Biodegradable scaffolds have significant potential for directed tissue regeneration in craniofacial and oral applications and bone regeneration. We designed an innovative cerium-altered/collagen-clacked polycaprolactone/poly-lactic-co-glycolic acid (PC/PL) electrospun scaffolds (PC/PL-PD-Ce-CL) to address the deficiencies of existing scaffold materials regarding osteogenic and antimicrobial characteristics. Novel scaffolds were developed by electrospinning a fundamental PCL/PLGA matrix, impregnating in situ reduction with cerium nanoparticles (CeNPs), applying a polydopamine coating, and clacking with collagen. The mechanical tests and scanning electron microscope demonstrated that the distinctive NSs preserved their tensile strength and elasticity modulus post-modification. The cell proliferation CCK-8 assay indicated that the PC/PL-PD-Ce-CL NSs exhibited a superior MG63 cell proliferation and cell adhesion ratio compared to the rest of the NSs. The alkaline phosphatase (ALP) activity and the expression levels of BMP2 and RUNX2 in MG63 cells cultivated on PC/PL-PD-Ce-CL NSs for 7 and 14 days were considerably elevated compared to the controls. The PC/PL-PD-Ce-CL and PC/PL-PD-Ce NSs exhibited a broader antibacterial zone compared to the control NSs, and bacterial fluorescence diminished on the Ce-altered NSs of incubation with S. mutans and S. aureus . Our innovative PC/PL-PD-Ce-CL NSs improved cytocompatibility, osteogenesis, and antibacterial capabilities, demonstrating their promising therapeutic applications for orofacial treatment.

The use of 3D printing technology has advanced the bone tissue engineering, and constructing large artificial bones for repairing large-scale bone defects is highly significant. This study aimed to construct large artificial bones in a precise and controllable manner, focusing on repairing critical-sized bone defects. The researchers used 3D printing technology to synthesize 3D PCL/β-TCP, and then evaluated its ability to promote MC3T3-E1 cell adhesion, proliferation, and new bone formation through a series of characterizations. The results confirmed that 3D PCL/β-TCP, presented as a lattice structure similar to natural bone, could be used to prepare personalized artificial bone blocks based on the actual range of bone defects. The researchers also experimented with dipping 3D PCL/β-TCP bone blocks in different proportions of β-TCP and found that PT2 showed better mechanical and hydrophilic properties. Further experiments showed that PT2 presented excellent biocompatibility and outstanding capacity to promote MC3T3-E1 cell proliferation and adhesion, and that 3D PCL/β-TCP bone blocks possessed good osteogenic activity. Finally, it was suggested that PT2 exhibited the property of promoting new bone formation. These findings provide experimental data support for the repair of critical-sized bone defects.

Periosteum is crucial to the initial healing of bone defects because it provides a stable periosteum-specific microenvironment and abundant osteogenic cells towards bone repair. However, the usage of autologous periosteum is restricted for limited availability. Xenogeneic decellularized periosteum (DP) preserves periosteum-specific cues but the hardly-controlled physicochemical properties render it inconvenient to use widely. Our study aimed to fabricate a tissue-engineered periosteum (TEP) with a continuous periosteum-specifically bioactive surface and tailored physicochemical properties by co-axial electrospinning of poly(ε-caprolactone) (PCL)/periosteal decellularized extracellular matrix (dECM) to promote bone defects healing. Before use, DP was confirmed for effective removal of residual DNA and well preservation of biological components, e.g., collagen and glycosaminoglycans . After optimization of fabrication processes, PCL/dECM co-axially electrospun membrane (PEC) bore an exquisite core-shell structure, which was effectively equipped with composite advantages of dECM and PCL. Notably, PEC stood out in terms of tensile strength and long-term durability within a physiological environment compared to that of the dECM uniaxial electrospun membrane (ECM). Besides, PEC exhibited remarkably better cell proliferation , migration, bio-mineralization, and osteogenic properties as compared to that of the electrospun PCL membrane. Moreover, PEC-TEP could significantly enhance the recovery of critical-sized bone defects in rats than that of PCL-TEP. To the best of our knowledge, biomimetic PEC-TEP was fabricated and used for the first time to repair bone defects. This novel strategy is promising to fabricate a dECM-based advanced membrane for tissue engineering .

