The combination of shape memory polymer composites and 3D printing technology provides novel opportunities for customized manufacturing and intelligentization of products. Here, we developed magneto-responsive shape memory polymer composites with well-balanced strengths and toughness by adding Fe 3 O 4 and cellulose nanofibers (CNFs) to a poly-hydroxybutyrate/poly(ε-caprolactone) blend as functional particles and reinforcing component. The influences of Fe 3 O 4 and CNFs on the properties including micromorphology, and rheological, melting-crystallizational, thermomechanical, static mechanical, and magneto-responsive shape-memory behaviors were investigated. The results displayed that the PHB/PCL (80:20) composites with 10 wt% Fe 3 O 4 and 0.5 wt% CNFs had the optimal comprehensive mechanical and magneto-responsive shape-memory properties. Moreover, 3D printing tests found that the addition of CNFs and printing parameters were crucial for the mechanical and magneto-responsive shape-memory properties of the resulting products. Finally, the fabrication of a scaffold with excellent load-carrying performance and a magneto-responsive self-recovery snowflake demonstrated the application feasibility of our work.

Polymeric micelles receive considerable attention as drug delivery vehicles, depending on the versatility in drug solubilization and targeting therapy. However, their use invariably suffers with poor stability both in in vitro and in vivo conditions. Here, we aimed to develop a novel nanocarrier (micellar emulsions, MEs) for a systemic delivery of genistein (Gen), a poorly soluble anticancer agent. Gen-loaded MEs (Gen-MEs) were prepared from methoxy poly(ethylene glycol)-block-(ε-caprolactone) and medium-chain triglycerides (MCT) by solvent-diffusion technique. Nanocarriers were characterized by dynamic light scattering, transmission electron microscopy, and in vitro release. The resulting Gen-MEs were approximately 46 nm in particle size with a narrow distribution. Gen-MEs produced a different in vitro release profile from the counterpart of Gen-ME. The incorporation of MCT significantly enhanced the stability of nanoparticles against dilution with simulated body fluid. Pharmacokinetic study revealed that MEs could notably extend the mean retention time of Gen, 1.57- and 7.38-fold as long as that of micelles and solution formulation, respectively, following intravenous injection. Furthermore, MEs markedly increased the elimination half-life (t1/2β) of Gen, which was 2.63-fold larger than that of Gen solution. Interestingly, Gen distribution in the liver and kidney for MEs group was significantly low relative to the micelle group in the first 2 hours, indicating less perfusion in such two tissues, which well accorded with the elongated mean retention time. Our findings suggested that MEs may be promising carriers as an alternative of micelles to systemically deliver poorly soluble drugs.

Core–shell structured cellulose acetate-polycaprolactone/chitosan (CA-PCL/CS) composite nanofibers were electrospun via coaxial electrospinning. Different solvents were used in core solutions, which led to different electrical conductivities in core solutions. The effect of the electrical conductivity on the morphology and structure of core–shell fibers was studied emphatically. As shown in the scanning electron microscopy (SEM) and transmission electron microscopy (TEM) images, the increasing electrical conductivity of core solutions would result in smaller diameter ratios of core to shell, thinner overall diameters and more inhomogeneous diameter distributions. Additionally, Fourier transform infrared spectroscopy (FTIR), X-ray photoelectron spectroscopy (XPS), X-ray diffraction (XRD) and differential scanning calorimetry (DSC) all demonstrated that the interactions between cores and shells weakened with the increase of electrical conductivity of core solutions, and this might lead to different diameter ratios of cores to shells in the fibers. The results provide a basis for further design and optimization of processing conditions to control the nanostructure of core–shell composite nanofibers.

