Highly Efficient Cyan-Green Emission in Self-Activated Rb3RV2O8 (R = Y, Lu) Vanadate Phosphors for Full-Spectrum White Light-Emitting Diodes (LEDs)

Peipei Dang,Dongjie Liu,Yi Wei,Guogang Li,Hongzhou Lian,Mengmeng Shang,Jun Lin

Phosphor-converted white-light-emitting diodes (pc-WLEDs) rely on combining a near-ultraviolet (n-UV) or blue chip with trichromatic and yellow-emitting phosphors. It is challenging to discover cyan-green-emitting (480–520 nm) phosphors for compensating the spectral gap and producing full-spectrum white light. In this work, we successfully discovered two unprecedented bright cyan-green emitting Rb3RV2O8 (R = Y, Lu) phosphors that gives emission bands centered at 500 nm upon 362 nm n-UV light excitation. Interestingly, the both self-activated compounds exhibit high internal quantum efficiencies (IQEs) of 71% for Rb3YV2O8 and 85% for Rb3LuV2O8, respectively. Moreover, controllable emission color can be successfully tuned from cyan-green to orange-red across the warm white light region by design strategy of VO43– → Eu3+ energy transfer. The thermal quenching of as-prepared phosphors could be effectively mitigated by this design strategy. Finally, the as-fabricated n-UV (λex = 370 nm) pumped phosphor-converted (pc) W-LED devices utilizing Rb3RV2O8 (R = Y, Lu) along with commercial phosphors demonstrate well-distributed warm white light with high color-rendering index (CRI) of 91.9 and 93.5, and a low correlated color temperature (CCT) of 5095 and 4946 K. It suggests that the both vanadate phosphors have potential applications in full-spectrum pc-WLEDs.

Lactobacillus Plantarum R-Fermented Phellodendron Amurense Extract Protects Against Pseudorabies Virus by Regulating Oxidative Stress and Inflammation in Vitro and in Vivo

Zhao Mengyao,Wu Feifan,Xu Zenghao,Han Xing,Mao Sai,Zhang Hongying,Zhang Yina,Yang Mingfan

Pseudorabies virus (PRV) infection poses a significant health challenge to swine production, inducing oxidative stress, inflammatory responses, and reduced growth performance. Probiotic fermentation of medicinal plants is a promising strategy for generating novel probiotic-derived products for improving animal health. In this study, Phellodendron amurense (PA), a plant with both medicinal and dietary applications, was fermented with Lactobacillus plantarum R (LpR). We investigated the potential of fermented extract (FPAE) as a probiotic-derived feed additive for swine and its underlying mechanisms. Nontargeted UPLC‒MS/MS metabolomics revealed that fermentation significantly reshaped the metabolic profile of PAs, with marked enrichment of phenolic acids, flavonoids, and alkaloids. Specifically, the berberine content increased 2.5-fold, and the levels of antioxidant components such as ferulic acid and kaempferol were significantly increased. In vitro, FPAE inhibited PRV replication in PK-15 cells in a concentration-dependent manner (84.15% inhibition at 5 mg/mL). It also alleviated virus-induced oxidative stress, as evidenced by restored glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) activities and reduced malondialdehyde (MDA) levels. The supernatant of LpR showed no antiviral activity, confirming that the enhanced efficacy resulted from biotransformation of the plant matrix. In vivo, daily oral administration of FPAE (crude drug concentration of 125 mg/mL, 200 µL/mouse, equivalent to 892 mg/kg) significantly improved the survival rate (80% vs. 53.33% in the model group, and higher than the 73.33% in the unfermented PAE group); increased the average daily weight gain and feed conversion ratio; reduced the viral loads in the brain, lung, spleen and kidney; and, compared with PAE, more effectively reduced viral loads in the brain and alleviated pathological damage; and alleviated clinical symptoms and pathological damage to the brain. Furthermore, FPAE suppressed systemic inflammatory responses (decreased serum IL-6, P  < 0.05) and enhanced antioxidant defense (increased GSH-Px activity, P  < 0.05, P  = 0.004). Through a multi-dimensional “constituent–cell–animal” approach, this study demonstrated that probiotic fermentation restructures the chemical composition of Phellodendron amurense and synergistically regulates host oxidative stress and inflammatory responses to confer protection against viral infection. These findings provide a theoretical basis for developing LpR-fermented Phellodendron amurense as a functional feed additive to control PRV infection in swine.

