nie,Yijun Liu,Zhonghui Shen,Zhihong Song,Xindi Wei,YanBo Wang,Hong Zeng,JianGuo Wang

In this study, pullulan-in-hydroxypropyl methylcellulose (PUL/HPMC) water-in-water Pickering emulsions stabilized by cellulose nanocrystals (CNCs) were prepared for the encapsulation and cultivation of L. plantarum . Results demonstrated that CNCs were mainly adsorbed at the PUL-HPMC interface, thereby effectively inhibiting the aggregation and coalescence of emulsion droplets. L. plantarum was encapsulated in emulsions and primarily distributed in the PUL phase. It was found that the optimal encapsulation concentration of L. plantarum was 9.67 log CFU/g, and the optimal addition of MRS broth was 1.0 wt%. Finally, the viable counts of L. plantarum increased from 4.44 to 7.42 log CFU/g during 30 h of cultivation. Furthermore, an initial pH 5.5 afforded the optimal cultivation performance for L. plantarum , with viable counts increasing from 4.00 to 7.06 log CFU/g during 24 h of cultivation. This study provides a novel microculture platform for efficient L. plantarum cultivation and fully aqueous emulsion systems with food-grade application potential for probiotics-based functional foods. Effect of the quorum sensing signal molecule auto-inducer-2 on fermentation performance of Lactobacillus delbrueckii subsp. Bulgaricus DPUL-F36

Xueqi Lu,Mengying Sun,Yuqing Chen,Dashuai He,Yanfeng Tuo,Guangqing Mu,Yinglong Song

Fermented milk is a typically product of co-fermentation by a variety of lactic acid bacteria (LAB). Quorum sensing (QS), as an intercellular communication mechanism of bacteria, may serve as a key factor influencing the quality of fermented milk. In this study, the fermentation performance of Lactobacillus delbrueckii subsp . bulgaricus DPUL-F36 were evaluated after supplementation with auto-inducer-2 (AI-2). The protein-protein interaction network (PPIN) was used to retrieve the interactions between QS genes and four major metabolic pathway genes. The expression of luxS gene in DPUL-F36 may affect amino acid metabolism, carbohydrate metabolism and cell wall formation. The key differential metabolites were identified by metabolomics analysis. A variety of amino acids including L-Glutamic, L-Tryptophan, L-Threonine, L-Histidine and L-Glutamine were found to be up-regulated in response to the addition of AI-2. Malic acid and succinate gradually accumulate in the tricarboxylic acid cycle. The consumption of citric acid increased with the fermentation process. In terms of rheological and texture characteristics of fermented milk, addition of AI-2 could effectively improve its shear stress resistance, firmness and index of viscosity. Our research provided a theoretical basis for improving the fermentation performance of DPUL-F36 strains, and support for the screening of excellent composite starter culture.
