The development of theranostic systems capable of diagnosis, therapy, and target specificity is considerably significant for accomplishing personalized medicine. Here, a multifunctional rattle-type nanoparticle (MRTN) as an effective biological bimodal imaging and tumor-targeting delivery system is fabricated, and an enhanced loading ability of hydrophobic anticancer drug (paclitaxel) is also realized. The rattle structure with hydrophobic Fe 3 O 4 as the inner core and mesoporous silica as the shell is obtained by one-step templates removal process, and the size of interstitial hollow space can be easily adjusted. The Fe 3 O 4 core with hydrophobic poly(tert-butyl acrylate) (PTBA) chains on the surface is not only used as a magnetic resonance imaging (MRI) agent, but contributes to improving hydrophobic drug loading amount. Transferrin (Tf) and a near-infrared fluorescent dye (Cy 7) are successfully modified on the surface of the nanorattle to increase the ability of near-infrared fluorescence (NIRF) imaging and tumor-targeting specificity. In vivo studies show the selective accumulation of MRTN in tumor tissues by Tf-receptor-mediated endocytosis. More importantly, paclitaxel-loaded MRTN shows sustained release character and higher cytotoxicity than the free paclitaxel. This theranostic nanoparticle as an effective MRI/NIRF bimodal imaging probe and drug delivery system shows great potential in cancer diagnosis and therapy.

Multifunctional nanomaterials with simple structure and good biosafety, integrating multimodal imaging and therapeutic functions, can facilitate the development of clinical cancer treatments. Here, a simple but powerful pure bismuth based nanoparticle (Gd-PEG-Bi NPs) was developed from pure Bi NPs and gadolinium-diethylenetriaminepentaacetic acid-bis-tetradecylamide, which not only shows high quality MRI/CT/PAI triple-modal imaging, but can also be a potent photothermal therapy agent under the guidance of the triple-modal imaging. The Gd-PEG-Bi NPs showed good stability and excellent biocompatibility. In vitro and in vivo study demonstrated that Gd-PEG-Bi NPs have ultrahigh X-ray attenuation coefficient, short T1 relaxation time in MRI, and strong PAI signal. Following the imaging diagnosis, the excellent light-to-heat conversion efficiency of Gd-PEG-Bi NPs was capable of suppressing the tumor growth effectively under near-infrared laser radiation in vivo . Such multifunctional nanoparticles were ideal candidates for cancer diagnosis and treatment.

Improving the effectiveness of cancer therapy will require tools that enable more specific cancer targeting and improved tumor visualization. Theranostics have the potential for improving cancer care because of their ability to serve as both diagnostics and therapeutics; however, their diagnostic potential is often limited by tissue-associated light absorption and scattering. Herein, we develop CuInSe2@ZnS:Mn quantum dots (QDs) with intrinsic multifunctionality that both enable the accurate localization of small metastases and act as potent tumor ablation agents. By leveraging the growth kinetics of a ZnS shell on a biocompatible CuInSe2 core, Mn doping, and folic acid functionalization, we produce biocompatible QDs with high near-infrared (NIR)-II fluorescence efficiency up to 31.2%, high contrast on magnetic resonance imaging (MRI), and preferential distribution in 4T1 breast cancer tumors. MRI-enabled contrast of these nanoprobes is sufficient to timely identify small metastases in the lungs, which is critically important for preventing cancer spreading and recurrence. Further, exciting tumor-resident QDs with NIR light produces both fluorescence for tumor visualization through radiative recombination pathways as well as heat and radicals through nonradiative recombination pathways that kill cancer cells and initiate an anticancer immune response, which eliminates tumor and prevents tumor regrowth in 80% of mice.

Prussian blue nanoparticles (PBNPs), which belong to the iron-based metal–organic frameworks, are important biomedical agents. Reducing the size of PBNPs can bring improved functional properties, but unfortunately, has been a long-standing challenge. Herein, sub-5 nm ultrasmall PBNPs (USPBNPs) were successfully synthesized by using ethanol/water mixture as the solvent and polyvinyl pyrrolidone (PVP) as the surface capping agent. Adjusting the ethanol/water ratio is not only able to control the nucleation time and size of PBNPs but also tune the conformation of PVP molecules so as to prevent interparticle attachment and enlargement. At an ethanol/water ratio of 3:1, highly stable USPBNPs with a size of ∼3.4 nm were synthesized. Due to their large specific surface area, they demonstrated high peroxidase-like and catalase-like activities, which outperform PBNPs synthesized by a conventional method. In addition, they also showed a high longitudinal relaxation rate (r1) of 1.3 mM–1 S–1, suggesting their potential to be used as T1 MRI agent.