The effects of R2O3 (R=La, Yb, Gd) on the microstructure, dielectric, ferroelectric, and energy storage properties of Ba0.65Bi0.07Sr0.245TiO3 relaxor ferroelectric ceramics

Fukang Chen,Yilin Zhang,Yang Li,Yan Yan,Lishun Yang,Xinyu Zeng,Tao Deng,Huanghui Nie,Gang Liu,Lixue Zhang

(1- x )Ba 0.65 Bi 0.07 Sr 0.245 TiO 3 - x R 2 O 3 (BBST- x R) ceramics embellished by lanthanides (La, Yb, Gd) were synthesized in this study, and the effects in properties were ascertained. The results show that lanthanides with smaller ionic radius differences and larger relative mass will effectively limit mass transport of other cations and restrain grain growth. It is also found that the paraelectric phase of ceramics can be stabilized by decreasing the ionic radius of lanthanides. In particular, the addition of La unfolds the exceptional recoverable storage density ( W rec ) up to 2.23 J/cm 3 and efficiency ( η ) of ∼76.7% under 280 kV/cm. Finite element simulation and PFM observation also further explain the reasons for the excellent performance. Concurrently, BBST-0.015La ceramic exhibits outstanding thermostability from 30 °C to 150 °C and frequency stabilization from 1 Hz to 300 Hz which indicates the potentiality of application in actual surroundings.

Vulgarisin-type diterpenoids from self-heal (Prunella vulgaris) and their neuroprotective effects against ischemia/reperfusion (I/R) via a mitochondria-related pathway

Fengwei Ma,Qingfang Deng,Huayong Lou,Jinyu Li,Su Xu,Wenneng Wu,Qihua Wen,Lei Tang,Xin Wang,Weidong Pan

Self-heal (Prunella vulgaris L.) is a perennial edible plant that is widely distributed across the world and is traditionally consumed as a food additive in soft drink beverages. In this study, to explore the functional components of P. vulgaris, an investigation of its ethanol extracts has been conducted by our group. As a result, twelve (1–12) vulgarisin-type diterpenoids with a special 5/6/4/5-fused ring skeleton, including six new ones (1–6), were obtained. Their structures including the absolute configuration were elucidated based on comprehensive spectroscopic evidence, ECD calculations, as well as single-crystal X-ray diffraction analyses. All the isolates were tested for neuroprotective effects against ischemia/reperfusion (I/R) on primary neuron cells through the oxygen and glucose deprivation and reperfusion (OGD/R) induced injury model. The results showed that all twelve vulgarisin-type diterpenoids possess promising neuroprotective activity at a concentration of 10 μM. Among them, compound 3 can significantly suppress cell apoptosis by regulating Bax/Bcl-2 protein expression and inhibiting cleaved caspase-3 and caspase-9 expression with a western blotting assay. Further research revealed that compound 3 could improve mitochondrial function by inhibiting mitochondrial cytochrome c release, reducing ROS levels, and maintaining the membrane potential. This work firstly reports vulgarisin-type diterpenoids possessing neuroprotective activity. These findings also suggest that daily consumption of P. vulgaris might prevent cerebral disorders via a mitochondria-related pathway.

