pH-sensitive prodrug conjugated polydopamine for NIR-triggered synergistic chemo-photothermal therapy

Huaihong Zhang,Yu Sun,Rong Huang,Hui Cang,Zhaosheng Cai,Baiwang Sun

Combination of chemotherapy with photothermal therapy (PTT) demonstrate highly desirable for efficient medical treatment of tumor. At present works, camptothecin (CPT)-containing polymeric prodrug (PCPT) were fabricated by polymerization of a pH-sensitive camptothecin (CPT) prodrug monomer and MPC using reversible addition-fragmentation transfer (RAFT) strategy. The pH-sensitive polymeric prodrug was tethered onto surface of polydopamine (PDA) nanoparticles by amidation chemistry for combination of chemotherapy with photothermal therapy. Specifically, the active CPT quickly released from the multifunctional nanoparticles in acidic microenvironment ascribe to the cleavage of bifunctional silyl ether linkage. Meanwhile, the PDA could convert the near infrared (NIR) light energy into heat with high efficiency, which makes the resulted nanoparticles an effective platform for photothermal therapy. In vitro analysis confirmed that the PDA@PCPT nanoparticles could be efficiently uptaked by HeLa cells and deliver CPT into the nuclei of cancer cells. The cell viability assays indicated an evident in vitro cytotoxicity to HeLa cancer cells under 808 nm light irradiation. Significant tumor regression was also observed in the tumor-bearing mice model with the combinational therapy provided from the PDA@PCPT nanoparticles. The PDA@PCPT multifunctional system which was achieved by a facile route should be a potential candidate in the anti-cancer field due to the synergistic therapeutic effect, which is superior to any single approach.

NIR-II fluorescence nanoprobe based on Erbium for fallopian tube diseases diagnosis

Guangxin Duan,Zhuxin Wei,Jingyu Zhang,Xuexiao Li,Shuwang Wu,Jianfeng Zeng,Ximing Wang,Hong Zhang,Ling Wen,Mingyuan Gao

Fallopian tube blockage-related diseases are the main cause of female infertility. Diagnosis of tubal patency and accurate determination of tubal obstruction lesions is of great value for infertility treatment. Conventionally, the common used hysterosalpingogram (HSG) has the disadvantages of radioactive hazard and high false positive rate. Thus, the development of a safe and accurate method which can replace traditional HSG for tubal patency diagnosis is urgently needed. Recently, fluorescence imaging in the second near-infrared region (NIR-II) was widely used in in-vivo imaging research due to its excellent tissue penetration and signal-to noise ratio. With the aid of fluorescence agents, NIR-II imaging has the potential to diagnose tubal patency, which, unfortunately, has not been explored. Herein, a kind of rare-earth Erbium-based nanoprobe (Er-RENPs) with outstanding NIR-II fluorescence property was synthesized. Based on the Er-RENPs, the potential of NIR-II fluorescence imaging was explored. Results show that the outline of fallopian tubes can be clearly visualized with the help of Er-RENPs. And the stenotic, occlusive fallopian tubes and hydrosalpinx can be diagnosed. Additionally, the Er-RENPs exhibit good biocompatibility. In this study, we reported that NIR-II rare-earth fluorescent nanoprobe can serve as a promising fluorescent agent, and exhibits considerable potential application for tubal diagnosis.

Injectable Nanomedicine–Hydrogel for NIR Light Photothermal–Chemo Combination Therapy of Tumor

Dashan Qi,Haowei Zhu,Yingjie Kong,Qingming Shen

Traditional hydrogels have drawbacks such as surgical implantation, large wound surfaces, and uncontrollable drug release during tumor treatment. In this paper, targeted nanomedicine has been combined with injectable hydrogel for photothermal–chemotherapy combination therapy. First, targeted nanomedicine (ICG—MTX) was fabricated by combining near-infrared (NIR) photothermal reagents (ICG) and chemotherapy drugs (MTX). The ICG—MTX was then mixed with the hydrogel precursor and radical initiator to obtain an injectable hydrogel precursor solution. Under the irradiation of NIR light, the precursor solution could release alkyl radicals, which promote the transition of the precursor solution from a liquid to a colloidal state. As a result, the nanomedicine could effectively remain at the site of the tumor and continue to be released from the hydrogel. Due to the targeted nature of MTX, the released ICG—MTX could target tumor cells and improve the accuracy of photothermal–chemo combination therapy. The results indicated that the injectable nanomedicine–hydrogel system has a favorable therapeutic effect on tumors. Keywords:injectable hydrogel;nanomedicine;near-infrared;photothermal therapy;chemotherapy

