Herein, a visual and luminescent dual-mode (colorimetric and fluorometric) method for the detection of P-phenylenediamine (PPD) in hair dye was successfully established based on cerium-nitrogen co-doped carbon dots (Ce, N-CDs) that displayed remarkable luminescence and peroxidase activity. Ce, N-CDs catalyzed H 2 O 2 to produce superoxide anion, which then oxidized the colorless 3,3,5,5-tetramethylbenzidine (TMB) into blue oxidized TMB (oxTMB), capable of quenching the fluorescence through fluorescence resonance energy transfer (FRET) between Ce, N-CDs and oxTMB. The reducing properties of PPD could reduce oxTMB back to TMB, leading to a decrease in the absorption intensity of oxTMB and a fluorescence recovery of Ce, N-CDs. As a result, the quantitative detection of PPD could be achieved by measuring the absorption values of oxTMB and the fluorescence signal of Ce, N-CDs. The detection limits for PPD were calculated as 0.36 µM and 0.10 µM for colorimetry and fluorimetry, respectively. Furthermore, smartphone application (ColorPicker) capable of measuring the RGB value of the color was utilized in the detection system, facilitating on-site quantitative detection. This approach effectively shortens the detection time and simplifies the operation, offering a powerful and convenient tool for real-time monitoring of PPD.

A far-red fluorescence “off-on” sensing strategy is described for sequential ratiometric determination of Cu 2+ and L-histidine (L-His) based on fluorescence resonance energy transfer (FRET) system. N,S,P co-doped carbon dots (N,S,P-CDs) and N-acetyl-L-cysteine functionalized gold nanoclusters (NAC-AuNCs) are used in the FRET system, which serve as energy donor and acceptor, respectively. After adding NAC-AuNCs into the solution of N,S,P-CDs, the fluorescence of N,S,P-CDs is effectively quenched, while the far-red fluorescence of NAC-AuNCs appears. Cu 2+ can decrease fluorescence of NAC-AuNCs, and then L-His can effectively recover the fluorescence of NAC-AuNCs. The possible reason is that the stronger affinity between Cu 2+ and L-His can pull Cu 2+ away from the surface of NAC-AuNCs. Through it all, the emission intensity of N,S,P-CDs remains nearly constant, so the ratio of fluorescence intensities at 485 and 625 nm exhibits a linear correlation to the Cu 2+ and L-His concentration, respectively. The sensing platform shows good selectivity towards Cu 2+ and L-His with a linear range of 0.65–26.58 μM and 3.13–56.25 μM and determination limits of 0.50 μM and 0.374 μM, respectively. The proposed method has been successfully used for Cu 2+ and L-His determination in real samples with satisfying results.

Early diagnosis and treatment of cancer are key problems for effective cancer therapy. Therefore, the development of multifunctional materials that can integrate imaging and therapeutic functions is of great importance. Lanthanide-doped nanoparticles@metal-organic frameworks (Ln-NPs@MOFs) offer an unprecedented opportunity to address this challenge, by combining the imaging capabilities of rare earth-doped nanoparticles with the photosensitizing properties of MOFs for photodynamic therapy (PDT). Here, we report a near-infrared (NIR) light-responsive nanoparticle based on a zirconium-porphyrin metal-organic framework (PCN-224) for synergistic photothermal and photodynamic therapy. Tetra(4-carboxyphenyl)porphyrin (TCPP) that constitutes the MOF framework serves as a natural photosensitizer, enabling efficient photodynamic therapy through Förster resonance energy transfer (FRET). The dye indocyanine green (ICG) was loaded into the MOF pores to sensitize the Ln nanoparticles, enhancing the upconversion fluorescence by 41 times and the downconversion fluorescence by 5 times, which is beneficial to the biological efficacy for imaging diagnosis. Meanwhile, ICG, a traditional photothermal agent, also imparts photothermal therapy capabilities to the system. Furthermore, the encapsulation of cancer cell membranes not only enhances biocompatibility but also improves the material's targeting towards cancer cells, thereby greatly enhancing the therapeutic effect. This dual-functional material provides a promising platform for the integration of cancer diagnosis and therapy.

Transcytosis-inducing nanomedicines have been developed to improve tumor extravasation. However, the fate during transcytosis across multicell layers and the structural integrity of the nanomedicines before reaching tumor cells could impact antitumor therapy. Here, a BAY 87–2243 (a hypoxia-inducible factor-1 inhibitor)-loaded liposomal system (HA-P-L BAY ) modified by low molecular weight protamine (LMWP) and crosslinked by hyaluronic acid (HA) was constructed. This system could accomplish differentiate cellular transport in endothelial and tumor cells by fine-tuning its structural integrity, i.e. transcytosis across the endothelial cells while preserving structural integrity, facilitating subsequent retention and drug release within tumor cells via degradation-induced aggregation. In vitro cellular uptake and transwell studies demonstrated that HA-P-L BAY were internalized by endothelial cells (bEnd.3) via an active, caveolin and heparin sulfate proteoglycan (HSPG)-mediated endocytosis, and subsequently achieved transcytosis mainly through the ER/Golgi pathway. Moreover, the fluorescence resonance energy transfer (FRET) study showed that HA-crosslinking maintained higher integrity of HA-P-L BAY after transcytosis, more efficiently than electrostatic coating of HA (HA/P-L BAY ). In addition, more HA-P-L BAY was retained in tumor cells (4T1) compared to HA/P-L BAY corresponding to its enhanced in vitro cytotoxicity. This may be attributed to better integrity of HA-P-L BAY post endothelial transcytosis and more degradation of HA in tumor cells, leading to more liposome aggregation and inhibition of their transcytosis, which was inferred by both TEM images and the HAase responsiveness assay proved by FRET. In vivo, HA-P-L BAY exhibited more potency in tumor suppression than the other formulations in both low and high permeability tumor models. This highlighted that fine-tuning of structural integrity of nanocarriers played a key role no matter whether the transcytosis of nanocarriers contributed to cellular transport. Collectively, this study provides a promising strategy for antitumor therapies by fine-tuning liposome integrity to achieve active trans-endothelial transport with structural integrity and selective aggregation for prolonged tumor retention.