Research on the Interaction Mechanism Between α Mino-Phosphonate Derivative Q-R and Harpin-Binding Protein 1 in Tobacco (Nicotiana tabacum) Plants

Huang Maoxi,Yan Yunlong,Wang Li,Chen Jun,Liu Tao,Xie Xin,Li Xiangyang

Amino-phosphonate derivative R-diphenyl-1-(4-methylbenzothiazole-2-amino)-1-(thiphene-2-yl)-methylphosphonate (Q-R) has a high protective anti-tobacco mosaic virus (TMV) activity. However, the mechanism responsible for Q-R’s effect on TMV infection is largely unknown. Here, we studied the expression levels of harpin-binding protein 1 (HrBP1) and pathogenesis-related protein-1a (PR-1a) in TMV-infected tobacco plants by using reverse transcription quantitative real-time PCR. Then, we verified the interactions between Q-R and the HrBP1 protein from Escherichia coli using isothermal titration calorimetry and studied the Q-R-associated assembly of HrBP1 using size-exclusion chromatography. The results showed that the expression levels of HrBP1 and PR-1a genes were significantly increased by Q-R at the transcriptional level in TMV-infected tobacco plants, and the E. coli-expressed HrBP1 protein was assembled into oligomers by Q-R via binding to HrBP1 with a dissociation constant of 1.19 μM. We, therefore, concluded that Q-R activated the HrBP1 and PR-1a genes and enhanced the ability of HrBP1 to assemble in tobacco plants.

Sol–gel combustion synthesis and antiferromagnetic properties of orthorhombic perovskite-type MFeO3:R3+ (M=La, Gd; R=Eu, Er, Ho) nanocrystalline powders

Peixi Hao,Linwen Jiang,Hanlei Zhao,Jiangtao Li,Shiwen Qiu,Hongbing Chen

The MFeO 3 : R 3+ (M = La, Gd; R = Eu, Er, Ho) nanocrystalline powders were successfully synthesized by the sol–gel combustion method using glycine-based gel at a low ignition temperature. The as-burnt products present honeycomb-like structure with many irregular continuous pores due to the releasing of a large amount of gas during the combustion reaction process. The lattice parameters and cell volumes of MFeO 3 :R 3+ (M = La, Gd; R = Eu, Er, Ho) nanocrystalline powders are compared in detail. The magnetic properties of all the samples were studied systematically. The results show that the doping of rare-earth ions enhanced greatly the coercivity of GdFeO 3 and LaFeO 3 as a result of the enhancement of magnetic anisotropy . The influence of doped rare-earth ions on the magnetic properties was summarized. The distinction of magnetism of MFeO 3 :R 3+ (M = La, Gd; R = Eu, Er, Ho) were explained in detail by the intrinsic magnetism of doped rare-earth ions. This work is significant for the application of rare-earth elements in other magnetic materials.

Stereoselective pharmacokinetics of (R)-(+)- and (S)-(−)-rabeprazole in human using chiral LC-MS/MS after administration of rabeprazole sodium enteric-coated tablet

Lu-Ning Sun,Yi-Wen Shen,Yu-Wen Ying,Duo Li,Teng-Fei Li,Xue-Hui Zhang,Ping Zhao,Li Ding,Yong-Qing Wang

Rabeprazole is an effective proton pump inhibitor to treat acid-related diseases. To achieve the simultaneous determination of rabeprazole enantiomers in human plasma, a chiral LC-MS/MS method was developed and validated. Acetonitrile including 0.1% ammonium were used as protein precipitating agent. Analytes were separated within 8 minutes on a Chiralpak IC column (4.6 mm × 150 mm, 5 μm). The mobile phase was 10 mM ammonium acetate including 0.2% acetic acid-acetonitrile (35:65, v/v). An API 4000 mass spectrometer was used as detector for the analysis, and the multiple reactions monitoring transitions of m/z 360.1 → 242.2 and 346.1 → 198.1 were opted for quantifying rabeprazole enantiomers and internal standard. Matrix effects were not apparent for each enantiomer and internal standard (esomeprazole), the calibration curves were linear over the concentration of 0.500 to 400 ng·mL −1 , the intra-run precisions were below 5.4%, the inter-run precisions were below 9.9%, and the accuracy was between −9.2% and 9.3%. There was no chiral inversion observed during sample storage, preparation procedure, and analysis, demonstrating that analytes were stable in this study. This method was applied to the stereoselective pharmacokinetic study of (R)-(+)- and (S)-(−)-rabeprazole after oral administration of 10-mg rabeprazole sodium enteric-coated tablet in healthy Chinese subjects.

