Nitroxide radical-modified CuS nanoparticles for CT/MRI imaging-guided NIR-II laser responsive photothermal cancer therapy

Chengwan Xia,Diya Xie,Lang Xiong,Qian Zhang,Yang Wang,Zezheng Wang,Yuxin Wang,Bin Li,Chao Zhang

Herein, we reported nitroxide radical-modified CuS nanoparticles (CuS–NO˙ NPs), and they exhibited a typical absorption peak at 1182 nm. Due to such a long wavelength absorbance, CuS–NO˙ NPs exhibited excellent therapeutic outcome and low damage to normal tissues. Besides, we simultaneously achieved CuS–NO˙ NPs for MRI and CT dual-modal imaging, which successfully provided a new strategy for imaging-guided tumor treatment, thus increasing potential clinical applications for cancer treatment.

Coumarin derivative dye sensitized NaYGdF4:Yb,Er nanoparticles with enhanced NIR II luminescence for bio-vascular imaging

Yaqun Ma,Zhan Wang,Yanxing Wang,Zhenghao Liu,Yukun Wang,Ruichan Lv

Lanthanide-doped biocompatible nanoparticles have promising applications in near-infrared second region imaging due to their high chemical stability, enhanced photostability and sharp emission bandwidth. However, the weak light absorption capacity limits the application of rare-earth nanoparticles (RENPs) for bioimaging. We prepared a coumarin-derived dye sensitized NaYGdF 4 :Yb,Er nanoparticle probe, in which the organic dye enhances photon absorption through the sensitization process, improving the luminescence efficiency of the rare earth particles near 1000 and 1500 nm. In addition, good water solubility and stability of the probe are imparted by coating the particles with amphiphilic polymers distearoyl phosphatidylethanolamine-polyethylene glycol (DSPE-PEG) and polyacrylic acid. This composite probe with good biocompatibility and NIR II luminescence properties can be used for vascular imaging, providing a tool for the detection of hematologic-related diseases.

Endoplasmic reticulum-targeted NIR-II phototherapy combined with inflammatory vascular suppression elicits a synergistic effect against TNBC

Guoyun Wan,Xuheng Chen,Jiayu Chen,Ruiling Gou,Haijiao Wang,Shuhao Liu,Mingyang Zhang,Hongli Chen,Dan Wang,Qiqing Zhang

Recurrence and metastasis are the main reasons for failure in the treatment of triple-negative breast cancer (TNBC). Phototherapy, one of the most well-known potent cancer treatment models is highlighted by ablating primitive tumors with immunogenic cell death (ICD) and is associated with endoplasmic reticulum (ER) stress to elicit long-lasting anti-tumor immunity. However, the provoked inflammatory response after phototherapy will stimulate angiogenesis, which provides nutrition for tumor recurrence. Here, an ER-targeted nanoplatform was constructed based on hollow mesoporous Cu2−XS (HMCu2−XS) nanoparticles to suppress recurrence and metastasis of TNBC by combining photo-ablation and microenvironment remodeling. Profiting from the metal ion coordination and large hollow space, HMCu2−XS can be easily modified with p-toluenesulfonamide for ER-targeting and quantitatively loaded celecoxib (CXB) as a vascular inhibitor, thus obtaining ER-HMCu2−XS/CXB. ER-HMCu2−XS showed great photothermal and photodynamic efficiency for ablating 4T1 tumors and inducing ICD under NIR-II laser irradiation. Compared with non-ER-targeted nanosystems, the ER-targeted nanosystem elicited stronger ICDs and recruited more immune cells. Moreover, the thermal-responsively released CXB successfully inhibited angiogenesis after photothermal therapy. The data showed that the ER-HMCu2−XS/CXB mediated the triplicate therapeutic effect of photo-ablation, immune response activation, and vascular suppression effectively, preventing the recurrence and metastasis of TNBC. In conclusion, this work provides a synergistic strategy to enhance therapeutic outcomes in TNBC.

Dextran 40 hybrid biomimetic bismuth-nanoflower designed for NIR II-triggered hypoxic tumor thermoradiotherapy via macrophage escape

Jing Wen,Chang Liu,Ji Liu,Lu Wang,Si Miao,Daquan Chen,Qiyue Wang,Meirong Huo,Yan Shen

