Compared to the first near-infrared (NIR-I) window (650–1000 nm), the second near-infrared (NIR-II) window (1000–1700 nm) is more valuable for photothermal treatment (PTT) due to its deeper tissue penetration and lower phototoxicity. However, the therapeutic efficacy of PTT remains suboptimal owing to the inherently low photothermal conversion efficiency (PCE) of existing materials. Therefore, the development of nanomaterials with high PCE is a promising strategy for improving anti-tumor therapy. In this study, hollow semimetal PtTe 2 nanorods were synthesized. Their unique electronic properties and hollow structure contribute to achieving high PCE under NIR-II irradiation, while simultaneously promoting reactive oxygen species (ROS) generation. Additionally, PtTe 2 catalyzed the endogenous conversion of hydrogen peroxide (H 2 O 2 ) into oxygen (O 2 ) and hydroxyl radicals (•OH), further enhancing anti-tumor effects via chemodynamic therapy (CDT). In vivo experiments confirmed the excellent biocompatibility of PtTe 2 , and laser treatment following PtTe 2 administration significantly inhibited tumor growth, demonstrating its favorable therapeutic efficacy. This study establishes a new nanoplatform for the treatment of malignant tumors through photothermal/photodynamic/chemodynamic therapy, providing valuable insights for future research on multifunctional cancer therapies.

Silver sulfide (Ag 2 S) quantum dots (QDs), which emit light in the near-infrared II window (NIR-II, 1000–1700 nm), exhibit unique and remarkable potential in bioluminescence. However, their widespread application in experimental research is currently limited by a relatively low quantum efficiency. Cation exchange reactions have emerged as a remarkably versatile approach for synthesizing nanomaterials with adjustable structures and customizable compositions. In this study, we developed a NIR-II fluorescent probe based on Sn-doped silver sulfide (Sn-Ag 2 S) QDs via a straightforward cation exchange method. The stoichiometric ratios of precursors play a crucial role in tailoring the optical properties of the resultant materials during structural modifications involving cation exchange processes. By optimizing the ratio of Ag and Sn, we obtained ca. 16-fold quantum yields enhancement of Sn-Ag 2 S QDs in the NIR-II range, specifically around strong emission peak at ca. 1200 nm. Following hydrophilic modification, the Sn-Ag 2 S-GSH/MPA QDs exhibited high NIR-II fluorescence signal signal-to-noise ratio (S/N) with a ca. 1.7-fold increase compared to Ag 2 S-GSH/MPA QDs in the femur of mice, showing significant potential for bone imaging. To further investigate the QDs biodistribution mechanisms within osseous microenvironments, we devised a comprehensive multimodal analytical approach incorporating: (1) optical spectral analysis, (2) ultrastructural validation through transmission electron microscopy (TEM), and (3) spatial resolution mapping via laser scanning confocal microscopy (LSCM). These integrated methodologies consistently demonstrated preferential QDs accumulation within trabecular bone marrow compartments, revealing distinct biodistribution patterns in mineralized tissues.

Blue-light-excitable near-infrared (NIR) emitting materials exhibit significant potential for compact light sources in bioimaging, nondestructive detection, and night vision. However, achieving efficient NIR-II/III (1000–1800 nm) emission under blue excitation remains challenging due to the large excitation–emission Stokes shift and the forbidden nature of f – f or d – d transitions. In this study, we report on the Ni 2 + -doped monoclinic double perovskite Ca 2 ScTaO 6 , which exhibits broadband NIR-II/III with a full width at half maximum of 240 nm and a photoluminescence quantum yield of approximately 21% when excited at 425 nm. The incorporation of Fe 3+ ions significantly improves the photoluminescence quantum yield to approximately 37% through energy transfer pathways from Fe 3+ to Ni 2+ and crystal field modulation. Structural and spectroscopic investigations reveal the presence of Fe 3+ ions at various lattice sites, resulting in localized distortions that facilitate the relaxation of d – d transitions in Ni 2+ . A prototype NIR light-emitting diode utilizing Ca 2 ScTaO 6 :Fe 3+ , Ni 2+ exhibits strong NIR-II/III emission, facilitating the visualization of subsurface damage in various materials, including fruits, capsules, meat, and silicon wafers. This work demonstrates an effective strategy for designing blue-light-excitable NIR materials through synergistic energy transfer and crystal field engineering.