Investigating the Mechanistic of Danhong Injection in Brain Damage Caused by Cardiac I/R Injury via Bioinformatics, Computer Simulation, and Experimental Validation

Jinfu Wang,Yan Guo,Huifen Zhou,Yanjie Hua,Haitong Wan,Jiehong Yang

Objective: Cardiac ischemia-reperfusion (I/R) injury has negative effects on the brain and can even lead to the occurrence of ischemic stroke. Clinical evidence shows that Danhong injection (DHI) protects the heart and brain following ischemic events. This study investigated the mechanisms and key active compounds underlying the therapeutic effect of DHI against brain damage induced by cardiac I/R injury. Methods: The gene expression omnibus database provided GSE66360 and GSE22255 data sets. The R programming language was used to identify the common differentially expressed genes (cDEGs). Gene ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analysis were performed, and protein–protein interaction network was constructed. Active compounds of DHI were collected from the Traditional Chinese Medicine Systems Pharmacology database. Molecular docking and molecular dynamics simulations were performed. The MMPBSA method was used to calculate the binding-free energy. The pkCSM server and DruLiTo software were used for Absorption, Distribution, metabolism, excretion, and toxicity (ADMET) analysis and drug-likeness analysis. Finally, in vitro experiments were conducted to validate the results. Results: A total of 27 cDEGs had been identified. The PPI and enrichment results indicated that TNF-α was considered to be the core target. A total of 80 active compounds were retrieved. The molecular docking results indicated that tanshinone I (TSI), tanshinone IIA (TSIIA), and hydroxyl safflower yellow A (HSYA) were selected as core active compounds. Molecular dynamics verification revealed that the conformations were relatively stable without significant fluctuations. MMPBSA analysis revealed that the binding energies of TSI, TSIIA, and HSYA with TNF-α were −36.01, −21.71, and −14.80 kcal/mol, respectively. LEU57 residue of TNF-α has the highest contribution. TSI and TSIIA passed both the ADMET analysis and drug-likeness screening, whereas HSYA did not. Experimental verification confirmed that DHI and TSIIA reduced the expression of TNF-α, NLRP3, and IL-1β in the injured H9C2 and rat brain microvascular endothelial cells. Conclusion: TNF-α can be considered to be a key target for BD-CI/R. TSIIA in DHI exerts a significant inhibitory effect on the inflammatory damage of BD-CI/R, providing new insights for future drug development.

Discovery of a new NADPH-dependent aldo-keto reductase from Candida orthopsilosis catalyzing the stereospecific synthesis of (R)-pantolactone by genome mining

Jiapao Wang,Pengfei Cheng,Yifeng Wu,Anming Wang,Fangming Liu,Xiaolin Pei

( R )-Pantolactone (PL) is a key chiral intermediate for the synthesis of calcium ( R )-pantothenate and ( R )-panthenol used as food additives. The commercial production of ( R )-pantothenate is performed by the resolution of racemic pantothenate, which is synthesized through an aldol condensation and a cyanation reaction. In this study, we investigated another synthetic method of ( R )-pantothenate through the stereoselective reduction of ketopantoyl lactone (KPL) by aldo-keto reductase (AKR). A series of conjugated polyketone reductases (CPRs) were discovered from GenBank database by genome mining approach. The putative CPR gene from Candida orthopsilosis Co 90–125 ( Cor CPR) was cloned and functionally expressed in Escherichia coli BL21 (DE3). The optimum pH and temperature of recombinant Cor CPR were 6.0–7.0 and 40 ℃, respectively. The K m and v max toward KPL were1.3 mM and 227.3 μ mol/min/mg protein, respectively. The conserved sequences suggest that Cor CPR belongs to AKR3C family of AKR superfamily. Furthermore, a catalytic tetrad was proposed, and the detailed mechanism was clarified by molecular docking. In a batch reaction, 50 mM KPL was reduced to ( R )-PL with 99% conversion and > 99% enantiomeric excess within 5 h. The recombinant Cor CPR from C. orthopsilosis shows potential application in the asymmetric synthesis of ( R )-pantothenate preparation.

