Early-life taurine supplementation confers long-term neuroprotection via NF-κB/FOXO-related nutritional programming

Luyao Zhang,Liting Guo,Bufan Xu,Di Wu,Yongning Wu,Guoliang Li

Taurine, an important nutritional fortifier in infant formula, is closely associated with neurodevelopment and immune homeostasis. However, existing studies have mainly focused on its immediate promoting effects on neurodevelopment, while whether early-life taurine supplementation confers long-term neuroprotective effects during later developmental stages remains unclear. In this study, a three-stage zebrafish model was established, including early-life taurine supplementation, cessation of supplementation, and subsequent exposure to acrylamide during later development, to evaluate the long-term neuroprotective potential of taurine. The results showed that early-life taurine supplementation significantly alleviated neurobehavioral deficits, oxidative damage in brain tissue, and inflammatory responses induced by acrylamide exposure during later developmental stages. Mechanistically, taurine may exert these effects through coordinated regulation of neuroimmune and metabolic pathways. At the immune level, taurine inhibited NF-κB-related inflammatory activation, suppressed M1-associated microglial markers, and promoted M2-associated responses, thereby improving the neuroimmune microenvironment. At the metabolic level, taurine regulated FOXO pathway-related gene expression and improved glycolysis-related metabolic disturbance, which was associated with reduced oxidative damage and improved energy homeostasis. Overall, these findings suggest that transient taurine supplementation during early life may reduce later susceptibility to foodborne neurotoxicant exposure, which is associated with nutritional programming-like changes in neuroimmune and metabolic pathways. This study provides a new perspective for early-life nutritional intervention and dietary risk assessment.

Targeting fabrication of a TFC NF membrane for the desalination of sulfate-type brackish water

Junliang Dong,Song Su,Qianzhi Sun,Ran Yu,Longyi Lv,Ruijun Zhang

Nanofiltration (NF) is a promising technology for brackish water desalination due to its tunable separation efficiency and effective removal of organic contaminants. However, the limited diversity of commercial NF membranes hinders optimal performance. To address this, we developed a tailored NF membrane by simultaneously increasing the piperazine (PIP) concentration and decreasing the trimesoyl chloride (TMC) concentration aiming for the treatment of a typical sulfate-type brackish water. The optimized membrane demonstrated superior performance: adequate desalination capability and improved anti-fouling properties. In particular, the NF membrane fabricated with 1.5 %( w / v ) PIP and 0.03 %(w/v) TMC exhibited a more compact polyamide structure and superior desalination performance compared to NF270. In comparison to NF90, it presented greater surface hydrophilicity (~40°) and nearly two-fold higher permeation flux. Moreover, it exhibited significantly improved anti-fouling ability, as evidenced by its 99.9 % flux recovery versus NF90's 23.5 % after brine cleaning. This enhancement is attributed to its optimized solute selectivity and surface chemistry, which reduce concentration polarization and foulant adhesion. Our work provides a practical and feasible strategy for customizing NF membrane structures to improve desalination efficiency and sustainability.

High-Performance Thin Film Composite Nanofiltration (NF) Membrane Constructed on Modified Polyvinylidene Fluoride (PVDF) Substrate

Junliang Dong,Qianzhi Sun,Xiaolin Feng,Ruijun Zhang

The inherent hydrophobic nature of PVDF material renders it challenging to establish a stable aqueous hydration layer, thereby limiting its suitability as a substrate for the preparation of nanofiltration (NF) membranes. In this study, we developed a novel modification approach that effectively enhances the hydrophilicity of PVDF substrates through the incorporation of sulfonic acid-doped polyaniline (SPANI) and hyperbranched polyester (HPE) into the PVDF casting solution, followed by cross-linking with trimesoyl chloride (TMC). The introduction of SPANI and HPE, which contain reactive polar amino and hydroxyl groups, improved the hydrophilicity of the substrate, while the subsequent cross-linking with TMC effectively anchored these components within the substrate through the covalent linking between TMC and the reactive sites. Additionally, the hydrolysis of TMC yielded non-reactive carboxyl groups, which further enhanced the hydrophilicity of the substrate. As a result, the modified PVDF substrate exhibited improved hydrophilicity, facilitating the construction of an intact polyamide layer. In addition, the fabricated TFC NF membrane demonstrated excellent performance in the advanced treatment of tap water, achieving a total dissolved solid removal rate of 57.9% and a total organic carbon removal rate of 85.3%. This work provides a facile and effective route to modify PVDF substrates for NF membrane fabrication.

