Study of chondroitin sulfate E oligosaccharide as a promising complement C5 inhibitor for osteoarthritis alleviation

Chen Yu,Hengchang Zang,Cui Yang,Dong Liang,Shuang Quan,Danyang Li,Yanni Li,Qin Dong,Fengshan Wang,Lian Li

Osteoarthritis (OA) is a degenerative joint disease which is highly prevalent worldwide. However, no therapy for blocking OA pathogenesis is available currently. In this study, chondroitin sulfate (CS) E oligosaccharides were prepared and we identified disaccharide as the functional unit showing the strongest anti-complement activity and screened out complement C5 as its target in the complement system. We determined that CS-E disaccharide produced anti-inflammatory effects to treat OA by regulating the complement system: it inhibited the formation of complement-dependent complexes such as the membrane-attack complex (MAC) by targeting C5 and suppressed MAC-induced protein expression and the activation of downstream MAPK and NF-κB signaling pathways accordingly. By identifying CS-E disaccharide which could be regarded as a complement regulator or inhibitor exhibiting high anti-complement activity and revealing its OA-alleviating mechanism, this study not only provides a new strategy for OA treatment and drug development, but also potentially offers a promising C5 target therapy for other associated diseases.

A portable, battery-powered photoelectrochemical aptasesor for field environment monitoring of E. coli O157:H7

Ting Zheng,Xiaoming Jiang,Naiwen Li,Xia Jiang,Chao Liu,Jing-Juan Xu,Peng Wu

Escherichia coli O157:H7 ( E. coli O157:H7 ) is typically associated with food- and water-borne outbreaks, thus making continuous monitoring essential. Photoelectrochemical (PEC) detection, which harvests the unique photo-to-electric property of photoactive species, has emerged as an exciting tool for bioanalysis. However, the current attempts are mostly carried out in lab, and less explored in field. Here, a simple PEC aptasensor for E. coli O157:H7 was designed and optimized (LOD: 200 cfu/mL). To promote its field application, a portable PEC analyzer was further constructed with a miniaturized LED (3 V, 3 W) for excitation and a USB-type electrochemical workstation for detection. Meanwhile, a re-chargeable power bank (5 V, 20,000 mAh, originally used for cell phone charging) was explored to power the LED through a self-designed electrical circuit. In this manner, the PEC analyzer could work continuously for more than 12 h. To facilitate transportation or the portable PEC analyzer, all components were mounted into a suitcase (size: 42 × 34 × 17 cm 3 ; wight: ∼3.5 kg). To validate the applicability of the portable PEC analyzer, on-site detection of E. coli O157:H7 in an in-land river was carried out (> 8 h), demonstrating the bright potential of the portable PEC analyzer for field environmental monitoring.

Early identification of Aspergillus spp. contamination in milled rice by E-nose combined with chemometrics

Shuang Gu,Zhenhe Wang,Wei Chen,Jun Wang

BACKGROUND Rice grains can be contaminated easily by certain fungi during storage and in the market chain, thus generating a risk for humans. Most classical methods for identifying and rectifying this problem are complex and time-consuming for manufacturers and consumers. However, E-nose technology provides analytical information in a non-destructive and environmentally friendly manner. Two-feature fusion data combined with chemometrics were employed for the determination of Aspergillus spp. contamination in milled rice. RESULTS Linear discriminant analysis (LDA) indicated that the efficiency of fusion signals (‘80 th s values’ and ‘area values’) outperformed that of independent E-nose signals. Linear discriminant analysis showed clear discrimination of fungal species in stored milled rice for four groups on day 2, and the discrimination accuracy reached 92.86% by using an extreme learning machine (ELM). Gas chromatography–mass spectrometry (GC–MS) analysis showed that the volatile compounds had close relationships with fungal species in rice. The quantification results of colony counts in milled rice showed that the monitoring models based on ELM and the genetic algorithm optimized support vector machine (GA-SVM) (R 2 = 0.924–0.983) achieved better performances than those based on partial least squares regression (PLSR) (R 2 = 0.877–0.913). The ability of the E-nose to monitor fungal infection at an early stage would help to prevent contaminated rice grains from entering the food chains. CONCLUSIONS The results indicated that an E-nose coupled with ELM or GA-SVM algorithm could be a useful tool for the rapid detection of fungal infection in milled rice, to prevent contaminated rice from entering the food chain. © 2021 Society of Chemical Industry