Aiming at achieving the concurrent performances of high loading, well controlled release and active targeted delivery, folate (FA) modified dual pH/reduction-responsive mixed polymeric micelles were rationally assembled using FA-PEG-PDEAEMA and PEG-SS-PCL by dissipative particle dynamics (DPD) simulations. The optimized polymers PEG112-PDEAEMA40, FA-PEG112-PDEAEMA40, and PEG112-SS-PCL70 were synthesized and characterized using 1H NMR, FT-IR and GPC, and their mixed micelles were applied for doxorubicin (DOX) delivery. The drug loading capacity (LC) and encapsulation efficiency (EE) values of the MIX1 (FA-PEG112-PDEAEMA40/PEG112-SS-PCL70) at a DOX/polymer feeding ratio of 15 mg/30 mg were 20.22% and 50.69%, which were higher than those of single polymer micelles and MIX2 (PEG112-PDEAEMA40/PEG112-SS-PCL70). Particle size distributions, mesoscopic morphologies, DPD simulations and in vitro drug release profiles all confirmed the well-controlled release performance of the DOX-loaded micelles formed by MIX1: slow DOX release with a cumulative release of 20.46% in the neutral environment and accelerated release with a cumulative release of 74.20% at pH 5.0 + 10 mM DTT within 120 h, which were similar to those of MIX2. Cytotoxicity assay found that both MIX1 and MIX2 blank micelles were biocompatible, and a superior inhibitory effect of the FA-modified DOX-loaded micelles MIX1 on HepG2 cells was found compared to that of free DOX and non-FA-modified DOX-loaded micelles MIX2. All of these confirmed the superiority of MIX1 micelles with high loading capacity, well controlled release, and enhanced inhibitory effects on HepG2 cells, which might be a prospective candidate for anticancer drug delivery.

In this study, the perfluoropolyether carboxylic acid (PFPE-COOH) was used to react with polycaprolactone (PCL) to synthesise the hydrophobic block copolymer PCL- b -PFPE. The esterification reaction was performed successfully with the terminated hydroxyl group in the PCL-OH after activation of PFPE-COOH with N, N′ -dicyclohexyl carbon diimide. The PCL- b -PFPE was dissolved in various solvents to prepare microspheres using electrostatic spraying technology, and the flow rate, voltage, temperature and humidity parameters were optimised. The findings demonstrated that the solvent and ambient humidity had a substantial influence on the microspheres' surface morphology . Through smooth surface morphology, chloroform (CF) single solvent formed an ideal microsphere and DMF/CF mixed solvent produced a ‘pit’ surface morphology. Furthermore, the surface roughness of microspheres increases with an increase in humidity. The static contact angles of electrostatic spraying polyester fabric were 147.1° ± 1.8°, 150.9° ± 1.5°, 156.3° ± 2.6° and 155.9° ± 3.3°, under 20 %, 40 %, 60 % and 80 % humidity, respectively. The findings demonstrated that during the electrostatic spraying, the PFPE chain's introduction into PCL helped construct the secondary rough morphology on the microsphere surface and substantially enhanced the hydrophobicity of coatings, providing the finished polyester fabric super-hydrophobicity. The PCL-b-PFPE microspheres were directly sprayed and coated with the developed spray solution via electrostatic spraying. The water contact angle of the finished polyester fabric was found to be ≥140°, which provided hydrophobicity after 5 cycles of laundry. Following the sandpaper abrasion and adhesion tests, the contact angle of the fabrics coated with the microspheres remained ≥145°, indicating a good mechanical stability.

Strong and tough β-TCP/PCL composite scaffolds with interconnected porosity were developed by combining digital light processing and vacuum infiltration. The composite scaffolds were comprised of pure β-TCP, β-TCP matrix composite and PCL matrix composite. The porous β-TCP/PCL composite scaffolds showed remarkable mechanical advantages compared with ceramic scaffolds with the same macroscopic pore structure (dense scaffolds). The composite scaffolds exhibited a significant increase in strain energy density and fracture energy density, though with similar compressive and flexural strengths. Moreover, the composite scaffolds had a much higher Weibull modulus and longer fatigue life than the dense scaffolds. It was revealed that the composite scaffolds with interconnected porosity possess comprehensive mechanical properties (high strength, excellent toughness, significant reliability and fatigue resistance), which suggests that they could replace the pure ceramic scaffolds for degradable bone substitutes, especially in complex stress environments. Keywords:porous composite scaffolds;mechanical properties;digital light processing;infiltration;gradient structure

In order to fabricate antibacterial nanofiber membranes for tissue engineering field, the oxidative damage of magnesium hydroxide nanoparticles (Mg(OH) 2 NPs) against the model system of Escherichia coli ( E. coli ) was researched by measuring the oxidative stress response, oxidative stress products and glucose metabolism. The toxicity of Mg(OH) 2 NPs to a normal biological system was evaluated in vivo through the acute oral toxicity, blood physiological and serum biochemical markers and acute skin irritation, indicating that the Mg(OH) 2 NPs exhibited great biosafety. The Mg(OH) 2 NPs with antibacterial property and biosafety were loaded on PCL/PVP nanofiber substrates through electrospinning technology to fabricate the Mg(OH) 2 /PCL/PVP nanofiber membranes. The results of characterization and antibacterial test showed that the Mg(OH) 2 /PCL/PVP nanofiber membranes with 8.0 wt% Mg(OH) 2 NPs present excellent antibacterial rate of 100% against E. coli that the Mg(OH) 2 NPs can be physically and evenly loaded on nanofiber substrates.