Herpetrione alleviates ANIT-induced cholestatic liver injury by targeting FXR to suppress NF-κB signaling

Lin Jing,Yixuan Liao,Honghong Zhan,Qianyu Chen,Xiaodong Wei,Yinda Qiu,Fancheng Meng,Zhihua Liao,Min Chen

Ethnopharmacology relevance Herpetrione, an active lignan isolated from Herpetospermum pedunculosum , a traditional Tibetan medicinal plant renowned for its use in hepatobiliary diseases, has been long applied in the treatment of jaundice, hepatitis, hepatic fibrosis and other conditions, demonstrating remarkable hepatoprotective efficacy. Aim of the study Based on the traditional efficacy of Herpetospermum pedunculosum as well as the hepatoprotective activity of herpetrione, we aim to investigate the therapeutic effects and molecular mechanisms of herpetrione against α-naphthylisothiocyanate (ANIT)-induced cholestasis in mice. Materials and methods Therapeutic effects of herpetrione were evaluated in ANIT-induced cholestatic mice and Fxr knockout mice. Molecular interactions with FXR were validated using CETSA, SIP, ITC, and SPR assays, with binding sites confirmed via molecular docking and amino acid mutation analyses. FXR-regulated signaling was explored in HepG2 cells treated with guggulsterone/lithocholic acid, using Co-IP, yeast one-hybrid, and dual-luciferase assays to clarify mechanisms. Results Herpetrione significantly alleviated ANIT-induced cholestatic liver injury, evidenced by reduced serum biomarkers of bile acid accumulation and liver damage, improved hepatobiliary pathology, and decreased hepatic inflammatory infiltration. Mechanistically, it directly bound the ligand-binding domain (Glu467/Ile317/Leu464) of FXR, then ligand-bound FXR recruit coactivator SRC-1 to trigger transcriptional upregulation of BSEP/MRP2/SHP to enhance bile acid excretion while inhibiting synthesis, thereby restoring hepatic and bile acid homeostasis. Notably, herpetrione-activated FXR directly binds the promoter region of NFKB1 to inactivate NF-κB signaling and suppress inflammation. Importantly, the therapeutic effects of herpetrione were abrogated in Fxr knockout mice. Conclusion This dual regulation of bile acid homeostasis and inflammatory signaling through targeting the FXR highlights the therapeutic potential of herpetrione in cholestatic injury.

Efficient nanosheet clusters Ni6Fe2Co2/NF-VC for promoting the OER in anion exchange membrane electrolysis

Zandong Zhang,Jing Xin,Zuobo Yang,Weixuan Liu,Hongchen Liu,Jimmy Yun,Jie Zhang

This study developed a ternary Ni₆Fe₂Co₂/NF-VC nanoparticle cluster material, aiming to enhance the efficiency of oxygen evolution reaction (OER) and achieve excellent water decomposition performance. A self-supported anode was fabricated via one-electrode reduction synthesis for the OER active site, which exhibits significant catalytic activity due to its unique surface restructuring process and controlled electronic structure. The results demonstrate that this anode has outstanding intrinsic catalytic activity in anion exchange membrane (AEM) electrolysis: at 10 mA/cm² current density, the overpotential is as low as 203 mV, with a Tafel slope of 19.69 mV/dec. Under industrial testing conditions (500 mA/cm²), the anode requires only 1.98 V to operate stably and maintains stable operation for 50 hours under continuous cycling. Pressure loss analysis and electrochemical impedance spectroscopy (EIS) studies reveal that the rate-controlling step of the OER reaction is the mass transfer process. The improved catalytic material achieves enhanced dynamic and transport performance through the construction of favorable gas-liquid transfer channels and electronic transfer pathways, effectively reducing kinetic and diffusion limitations in the electrolysis system. This study provides a novel approach for designing highly efficient and stable water decomposition catalysts.