Biocompatible Multifunctional E-Skins with Excellent Self-Healing Ability Enabled by Clean and Scalable Fabrication

Lin Xiuzhu,Li Fan,Bing Yu,Fei Teng,Liu Sen,Zhao Hongran,Zhang Tong

A printable multifunctional electronic skin (e-skin) with an excellent self-healing ability was developed via a simple, clean and universally applicable method. Water served as both the only solvent throughout the fabrication process and the trigger of the self-healing process. Each e-skin component was biocompatible. Combined with conventional electronics, signals collected by the e-skin could be transmitted to smartphones via Bluetooth for post-processing.

E-Jet 3D-Printed Scaffolds as Sustained Multi-Drug Delivery Vehicles in Breast Cancer Therapy

Qiao Xiaoyin,Yang Yikun,Huang Ruiying,Shi Xuelei,Chen Haoxiang,Wang Jian,Chen Yanxiang,Tan Yongjun,Tan Zhikai

Purpose Combination chemotherapy is gradually receiving more attention because of its potential synergistic effect and reduced drug doses in clinical application. However, how to precisely control drug release dose and time using vehicles remains a challenge. This work developed an efficient drug delivery system to combat breast cancer, which can enhance drug effects despite reducing its concentration. Methods Controlled-release poly-lactic-co-glycolic acid (PLGA) scaffolds were fabricated by E-jet 3D printing to deliver doxorubicin (DOX) and cisplatin (CDDP) simultaneously. Results This drug delivery system allowed the use of a reduced drug dosage resulting in a better effect on the human breast cancer cell apoptosis and inhibiting tumor growth, compared with the effect of each drug and the two drugs administrated without PLGA scaffolds. Our study suggested that DOX-CDDP-PLGA scaffolds could efficiently destroy MDA-MB-231 cells and restrain tumor growth. Conclusions The 3D printed PLGA scaffolds with their time-programmed drug release might be useful as a new multi-drug delivery vehicle in cancer therapy, which has a potential advantage in a long term tumor cure and prevention of tumor recurrence.

Roles of additional emulsifiers in the structures of emulsion gels and stability of vitamin E

Hongqiang Chen,Like Mao,Zhanqun Hou,Fang Yuan,Yanxiang Gao

The role of the oil droplets in emulsion gelation (as active or inactive fillers) was modified by additional emulsifiers, and structures and functionalities of emulsion gels were evaluated. Emulsion gels based on denatured protein (whey protein isolate) were prepared via an acid-induced approach, and four emulsifiers, including glycerol monostearate (MG), tween 20 (TW), modified starch (OSA) and gum Arabic (GA) were tested. The including of MG or TW led to delayed onset of gelation, and the emulsion gels formed were of lower gel strength (described as storage modulus and fracture force), compared with the emulsion gels without additional emulsifiers. Increase in the content of MG or TW resulted in the gels with lower water holding capacity , and slight changes in swelling ratio. The presence of OSA or GA allowed faster gelation of the emulsions, forming gels with higher storage modulus and fracture force. Increase in the content of OSA or GA did not affect WHC of the gels, but contributed to higher swelling ratios. Microstructural observation indicated that the emulsion gels with OSA or GA had smaller pore size. When vitamin E was incorporated within the gels, the emulsifiers presented different effects on its degradation kinetics. Vitamin E degraded much faster in the systems with higher content of MG or TW, and lower retention ratios were obtained after the storage (55 °C for 12 days). On the contrary, the emulsion gels with OSA or GA provided good protection for vitamin E, and increase in the content of the emulsifiers allowed higher retention ratios.