pH and NIR responsive polydopamine-doped dendritic silica carriers for pesticide delivery

Menghui Wan,Yanbao Zhao,Hui Li,Xueyan Zou,Lei Sun

The stimuli-responsive pesticide delivery system provides a powerful strategy for enhancing the effective utilization of pesticides and reducing environmental pollution. Here, we prepared a new polydopamine doped dendritic silica (SiO 2 /PDA) nanocarriers for pesticide delivery. The SiO 2 /PDA nanocarriers present uniform spheres with an average diameter of 250 nm and carry center-radial inner pores. After loading dinotefuran (DNF) insecticide, polyethyleneimine (PEI) was used to cap the loaded-pesticide pores. The resulting SiO 2 /PDA@PEI displays high photothermal conversion effect ( η  = 35.1 %) and pH and near infrared (NIR) light-responsive release behavior. Meanwhile, the SiO 2 /PDA and SiO 2 /PDA@PEI carriers displayed high adhesion and wettability to leaves, and the photostability of DNF encapsulated in SiO 2 /PDA@PEI was improved by nearly 10 times greater than for free DNF. Importantly, the SiO 2 /PDA carriers possessed a benign biocompatibility on Escherichia coli ( E. coli ), seed and cells. Therefore, this work provides a promising approach to improve the utilization of pesticide.

A glutathione-activated NIR-II fluorescent probe for precise localization of micrometastases

Yao Huang,Fengyu Tian,Leilei Sun,Chenhui Ji,Craig A. Grimes,Qingyun Cai

Accurate detection and localization of micrometastases are extremely important for tumor diagnosis and comprehensive treatment. However, it is clinically difficult to finely localize micrometastases, especially focal lesions smaller than 1 mm. To that purpose, this work designed a tumor microenvironment factor (glutathione, GSH)-activated second near-infrared transparency window (NIR-II) fluorescent probe for visualization of micrometastases. It is constructed by complexing copper ions with 3-mercaptopropionic acid-modified silver sulfide (Ag 2 S-MPA) quantum dots . The experimental results showed that the probe has good and stable GSH activation properties and can achieve tumor-specific NIR-II fluorescence imaging. Further, in vivo imaging experiments demonstrated that the probe can precisely localize melanomas with a diameter of < 0.1 mm in lung micrometastases. This work provides a potential strategy for the development of novel tools for localization and visualization of micrometastases.

CRISPR/Cas9 delivery by NIR-responsive biomimetic nanoparticles for targeted HBV therapy

Wang Dan,Chen Ling,Li Chengbi,Long Quanxin,Yang Qing,Huang Ailong,Tang Hua

Background Currently, there are no curative drugs for hepatitis B virus (HBV). Complete elimination of HBV covalently closed circular DNA (cccDNA) is key to the complete cure of hepatitis B virus infection. The CRISPR/Cas9 system can directly destroy HBV cccDNA. However, a CRISPR/Cas9 delivery system with low immunogenicity and high efficiency has not yet been established. Moreover, effective implementation of precise remote spatiotemporal operations in CRISPR/Cas9 is a major limitation. Results In this work, we designed NIR-responsive biomimetic nanoparticles (UCNPs-Cas9@CM), which could effectively deliver Cas9 RNP to achieve effective genome editing for HBV therapy. HBsAg, HBeAg, HBV pgRNA and HBV DNA along with cccDNA in HBV-infected cells were found to be inhibited. These findings were confirmed in HBV-Tg mice, which did not exhibit significant cytotoxicity and minimal off-target DNA damage. Conclusion sThe UCNPs-based biomimetic nanoplatforms achieved the inhibition of HBV replication via CRISPR therapy and it is a potential system for efficient treatment of human HBV diseases.Graphical Abstract

