Fabrication of Folic Acid-Decorated Hollow ZIF-8/Au/CuS Nanocomposites for Enhanced and Selective Anticancer Therapy

Le Gao,Zhitao Wu,Abdul-Rauf Ibrahim,Shu-Feng Zhou,Guowu Zhan

Hollow nanomaterials have been used as an attractive platform for the integration of multiple bioactive components for effective anticancer therapy. Herein, we report a novel and facile strategy for the fabrication of hollow and monodispersed zeolitic imidazolate framework-8 (ZIF-8) by the self-template method with folic acid (FA) as a bioetcher. Gold nanocluster and folic acid were critical for the formation of the hollow ZIF-8 (thickness of 38 nm) during solvothermal synthesis. By integrating CuS nanoparticles (size of 4.9 nm), the resultant quadruple ZIF-8/Au/CuS/FA nanocomposites (denoted as FACZ) exhibited effective anticancer activities on FA receptor-positive MCF-7 and HepG-2 tumor cells but a weak killing effect on HCMEC/D3 cells. Folic acid molecules were conjugated to the external surface of FACZ, which simultaneously offered an excellent tumor-targeting ability and fluorescence imaging property. Although the photothermal therapy caused by CuS was not so obvious due to partial reduction, the nanosized FACZ after cellular uptake was able to release Cu(I) to enable chemodynamic therapy. This catalytically decomposed H2O2 to produce highly reactive oxygen species via the Fenton-like reaction as determined by the extracellular and intracellular hydroxyl radical. Our work offers a simple route for the fabrication of hollow ZIF-8 nanocomposite with active and selective anticancer activity. This is envisaged to have great potentials in biomedical applications.

Injectable Self-Healing Hydrogels Containing CuS Nanoparticles with Abilities of Hemostasis, Antibacterial activity, and Promoting Wound Healing

Yue Kong,Zishuo Hou,Liangqin Zhou,Panfeng Zhang,Yaowen Ouyang,Peiwen Wang,Yuanwei Chen,Xianglin Luo

Injectable self-healing hydrogels containing functional nanoparticles (NPs) have attracted much attention in many fields of biomedicine. A series of injectable self-healing hydrogels containing PEGylation CuS NPs based on N-carboxyethyl chitosan (CEC) and oxidized sodium alginate (OA) were developed by taking advantages of the unique functions of CuS NPs and chitosan, referred to as CuS NP hydrogels or CEC–OAm–CuSn, where “m” stands for the concentration percentage of the added OA solution (w/v) and “n” represents the molar concentration of CuS NPs in the hydrogels. The physical properties of CuS NP hydrogels, syringeability, rapid self-repair ability, and photothermal performance were systematically investigated. The multiple functions for CuS NP hydrogels requested in the skin healing process were explored. The results showed that CuS NP hydrogels had not only adjustable physical properties and good injectable self-healing characteristics but also excellent functionalities, concurrently including hemostatic ability, bacteria killing capability, and cell migration and proliferation promotion. In vivo wound healing and histomorphological examinations of immunofluorescence staining in a mouse full-thickness wound model demonstrated good acceleration effects of these hydrogels for infected wound healing. Therefore, these injectable self-healing CuS NP hydrogels which possess the abilities of hemostasis, antibacterial activity, and infected-wound healing promotion exhibit great potential as in situ wound dressings.

Cu-MOF assisted synthesis of CuS/CdS(H)/CdS(C): Enhanced photocatalytic hydrogen production under visible light

Liangfeng Luo,Yidi Wang,Siping Huo,Peng Lv,Jun Fang,Yang Yang,Bin Fei

CuS/CdS(H)/CdS(C) photocatalysts were synthesized via the hydrothermal method by employing thiourea , Cd(CH 3 COO) 2 ·3H 2 O and copper 1,4-benzenedicarboxylate MOF (CuBDC). The photocatalysts were characterized by XRD , XPS , BET, TEM and UV–vis diffuse reflectance spectra. Interestingly, hexagonal CdS (CdS(H)) and cubic CdS (CdS(C)) were formed with phase junctions in one step when CuBDC was introduced in the synthesis process, in addition, CuS nanoparticles were deposited on CdS. However, only hexagonal CdS was obtained without CuBDC. It demonstrated that CuBDC was not only the precursor of CuS but also the structural modifier for CdS. With the reduction of re-combination of photo-induced electrons and holes caused by phase junctions and the enhancement of visible-light absorptions due to the loading of CuS, all CuS/CdS(H)/CdS(C) photocatalysts had higher photocurrent densities under visible-light irradiation, and consequently the higher rates of H 2 production than pure CdS(H). Typically, the catalyst with 2.89 wt% of Cu showed a highest rate of H 2 evolution at 2042 μmol/g/h.