Impact of pore-forming agents on the performance and structure of TFC polyamide NF membrane

Jinxiong Song,Lijun Song,Biao Ai,Youcai Chen,Ying Liu,Yuzhang Zhu,Jian Jin

Thin-film composite (TFC) polyamide (PA) nanofiltration (NF) membranes are essential for separating ions and small molecules in the industries, including water treatment, chemical processing, energy production, and pharmaceuticals. These membranes are typically fabricated via interfacial polymerization (IP) on polysulfone (PSf) or polyethersulfone (PES) ultrafiltration (UF) supports. Apparently, optimizing the physicochemical properties of the UF support is very important for achieving TFC-PA NF membranes with superior separation performance. This study systematically investigated the influence of common pore-forming agents used in UF membrane fabrication, specifically polyvinylpyrrolidone (PVP), polyethylene glycol (PEG), and lithium chloride (LiCl), on the desalination performance of resulting TFC-PA NF membranes. We characterized the zeta potential, pore structure, and surface wettability of PSf UF membranes prepared with each pore agent. These characteristics were then correlated with the adsorption and desorption behavior of piperazine (PIP) monomers during the IP process. Our findings indicate that employing PVP as a pore-forming agent not only enhanced PIP adsorption within the UF support but also facilitated its rapid release during the IP reaction. This dual effect resulted in a PA selective layer with reduced thickness and smaller pore size. Consequently, the TFC-PA NF membrane fabricated on the PVP-modified UF support exhibited both higher water permeance and improved salt rejection. This research highlights the significant role of the pore-forming agent in determining the performance of TFC-PA NF membranes. The results suggest that UF supports prepared using PVP as a pore agent may be optimal for creating NF membranes with enhanced overall performance.

Fluorine-doped Mo-Ni-F/NF bifunctional electrode for efficient electrochemical hydrogen storage via quinoxaline

Qiyuexin Wang,Hanyu Li,Huan Li,Xueying Yang,Fangcheng Qiu,Dacheng Zhou,Zhengwen Yang,Xin Zhen,Jianbei Qiu

The implementation of liquid organic hydrogen carriers (LOHCs) is constrained by the high energy consumption and strong dependence on precious‑metal catalysts in conventional thermal processes. This work develops a fluorine-doped Mo-Ni-F/NF bifunctional electrode through a simple electrochemical deposition-sintering method for efficient electrochemical hydrogen storage using quinoxaline under ambient conditions. XRD and TEM characterization revealed a unique structure comprising a MoNi₄ shell with embedded Ni nanoparticles, while SEM and EDS mapping confirmed uniform fluoride distribution and enhanced surface morphology. XPS analysis indicated that fluoride modification improved surface characteristics without altering chemical states. The catalyst demonstrates exceptional performance in reversible quinoxaline hydrogenation (92.1 % conversion after 5 h, 95.6 % FE) and dehydrogenation (100 % conversion after 4 h, 97.2 % FE). DFT calculations showed that electron transfer between MoNi₄ and nickel nanoparticles enhanced catalytic activity, while fluorine modulation reduced the adsorption energy of quinoxaline on nickel (111) from −2.84 eV to −0.55 eV, and tetrahydroquinoxaline from −2.08 eV to −0.84 eV, mitigating catalyst poisoning. In situ Raman spectroscopy identified dynamically formed NiOOH as the active phase, which facilitates proton-coupled dehydrogenation via -OH/μ-O-Ni intermediates. This work provides novel catalytic materials and mechanistic insights for the development of low-cost, room-temperature, and highly effective electrochemical LOHC systems.