Enhancing NIR-II luminescence of erbium sublattice through lanthanide-mediated energy modulation

Zhiyong Hu,Jinshu Huang,Long Yan,Bo Zhou

Lanthanide-doped near-infrared-II (NIR-II) luminescent nanoparticles have shown great potential in many fields. However, most previous studies were focused on the sensitizer-activator coupled systems in which the NIR-II luminescence intensity is seriously restricted by the low dopants concentration . Improving the NIR-II emission intensity in lanthanide-doped nanoparticles has remained a daunting challenge. Herein, we report a systematic research on the enhancement of the NIR-II luminescence of Er 3+ through lanthanide-mediated energy modulation. It is found that introducing proper content of Yb 3+ , Ce 3+ , Tm 3+ , and Ho 3+ in the erbium sublattice can indeed enhance the NIR-II emission under 980 and 808 nm excitations by mediating the energy transfer processes. Doping of other lanthanide ions (e.g., Pr 3+ , Tb 3+ , Dy 3+ , and Sm 3+ ) only led to a quick decline in luminescence intensity. The NIR-II emission was further enhanced by designing a core-shell-shell nanostructure which consists of an NaYbF 4 interlayer to maximize the absorption of 980 nm excitation. The NIR-II luminescence exhibits superior penetration depth in biological tissue, showing great promise in biological applications .

Y. Chen,Y. Wang,Z. Chen,J. Cai,K. Li,H. Huang,F. Song,M. Gao,Y. Yang,L. Zheng,J. Zhao

The excessive reactive oxygen species (ROS) is always associated with the degradation of cartilage matrix during osteoarthritis (OA). The development of novel nanoenzymes with excellent ROS scavenging ability and biosafety shows great potentials for improving OA treatment. Herein a novel functional polydopamine (P) loading with ultrasmall Pt and Cu nanoparticles ( [email protected] @Cu) was developed to act as nanoenzymes to scavenge ROS for OA treatment, wherein the excellent catalytic activities of Pt and Cu were employed. Results showed that [email protected] @Cu simultaneously presented obvious enzyme-mimicking properties like superoxide dismutase , catalase , and glutathione peroxidase , leading to excellent antioxidant and anti-inflammatory activities in H 2 O 2 stimulated chondrocytes and OA joints with low cytotoxicity, leading to significant alleviation of OA progression. Besides, near infrared stimulation strengthened the antioxidant and anti-inflammation ability of [email protected] @Cu, resulting in the most prominent therapeutic effects. The proposed polydopamine-based nanoenzymes inspired a novel therapeutic strategy for OA therapy in clinic.

Sulourea-coordinated Pd nanocubes for NIR-responsive photothermal/H2S therapy of cancer

Guo Xiaoyang,Liu Jia,Jiang Lingdong,Gong Wanjun,Wu Huixia,He Qianjun

Background Photothermal therapy (PTT) frequently cause thermal resistance in tumor cells by inducing the heat shock response, limiting its therapeutic effect. Hydrogen sulfide (H2S) with appropriate concentration can reverse the Warburg effect in cancer cells. The combination of PTT with H2S gas therapy is expected to achieve synergistic tumor treatment. Methods Here, sulourea (Su) is developed as a thermosensitive/hydrolysable H2S donor to be loaded into Pd nanocubes through in-depth coordination for construction of the Pd-Su nanomedicine for the first time to achieve photo-controlled H2S release, realizing the effective combination of photothermal therapy and H2S gas therapy. Results The Pd-Su nanomedicine shows a high Su loading capacity (85 mg g−1), a high near-infrared (NIR) photothermal conversion efficiency (69.4%), and NIR-controlled H2S release by the photothermal-triggered hydrolysis of Su. The combination of photothermal heating and H2S produces a strong synergetic effect by H2S-induced inhibition of heat shock response, thereby effectively inhibiting tumor growth. Moreover, high intratumoral accumulation of the Pd-Su nanomedicine after intravenous injection also enables photothermal/photoacoustic dual-mode imaging-guided tumor treatment. Conclusion sThe proposed NIR-responsive heat/H2S release strategy provides a new approach for effective cancer therapy.Graphic abstract