Lanthanide NIR-II luminescence free from thermal quenching is attractive for various applications at elevated temperatures. Here, we present a thermal enhancement of Er 3+ NIR-II luminescence based on the synergy of phonon assisted Er 3+ -Er 3+ cross relaxations and controllable Er 3+ -Ho 3+ energy transfer. The lattice expansion of NaYGeO 4 olivine phosphor upon heating (298–823 K) suppresses the Er 3+ -Ho 3+ energy transfer and leads up to 46.1 times enhancement of Er 3+ NIR-II luminescence under 379 nm excitation. The NIR-II luminescence evolution as increasing temperature allows dual-mode ratiometric thermometry, i.e., luminescence intensity ratios (LIRs) based on Er 3+ 4 I 13/2 Stark sublevels and on Er 3+ 4 I 13/2 and Ho 3+ 5 I 6 levels. Obtained relative sensitivities above 0.46% K ‒1 over a broad temperature range of 298–673 K with a maximum of 2.32% K -1 suggest NaYGeO 4 :Er 3+ /Ho 3+ phosphor as a promising NIR-II luminescence thermometer. Moreover, this research offers an available perspective for designing thermal-enhanced NIR luminescence in ordinary thermal expansion materials.

Tumor hypoxia limits the efficacy of photodynamic therapy (PDT), necessitating photosensitizers with hypoxia-adaptive therapy and imaging. Here, we present a fluorinated N -oxide aza-BODIPY ( OFBD ) nanoemulsion ( OFBD-NP ) for hypoxia-responsive, multimodal imaging-guided phototherapy. OFBD generates robust singlet oxygen under normoxia for PDT, but is reduced by CYP450 enzymes in hypoxic cells to photothermal-potent FBD , enabling switchable PDT/PTT. Co-assembly with fluorinated oil enhances oxygen delivery, boosts 19 F MRI sensitivity, and promotes J -aggregation, shifting fluorescence into the NIR-II window for deep-tissue imaging. The redox conversion also activates photoacoustic signals, enabling responsive tri-modal imaging (NIR-II FLI/ 19 F MRI/PAI). OFBD-NP shows potent cytotoxicity in vitro under both normoxic and hypoxic conditions via apoptosis. In vivo, it selectively accumulates in tumors, offers high-contrast imaging, and leads to complete tumor regression in subcutaneous A549 models after laser irradiation, without systemic toxicity. This work demonstrates a smart nanoplatform that integrates deep-tissue imaging and hypoxia-triggered therapeutic switching, addressing major limitations of conventional photosensitizers in cancer imaging and phototherapy.

Therapy based on heat and reactive oxygen species (ROS) generation have attracted increasing attention, while the weak photothermal ability and low ROS production have severely limited the efficiency of the tumor treatment. Here, 2D ultrathin CuS-phosphomolybdic acid (PMA) nanosheets are synthesized at sub-nanoscale. Typically, the improved second near-infrared light (NIR-II) light absorption, small bandgap, fast hot carrier response, abundant oxygen vacancies, and the electron delocalization between closely connected PMA and CuS units in sub-nanoscale result in the notably enhanced NIR-II photothermal therapy (PTT) performance of CuS-PMA. The reversible redox of Mo active sites in PMA provides basis for glutathione (GSH) depletion while triggering chemodynamic therapy (CDT) in the tumor microenvironment (TME), and the heat produced by the PTT greatly promotes the CDT and GSH depletion, where CuS-PMA presents sevenfold more ROS generation and fourfold more GSH depletion under PTT. In addition, the ROS generation/GSH depletion can downregulate heat shock proteins, weakening the heat resistance of tumor cells and thereby amplifying the killing effect of PTT. Moreover, the combination of antiprogrammed death-ligand 1 (αPD-L1) with the treatment of CuS-PMA promote the immunotherapy, resulting in the suppression of untreated abscopal tumors.