The construction of functional biomimetic scaffolds that recapitulate the topographical and biochemical features of bone tissue extracellular matrix is now of topical interest in bone tissue engineering. In this study, a novel surface-functionalized electrospun polycaprolactone (PCL) nanofiber scaffold with highly ordered structure was developed to simulate the critical features of native bone tissue via a single step of catechol chemistry. Specially, under slightly alkaline aqueous solution, polydopamine (pDA) was coated on the surface of aligned PCL nanofibers after electrospinning, followed by covalent immobilization of bone morphogenetic protein-7-derived peptides onto the pDA-coated nanofiber surface. Contact angle measurement, Raman spectroscopy, and X-ray photoelectron spectroscopy confirmed the presence of pDA and peptides on PCL nanofiber surface. Our results demonstrated that surface modification with osteoinductive peptides could improve cytocompatibility of nanofibers in terms of cell adhesion, spreading, and proliferation. Most importantly, Alizarin Red S staining, quantitative real-time polymerase chain reaction, immunostaining, and Western blot revealed that human mesenchymal stem cells cultured on aligned nanofibers with osteoinductive peptides exhibited enhanced osteogenic differentiation potential than cells on randomly oriented nanofibers. Furthermore, the aligned nanofibers with osteoinductive peptides could direct osteogenic differentiation of human mesenchymal stem cells even in the absence of osteoinducting factors, suggesting superior osteogenic efficacy of biomimetic design that combines the advantages of osteoinductive peptide signal and highly ordered nanofibers on cell fate decision. The presented peptide-decorated bone-mimic nanofiber scaffolds hold a promising potential in the context of bone tissue engineering.

Optimized formulations produce RGB structural colors and dual visible‑UV encryption signals from fluorescent PDLC. • Wavelength-selective Bragg reflection enables multi-stage decryption for high-security anti-counterfeiting. • A ternary PSCLC/PDLC/PCL composite is prepared via two‑step polymerization to achieve tunable cholesteric helical pitch. Liquid crystals (LC) are regarded as promising candidates for optical encryption on account of their distinctive stimulus-responsive structural coloration. However, single-component polymer-stabilized cholesteric liquid crystal (PSCLC) systems suffer from limited information encoding dimensions. Polymer-dispersed liquid crystals (PDLC) can only achieve basic light scattering modulation and cannot meet the requirements of complex multi-channel encryption. To circumvent these drawbacks, a synergistic PSCLC/PDLC/polycaprolactone (PCL) composite thin film is fabricated for optical anti-counterfeiting. Laser-patterned PDLC templates are filled with PSCLC precursors, and S5011-loaded PCL thin films laminated on independent glass substrates serve as sustainable chiral donors. Regulated interlayer chiral diffusion enables continuous tuning of structural hues within predefined laser-etched cavities. Incorporation of red fluorescent powder imparts dual visible and ultraviolet imaging capacity to the composite. Tunable color variation in designated regions is realized via precise modulation of diffusion temperature and duration. Benefiting from the wavelength-selective Bragg reflection effect of cholesteric liquid crystals (CLCs), multi-layered information encryption is accomplished. Colored patterned regions can only be visualized under matching monochromatic illumination. Benefiting from facile manufacturing, large information storage capacity and diversified encryption modes, the proposed composite presents great application potential in premium anti-counterfeiting tags, encrypted two-dimensional codes and intelligent display devices. Scaffold-based three-dimensional (3D) cell culture systems have gained increased interest in cell biology, tissue engineering, and drug screening fields as a replacement of two-dimensional (2D) monolayer cell culture and as a way to provide biomimetic extracellular matrix environments. In this study, microscale fibrous scaffolds were fabricated via electrohydrodynamic printing, and nanoscale features were created on the fiber surface by simply leaching gliadin of poly(ε-caprolactone) (PCL)/gliadin composites in ethanol solution. The microstructure of the printed scaffolds could be precisely controlled by printing parameters, and the surface nanotopography of the printed fiber could be tuned by varying the PCL/gliadin ratios. By seeding mouse embryonic fibroblast (NIH/3T3) cells and human nonsmall cell lung cancer (A549) cells on the printed scaffolds, the cellular responses showed that the fiber nanotopography on printed scaffolds efficiently favored cell adhesion, migration, proliferation, and tissue formation. Quantitative analysis of the transcript expression levels of A549 cells seeded on nanoporous scaffolds further revealed the upregulation of integrin-β1, focal adhesion kinase, Ki-67, E-cadherin, and epithelial growth factor receptors over what was observed in the cells grown on the pure PCL scaffold. Furthermore, a significant difference was found in the relevant biomarker expression on the developed scaffolds compared with that in the monolayer culture, demonstrating the potential of cancer cell-seeded scaffolds as 3D in vitro tumor models for cancer research and drug screening.