At present, NIR-II-triggered photothermal biomedical applications are limited by complex synthesis reactions , mediocre photothermal conversion efficiency, and difficult degradation. Herein, we prepared biodegradable Bi flower-like nanoparticles (phospholipid-modified Bi nanoflowers , BNFs) with high photothermal conversion efficiency (∼33.52 %) in NIR-II by a simple method and then modified them with the red blood cell membrane and dextran 40 (DRBCM) to improve their in vitro stability, to escape macrophages clearance and to enhance tumor accumulation. Dextran coating onto the surface of particles as a dispersant shell stabilizes inorganic particles by maintaining the surface charges and creating steric repulsions upon compression of neighboring polymer chains. In vitro and in vivo experiments proved that combined thermoradiotherapy of DRBCM-BNFs exhibited significantly enhanced tumor inhibition efficacy than monotherapy with good biocompatibility and low toxicity due to its biodegradability . Furthermore, the mechanism studies demonstrated that DRBCM-BNFs could serve as a nano sensitizer to promote the thermoradiotherapy under NIR-II illumination and X-ray irradiation, by downregulating heat shock protein 70 (HSP70) and phosphorylated-p65 (p-p65) to reduce the thermal resistance and radioresistance of tumor cells and increasing the expression of apoptosis-related protein cleaved caspase-3. In conclusion, DRBCM-BNFs could be a promising green delivery platform for the sensitization of synergistic thermoradiotherapy.

Gold Nanocluster Encapsulated Nanorod for Tumor Microenvironment Simultaneously Activated NIR-II Photoacoustic/Photothermal Imaging and Cancer Therapy

Zhifang Li,Shulong Wang,Jingjin Zhao,Yanni Luo,Hong Liang,Shulin Zhao,Liangliang Zhang

Tumor microenvironment (TME)-responsive imaging and therapy has great potential for application towards precision diagnosis and treatment of cancer. Photoacoustic (PA) imaging with the second near-infrared window (NIR-II) and photothermal therapy are important ways to address high-resolution imaging of in vivo deep tumor tissue and high-effect therapy of cancer. A gold nanocluster (AuNCs) encapsulated nanorod (AuNRs) (AuNRs@MEA/DTGA@AuNCs, hereafter AMDAs) is prepared. The AuNRs are first modified using β-mercaptoethylamine (MEA) to form AuNRs@MEA. Dithioglycolic acid (DTGA) is coupled with AuNRs@MEA to obtain AuNRs@MEA/DTGA (AMD). Finally, AuNCs are attached to AuNRs@MEA/DTGA surface to form AMDAs. In this nanocomposite materials, the encapsulation of the AuNRs by the AuNCs obscures NIR-II PA signal of AuNRs. The high expressed glutathione in the TME react with the AMDAs via redox reaction, the detachment of the AuNCs from the AuNRs surface, and detection of the NIR-II PA signal from the AuNRs, thus enabling NIR-II PA imaging of tumor tissues. In addition, due to the high intensity light absorption of AuNRs in the NIR-II region, the release of AuNRs also enhance tumor photothermal imaging and therapy. This work establishes an accurate and efficient integrated platform for nanodiagnosis and treatment of cancer.

Real-time screening of hepatotoxins in natural medicine by peroxynitrite responsive lanthanide-based NIR-II luminescent probes

Yang Zhong,Jiayu Gu,Yaoquan Su,Lingzhi Zhao,Yunyun Zhou,Juanjuan Peng

Herbal medicines and dietary supplements are widely used worldwide. However, reports have revealed that a large number of herbal medicines are implicated in the hepatic damage, which is referred to as Herbal medicines-induced liver injury (HILI). Traditional diagnostic methods for HILI include histological and serological assessments, which fail to offer real-time and unambiguous visualization of hepatotoxicity in vivo . Herein, we constructed a second near-infrared (NIR-II) luminescent nanoprobes to achieve accurate, high-efficiency, and non-invasive detection of HILI by the assembly of lanthanide-based NIR-II luminescence nanoparticles with a chromophore with absorption band in the NIR-II region. Specifically, the NIR-II luminescence of the lanthanide nanoparticles can be effectively quenched by the chromophores on the surface of nanoparticles via an absorption competition-induced quenching effect, and subsequently recovered by the pathophysiological biomarker peroxynitrite (ONOO − ) generated in early HILI. It is demonstrated that the nanoprobes are capable for the screening of liver toxins in a noninvasive pattern, which is more convenient than the conventional diagnostic approaches. Finally, our luminescence detection approach offers an immediate testing technique for the identification of hepatotoxic components of herbal medicine such as colchicine, monocrotaline and stibene glucoside, providing a high-throughput screening strategy for herbal medicine-induced hepatotoxicity.