In this work, well-defined pH-responsive methoxy poly(ethylene glycol)-block-poly(ε-caprolactone)-block-poly[2-(dimethylamino)ethyl methacrylate]-g-7-propinyloxy coumarin triblock amphiphilic copolymers (mPEG-b-PCL-b-PDMAEMA-g-PC) were synthesized using a combination of atom transfer radical polymerization (ATRP), ring opening polymerization (ROP) and click chemistry. The chemical structures and compositions of these copolymers were characterized using Fourier transform infrared spectroscopy (FT-IR) and proton nuclear magnetic resonance (1H NMR). The molecular weights of the copolymers were obtained using 1H NMR spectroscopy and gel permeation chromatography (GPC) measurements. Subsequently, the polymers could self-assemble into micelles which were investigated using dynamic light scattering (DLS), transmission electron microscopy (TEM), and fluorescence spectroscopy. The pH-responsive self-assembly behavior of these triblock copolymers in water were investigated at different pH values of 5 and 7.4 for controlled doxorubicin release, the results indicated that the release rate of DOX could be effectively controlled by altering the pH, DOX was sealed in neutral surroundings and DOX release was triggered in acidic surroundings. CCK-8 assays and confocal laser scanning microscopy (CLSM) against HeLa cells indicated that the micelles had no associated cytotoxicity, possessed good biodegradability and biocompatibility, and identified the location of the DOX in HeLa cells. The DOX-loaded micelles possessed high cytotoxicity to HeLa cells and exhibited inhibition of the proliferation of HeLa cells. Moreover, these flexible micelles with an on–off switched drug release may offer a promising pattern to deliver a wide variety of hydrophobic payloads to tumor cells for cancer therapy.

A well-defined thermoresponsive poly(ethylene glycol)-block-poly(N-isopropylacrylamide)-block-poly(ε-caprolactone) (PEG43-b-PNIPAM82-b-PCL87) triblock copolymer was synthesized by combination of atom transfer radical polymerization (ATRP), ring-opening polymerization (ROP), and click chemistry. The synthesis included the four steps, and all the structures of the polymers were determined. The thermoresponsive triblock copolymer can disperse in water at room temperature to form core–shell–corona micelles with the hydrophobic PCL block as core, the thermoresponsive PNIPAM block as shell, and the hydrophilic PEG block as corona. At temperatures above the lower critical solution temperature (LCST) of the PNIPAM block, the PNIPAM chains gradually collapse on the PCL core to shrink the size and change the structure of the resultant core–shell–corona micelles with temperature increasing.

In this work, we report that well-defined amphiphilic star-shaped polyhedral oligomeric silsesquioxane (POSS)-based inorganic/organic hybrid block copolymers of poly(ε-caprolactone)–poly(2-(2-methoxyethoxy)ethyl methacrylate)-co-poly(ethylene glycol) methacrylate (POSS–(PCL–P(MEO2MA-co-PEGMA))16) were synthesized via click chemistry, ring opening polymerization (ROP) and atom transfer radical polymerization (ATRP), and their synthesis was confirmed by Fourier transform infrared spectroscopy (FT-IR), nuclear magnetic resonance (1H NMR), gel permeation chromatography (GPC) and X-ray photoelectron spectroscopy (XPS). The obtained polymers could self-assemble into ellipsoidal aggregates with a moderately uniform size, owing to the amphiphilic properties of the hydrophobic inorganic POSS and PCL cores and the hydrophilic P(MEO2MA-co-PEGMA) segments. These thermo-responsive organic/inorganic hybrid polymeric micelles exhibited critical phase transition temperatures in water, which could be finely tuned by changing the feed ratios of PEGMA and MEO2MA. The lower critical solution temperature (LCST) of star-shaped POSS–(PCL–P(MEO2MA-co-PEGMA))16 increased from 34 to 57 °C with the increase of PEGMA content. The results were investigated by ultraviolet-visible (UV-vis) spectroscopy, dynamic light scattering (DLS), and transmission electron microscopy (TEM). When the temperature was higher than the corresponding LCSTs, the micelles started to associate and form spherical nanoparticles. Therefore, POSS–(PCL–P(MEO2MA-co-PEGMA))16 block copolymers, a class of novel organic/inorganic hybrid materials with advantageous properties, are amphiphilic hybrid polymers with potential applications as nano-carriers, nano-reactors, and smart materials and in the biomedical field.

Solid propellants are essential for rocket engine efficiency and safety. The introduction of energetic self-healing adhesives can improve its energy density, mechanical properties, and safety, while extending its service life. Therefore, this paper proposes a novel energetic self-healing adhesive that utilizes asymmetric alicyclic isophorone diisocyanate (IPDI) and 2-(aminophenyl) disulfide (2-AFD), which has a bent biphenyl ring structure, as hard segments, combined with semicrystalline polymer polycaprolactone (PCL) and energetic adhesive glycidyl azide polymer (GAP) as soft segments to synthesize a series of self-healing adhesives. By adjusting the hard segment content, a balance between mechanical properties and self-healing performance is achieved. Results show that increasing the hard segment content enhances toughness (from 26.60 MJ·m −3 to 58.54 MJ·m −3 ), but decreases self-healing efficiency (from 90 % to 58 %). GPPU-2 exhibits 38.95 MJ·m −3 toughness and recovers 86 % of its toughness within 90 min at 80 °C via dynamic disulfide and hydrogen bond interactions. The GPPU-2-based propellant, GPPU80, recovers 88.22 % of tensile strength after 24 h at 80 °C, demonstrating effective crack propagation inhibition. Compared to inert adhesive-based propellants, GPPU80 shows superior combustion performance and energy release. This study offers insights for designing high-performance self-healing adhesives to enhance propellant safety and energy density.