Freestanding CuS nanowalls: ionic liquid-assisted synthesis and prominent catalytic performance for the decomposition of ammonium perchlorate

Kaisheng Yao,Chenchen Zhao,Nannan Sun,Weiwei Lu,Yuan Zhang,Huiyong Wang,Jianji Wang

Rational design of ingenious strategies to build hierarchical architectures of nanomaterials is highly desirable and technically important due to their excellent properties and widespread application in varied fields. Here, we report a facile one-pot approach for the solvothermal synthesis of freestanding CuS nanowalls with the assistance of a common ionic liquid, 1-decyl-3-methylimidazolium bromide ([C10mim]Br). In the presence of [C10mim]Br, intriguing CuS nanowalls have been fully constructed from the as-generated CuS thin nanosheets at the liquid–liquid interface of chloroform and water. An outstanding feature is that no hard template is utilized to support and/or assist the growth of CuS nanowalls. The as-prepared CuS nanowalls have been characterized and analyzed systematically and a series of factors affecting the morphology have been investigated. It is found that the alkyl chain length, anionic nature and heterocyclic structure of ionic liquids play an important role in directing the formation of freestanding CuS nanowalls. A possible growth mechanism for the formation of CuS nanowalls is presented based on control experiments. In addition, the as-synthesized CuS nanowalls demonstrate prominent catalytic activity for the thermal decomposition of ammonium perchlorate (AP).

Interfacial Electric State Reconstruction of CuS Nanosheets via Indium-Doping for Electro-Removal of Heavy Metal Anions

Guangzhen Liu,Zhenglin Chen,Tian Liu,Xunsheng Guo,Xianchuan Xie,Liming Yang,Xubiao Luo

Electro-reductive removal of heavy metal anions holds great potential for clean production. However, its application is hindered by a low removal efficiency and high energy consumption, due to inefficient electron and mass transfer under strong electrostatic repulsion and the occurrence of undesirable side reactions. In this study, we doped atomic In into the CuS nanoparticles to modify the built-in electric state of the CuS lattice, thus enhancing the reduction and removal of Cr2O72– (Cr(VI)). The In-doped CuS electrode (0.08In-CuS) achieves a 100% reduction efficiency for Cr(VI) within 20 min, with a Faradaic efficiency of 97.42%, and completely removes total Cr within 100 min, with kinetic constants five times higher than those of CuS. Detailed characterization and theoretical simulations revealed that the introduction of atomic In results in the deformation of the triangularly coordinated and tetrahedrally coordinated Cu layers and the displacement of atoms in the CuS lattice. The electron-deficient state of In resulted in a polarization of the electron distribution of Cu, thus promoting strong adsorption of both Cr(VI) and Cr(III). This work highlights the necessity of modulating the intrinsic electric field of the electrode surface in order to achieve the effective removal of heavy metal anions.

Interfacial electric field of CdIn2S4/CuS heterostructure induced S-scheme charge transfer for efficient photoelectrochemical water splitting

Zonghan Hu,Qi Wang,Zihang Wang,Lanlan Zhang,Chengwei Hu,Yuanhu Lei,Yupei Qiao,Bing Lv

Currently, the two predominant heterojunction types, namely type II and S-scheme heterojunctions, are extensively employed in photoelectrochemical (PEC) water splitting for hydrogen production. However, type II heterojunctions suffer from the drawback of diminished oxidation-reduction capability. In this work, S-scheme CdIn 2 S 4 /CuS (CIS/CuS) heterojunction nanoblocks (NBs) photoanode is constructed for the first time by a hydrothermal and the successive ionic layer adsorption and reaction (SILAR) methods. The heterojunction achieves a photocurrent density of up to 4.0 mA/cm 2 at 0 V vs. Ag/AgCl, 2.37 times that of the pure CIS NBs photoanode. X-ray photoelectron spectroscopy (XPS), Tauc plots, Mott-Schottky (M − S) tests and density functional theory (DFT) calculations reveal the formation of S-scheme band structure between the interface of CIS and CuS. As a result, the significantly enhanced PEC water splitting performance of CIS/CuS NBs photoanode is attributed to the superior light absorption ability of CuS and the effective charge separation achieved by S-scheme band structure between the interface of CIS and CuS. This work also provides new ideas and strategies for designing efficient S-scheme heterostructures for efficient PEC water splitting.