Electroacupuncture alleviates pain by activating the MD2/TLR4/NF-κB pathway in the ST36 acupoint

Jiangjiang Fu,Gangchen Cheng,Zhongxi Lyu,Zezhi Fan,Feiyang Li,Zhifang Xu,Bo Chen,Yongming Guo,Yuhan Liu,Hui Zhang,Yuan Xu,Yi Guo

Introduction: The acupuncture acupoint is the critical initial site for the therapeutic efficacy of electroacupuncture (EA). Previous studies have confirmed that the NF-κB pathway within the acupoint region mediates the therapeutic effects of acupuncture. Therefore, this study focuses on an in-depth investigation of the MD2/TLR4/NF-κB axis. Methods: The mouse model of adjuvant-induced arthritis (AIA) was established via intraplantar injection of Complete Freund’s Adjuvant (CFA). EA intervention was applied bilaterally to the Zusanli (ST36) acupoints, and behavioral, molecular, and immunological approaches were integrated to investigate the role of MD2 in EA-mediated analgesia. Results: EA increased paw thermal withdrawal thresholds (PTWTs) in AIA mice (P < 0.05), accompanied by higher levels of MD2, TLR4, p65, and the phosphorylated form of p65 (p-p65) at the acupoint. Co-immunoprecipitation (Co-IP) confirmed binding between MD2 and TLR4 in ST36 acupoint, while immunofluorescence (IF) revealed co-localization of TLR4 with fibroblasts and mast cells in ST36, suggesting these immune cells are critical targets for signal activation. Lentivirus-mediated knockdown of MD2 in the acupoint partially reversed EA’s analgesic effects and suppressed downstream TLR4/NF-κB pathway activation, whereas MD2 overexpression elicited a partial analgesic effect and promoted pathway activation. Together with spinal cord proteomics data, these findings indicate that modulating MD2 in the acupoint can regulate spinal cord-related signaling pathways. Mechanistically, EA dynamically regulates the equilibrium of the Grem1/BMP4/COX2 axis in the spinal dorsal horn via activation of the MD2/TLR4/NF-κB pathway cascade, achieving systemic analgesia. Conclusion: This study provides molecular evidence supporting the "acupoint priming" theory in acupuncture and highlights MD2 as a potential therapeutic target for pain management.

Macrophage-biomimetic nanomedicine for targeted therapy of abdominal aortic aneurysm via Nrf2/NF-κB pathway

Qiujie Luo,Wei Meng,Jiahui Wang,Xiang Zhang,Qingbo Meng,Mengjie Hu,Shunbo Wei,Xiaobo Yu,Dawei Deng,Yuqin Zhang,Zixuan Ma,Shentao Li,Shuang Wang,Binhao Zhang,Jingli Ding,Jinping Liu,Jianliang Zhou

Rationale: Abdominal aortic aneurysm (AAA) is a significant cause of death worldwide, with both mortality and incidence rates gradually increasing. Its complex pathological mechanisms hinder drug development. This work aims to develop a biomimetic multifunctional nanoparticle for targeted AAA therapy. Methods: Leveraging the multi-pore property of mesoporous Prussian blue nanoparticles (MPB NPs), we encapsulated the rosiglitazone (RLZ) into MPB NPs to synthesize MPB-RLZ NPs. Then, macrophage-biomimetic nanoparticles (MPB-RLZ@MM NPs) were prepared by coating MPB-RLZ NPs with macrophage membranes (MM). The characterization and reactive oxygen species (ROS)-scavenging ability of MPB-RLZ@MM NPs were evaluated. Next, the biocompatibility and biological functions of MPB-RLZ@MM NPs were evaluated. Finally, we assessed the targeting efficacy and therapeutic efficacy of MPB-RLZ@MM NPs in vivo . Results: MPB-RLZ@MM NPs reduced ROS levels in human umbilical vein endothelial cells (HUVECs) and apoptosis in vascular smooth muscle cells (VSMCs). MPB-RLZ@MM NPs inhibited M1-like macrophage polarization via the Nrf2/NF-κB pathway. In addition, MPB-RLZ@MM NPs accumulated in dilated aneurysms. After 4 weeks of treatment, MPB-RLZ@MM NPs effectively delayed aneurysm dilation. Conclusions : MPB-RLZ@MM NPs exhibited excellent biosafety and therapeutic efficacy against AAA. This macrophage-biomimetic strategy presents a promising therapeutic approach for AAA treatment.