Although artificial polymeric scaffolds act as vital characters in bone repair, their application is limited due to their inferior bioactivity. Herein, osteoinductive poly(ε-caprolactone) (PCL) composite scaffolds were prepared by synchronously enlisting bioactive nanohydroxyapatite (nHA), bioglass (BG), and bone morphogenetic protein-2 (BMP-2), which was bound up with polydopamine (pDA). It was found that pDA deposition not only significantly enhanced hydrophilicity and cell affinity of composite scaffolds but also endowed steady immobilization of BMP-2 with long-term yet sustained release. Compared to pure PCL and PCL/nHA/BG (PHB) scaffolds, the designed PHB-pDA-BMP-2 scaffolds significantly induced the differentiation of bone marrow stromal cells toward an osteogenic lineage. Meanwhile, in vivo examinations revealed the prominent osteogenic capability of PHB-pDA-BMP-2 scaffolds, which enabled complete bone healing of calvarial bone defects in rabbits by 12 weeks. This finding indicates that the developed porous composite scaffolds hold great potential for bone regeneration.

A pivotal approach in engineering artificial peripheral nerve sheaths encompasses the augmentation of the regenerative microenvironment via the manipulation of Schwann cells (SCs). Our investigation employed single-cell sequencing analysis to elucidate the potential functions of Schwann cells and the Wnt pathway in facilitating peripheral nerve regeneration. In vitro studies showed that activating the Wnt signaling pathway promotes the transition to repair SCs, boosting their growth, movement, and immune functions. To better understand the peripheral nerve regeneration environment, we created a polymer scaffold using ammonization and electrospinning. The Wnt3a protein was incorporated into the polycaprolactone (PCL) electrospun fiber surface. In a rat sciatic nerve defect model, the Wnt3a-modified scaffold showed better nerve repair outcomes than traditional electrospun scaffolds. After a week, the test group showed better immune regulation and angiogenesis, with a significant increase in axon growth rate observed after 3 weeks. Three-month-long animal experiments revealed notable improvements in neuroelectrophysiology, reduced organ atrophy, and enhanced sciatic nerve recovery. In this nerve defect model, Wnt3a-modified neural scaffolds achieved repair effects.

Nitric oxide (NO) gas therapy has aroused intense interest in recent years. l-Arginine (l-Arg) reacts with reactive oxygen species (ROS) in tumor cells to generate NO. This phenomenon represents an effective method for tumor therapy. However, endogenous ROS levels in most types of tumor cells cannot enable an effective reaction. β-Lapachone is generally used to increase H2O2, which can oxidize guanidine derivatives to form nitric oxide in tumor cells. In addition, based on the ferrocene (Fc)-catalyzed Fenton reaction, ·OH is generated from H2O2, and the ONOO– could be generated from an interaction between ·O2– (generated through the Haber–Weiss reaction) and NO. Arg-rich poly(ε-caprolactone) (PCL)-b-PArg, a macromolecular NO donor, was accurately synthesized to avoid premature l-Arg leakage during in vivo transport. In this design, the self-assembled PCL-b-PArg nanoparticles were dressed with the tumor-shreddable masking (PEG-b-PDMA, a negatively charged pH-sensitive hydrophilic diblock polymer), to prepare P-lapa-Fc nanoparticles and hide penetrative capability in the circulation. The experimental results confirmed that this synergistic therapy based on ROS and NO had a significant inhibitory effect on cancer cells, thereby providing new inspiration for NO gas treatment.

Tissue engineered heart valves (TEHVs) demonstrates the potential for tissue growth and remodel, offering particular benefit for pediatric patients. A significant challenge in designing functional TEHV lies in replicating the anisotropic mechanical properties of native valve leaflets. To establish a biomimetic TEHV model, we employed melt-electrowriting (MEW) technology to fabricate an anisotropic PCL scaffold. By integrating the anisotropic MEW-PCL scaffold with bioactive hydrogels (GelMA/ChsMA), we successfully crafted an elastic scaffold with tunable mechanical properties closely mirroring the structure and mechanical characteristics of natural heart valves. This scaffold not only supports the growth of valvular interstitial cells (VICs) within a 3D culture but also fosters the remodeling of extracellular matrix of VICs. The in vitro experiments demonstrated that the introduction of ChsMA improved the hemocompatibility and endothelialization of TEHV scaffold. The in vivo experiments revealed that, compared to their non-hydrogel counterparts, the PCL-GelMA/ChsMA scaffold, when implanted into SD rats, significantly suppressed immune reactions and calcification. In comparison with the PCL scaffold, the PCL-GelMA/ChsMA scaffold exhibited higher bioactivity and superior biocompatibility. The amalgamation of MEW technology and biomimetic design approaches provides a new paradigm for manufacturing scaffolds with highly controllable microstructures, biocompatibility, and anisotropic mechanical properties required for the fabrication of TEHVs.