To improve bone regeneration in oral microenvironment, we generated a novel biodegradable, antibacterial, and osteoconductive electrospun PLGA/PCL membrane as an ideal osteogenic scaffold. The novel three-layer membranes were structured with serial layers of electrospun chlorhexidine-doped-PLGA/PCL (PPC), PLGA/PCL (PP), and β-tricalcium phosphate-doped-PLGA/PCL (PPβ). To characterize osteoconductive properties of these membranes, MC3T3-E1 (MC) cultures were seeded onto the membranes for 14 days for evaluation of cell proliferation, morphology and gene/protein expression. In addition, MC cells were cultured onto different surfaces of the three-layer membranes, PPC layer facing MC cells (PPβ-PP-PPC) and PPβ layer facing MC cells (PPC-PP-PPβ) to evaluate surface–material effects. Membrane properties and structures were evaluated. Antibacterial properties against Streptococcus mutans and Staphylococcus aureus were determined. Scanning electron microscope demonstrated smaller interfiber spaces of PPC and PPβ-PP-PPC compared to PPβ, PPC-PP-PPβ, and PP. PPC and PPβ-PP-PPC exhibited hydrophilic property. The three-layer membranes (PPC-PP-PPβ and PPβ-PP-PPC) demonstrated significantly higher Young’s modulus (94.99 ± 4.03 MPa and 92.88 ± 4.03 MPa) compared to PP (48.76 ± 18.15 MPa) or PPC (7.92 ± 3.97 MPa) (p < 0.05). No significant difference of cell proliferation was found among any groups at any time point (p > 0.05). Higher expression of integrins were detected at 12 h of cultures on PPC-PP-PPβ compared to the controls. Promoted osteoconductive effects of PPC-PP-PPβ were revealed by alkaline phosphatase assays and Western blot compared with the controls at 7 and 14 days. PPC, PPC-PP-PPβ and PPβ-PP-PPC exhibited a significantly wider antibacterial zone against the tested bacteria compared to PP and PPβ (p < 0.05). These results suggested that the three-layer electrospun membranes demonstrated superior properties: higher strength, better cell adhesion, and promoted osteoconductive properties compared to single-layer membrane: however, antibacterial properties were exhibited in three-layer electrospun membranes and chlorhexidine-doped single-layer membrane. We concluded that the novel three-layer membranes could be used as a biocompatible scaffold for intraoral bone regeneration due to its enhanced osteoconductive activity and antibacterial effect.

The side effects of doxorubicin (DOX) extremely limit its application in the treatment of malignant tumors. Nano-sized polymeric drugs based on the acidic microenvironment of tissular- or intra- tumor have attracted ample attention because of their potential in reducing side effects. In this research, an amphiphilic diblock copolymer based on poly (ethylene glycol) (PEG) and functionalized polycaprolactone (PCL) was synthesized and utilized as the drug carrier. DOX was chemically conjugated with the polymer via acid-cleavable imine bonds to obtain a novel pH-sensitive DOX prodrug (mPEG-PCL-Imi-DOX). mPEG-PCL-Imi-DOX (24.2 wt % DOX content) formed micelles with an average diameter of 125 nm through a simple solvent evaporation method. The in vitro release profile demonstrated that DOX release of the prodrug micelles was pH-responsive and able to be accelerated with the decrease of pH. In vitro cytotoxicity assay tests revealed that the pH-sensitive DOX prodrug micelles exhibited relatively lower toxicity and similar antitumor efficacy towards MCF-7 cells compared with free DOX. Hence, the DOX prodrug micelles with imine bonds can offer a carrier with great potential for chemo-therapeutics.Keywords:doxorubicin;pH-sensitive;prodrug;micelles;imine