Protective effects of vitamin E against bisphenol A-induced reproductive toxicity in male Kunming mice

Yu Wang,Jiayi Li,Rui Zhou,Lincai Yang,Peisheng He,Lixuan Chen,Liying Luo,Haowen Chen,Lian Wei

The protective effects of vitamin E (45, 150, and 500 mg/kg/d) against reproductive toxicity induced in male Kunming mice by intragastric administration of 120 mg/kg/d bisphenol A was investigated. After two weeks, relative organ weights, sperm counts, sperm abnormality rates and serum testosterone concentrations were determined. Epididymal sperm counts were significantly decreased, sperm abnormality was increased, and serum testosterone concentrations were dramatically reduced following bisphenol A exposure. Vitamin E treatment led to a marked increase in sperm numbers and a decrease in the proportion of malformed sperm in a dose-related manner; serum testosterone levels were also restored. This suggests that vitamin E could be used as prophylactic agent for preventing bisphenol A-induced reproductive toxicity.

Highly Integrated Nanoplatform Based on an E-Selectin-Targeting Strategy for Metastatic Breast Cancer Treatment

Xiao-Ling Xu,Meng-Lu Zhu,Di Liu,Gao-Feng Shu,Jing Qi,Yan Lu,Fei Wang,Xiao-Ying Ying,Jian Chen,Yong-Zhong Du

Therapeutic goals for metastatic breast cancer, including shrinkage of established metastasis and suppression of movement of tumor cells, are often hard to achieve and remain the main obstacles restricting the antimetastatic efficacy of targeted drug delivery systems (TDDSs). Herein, we proposed an E-selectin-targeting nanoplatform for the systemic treatment of metastatic breast cancer. Versatile functions, including killing the circulating tumor cells, shrinking the established lesions, as well as inhibiting the movement of tumor cells, were integrated into doxorubicin-loaded sialic acid-dextran-octadecanoic acid (SDO) micelles (SDD). The prepared SDD micelles could not only inhibit lung and liver metastasis in the orthotopic 4T1 tumors model, but also decrease the metastatic lesions in the metastatic 4T1 cell model, resulting in 27.33% reduced number of metastatic nodules when compared to those without sialic acid modification. It was found that the good antimetastatic effect of SDD was only partially attributed to its ability on removing metastatic cells and metastases. Most importantly, the blank SDO micelles left in the lesion could further inhibit the cell migration and cell–cell binding. These results suggest that SA-driven TDDS has the potential for specific targeting and effective treatment of cancer metastasis.

In situ quantitative bioanalysis of monomethyl auristatin E-conjugated antibody-drug conjugates by flow cytometry

Wen-Bin Zhao,Chi-Xiao Qiu,Ying Shen,Wen-Hui Liu,Jie Zhou,Ying-Chun Xu,Zhan Zhou,Shu-Qing Chen

Antibody-drug conjugates (ADCs) consist of cytotoxic agents covalently conjugated to monoclonal antibodies that substantially improve antitumour activity and reduce systemic toxicity. With the growing number of ADCs in clinical applications, more accurate bioanalysis data are urgently needed to facilitate the development and rational use of ADCs. Herein, we used antigen-positive cells as antigen carriers and ofatumumab (OFA-HL) and ofatumumab-based ADC (OFA-HL-MMAE) as examples to establish a new ligand-binding assay (LBA) method based on flow cytometry. We proved that the new method met the required analytical performance criteria and the lower limit of quantitation (LOQ) was 0.2 μg/mL. In addition, the LOQ of the quantitative OFA-HL flow cytometry method was reduced to 0.025 μg/mL by choosing an optimized fluorescent antibody, which indicated that the LOQ of the new method can be improved. What's more, the new method showed good stability and specificity when we used it to determine the concentrations of OFA-HL and OFA-HL-MMAE in mouse serum. During the bioanalysis of ADCs, various factors should be considered. Therefore, choosing optimal methods for ADC bioanalysis is necessary. This new method using in situ antigens not only extends the scope of application of the conventional LBA methods by avoiding the need for soluble antigens, but also improves the authenticity of ADC bioanalysis as a supplementary approach, which is valuable for developing accurate ADC assays.

