Properties of a novel carboxymethyl chitosan derived from silkworm pupa

Lin Zhu,De-Qing Zou,Zuo-Qing Fan,Na Wang,Ying-Ying Bo,Yu-Qing Zhang,Guang Guo

In this study, a carboxymethyl chitosan derived from silkworm pupa (SP-carboxymethyl chitosan) was prepared. The physical characteristics of the SP chitin, chitosan, and carboxymethyl chitosan were analyzed. The scanning electron microscopy results showed that the surfaces of the samples from SP were more uneven, with more surface fractures compared with those of the reference substance (RS). Thermal analysis, X-ray photoelectron spectroscopy, and Fourier transform infrared spectroscopy analysis showed that the main molecular chain structures of SP samples and RSs had no substantial differences. However, the crystallinity and thermal decomposition temperature of the SP samples were lower compared with those of the RSs. All of these results provide a theoretical basis for the development of applications for the SP-carboxymethyl chitosan.

Preparation of Chitosan Stacking Membranes for Adsorption of Copper Ions

Xiaoxiao Zhang,Xuejuan Shi,Liang Ma,Xuan Pang,Lili Li

Chitosan (CS) stacking mats with excellent performance for adsorption of copper ions (Cu(II)) in wastewater were fabricated by alternating electrospinning/electrospraying. The hierarchical structure of the stacking membranes was designed by CS micro-hemispheres sandwiched between CS fibers. The CS stack membranes prepared by the electrospinning technology could effectively increase the specific surface area, and thus, facilitate the adsorption of copper ions. CS stacking membranes with three layers reached adsorption equilibrium within 60 min, and had a maximum absorbance of 276.2 mg/g. The absorbance performance was superior to most of the reported CS adsorbents. Compared with CS fiber mats which were dominated by CS chemical structure during adsorption, the stacking structure of CS membranes contributed to the high efficient capability, and exhibited the multilayer adsorption behavior. This study may develop a promising method for the design of environmentally-friendly natural polymer adsorbents to remove Cu(II) in wastewater. Keywords:chitosan;sandwiched stacking structure;copper ions;adsorption

Preparation and Characterization of Magnetic Chitosan Microspheres for Endotoxin Adsorption

Yu Yi,Canlin Lai,Yuqi Jiang,Wenyue Xiong,Jianfeng Mei,Guoqing Ying

A novel magnetic chitosan microsphere (MCM) was prepared from chitosan and Fe3O4 through inverse suspension crosslinking, epichlorohydrin activation, and grafted with ligands. The magnetometer measurements data indicated that the magnetic microspheres had superparamagnetic property as well as fast magnetic response without the magnetic hysteresis effect and thus can be well used in magnetic separation. The adsorption results showed that the magnetic chitosan microsphere grafted with ligand had good adsorption capacity on endotoxin up to 554.1 EU/g under the condition of 25.0 mg/mL adsorbent, 17.2 EU/mL endotoxin. It is expected that this new microsphere will have a feasible and useful application in adsorbing endotoxin from biological products.

Preparation and Evaluation of Vancomycin-Loaded N-trimethyl Chitosan Nanoparticles

Jiaojiao Xu,Beihua Xu,Dan Shou,Xiaojing Xia,Ying Hu

Chronic intracellular infections caused by drug-resistant pathogens pose a challenge to the treatment of chronic osteomyelitis. Such treatment requires an intracellular delivery system for the sustained release of antibiotics such as vancomycin (VCM), which is an antibiotic of last resort used against many clinically resistant bacteria. In this work, we report VCM-loadedN-trimethyl chitosan (TMC) nanoparticles and their potential application for drug delivery. The results showed that the prepared nanoparticles were predominantly spherical in shape with an average particle diameter of 220 nm, a positive zeta potential, and a loading efficiency of 73.65% ± 1.83%. Furthermore, their drug release profile followed the Higuchi model for sustained release, with non-Fickian diffusion. Over a 24-h period, 6.51% ± 0.58% of the drug within the optimized nanoparticles was released. In vitrocytology showed that osteoblasts (OBs) exhibited higher alkaline phosphatase activity (ALP) after exposure to TMC nanoparticle material. Furthermore, TMC nanoparticles increased the uptake of water-soluble quantum dots (QDs) by OBs, and both nanoparticles and VCM/TMC mixtures improved OB proliferative activity. We also investigated the minimum inhibitory concentration (MIC, 60 μg/mL), half maximal inhibitory concentration (IC50, 48.47 μg/mL), diameter of inhibition zone (DIZ, 1.050 cm), and turbidimetric (TB) assay of nanoparticles. All data demonstrated that VCM/TMC nanoparticles had excellent antibacterial activity against the Gram-positive bacteriumStaphylococcus aureus. These findings suggest that VCM-loaded TMC nanoparticles have good potential for the sustained delivery of antibiotics to bone infections. Keywords:vancomycin (VCM);N-trimethyl chitosan (TMC);intracellular infections;nanoparticles

