Background Co-delivery all-trans-retinoic acid (ATRA) and paclitaxel (PTX) is an effective strategy for cancer therapy. However, in many previous reported ATRA conjugated co-delivery systems, the ATRA was released slower than PTX, and the total drug release of ATRA far lower than that of PTX.Purpose We designed and prepared a pH and redox dual responsive drug delivery system (DA-ss-NPs) co-delivery ATRA and PTX for cancer therapy. The surface charge of DA-ss-NPs could change from negative to positive under tumor slightly acidic microenvironment, and both drugs could be quickly released from DA-ss-NPs under intracellular high concentration of glutathione (GSH).Methods The DA-ss-NPs were constructed by encapsulating PTX into the hydrophobic core of the polymer micelles, in which the polymer was synthesized by conjugating ATRA and 2,3-Dimethylmalefic anhydride (DMA) on side chains of Cystamine dihydrochloride (Cys) modified PEG-b-PAsp (named DA-ss-NPs). The surface charge of DA-ss-NPs under different pH conditions were detected. And the drug release was also measured under different concentration of GSH. The therapeutic effect of DA-ss-NPs were investigated in Human lung cancer A549 cells and A549 tumor-bearing mice. Results The zeta potential of DA-ss-NPs was -16.3 mV at pH 7.4, and which changed to 16 mV at pH 6.5. Cell uptake experiment showed that more DA-ss-NPs were internalized by A549 cells at pH 6.5 than that at pH 7.4. In addition, in presence of 10 mM GSH at pH 7.4, about 75%-85% ATRA was released from DA-ss-NPs within 48 h; but less than 20% ATRA was released without GSH. In vivo antitumor efficiency showed that the DA-ss-NPs could affectively inhibite the tumor in compared with control groups. Conclusion The charge-reversal and GSH-responsive DA-ss-NPs provide an excellent platform for potential tumor therapy.

This study obtained trans -free fats containing nervonic acid, a fatty acid with beneficial effects for mental health. Four fats containing 1.28–2.41% (w/w) nervonic acid were prepared by chemical or enzymatic interesterification of palm stearin (PS), Acer truncatum oil (ATO) and palm kernel oil (PKO). The chemical procedure was catalyzed using MeONa, whereas the enzymatic procedure was catalyzed with a 1,3–specific lipase. As a result, the interesterified fats showed a similar fatty acid profile to their corresponding physical blends measured by gas chromatography , but contained more nervonic acids at the sn -2 position, which was more conducive to metabolism and absorption. The content of tripalmitoylglycerol (PPP) and 1,3-dipalmitoyl-2-oleoyl glycerol (POP) or 1,2-dipalmitoyl-3-oleoyl glycerol (PPO) in the interesterified fats decreased significantly, resulting in improved plasticities. All interesterified fats showed a satisfactory content of tocopherols and oxidative stability. A combination of differential scanning calorimetry , X-ray diffraction spectroscopy and polarized light microscopy results suggested that the interesterified fats formed spherulitic needle-like crystals in the β′ form, while the physical blends formed dendritic crystals in the β form. These data indicated that the trans -free interesterified fats rich in nervonic acid may have a great potential in margarine applications.

Novel chiral core–shell silica microspheres with trans -(1 R ,2 R )-diaminocyclohexane (DACH) moiety bridged in the mesoporous shell were synthesized using layer-by-layer method. The chiral mesoporous shell around the nonporous silica core was formed by the co-condensation of N , N ′- bis -[(triethoxysilyl)propyl]- trans -(1 R ,2 R )- bis -(ureido)-cyclohexane (DACH-BS) and tetraethoxysilane (TEOS) using octadecyltrimethylammonium chloride (C 18 TMACl) and triblock poly(ethylene oxide)-b-poly(propylene oxide)-b-poly(ethylene oxide) copolymer (P123) as the templates. The functionalized core–shell silica microspheres were characterized and tested as chiral stationary phases for high performance liquid chromatography (HPLC). R / S -1,1′-bi-2,2′-naphthol, R / S -6,6′-dibromo-1,1′-bi-2-naphthol and R / S -1,1′-bi-2,2′-phenanthrol were enantioseparated rapidly on the column packed with the DACH core–shell silica particles. Moreover, the column packed with core–shell particles exhibited better performance than the column packed with the DACH functionalized periodic mesoporous organosilicas.