Luminescent metal nanoclusters featuring near-infrared (NIR) emission hold promise for diverse applications. Although ligand engineering can modulate their NIR optical properties, separating the effects of single-ligand in dual-ligand systems remains challenging. In this study, we employed a controlled variable approach to prepare three structurally comparable silver-rich clusters-Ag11Cu3, Cu@Ag14-xCux, and Ag18. By maintaining a constant thiolate ligand and solely varying the phosphines, we elucidated the synergistic regulatory impacts of the ligands on the core configuration and excited-state dynamics. Structural and optical analyses reveal that the monodentate phosphine (TPP) stabilizes an Ag6@Ag5Cu3 core of the cluster Ag11Cu3, leading to NIR-I emission (720 nm) originating from mixed triplet state metal-centered and charge transfer transitions. In contrast, the bidentate phosphine (trans-dppe) facilitates the formation of a nested Ag6@Ag8@Ag4 core of the cluster Ag18, triggering NIR-II emission (1424 nm) associated with highly delocalized core-based excited states. Moreover, the spatial influence of the chiral bidentate phosphine ((S,S)-BDPP) can induce core-structural distortion, which promotes nonradiative decay and results in luminescence quenching of the cluster Cu@Ag14-xCux. This work clarifies the relation between the structure and luminescence properties of ligand-mediated metal clusters, offering valuable insights for the design of tailored NIR optical materials.

Bioadhesives are being increasingly used in clinical practice, but pose significant health risks due to their uncontrollable intraoperative leakage. Nevertheless, there is currently a critical lack of effective solutions. Herein, we propose a second near-infrared window (NIR-II) fluorescence-visible indocyanine green (ICG)-BioGlue adhesive for precision surgical adhesion with intraoperative leakage prevention. The fabricated ICG-BioGlue adhesive based on the non-covalent interaction exhibits nearly 100% fluorescence labeling efficiency and maintains long-term stable NIR-II fluorescence for up to 3 months. During surgery, ICG-BioGlue adhesive enables real-time visualization of wound adhesion in various fields such as aortic dissection adhesion and liver or kidney wound hemostasis, preventing leakage into surrounding tissues. Moreover, the stable fluorescence of ICG-BioGlue adhesive allows effective imaging-guided removal of long-term adhesive fragments in the body when they cause compression symptoms after surgery. Considering it consists solely of Food and Drug Administration-approved drugs ICG and BioGlue, with a straightforward synthesis method and versatile applicability, ICG-BioGlue adhesive holds excellent potential for clinical translation. Our study provides a leakage-preventing strategy to enhance biosafety and promote the widespread application of bioadhesives in clinical settings.

The emission peak of Mn 5+ lies in the second near-infrared window (NIR-II) biological transparency window, and it exhibits outstanding application prospects in deep-tissue bioimaging, non-contact optical thermometry, and related fields. In this work, a novel Mn 5+ -activated Ba 1.9 Sr 0.1 TiO 4 NIR-II phosphor is designed and synthesized via a high-temperature solid-state reaction, where partial Sr 2+ substitution for Ba 2+ enables precise crystal field modulation around Mn 5+ ions. Mn K-edge X-ray absorption near-edge structure (XANES) spectroscopy confirms that Mn ions are stabilized in the +5 oxidation state. The optimized BSTO: 0.1 % Mn 5+ phosphor shows efficient 680 nm red light excitation and a characteristic narrowband NIR-II emission at 1190 nm from the 1 E → 3 A 2 transition of Mn 5+ , with a long luminescence lifetime of 184.9 μs and excellent thermal stability. Density functional theory with Hubbard U correction (DFT + U) calculations further reveal that Sr 2+ substitution induces controllable lattice contraction and local structural distortion, which modulates the host electronic structure and enhances the stability of Mn 5+ luminescent centers. Furthermore, the phosphor delivers a maximum relative temperature sensitivity of 2.24 % K −1 via the luminescence intensity ratio technique. A proof-of-concept NIR phosphor-converted light-emitting diode fabricated with the as-prepared phosphor realizes clear NIR penetration imaging.