Hypromellose succinate-crosslinked chitosan hydrogel films for potential wound dressing

Qiong Jiang,Wei zhou,Jun Wang,Rupei Tang,Di Zhang,Xin Wang

The objective of this study was to develop novel hydrogel films based on carboxyl-modified hypromellose-crosslinked chitosan for potential wound dressing. Hypromellose (HPMC) was grafted with succinic acid to yield hypromellose succinate (HPMCS), and then the reinforced hydrogel films of HPMCS-crosslinked chitosan (HPMCS-CS) were prepared through amide bond formation using 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide (EDC) and N - hydroxysuccinimide (NHS) as a catalyst. Compared to that of blend film, mechanical properties of HPMCS-CS hydrogel films were significantly enhanced both in dry and swollen state. To assess the applicability of HPMCS-CS hydrogel films as wound dressing, the swelling behavior, water vapor transmission rate (WVTR), oxygen permeability, biocompatibility (cytotoxicity and hemolysis), in vitro drug release and bactericidal properties were analyzed. The results indicated that HPMCS-CS hydrogel films with good biocompatibility possess high swelling ratio, proper WVTR, and oxygen permeability, which might accelerate tissue regeneration. Meanwhile, gentamycin sulfate release from drug-loaded HPMCS-CS hydrogel films were sustained, which would help to protect wound from infection.

A recyclable and regenerable magnetic chitosan absorbent for dye uptake

Weifeng Zhao,Xuelian Huang,Yilin Wang,Shudong Sun,Changsheng Zhao

A recyclable and regenerable magnetic polysaccharide absorbent for methylene blue (MB) removal was prepared by coating magnetic polyethyleneimine nanoparticles (PEI@MNPs) with sulfonated chitosan (SCS) and further cross-linked with glutaraldehyde. The driving force for coating is the electrostactic interaction between positively charged PEI and negatively charged SCS. Infrared spectra, zeta potential, thermal gravimetric analysis and X-ray diffraction demonstrated the successful synthesis of magnetic polysaccharide absorbent. The self-assembly of polysaccharide with magnetic nanopartices did not alter the saturation magnetization value of the absorbent confirmed by vibrating sample magnetometer. The nanoparticles showed fast removal (about 30   min reached equilibrium) of MB. In particular, the removal ability of MB after desorption did not reduce, demonstrating an excellent regeneration ability. Our study provides new insights into utilizing polysaccharides for environmental remediation and creating advanced magnetic materials for various promising applications.

Preparation and sustainable release of modified konjac glucomannan/chitosan nanospheres

Congjiao Shi,Pei Zhu,Na Chen,Xiaozhou Ye,Yun Wang,Shaobo Xiao

Biodegradable and biocompatible polymer nanospheres are useful materials for controlled drug delivery. In the present study, novel composite nanospheres were prepared from carboxymethyl konjac glucomannan (CKGM) and 2-hydroxypropyl trimethyl ammonium chloride chitosan (HACC) as a vaccine delivery vehicle by electrostatic complexation in a neutral aqueous solution without the use of chemical crosslinkers. By altering the CKGM and HACC concentrations, the average CKGM/HACC particle size could be tuned from approximately 600   nm to 1460   nm and the zeta potential from 39   mV to 50   mV. Furthermore, using ovalbumin (OVA) as a model molecule for vaccines, various parameters were determined to affect the CKGM/HACC nanosphere encapsulation efficiency and in vitro controlled release properties. Under optimum conditions, the OVA encapsulation efficiency of CKGM/HACC nanospheres was 71.8%, while sustained and continuous in vitro OVA release over a period of more than 24   h was observed. Therefore, CKGM/HACC nanospheres are novel drug delivery carriers with great potential for medical applications.

Ultrasonic-assisted synthesis and enhancement of chitosan/graphene nanosheet composites

Guangyu Wu,Tianqi Huang,Jianguo Huang

This study investigated a simple and low-cost method for synthesizing graphene nanosheet/chitosan (GNS/CS) composites. The method involved (1) exfoliating graphite to prepare graphite intercalation compounds (GICs) using environment-friendly ultrasonic energy and intercalating techniques, (2) preparing a high-concentration GNS/CS dispersion from a mixture of GICs and CS, and (3) producing GNS/CS composites with varying GNS levels by adding the GNS/CS dispersion to CS. By incorporating 0.6 wt% GNS into a CS matrix, the mechanical and electrochemical properties of the resultant GNS/CS composite were markedly improved. Maximum enhancements of 29% and 39% in the elastic modulus and hardness of CS, respectively, were obtained. Furthermore, GNS/CS redox peak currents increased sevenfold relative to a CS-modified electrode. POLYM. COMPOS., 39:4217–4223, 2018. © 2017 Society of Plastics Engineers