Multimodal imaging has attracted tremendous attention in biotechnology and nanomedicine applications. Monoclinic Gd 2 O 3 :Dy 3+ nanoparticles (NPs) with dual-color fluorescence imaging (FI) and magnetic resonance imaging (MRI) have been successfully synthesized by employing laser ablation in liquid (LAL) technique. The synthesized nanoparticles can be used as yellow fluorescence nanoprobe as well as blue ones due to their sharp and strong yellow and blue emissions. Magnetic resonance imaging (MRI) measurement shows that the longitudinal relaxivity (r 1 ) of the Gd 2 O 3 :1%Dy 3+ NPs is 4.5 times higher than that of the commercial Gd-DTPA. The experiment of in vitro cell fluorescence imaging and magnetic resonance imaging of female Balb/c mouse indicates that the Gd 2 O 3 :Dy 3+ NPs synthesized by LAL technique are promising candidates of dual-color FI nanoprobe and MRI contrast agent in further clinical applications.

Background Postoperative chemotherapy for gastric cancer often causes multidrug resistance (MDR), which has serious consequences for therapeutic effects. Individualized treatment based on accurate monitoring of MDR will greatly improve patient survival. Results In this article, a self-enhanced Mn3O4 nanoplatform (MPG NPs) was established, which can react with glutathione to produce Mn2+ to enhance T1-weighted magnetic resonance imaging (MRI) and mediate in vivo real-time MDR monitoring. In vitro MRI results showed that MRI signals could be enhanced in the presence of hydrogen peroxide and glutathione and at acidic pH. In vivo MRI results indicated that MPG NPs could specifically target MDR cells, thereby realizing real-time monitoring of MDR in gastric cancer. Furthermore, MPG NPs have good chemodynamic activity, which can convert the endogenous hydrogen peroxide of tumor cells into highly toxic hydroxyl radical through Fenton-like reaction at acidic pH to play the role of chemodynamic therapy. In addition, Mn3O4 can significantly enhance the chemodynamic therapy effect because of its good photothermal conversion effect. Furthermore, in situ photothermal/chemodynamic synergistic therapy obtained remarkable results, the tumors of the mice in the synergistic therapy group gradually became smaller or even disappeared. Conclusion sMPG NPs have good biocompatibility, providing a good nanoplatform for real-time monitoring and precise diagnosis and treatment of MDR in gastric cancer.Graphical Abstract

Magnetic resonance (MR) angiography is one of the main diagnostic approaches for cardiac-cerebral vascular diseases. Nevertheless, the non-contrast-enhanced MR angiography suffers from its intrinsic problems derived from the blood flow-dependency, while the clinical Gd-chelating contrast agents are limited by their rapid vascular extravasation. Herein, we report a hypersensitive MR angiography strategy based on interlocking stratagem of zwitterionic Gd-chelate contrast agents (PAA-Gd). The longitudinal molar relaxivity of PAA-Gd was 4.6-times higher than that of individual Gd-chelates as well as appropriate blood half-life (73.8 min) and low immunogenicity, enabling sophisticated micro-vessels angiography with a resolution at the order of hundred micrometers. A series of animal models of cardiac-cerebrovascular diseases have been built for imaging studies on a 7.0 T MRI scanner, while the clinical translation potential of PAA-Gd has been evaluated on swine on a 3.0 T clinical MRI scanner. The current studies offer a promising strategy for precise diagnosis of vascular diseases.

Triple-negative breast cancer (TNBC), a highly refractory malignancy characterized by aggressive clinical behavior, poses a significant therapeutic challenge due to its resistance to conventional treatments. While microwave thermal therapy (MWTT) emerges as a promising modality for TNBC treatment through deep-tissue penetration and uniform energy delivery, its reliance on prolonged hyperthermia exposure risks collateral damage to adjacent healthy tissues. Herein, we present a rationally engineered iron-doped zeolitic imidazolate framework-8 nanoplatform (Fe@Zif-8). This study aimed to develop a theranostic agent for MRI-guided combination therapy by pursuing the following objectives: (1) synthesizing and characterizing Fe-doped ZIF-8; (2) evaluating the enhancement of microwave thermal conversion efficiency via iron doping; (3) validating the nanoparticle's Fenton catalytic activity for reactive oxygen species (ROS) generation and glutathione depletion in the tumor microenvironment; and (4) demonstrating the synergistic antitumor efficacy of combined MWTT and CDT both in vitro and in vivo . Accordingly, Fe@Zif-8 was synthesized via a one-pot solvothermal method and systematically characterized. Its therapeutic performance was then rigorously assessed through a series of physicochemical and biological experiments. The iron dopants substantially enhance microwave absorption and dielectric loss effects in Zif-8 matrices, enabling Fe@Zif-8 to generate intensified hyperthermia under MW irradiation. Concurrently, iron ions released from the nanoplatform react with endogenous hydrogen peroxide (H 2 O 2 ) via Fenton catalysis to produce substantial ROS. The concerted action of MWTT and CDT achieves rapid tumor ablation while significantly reducing therapeutic energy requirements. Furthermore, the platform served as an effective T2 contrast agent for magnetic resonance imaging (MRI), enabling non-invasive monitoring. This work provides a new approach for the preparation of nanomaterials with highly efficient anti-TNBC performance.