Multifunctional cascade nanocatalysts for NIR-II-synergized photonic hyperthermia-strengthened nanocatalytic therapy of epithelial and embryonal tumors

Xiangqi Liu,Wei Feng,Huijing Xiang,Baihui Liu,Mujie Ye,Meng Wei,Rui Dong,Yu Chen,Kuiran Dong

Chemodynamic therapy, which produces cytotoxic hydroxyl radicals through intratumoral Fenton reactions, has been extensively explored in nanomedicine for cancer treatment. However, the limited endogenous hydrogen peroxide (H 2 O 2 ) levels, which serve as the reactant, weakens the anticancer effect of the Fenton reaction. In this work, a cascade catalytically reactive nanosystem has been constructed for the treatment of epithelial and embryonal tumors with high synergetic efficacy through the integration of natural glucose oxidase (GOD) and polyethylene glycol (PEG) onto the surface of Fe 3 S 4 nanoplates (abbreviated as Fe 3 S 4 -PEG-GOD NPs). The GOD component consumes glucose in tumor and produces a large amount of H 2 O 2 , which provides rich substrates for the Fenton reaction catalyzed by the Fe catalytic center of the nanoplates. The intriguing photothermal conversion efficiency (45%) of the constructed composite nanocatalysts in the second near infrared (NIR-II) biowindow allows for photothermal treatment to substantially strengthen and synergize the anticancer effects of nanocatalysts both in vitro and in vivo. This synergy occurs through generation of a hyperthermic effect necessary for promoting the sequential catalytic reaction. The synergistic anticancer effect has been verified in both epithelial and embryonal tumor mouse models, and provides justification for the biomedical use of multifunctional nanocatalysts for versatile tumor nanotherapeutics.

Activatable NIR-II photoacoustic imaging and photochemical synergistic therapy of MRSA infections using miniature Au/Ag nanorods

Zihan Mei,Duyang Gao,Dehong Hu,Huichao Zhou,Teng Ma,Liang Huang,Xin Liu,Rongqin Zheng,Hairong Zheng,Ping Zhao,Jianqiao Zhou,Zonghai Sheng

Multidrug-resistant Staphylococcus aureus (MRSA) seriously endanger human health. The development of efficient methods to eliminate the infections and monitor the treatment process are of great significance. Near-infrared-II (NIR-II) photoacoustic (PA) imaging and photothermal therapy (PTT) are highly integrated theranostic platforms with superior performance including a low imaging background, increased tissue penetration depth, and high photothermal threshold. Herein, we report an activatable near-infrared II (NIR-II) phototheranostic strategy using miniature Au/Ag nanorods (NRs) for the photochemical synergistic therapy of MRSA infections and in situ monitoring of the treatment progress. Au/Ag NRs were efficiently activated by ferricyanide solution and allowed to continuously release free Ag + to eliminate MRSA, triggering NIR-II photothermal and PA performance enhancement. The activated NIR-II photothermal effect in turn accelerated the release of free Ag + from Au/Ag NRs for the synergistic elimination of gram-positive Staphylococcus aureus and promoted wound healing. No photothermal damages or free Ag + -induced side effects were observed in treated mice. After synergistic treatment, a 20-fold NIR-II PA signal increase with a maximum signal-to-noise measurement of 9.5 was observed between the implanted site and normal tissue, enabling sensitive monitoring of Ag + release process. The prepared Au/Ag NRs were stable and biocompatible, showing great potential for activatable NIR-II phototheranostic application.

Defect-engineered transition metal hydroxide nanosheets realizing tumor-microenvironment-responsive multimodal-imaging-guided NIR-II photothermal therapy

Li Yang,Xiaojiao Zhu,Tianren Xu,Fusen Han,Gang Liu,Yingcui Bu,Jie Zhang,Feng Zhang,Hongping Zhou,Yi Xie

Exploiting two-dimensional nanomaterials as photo-based theranostic agents is promising for the highly efficient ablation of deep-tissue-buried tumors. However, they are limited by their poor absorption in the second near-infrared-light (NIR-II) bio-window (1000–1300 nm) and intrinsic nonbiodegradability. Herein, defect-rich sulfur-doped Ni(OH)2 (S-Ni(OH)2) nanosheets decorated with bovine serum albumin (BSA) as a novel theranostic agent is developed, which can accomplish multimodal-imaging-guided photothermal ablation of mouse cancers in the NIR-II bio-window. Sulfur doping extends the absorption spectra of Ni(OH)2 nanosheets from the visible to NIR-II bio-window, affording highly efficient photothermal conversion (58.20% for 1064 nm), entailing it to become an excellent contrast agent for photoacoustic imaging. Further, because of their intrinsic paramagnetic property, they can be applied for magnetic resonance imaging. Owing to the abundant defective sites in S-Ni(OH)2 nanosheets, they exhibit response to the tumor microenvironment, resulting in effective biodegradation and excretion from the body. In vivo toxicity experiments indicated that S-Ni(OH)2-BSA NSs delivered no appreciable toxicity and good biocompatibility. This work provides an avenue for the rational design of effective theranostics agents.