Fabrication of chitosan microspheres for efficient adsorption of methyl orange

Linlin Zhai,Zhishan Bai,Yong Zhu,Bingjie Wang,Wenqiang Luo

In this article, morphology, structure and size controllable chitosan microspheres with high mechanical strength were synthesized by microfluidic technology combining chemical crosslinking and used as an adsorbent for methyl orange. The synthesized adsorbents were characterized using scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), and an Energy Dispersive Spectrometer (EDS). The effect of pH revealed that the adsorption process depended on pH and the pH variation of methyl orange solution after adsorption indicated that adsorption capacity was affected through the associated role of chitosan nature and pH variation. Experimental results suggested that the as-prepared chitosan microspheres were controlled within a narrow size distribution (coefficients of variation is 1.81%), whose adsorption capacity reached to 207   mg·g −   1 and mechanical strength was suitable to resist forces. In addition, the adsorption isotherm was well fitted with the Langmuir model, and the adsorption kinetic was best described by the pseudo-second-order kinetic model. The high performance microfluidic-synthesized chitosan microspheres have promising potentials in the applications of removing dyes from wastewater.

PEGylated chitosan microspheres as mucoadhesive drug-delivery carriers for puerarin

Xiaofen Hu,Yuyan Zhang,Huifen Zhou,Haitong Wan

PEGylated chitosans with different degrees of grafting were synthesized, and the application potential of microspheres based on PEGylated chitosan as mucoadhesive drug-delivery carriers for puerarin was investigated. Compared with chitosan microspheres, PEGylated chitosan microspheres (PCMs) exhibited better physical stability and higher swelling capacity, and the amount of water uptake increased as the content of poly(ethylene glycol) methyl ether in the microspheres increased. PCMs showed obviously improved mucoadhesive behavior on a mucosa-like surface. Puerarin was incorporated into the microspheres, and the release experiments in vitro showed that the PEGylation of chitosan accelerated puerarin release from the particles and decreased the retention of the drug. The abilities of all of the tested microspheres to open tight junctions and improve the permeability of puerarin were demonstrated with a Caco-2 cell monolayer as an in vitro model. The amount of puerarin permeating across the Caco-2 cell monolayer was significantly increased by the incorporation of puerarin into the PCMs. © 2015 Wiley Periodicals, Inc. J. Appl. Polym. Degradation and compatibility behaviors of poly(glycolic acid) grafted chitosan

Luzhong Zhang,Sufeng Dou,Yan Li,Ying Yuan,Yawei Ji,Yaling Wang,Yumin Yang

The films of poly(glycolic acid) grafted chitosan were prepared without using a catalyst to improve the degradable property of chitosan. The films were characterized by Fourier transform-infrared spectroscopy and X-ray photoelectron spectroscopy (XPS). The degradation of the poly(glycolic acid) grafted chitosan films were investigated in the lysozyme solution. In vitro degradation tests revealed that the degradation rate of poly(glycolic acid) grafted chitosan films increased dramatically compared with chitosan. The degradation rate of poly(glycolic acid) grafted chitosan films gradually increased with the increasing of the molar ratio of glycolic acid to chitosan. Additionally, the poly(glycolic acid) grafted chitosan films have good biocompatibility, as demonstrated by in vitro cytotoxicity of the extraction fluids. The biocompatible and biodegradable poly(glycolic acid) grafted chitosan would be an effective material with controllable degradation rate to meet the diverse needs in biomedical fields.

Shear induced self-thickening in chitosan-grafted polyacrylamide aqueous solution

Lei Jin,Yonggang Shangguan,Tao Ye,Hu Yang,Quanfu An,Qiang Zheng

A remarkable shear induced self-thickening of chitosan-graft-polyacrylamide aqueous solution was observed. After the polyelectrolyte solution presenting shear thinning was subjected to a high-rate shear for several minutes, their viscosities recovered and then a much higher zero shear viscosity than the original one appeared. Obviously, the self-thickening differs from conventional shear thickening or viscous recovery, as reported previously. The mechanism of self-thickening was investigated by rheological methods together with TEM, 1H NMR and DLS, etc. It was found that some aggregates exist in original chitosan-graft-polyacrylamide aqueous solution and the scale of such aggregations would become larger within several minutes after a strong shear. The thickening was proven to be the result of an enhanced scale of GPAM aggregation in aqueous solution, and the mechanism of aggregation was proven to be intermolecular hydrogen bonding effects. Besides, the shear-induced self-thickening appears to be facile, maintainable and easily controllable by changing the shear conditions.