CuS nanosheets as additives in the hole transport layers for stable p-i-n perovskite solar cells

Qiaoyun Chen,Hatameh Asgarimoghaddam,Wenxi Ji,Wenting Wu,Ji Cao,Rui Wang,Wei Yu,Xiaoting Nie,Yi Zhou,Bo Song,Kevin P. Musselman

Although poly[bis(4-phenyl) (2,4,6-trimethylphenyl) amine] (PTAA) is a widely used hole transport layer (HTL) in perovskite solar cells, its poor conductivity and the mismatched energy levels between PTAA and formamidinium-based perovskites increase interfacial carrier recombination and charge-transport resistance. To solve these problems, inorganic CuS nanosheets were synthesized and applied as additives in PTAA for the first time. After the addition of CuS, the conductivity of the HTL improves, and the energy levels are better aligned. In addition, perovskite growth is controlled through the interaction between CuS and PbI 2 , thus improving the quality of the perovskite films, which reduces nonradiative recombination. The addition of CuS into PTAA improved the power conversion efficiency from 21.99 % to 22.92 %. Moreover, both the thermal and humidity stability improved.

Nanobody and CuS Nanoflower-Au-Based Lateral Flow Immunoassay Strip to Enhance the Detection of Aflatoxin B1

Yiming Zhao,Baoshan He,Danyang Li,Leyan Gao,Wenjie Ren

In the realm of analysis, the lateral flow immunoassay (LFIA) is frequently utilized due to its capability to be fast and immediate. However, the biggest challenge of the LFIA is its low detection sensitivity and tolerance to matrix interference, making it impossible to enable accurate, qualitative analyses. In this study, we developed a new LFIA with higher affinity and sensitivity, based on a nanobody (G8-DIG) and CuS nanoflowers-Au (CuS NFs-Au), for the detection of aflatoxin B1(AFB1) in maize. We synthesized the immunoprobe G8-DIG@CuS NFs-Au, stimulated the in situ development of Au nanoparticles (Au NPs) on Cu NFs by electrical displacement, and obtained Cu NFs-Au for fixing the G8-DIG. G8-DIG@CuS NFs-Au probe-based LFIAs may, in ideal circumstances, use a strip chromatography reader to accomplish sensitive quantitative detection and qualitative visualization. AFB1has a detection range of 2.82–89.56 µg/L and a detection limit of 0.87 µg/L. When compared with an LFIA based on CuS NFs, this sensitivity is increased by 2.76 times. The practical application of this method in corn flour demonstrated a recovery rate of 81.7% to 117%. Therefore, CuS NFs-Au show great potential for detecting analytes.

NIR-activatable proenzyme DSF/CuS@M for synergistic in situ chemotherapy/CDT/PTT/cuproptosis-based antitumor therapy

Lushuang Xu,Qinyue Chen,Yangwei Qian,Jin Wang,Chenxi Wang,Rui Fan,Jia-Wei Shen,Qiying Shen

Nanozymes provide therapeutic benefits against a diverse range of malignant tumors. However, their antitumor potencies are seriously impaired by their low catalytic efficiencies and the off-target toxicities caused by their “always-on” catalytic activity. Herein, we developed multifunctional nanoparticles co-loaded with ultra-small copper sulfide nanoparticles (CuS NPs) and disulfiram (DSF) (DSF/CuS@M). These nanoparticles had peroxidase-like activity and served as (near-infrared) NIR-activatable proenzymes to achieve synergistic in situ chemotherapy, chemodynamic therapy (CDT), photothermal therapy (PTT), and cuproptosis-based antitumor therapy. DSF/CuS@M not only provided PTT as NIR-activatable proenzymes but also enabled complete DSF and Cu 2+ release. Non-toxic DSF was converted into diethyldithiocarbamic acid (DDC) through a glutathione (GSH)-mediated metabolic reaction, leading to the generation of cytotoxic Cu(DDC) 2 in situ , which enhanced cell apoptosis. Cu 2+ was converted into Cu + by GSH, and this Cu + catalyzed the conversion of H 2 O 2 into hydroxyl radicals (·OH) to induce CDT. Moreover, Cu + could also bind to dihydrolipoyl transacetylase (DLAT) in the tricarboxylic acid (TCA) cycle, thereby inducing cuproptosis. These results demonstrate that the NIR-activatable proenzyme developed in this study is effective and safe for synergistic therapy against malignant tumors.