Constructing ZnCoFe@NiS/NF Micro-flower Structure as Highly Efficient Electrocatalysts for Overall Water Splitting

Zhou Shilong,Li Dongbo,Zhang Guo,Xing Jinglong,Ying Chengqiang,Li Kai,Sun Yi,Hong Yan,Li Yueming

The development of efficient and stable bifunctional electrocatalysts composed of Earth-abundant elements to facilitate both the oxygen evolution reaction (OER) and hydrogen evolution reaction (HER) in water splitting still presents considerable challenges. In this work, a ZnCoFe@NiS/NF catalyst featuring a three-dimensional spherical micro-flower structure supported on nickel foam was successfully prepared through a simple, multi-step hydrothermal synthesis method. Owing to its significantly enhanced specific surface area, abundance of active sites, and superior electron conductivity, the ZnCoFe@NiS/NF catalyst demonstrates outstanding bifunctional catalytic performance in 1 M KOH. At a current density of 10 mA cm − 2 , the overpotential is as low as 170 mV for the OER and 178 mV for the HER. Specifically, an overall water splitting electrolyzer constructed from ZnCoFe@NiS/NF requires only a remarkably low operating voltage of 1.52 V to achieve a current density of 10 mA cm − 2 , surpassing the performance of the majority of reported bifunctional metal sulfide electrocatalysts. Furthermore, the catalyst operates at a similarly low voltage of 1.72 V and maintains stability for more than 100 h at a current density of 50 mA cm − 2 . This work not only reveals the critical importance of controlling morphology and electronic properties for enhancing the kinetics of electrocatalytic reactions, but also offers a simple and effective synthetic route for the large-scale production of high-performance and highly stable overall water splitting electrocatalysts. MOF-derived carbonized C@FeCoNiMnMo/NF high-entropy alloy catalyst for enhanced urea oxidation reaction

Xiaojing Liu,Fozia Sultana,Ting Xiao,Qiurong Wang,Meijie Shi,Xiaoqing Yan,Tongtong Li,Renhong Li

High-entropy alloy (HEA) catalysts derived from metal-organic frameworks (MOFs) pave the way for efficient electrochemical energy conversion. Herein, we report the synthesis of C@FeCoNiMnMo/NF, C@FeCoNi/NF and C@FeCoNiMn/NF catalysts via a one-step hydrothermal process coupled with subsequent calcination. Structural and morphological characterizations (XRD, XPS, SEM, and TEM) verifies the successful formation of a carbonized MOF-derived high-entropy alloy with uniform metal dispersion. Among these catalysts, C@FeCoNiMnMo/NF exhibits superior urea oxidation reaction (UOR) activity, achieving E 10  = 1.24 V and E 100  = 1.32 V (corresponding to 10 and 100 mA·cm −2 , respectively), with excellent durability over 30 h of continuous operation. The enhanced adsorption of key intermediates induced by Mo doping, as evidenced by both electrochemical analysis and DFT calculations, is identified as the primary factor responsible for the accelerated UOR kinetics. This work establishes an innovative approach for the rational design of MOF-derived HEA catalysts, which boosts the efficiency of urea-assisted hydrogen production from water.

Co9S8-Ni3S2@CoFe LDH/NF Heterostructure as an Efficient Electrocatalyst for Enhanced Oxygen Evolution Performance

Xinquan Zhao,Shisheng Li,Xinshu Zhang,Zhi Gao,Yi Du,Wanying Lei

Exploring efficient, stable, and earth-abundant electrocatalysts toward the oxygen evolution reaction (OER) is a central theme for realizing industrial application of water splitting. In this study, a 1D@2D heterostructure of Co 9 S 8 -Ni 3 S 2 @CoFe LDH/NF is constructed via hydrothermal and electrodeposition approaches, where ultrathin CoFe LDH nanosheets are interconnected and deposited onto Co 9 S 8 -Ni 3 S 2 nanorod arrays. This 1D@2D microstructure benefits from the exposure of a large number of accessible reactive sites. The strong interaction between the sulfide hybrid and LDH material could modulate the electronic structure of Co 9 S 8 -Ni 3 S 2 @CoFe LDH and generate a high concentration of high-valence metal sites. As a result, the optimal catalyst exhibits superior OER performance with a low overpotential of 189 mV at 10 mA cm −2 , a small Tafel slope of 30.22 mV dec −1 , and good stability in alkaline solution. This work opens up a possibility for obtaining efficient and cost-effective electrocatalysts for clean energy production through water splitting.