Intravital NIR-II three-dimensional photoacoustic imaging of biomineralized copper sulfide nanoprobes

Hui-Chao Zhou,Jie Ren,Yuejun Lin,Duyang Gao,Dehong Hu,Tinghui Yin,Chen Qiu,Xiaoyan Miao,Chengbo Liu,Xin Liu,Hairong Zheng,Rongqin Zheng,Zonghai Sheng

Photoacoustic (PA) imaging with functional nanoprobes in the second near-infrared region (NIR-II, 1000–1700 nm) has aroused much interest due to its deep tissue penetration and high maximum laser permissible exposure. However, most NIR-II PA imaging is performed using the two-dimensional (2D) imaging modality, which impedes the comprehension of the in vivo biodistribution, angiography and molecular-targeted performance of NIR-II nanoprobes (NPs). Herein, we report the systematic monitoring of biomineralized copper sulfide (CuS) NPs, typical NIR-II NPs, in mouse models by employing NIR-II three-dimensional (3D) PA imaging. The advanced imaging modality provides dynamic information about the 3D biodistribution and metabolic pathway of CuS NPs. We also achieved contrast-enhanced 3D PA imaging of abdominal and cerebral vessels at a high signal-to-background ratio. Moreover, the tumor-targeted CuS NPs conjugated with the bombesin peptide endowed NIR-II 3D PA with superior performance in imaging orthotopic tumors both deep in the prostate and in the brain beneath the intact scalp and skull. Our results highlight the potential of NIR-II 3D PA imaging for the evaluation of the in vivo behavior of NPs, thus providing a promising strategy for screening NPs in clinical translational studies.

NIR Photosensitizer for Two-Photon Fluorescent Imaging and Photodynamic Therapy of Tumor

Chen Lujia,Chen Meijuan,Zhou Yuping,Ye Changsheng,Liu Ruiyuan

Preparation of near-infrared (NIR) emissive fluorophore for imaging-guided PDT (photodynamic therapy) has attracted enormous attention. Hence, NIR photosensitizers of two-photon (TP) fluorescent imaging and photodynamic therapy are highly desirable. In this contribution, a novel D-π-A structured NIR photosensitizer (TTRE) is synthesized. TTRE demonstrates near-infrared (NIR) emission, good biocompatibility, and superior photostability, which can act as TP fluorescent agent for clear visualization of cells and vascular in tissue with deep-tissue penetration. The PDT efficacy of TTRE as photosensitizer is exploited in vitro and in vivo. All these results confirm that TTRE would serve as potential platform for TP fluorescence imaging and imaging-guided photodynamic therapy.

NIR Light-Driven Photocatalysis on Amphiphilic TiO2 Nanotubes for Controllable Drug Release

Junjian Zhao,Jingwen Xu,Xiaoxia Jian,Jing Xu,Zhida Gao,Yan-Yan Song

Titanium dioxide (TiO2) nanomaterials have attracted much interest in life science and biological fields because of their excellent photocatalytic activity and good biocompatibility. However, owing to its wide band gap, photocatalysis of TiO2 can be only triggered by UV light. The limited transparent depth of UV light and the generated reactive oxygen species (ROSs) cause inflammation response of skin tissue, thus posing two major challenges in the photocatalytic application of TiO2-based materials in drug delivery and other biotechnology fields. Here, we propose an upconversion-related strategy to enable the photocatalytic activity of TiO2 nanotubes in near-infrared light and apply the system as a controllable drug delivery platform. More importantly, the ROS-induced cytotoxicity and the preleaching of payloads are significantly reduced on the as-proposed amphiphilic TiO2 nanotubes. The hydrophobic monolayers are served as a “cap” to provide protection for ROS-induced inflammation and long-term storability. This amphiphilic drug delivery system broadens the potential applications of TiO2-based nanomaterials in biomedicine.