UV-Visible and Raman Spectroscopic Studies of Lithocholic Acid on E-2-Butenal for AntiGlioma

Bo Zhang,Yanbin Su,Lei Chen,Zhuofu Wu,Fengrong Zhang,Yanwen Su,Gang Zhao,Zhengqiang Li

The characteristic changes in UV-visible and Raman spectra of lithocholic acid on E-2-Butenal for antiglioma are the maximum absorption peak of E-2-Butenal from 302 to 319 nm, the decreases of Raman signal of 1688 cm−1 and 1641 cm−1 in E-2-Butenal by lithocholic acid with the phenomenon of 1688 cm−1 splitting, and the disappearance of the band at 932 cm−1, which clearly displays the change of wagging in methyl. In the presence of mitochondrial protein deriving from glioma, the above mentioned effects of lithocholic acid on E-2-Butenal are more significant by Raman spectra detections. The molecular targets of lithocholic acid on E-2-Butenal are carbonyl group, conjugated double bonds, and methyl, respectively. The study concludes that the interaction between lithocholic acid and E-2-Butenal decreases the carbonyl group bond strength in E-2-Butenal and then changes the structure and function of E-2-Butenal. The internal cause of the interaction between lithocholic acid and E-2-Butenal leading to the change of the carbonyl group bond strength is the electronic delocalization near the carbonyl group bond in E-2-Butenal. Since E-2-Butenal derives from glioma in the process of lipid peroxidation of mitochondria, our investigation significantly contributes to understanding the mechanism of action of lithocholic acid as a potential antiglioma drug.

Bioimpedance Measurement under Feature Combination of Time-Frequency Domain and Application for E. coli Detection

Ning Lu,Boyu Guo,Zhikang Rao,Juntao Chen,Ying Xu

Electrochemical impedance sensors are widely used for bacterial detection and interpreted by the equivalent circuit model (ECM) to evaluate the concentration changes on the electrode interface. However, this method is mainly based on prior models and focuses on frequency domain analysis which cannot precisely describe unknown dynamic processes and a fractional surface coverage of electrode. Distribution of Relaxation Times (DRT) method, as an alternative to interpret impedance spectra can be applied to solve the problem. In this research, two electrode schemes with different modification steps were designed to collect impedance data. An Ensemble Tree-SHAP model was then adopted to quantitatively analyze the impedance data processed by ECM-DRT method in time-frequency domain and applied for the detection of Escherichia coli (E.coli). The results showed that using a dataset integrated with time-frequency domain features significantly improved the accuracy of bacterial concentration prediction. As a result, the RMSE of untrained concentration with LightGBM model was 6.4\times 1{0}^{-4}\,log CFU ml−1, with the sensor detection limit about 2.69 log CFU ml−1. The research provided a tool combining time-frequency analysis for accurate prediction of bacterial concentration, which is crucial for evaluation of the antibacterial effect of long-term antibiotics on E.coli in future.

Ameliorative effects of zinc and vitamin E against phthalates-induced reproductive toxicity in male rats

Ling-Zi Xia,Li-Lan Liu,Jun-Zhe Yue,Zhen-Yu Lu,Ru-Ya Deng,Xi He,Can-Can Li,Burong Hu,Hai-Tao Gao

Objective Phthalates (PEs) could cause reproductive harm to males. A mixture of three widely used PEs (MPEs) was used to investigate the ameliorative effects of zinc (Zn) and vitamin E (VE) against male reproductive toxicity. Methods Fifty male SD rats were randomly divided into five groups ( n = 10). Rats in MPEs group were orally treated with 160 mg/kg/d MPEs, while rats in MPEs combined Zn and/or VE groups were treated with 160 mg/kg/d MPEs plus 25 mg/kg/d Zn and/or 25 mg/kg/d VE. After intervention for 70 days, it's was measured of male reproductive organs' weight, histopathological observation of sperms and testes, serum hormones, PIWI proteins and steroidogenic proteins. Results Compared with control, anogenital distance, testes weight, epididymides weight, and sex hormones were significantly decreased, while the sperm malformation rate was markedly increased in MPEs group ( p < .05); the testicular tissues were injured in MPEs group with disordered and decreased spermatids, and arrested spermatogenesis. PIWIL1, PIWIL2, StAR, CYP11A1 and CYP19A1 were down-regulated in MPEs group ( p < .05). However, the alterations of these parameters were restored in MPEs combined Zn and/or VE groups ( p < .05). Conclusion Zn and/or VE improved steroid hormone metabolism, and inhibited MPEs' male reproductive toxicity.