Changes in the content of trans -caryophyllene could identify whether sweetpotatoes are infected with black spot disease. Therefore, in this study, a quartz crystal microbalance (QCM) sensor based on modified CAU-1@ZIF-8 was used for non-destructive detection of sweetpotato black spot. The experimental results showed that the constructed sensor performed best when both modified CAU-1 and ZIF-8 were present. The sensor showed a linear trend (R 2 = 0.996, limit of detection (LOD) = 0.55 ppm) in response to trans -caryophyllene in the concentration range of 20–340 ppm with a sensitivity of −0.489 Hz (ppm) −1 . The developed gas sensor was successfully used for the detection of trans -caryophyllene in real samples of sweetpotato. The experimental data showed a significant correlation with the results of gas chromatography-mass spectrometry (GC–MS) with a correlation coefficient of 0.970 ( p < 0.01). Therefore, it could be used for early warning of black spot disease in sweetpotato.

Chronic exposure to lead ions (Pb2+) disrupts intestinal flora homeostasis, underscoring the need for sensitive and practical detection methods. Herein, we report a dual-mode analytical strategy combining split-DNA re-assembly-driven colorimetric reaction with CRISPR/Cas12a trans-cleavage activity for Pb2+ analysis. A bifunctional magnetic probe is constructed, where Pb2+-specific DNAzyme recognition triggers cleavage and release of crRNA, which subsequently activates Cas12a to generate fluorescence by cleaving a fluorophore-quencher reporter. Concurrently, the remaining magnetic bead-anchored trigger initiates split G-quadruplex re-assembly, catalyzing a visible colorimetric reaction. Under optimized conditions, the fluorescence mode achieves a detection limit of 1.03 fM with a linear range of 5 fM to 100 pM, while the colorimetric mode offers a limit of 62.1 fM from 100 fM to 500 pM. Both modes exhibit excellent specificity against competing metal ions. The platform's clinical feasibility is validated using human serum samples, showing strong correlation with ICP-MS, satisfactory recoveries (97.1–103.4%), and good repeatability (CV = 4.1%). Magnetic separation effectively minimizes matrix interference, making the assay suitable for complex biological specimens such as intestinal contents and fecal samples. This dual-mode design integrates fluorescence for precise quantification and colorimetry for equipment-free visual detection, holding great promise for point-of-care testing, early risk assessment of lead-induced gut microbiota dysbiosis, and environmental monitoring.

Although the impact of postharvest trans-2-hexenal (E2H) application in maintaining the quality of horticultural products has been widely studied, its effect on fresh common beans and the preservation mechanism remain unexplored. In this study, the impacts of E2H on reactive oxygen species (ROS) and cell wall metabolisms, and lignin biosynthesis in common beans were investigated. Fresh common beans fumigated with E2H (0, 0.5, 1.0, 1.5, and 2.0 μL L −1 ) were stored at 20 ± 1 °C for 12 days. Results showed that 1.0 μL L −1 E2H remarkedly repressed the decline of chlorophyll content to delay the yellowing, maintained ROS homeostasis, modulated cell wall metabolism-related enzyme activities to regulate cell wall components, and inhibited lignin biosynthesis-related enzyme activities to reduce lignin accumulation, thereby alleviating softening and lignification in common beans. These findings indicated that E2H could improve antioxidant activity, delay softening, and attenuate lignification to mitigate the quality deterioration in common beans.