Synergistic purification of chromium-containing wastewater via adsorption-photocatalysis induced by multiple defect-containing CuS/Cv-CNNs

Luhua Wang,Yi Ren,Hongfang Zhao,Hongxia Li,Xiaohui Niu,Deyi Zhang,Haiyan Fan,Kunjie Wang

To address the water pollution problem caused by hexavalent chromium, this study took advantage of a multi-defect adsorption mechanism and constructed a multiple defect-containing type-II heterojunction material, CuS/Cv-CNNs. This material has simultaneous sulfur vacancies, copper vacancies, and carbon vacancies, which were used to accelerate the removal of chromium from wastewater. Attributed to the formation of electron-rich structures and free-moving ions by the construction of multiple defect sites, the photogenerated carriers generated by CuS/Cv-CNNs-70 were efficiently separated, and a high amount of physical Cr(VI) adsorption was achieved by electrostatic attraction and ion exchange, consistent with a Freundlich isothermal adsorption model. Studies on the adsorption thermodynamics demonstrate that CuS/Cv-CNNs-70 prefers to adsorb Cr(VI) when temperatures are raised. In addition, the removal of Cr(VI) (120 mg L−1) by CuS/Cv-CNNs-70 reached 97.10% after 60 minutes through the synergistic effect of adsorption and visible-light photocatalysis. This finding indicates that the synergistic effect between the multilocation defects stimulated the combined potential of adsorption and photocatalysis of CuS/Cv-CNNs-70 to realize the full utilization of the defects. In conclusion, this study confirms the prominent role of a multiple defect mechanism in wastewater treatment and environmental protection.

Synergizing photothermal and pyroelectric effects: Coal gangue assisted simultaneous ZnS/CuS for degradation of mineral processing wastewater

Meng Li,Weiji Sun,Wangkai Hui,Lang Liu

Photocatalytic oxidation technology is a green and low carbon approach for degradation of mineral processing wastewater, which is extensively applied to ecological environment restoration. In this research, coal gangue-ZnS/CuS pyroelectric/photothermal composite photocatalyst is constructed by high temperature calcination method. The temperature difference between heat converted from light energy absorbed by CuS and the circulating condensate water provides the impelling force for the pyroelectric effect. The porous structure of coal gangue acts as a “heat capacitor”, which furnishes greater temperature fluctuation for pyroelectric effect. Furthermore, the hydroxyl groups on the surface of coal gangue can availably adsorb oxygen and facilely form superoxide radicals with electrons. The degradation efficiency of xanthate is as high as 99.5 % within 50 min. This research has significant implications for designing and developing highly efficient photocatalysts for the degradation of mining wastewater.

Sulfated Chitosan-Modified CuS Nanocluster: A Versatile Nanoformulation for Simultaneous Antibacterial and Bone Regenerative Therapy in Periodontitis

Xiaoyu Chen,Ning Huang,Danyang Wang,Miao Zhang,Xuyang Deng,Fangze Guo,Bingcheng Yi,Changqing Yuan,Qihui Zhou

Periodontitis, a prevalent chronic inflammatory disease worldwide, is triggered by periodontopathogenic bacteria, resulting in the progressive destruction of periodontal tissue, particularly the alveolar bone. To effectively address periodontitis, this study proposed a nanoformulation known as CuS@MSN-SCS. This formulation involves coating citrate-grafted copper sulfide (CuS) nanoparticles with mesoporous silica (MSNs), followed by surface modification using amino groups and sulfated chitosan (SCS) through electrostatic interactions. The objective of this formulation is to achieve efficient bacteria removal by inducing ROS signaling pathways mediated by Cu2+ ions. Additionally, it aims to promote alveolar bone regeneration through Cu2+-induced pro-angiogenesis and SCS-mediated bone regeneration. As anticipated, by regulating the surface charges, the negatively charged CuS nanoparticles capped with sodium citrate were successfully coated with MSNs, and the subsequent introduction of amine groups using (3-aminopropyl)triethoxysilane was followed by the incorporation of SCS through electrostatic interactions, resulting in the formation of CuS@MSN-SCS. The developed nanoformulation was verified to not only significantly exacerbate the oxidative stress of Fusobacterium nucleatum, thereby suppressing bacteria growth and biofilm formation in vitro, but also effectively alleviate the inflammatory response and promote alveolar bone regeneration without evident biotoxicity in an in vivo rat periodontitis model. These findings contribute to the therapeutic effect on periodontitis. Overall, this study successfully developed a nanoformulation for combating bacteria and facilitating alveolar bone regeneration, demonstrating the promising potential for clinical treatment of periodontitis.