Real-time monitoring of intestinal movement is critical for the clinical diagnosis of bowel diseases, however, the application of organic fluorophores in non-invasive imaging of the intestinal tract via systemic administration is still challenging. We herein report a series of albumin-chaperoned chloro-cyanine dyes for high spatiotemporal tracking of intestinal peristalsis and bowel disease in vivo. By adjusting the size of the central rings, Cy7–5 , with a five-membered ring and meso-chlorine, could covalently bind with circulating endogenous albumin in the blood, generating a bright and stable cyanine-albumin fluorescent complex with ∼1055 nm fluorescence in the near-infrared-II window in situ. Notably, Cy7–5 was able to clearly view intestinal peristalsis by harnessing the inherent intestine tropism of Cy7–5 -albumin complex following intravenous (i.v.) administration. Furthermore, a hydroxyl radical (·OH)-activatable fluorophore ( Hydro-Cy7–5 ) was created, allowing for the selective detection of overproduced ·OH in bowel disease as well as the successful visualization of the inflammatory intestinal tract in vivo after i.v. injection. This work provides a facile strategy for the investigation of intestinal movement and related diseases.

Immune checkpoint blockade therapy has revolutionized clinical oncotherapy by intercepting immunosuppressive pathways in tumor tissues. Nevertheless, the therapeutic efficacy is limited by a multidimensional immunosuppressive tumor microenvironment, which impedes the activity of tumor-infiltrating immune cells and undermines treatment outcomes. In this study, a zwitterionic polymer is designed for the conjugation and traceless release of a checkpoint inhibition antibody (Atezolizumab), while concurrently enabling the restoration of membrane redox homeostasis on T cell membranes within the oxidative tumor microenvironment. Contrast-enhanced fluorescence imaging in the near-infrared II spectral region with a deep learning network is employed to visualize in vivo dynamics of the antibody conjugates. The antibody conjugates demonstrate enhanced and accelerated tumor accumulation, thereby promoting T cell proliferation and augmenting the antitumor efficacy. Mechanistic investigations reveal that the conjugate-mediated regulation of the T cell surface redox state protects against damage induced by extracellular reactive oxygen species, subsequently mitigating ferroptosis of T cells. In murine models, the antibody conjugates effectively suppressed colorectal tumor growth and recurrence, leading to a significant extension of animal survival. This work thus realizes reversing suppressive immune microenvironment from multiple dimensions with a simple checkpoint inhibitor conjugate, promising for clinical translation and next-generation immune checkpoint blockade therapy.

Combined photothermal/photodynamic therapy is a promising strategy to achieve an enhanced anticancer effect. However, hypoxia is one of the representative characteristics of the microenvironment of solid tumors, which not only attenuates the therapeutic effects but also promotes tumor invasion and metastasis. Herein, a PtBi-β-CD-Ce6 nanoplatform for the generation of sustained O2 was constructed for more effective tumor therapy. In detail, the catalase (CAT)-like nanozyme, PtBi, which could decompose H2O2 to produce O2, was modified with β-cyclodextrin (β-CD). O2 would be converted into 1O2 by PtBi-β-CD-Ce6 for enhanced photodynamic therapy (PDT) under 650 nm laser irradiation. In addition, by reason of excellent absorption in the near-infrared-II (NIR-II) region, PtBi-β-CD-Ce6 was used for photoacoustic imaging (PA) and photothermal imaging (PT)-guided photothermal therapy (PTT) in the NIR-II biowindow. Furthermore, PtBi-β-CD-Ce6 could be elected to serve as a contrast agent for X-ray computed tomography (CT) imaging due to the apparent X-ray attenuation capability of the Pt and Bi elements themselves. Therefore, by integrating the advantages of overcoming the hypoxia function and photothermal effect into a single nanoplatform, PtBi-β-CD-Ce6 showed an immense possibility in multimodal imaging-guided combined PDT/PTT.