In this study, a water falling film dielectric barrier discharge (DBD) reactor was used to degrade trans-ferulic acid (FA) in water. The results show that 98.4% of FA was degraded by 10 min treatment with the discharge voltage of 19 kV. The reactor displayed an excellent energy efficiency, which could reach to 13.4 g/kWh for FA degradation. Degradation characteristics of the water falling film DBD for FA was studied by single factor experiments and response surface methodology. The optimal operating condition and a quadratic polynomial equation for predicting FA degradation efficiency were obtained by the response surface model. The reactive species analysis unveils that the peroxone reaction between H 2 O 2 and O 3 generated by DBD could promote the formation of OH, causing a synergistic effect for FA degradation. Hydrated electron (e aq − ) converted into H in FA solution. In the present reaction system, H preferred to react with FA rather than merge with OH, and H showed a well ability to degrade FA. FA solution toxicity was weakened after DBD treatment.

Stemness and metastasis are the two main challenges in cancer therapy and are related to disease relapse post-treatment. They both have a strong correlation with chemoresistance and poor prognosis, ultimately leading to treatment failure. It has been reported that chemotherapy can induce stemness and metastasis in many cancer types, especially treatment with the chemotherapeutic agent doxorubicin (DOX) in breast cancer. A combination treatment is an efficient and elegant approach in cancer therapy through simultaneous delivery of two or more drugs with a delivery system for its synergistic effect, which is not an additive of two individual drugs. Herein, we report a combinatorial system with DOX and all-trans retinoic acid (ATRA) to address both of the above issues. As a common critical regulatory factor for oncogenic signal transduction pathways, Pin1 is a specific isomerase highly expressed within various tumor cells. ATRA, a newly identified Pin1 inhibitor, can abolish several oncogenic pathways by effectively inhibiting and degrading overexpressed Pin1. We successfully developed a folic acid (FA)-modified chitosan (CSO)-derived polymer (FA-CSOSA) and obtained FA-CSOSA/DOX and FA-CSOSA/ATRA drug-loaded micelles. FA modification can improve the uptake of the nanoparticles in tumor cells and tumor sites via folate receptor-mediated cell internalization. Compared to treatment with DOX alone, the combined treatment induced 4T1 cell apoptosis in a synergistic manner. Reduced stemness-related protein expression and inhibited metastasis were observed during treatment with FA-CSOSA/DOX and FA-CSOSA/ATRA and were found to be associated with Pin1. Further in vivo experiments showed that treatment with FA-CSOSA/DOX and FA-CSOSA/ATRA resulted in 85.5% tumor inhibition, which was 2.5-fold greater than that of cells treated with DOX·HCl alone. This work presents a new paradigm for addressing chemotherapy-induced side effects via degradation of Pin1 induced by tumor-targeted delivery of DOX and ATRA.

Herein, we demonstrated the synthesis and characterization of a well-defined diblock copolymer consisting of isotactic poly( p -hydroxystyrene) ( i P( p -HOS)) and 1,4- trans -polybutadiene (1,4- trans -PBD), i P( p -HOS)- b -1, 4- trans -PBD, by the combination of sequential monomer addition and deprotection. Isospecific living polymerization of p - tert - butyldimethylsilyloxystyrene and living trans -1,4-polymerization of 1,3-butadiene were catalyzed by 1, 4-dithiabutandiyl-2, 2ʹ-bis(6-cumenyl-4-methylphenoxy) titanium dichloride (complex 1) activated by methylaluminoxane (MAO) at room temperature to provide highly stereospecific isotactic poly( p - tert -butyldimethylsilyloxystyrene) ( i P( p -TBDMSOS)) and 1,4- trans -polybutadiene (1,4- trans -PBD) with narrow molecular weight distribution. Furthermore, the i P( p -TBDMSOS)- b -1, 4- trans -PBD was prepared via sequential monomer addition in the presence of complex 1 and MAO. The deprotection of the i P( p -TBDMSOS) block was promoted by concentrated hydrochloric acid to produce i P( p -HOS)- b -1, 4- trans -PBD. In addition, the resultant diblock copolymer and intermediate polymers were characterized by gel permeation chromatography, nuclear magnetic resonance and differential scanning calorimeter.