Accurate diagnosis and effective treatment of pancreatic cancer are essential to improving survival rates, yet current strategies are highly limited by toxicity of therapeutic drugs and efficacy challenges. Herein, we proposed a biodegradable nanosystem (denoted as MnCO3–BSA@DOX/IR820) based on manganese carbonate (MnCO3) loaded with doxorubicin (DOX) and photothermal agent IR820. This platform integrates chemotherapy with photothermal/photodynamic therapy (PTT/PDT) under 808 nm laser irradiation, achieving synergistic antitumor efficacy against pancreatic cancer. Specifically, the MnCO3-based nanosystem can enhance tumor accumulation, while the hydrogen peroxide (H2O2)-enriched acidic tumor microenvironment (TME) triggers oxidative stress pathways to generate reactive oxygen species (ROS). Upon 808 nm laser irradiation, IR820 is activated to exert a combined therapeutic effect. It mediates PDT to generate ROS, while the localized hyperthermia induced by PTT causes direct cellular apoptosis and facilitates DOX release, thereby enhancing therapeutic efficacy. Additionally, the nanosystems integrate magnetic resonance imaging (MRI) and near-infrared fluorescence (NIR) imaging capabilities, enabling real-time treatment monitoring and precise diagnosis. In vivo studies demonstrated that MnCO3–BSA@DOX/IR820 nanosystems effectively inhibit tumor growth without causing notable damage to normal tissues. Therefore, this pH-responsive and MRI/NIR imaging-guided nanoplatform represents a targeted theranostic system for pancreatic cancer, enabling chemo-PTT/PDT synergy.

The Warburg effect drives cancer cells to rely preferentially on glycolysis, resulting in diminished mitochondrial oxidative phosphorylation (OXPHOS). This metabolic phenotype limits the ability of excess copper (Cu) ions to trigger cuproptosis in dysfunctional mitochondria, leading to suboptimal therapeutic outcomes. Therefore, remodeling tumor energy metabolism to counteract the Warburg effect represents a pivotal strategy to enhance cuproptosis efficacy. Herein, we developed a Cu(I)-stabilized metal-organic framework nanoplatform (CuTz-1) encapsulated by Azurin (CuTz-1@Azurin) as a cuproptosis sensitizer. Mechanistically, Azurin increases p53 protein levels by intercepting MDM2-mediated ubiquitin-proteasome degradation. Elevated p53 subsequently inhibits pyruvate kinase M2 (PKM2) activity, suppressing glycolysis, while simultaneously promoting mitochondrial OXPHOS—collectively shifting cellular metabolism from glycolysis toward mitochondrial respiration and sensitizing tumor cells to Cu-induced proteotoxic stress. In addition, CuTz-1@Azurin triggers oxidative stress via reactive oxygen species (ROS) generation and glutathione (GSH) depletion, which act together with p53-mediated apoptosis to suppress tumor growth. Meanwhile, the oxidation of Cu(I) to paramagnetic Cu(II) enables real-time monitoring of the therapeutic process by magnetic resonance imaging (MRI). Both in vitro and in vivo results demonstrate good antitumor performance of CuTz-1@Azurin. This study highlights the potential of Azurin-mediated metabolic regulation as a promising strategy to potentiate cuproptosis for combined cancer therapy.

Liver cancer remains a major global public health threat, with high mortality. Lenvatinib is a tyrosine kinase inhibitor that has been used for the treatment of advanced liver cancer. However, its therapeutic efficacy is often limited by severe adverse effects and drug resistance. To overcome these challenge, we developed a biomimetic versatile nanoplatform based on Mn-doped polydopamine (PML@CM). Owing to their homologous tumor-targeting ability and pH-responsive drug release properties, PML@CM nanoparticles effectively improved the biocompatibility of lenvatinib and enhanced its tumor accumulation. Upon exposure to the tumor microenvironment, the released Mn2+ not only triggered chemodynamic therapy through Fenton-like reactions but also enabled magnetic resonance imaging (MRI). In addition, PML@CM nanoparticles exhibited an efficient photothermal conversion performance for tumor photothermal therapy. In conclusion, this nanoplatform provides a promising therapeutic strategy for MRI-enabled synergistic therapy against liver cancer.