A Cr3+-Yb3+ co-doped high-efficiency near-infrared luminescent phosphor for NIR-II light source applications

Tong Feng,Yuanlin Jin,Zhangwen Long,Dacheng Zhou,Xiaqing Jiang,Bitao Liu,Jianbei Qiu

Phosphor conversion light-emitting diode (PC-LED) technology has become a focal point of research for portable near-infrared light sources in recent years. One of the crucial aspects is to develop phosphor materials that can be effectively stimulated by commercial available blue light. Unfortunately, efficient phosphors in the NIR-II that can be excited by commercial blue light are particularly scarce. In this study, Mg 4 Ta 2 O 9 :0.04Cr 3+ ,0.03Yb 3+ ,0.07Li + (MTO:0.04Cr 3+ ,0.03Yb 3+ ,0.07Li + ) phosphor was synthesized using the high temperature solid phase method, and its emission spectrum covered the first and second near-infrared windows (700–1200 nm). The internal quantum efficiency (IQE) and external quantum efficiency (EQE) are 88.7 % and 45.6 %, respectively, due to the synergistic effect of energy transfer and Li-ion modification. Additionally, the doping of Yb 3+ ions and Li + ions enhances the thermal stability of luminescence. The thermal stability of MTO:0.04Cr 3+ ,0.03Yb 3+ ,0.07Li + and MTO:0.04Cr 3+ ,0.03Yb 3+ is 9.81 % and 14.43 % higher, respectively, than that of MTO:0.04Cr 3+ ,0.03Yb 3+ and MTO:0.04Cr 3+ . To evaluate its potential applications, we developed a prototype PC-LED utilizing MTO:0.04Cr 3+ ,0.03Yb 3+ ,0.07Li + phosphor as an emission emitter which demonstrated advantages in biomedical imaging, night vision, and non-destructive testing.

Full-course NIR-II imaging-navigated fractionated photodynamic therapy of bladder tumours with X-ray-activated nanotransducers

He Liangrui,Wang Liyang,Yu Xujiang,Tang Yizhang,Jiang Zhao,Yang Guoliang,Liu Zhuang,Li Wanwan

The poor 5-year survival rate for bladder cancers is associated with the lack of efficient diagnostic and treatment techniques. Despite cystoscopy-assisted photomedicine and external radiation being promising modalities to supplement or replace surgery, they remain invasive or fail to provide real-time navigation. Here, we report non-invasive fractionated photodynamic therapy of bladder cancer with full-course real-time near-infrared-II imaging based on engineered X-ray-activated nanotransducers that contain lanthanide-doped nanoscintillators with concurrent emissions in visible and the second near-infrared regions and conjugated photosensitizers. Following intravesical instillation in mice with carcinogen-induced autochthonous bladder tumours, tumour-homing peptide-labelled nanotransducers realize enhanced tumour regression, robust recurrence inhibition, improved survival rates, and restored immune homeostasis under X-ray irradiation with accompanied near-infrared-II imaging. On-demand fractionated photodynamic therapy with customized doses is further achieved based on quantifiable near-infrared-II imaging signal-to-background ratios. Our study presents a promising non-invasive strategy to confront the current bladder cancer dilemma from diagnosis to treatment and prognosis.

Aggregation control of anionic pentamethine cyanine enabling excitation wavelength selective NIR-II fluorescence imaging-guided photodynamic therapy

Li Yibin,Qu Fei,Wan Fang,Zhong Cheng,Rao Jingyi,Liu Yijing,Li Zhen,Zhu Jintao,Li Zhong’an

Near-infrared (NIR)-II fluorescence imaging-guided photodynamic therapy (PDT) has shown great potential for precise diagnosis and treatment of tumors in deep tissues; however, its performance is severely limited by the undesired aggregation of photosensitizers and the competitive relationship between fluorescence emission and reactive oxygen species (ROS) generation. Herein, we report an example of an anionic pentamethine cyanine (C5T) photosensitizer for high-performance NIR-II fluorescence imaging-guided PDT. Through the counterion engineering approach, a triphenylphosphine cation (Pco) modified with oligoethylene glycol chain is synthesized and adopted as the counterion of C5T, which can effectively suppress the excessive and disordered aggregation of the resulting C5T-Pco by optimizing the dye amphipathicity and enhancing the cyanine-counterion interactions. Dynamic tuning of fluorescence characteristics and ROS generation is achieved at the aggregate level, resulting in an impressive type I ROS generation under 760 nm light irradiation, accompanied by efficient NIR-II fluorescence emission excited at 808 nm. As a result, excitation wavelength selective NIR-II fluorescence imaging-guided PDT has been successfully demonstrated for tumor diagnosis and therapeutics of female mice.