Fresh-cut potatoes (Solanum tuberosumL.) are susceptible to browning and microbial contamination during storage. In this study, the effects of trans-2-hexenal (E2H), ascorbic acid (VC), dimethyl dicarbonate (DMDC), and the combined treatment of E2H, VC, and DMDC on quality deterioration in fresh-cut potatoes were investigated. The response surface methodology (RSM) demonstrated that E2H, VC, and DMDC concentrations of 0.010%, 0.65%, and 240 mg/L, respectively, were the optimum conditions for fresh-cut potato preservation. Further analysis showed that the combined treatment of E2H, VC, and DMDC was the most effective method of reducing quality deterioration in potatoes compared to the control and individual treatments. Furthermore, the combined treatment of E2H, VC, and DMDC could decrease the accumulation of reactive oxygen species (ROS) via improving antioxidant enzyme activities. Meanwhile, energy-metabolism-related enzyme activities and glutamate decarboxylase (GAD) activity were enhanced, while γ-aminobutyric acid transaminase (GABA-T) activity was reduced via the combined treatment of E2H, VC, and DMDC, which contributed to maintaining high energy levels and GABA content in potatoes. These findings suggested that the combined treatment of E2H, VC, and DMDC could protect membrane integrity through enhancing antioxidant capacity, energy levels, and GABA content to maintain quality in fresh-cut potatoes.

All‑trans retinoic acid (ATRA) has been implicated in the differentiation of hepatic stellate cells (HSCs). In the present study, the liver‑targeting hyaluronic acid micelles (ADHG) were prepared for co‑delivery of ATRA and doxorubicin (DOX) to block the HSC‑hepatoma interrelation. To simulate the tumor microenvironment, an in vitro dual‑cell model and an in vivo co‑implantation mouse model were established for anticancer studies. The experimental methods involved the MTT assay, wound‑healing assay, cellular uptake, flow cytometry and and in vivo antitumor study. The results revealed that the HSCs in the research models notably promoted tumor proliferation and migration. Furthermore, ADHG were readily internalized by cancer cells and HSCs simultaneously, and widely distributed in cancer regions. The in vivo antitumor studies demonstrated that ADHG could notably decrease HSC activation and extracellular matrix deposition, as well as constrain tumor growth and metastasis. Therefore, ATRA could facilitate DOX‑induced anti‑proliferation and anti‑metastasis effects, and ADHG are a promising nano‑sized formulation for the combination therapy of hepatocellular carcinoma.

The solubility of trans-3-hydroxycinnamic acid in pure solvents (methanol, ethanol, n-propanol, isopropanol, n-butanol, isobutanol, methyl acetate, ethyl acetate, and n-propyl acetate) and (ethanol + ethyl acetate) cosolvent mixtures was measured by the gravimetric method from 293.15 to 333.15 K in normal pressure. The experimental results show that the solubility of 3-HCA(E) in alcohol solvents is greater than in ester solvents, which conforms to “like dissolves like”. On this basis, the experimental result was correlated with semiempirical models (the modified Apelblat, van’t Hoff, λh, Jouyban–Acree–van’t Hoff equation, and combined model) and activity coefficient models (Wilson, NRTL, and UNIQUAC equation). The modified Apelblat model has the best correlation of the experimental data, with an ARD of less than 0.5% for pure solvents and cosolvent mixtures. The solvent effect was calculated by the KAT-LSER equation, showing that the hydrogen bond alkalinity of solvents–solutes (β) and the solvent self-association (δH) significantly influence the solubility of 3-HCA(E), accounting for 32.46 and 55.02%, respectively. Finally, the thermodynamic parameters (ΔHmix, ΔSmix, ΔGmix,%ξH, and %ξTS) were calculated. Negative values of both ΔHmix and ΔGmix indicate that the mixing process of 3-HCA(E) is exothermic and spontaneous. The Gibbs energy of the mixing process is jointly contributed by enthalpy and entropy.