Sporopollenin-chitosan microspheres loaded with an endophytic fungus Talaromyces neorugulosus R-209 for promoting development and controlling root rot in pigeon pea

Jin-Xian Fu,Jiao Jiao,Qing-Yan Gai,Yu-Jie Fu,Mo-Nan Wen,Xiao-Qing Wang,Jing He

Plant-beneficial microbes can be effective as biological agents for promoting development and controlling diseases in plants. However, direct inoculation of non-encapsulated plant-beneficial microbes into the soils can affect their vitality and efficacy. A novel bio-based encapsulant, sporopollenin-chitosan microspheres (SCMs), was developed to load an endophytic fungus Talaromyces neorugulosus R-209 with antagonistic activities against the root rot pathogen ( Rhizoctonia solani AG4) and plant growth-promoting functions. The results showed that T. neorugulosus R-209 encapsulated in SCMs ( Tn R-209−SCMs) could significantly enhance fungal spore germination rates and available nitrogen/phosphorus levels in the soil compared to the non-encapsulated fungus. In addition, the preliminary evidence suggests that Tn R-209−SCMs have a basic safety profile for practical applications. Inoculation with Tn R-209−SCMs could effectively promote development and enhance resistance in pigeon pea seedlings by promoting chlorophyll synthesis, improving photosynthesis, and enhancing phenolic compound accumulation. Meanwhile, T. neorugulosus R-209 was found to endogenously colonize root intercellular spaces. Moreover, co-inoculation of Tn R-209−SCMs and R. solani AG4 could reduce host defense responses compared to R. solani AG4-infected roots, as reflected by lower levels of phenolic compound accumulation and pathogenesis-/biosynthesis-related gene expression. Overall, Tn R-209−SCMs is a promising biological agent that can promote development and control root rot in plants, which also provides an innovative approach to biomaterial-supported agricultural practices.

Strengthening core-region hydrogen-bond networks and rigidifying surface loop to enhance thermostability of an (R)-selective transaminase converting chiral hydroxyl amines

Yuwen Wei,Fulong Li,Yukun Zheng,Youxiang Liang,Yan Du,Huimin Yu

Transaminases have important applications in the synthesis of drug intermediates such as chiral amines. However, natural transaminases exhibit suboptimal thermal stability, limiting their further applications. Building upon an Rhodobacter sp. -derived ( R )-selective transaminase ( Rb TA), we report a dual-region coupling engineering approach to improve thermostability of Rb TA by strengthening the core hydrogen-bond networks and rigidifying the flexible surface loop. Through single strategy, we identified 4 thermostability improved single mutations, among which I249Q demonstrated the most substantial improvement, achieving a 18-fold increase in half-life ( t 1 / 2 40 ) and a 11.2 ℃ increase in T 50 10 . Then in strategic coupling, the synergistic effect of dual-region modification was observed in both thermal stability and activity enhancement, as mutant with the best high-temperature catalytic performance, R136P/F228Y, had its T 50 10 improved by 7.1℃ and exhibited a 4.2-fold increase in k cat / K m towards ( R )-3-amino-1-butanol. Finally, R136P/F228Y achieved a 20.5 % improvement in conversion over WT in an analytical-scale synthesis in 72 h at a 5 ℃ elevated catalytic temperature. Molecular dynamics simulations demonstrated that the synergy of the formation of new hydrogen bonds and decrease in flexibility accounted for the thermostability improvements. This study provides guidance for enhancing thermostability of similar fold-type enzymes without impairing enzymatic activity in an efficient manner.
