Dipicolinic acid reduces Epicoccum sorghinum symptoms on maize and inhibits tenuazonic acid biosynthesis

Xin-Yu Ji,Bing-Yi Wang,Yi-Feng Zhang,Yu-Jing Zhang,Ya-Jie Lai,Yang Yang,Xiang-Chen Wang,Su-Yan Wang,Pedro Laborda,Xin-Chi Shi

BACKGROUND Epicoccum sorghinum is a pathogenic fungus that causes leaf spot in a wide range of plants, including maize, and synthesizes the mycotoxin tenuazonic acid (TEA), which is carcinogenic. Despite the relevant economic and yield losses caused by E. sorghinum worldwide, methods for the control of this pathogen are lacking. RESULTS In this work, the efficacy of Bacillus -produced dipicolinic acid (DPA) for control of E. sorghinum was evaluated using in vitro and in vivo assays, and compared with the efficacy of three commercial fungicides, including carbendazim, prochloraz, and thiram. DPA inhibited E. sorghinum mycelial growth, and conidia germination, and produced important alterations in E. sorghinum hyphae. Interestingly, 10 mM DPA reduced TEA biosynthesis by 86.6%. Although DPA rapidly degraded on maize leaves, 10 mM DPA showed higher preventive (67.4% lesion length inhibition) and inhibitory (89.5% lesion length inhibition) efficacies for the control of E. sorghinum on maize leaves compared to the commercial fungicides. CONCLUSIONS Collectively, this study presents the first method for the control of E. sorghinum on maize and demonstrates that DPA application is a suitable approach to inhibit E. sorghinum symptoms in plants and TEA biosynthesis. © 2024 Society of Chemical Industry.

Preparation of a long-lasting tablet of spinosad microspheres and its residual insecticidal efficacy against the yellow fever mosquito, Aedes aegypti (Diptera: Culicidae) larvae

Hongxiao Yu,Yunqi Li,Hong Wang,Lei Zhang,Penghui Suo,Tianyun Su,Qian Han

BACKGROUNDS In order to provide a long-lasting formulation for spinosad (SP) targeting larval stages of Aedes aegypti (Linnaeus) and others alike, a SP tablet was developed based on microspheres, using polylactic acid as inside coating material. The microspheres were encapsulated using polyethylene glycol and 1-hexadecanol to form a sustained-release SP tablet. Micromorphology, active ingredient loading, structure identification, photolysis resistance and biological activity were evaluated in this report. RESULTS (i) The SP microspheres had an average particle size of 6.16 ± 2.28 μm, low adhesion and good dispersion as evaluated by scanning electron microscopy and morphology. (ii) The average active ingredient loading and encapsulation of SP microspheres were 32.80 ± 0.74% and 78.41 ± 2.22%, respectively. (iii) The chemical structure of encapsulated SP was confirmed by Fourier transform infrared and 1 H-nuclear magnetic resonance. (iv) The photostability of the microspheres and the tablets were evaluated. The results showed that DT 50 (time required to dissipate 50% of the mass originally present) of SP was 0.95 days in microspheres and 6.94 days in tablets. (v) The long-term insecticidal activity of SP tablets was investigated, and the tablet had a long-lasting activity against the mosquito larvae, showing 100% larval mortality for 63 days. CONCLUSIONS The study provided a new long-lasting formulation of SP, which displayed good efficacy in the control of Ae. aegypti larvae. © 2024 Society of Chemical Industry.

Point mutations in the voltage-gated sodium channel gene conferring pyrethroid resistance in China populations of the Dermanyssus gallinae

Penglong Wang,Qi Liu,Xu Wang,Tiancong Sun,Boxing Liu,Bohan Wang,Huan Li,Chuanwen Wang,Weiwei Sun,Baoliang Pan

BACKGROUND Dermanyssus gallinae , the poultry red mite (PRM), is a worldwide ectoparasite posing significant economic challenges in poultry farming. The extensive use of pyrethroids for PRM control has led to the emergence of pyrethroid resistance. The objective of this study is to detect the pyrethroid resistance and explore its associated point mutations in the voltage-gated sodium channel (VGSC) gene among PRM populations in China. RESULTS Several populations of D. gallinae , namely CJF-1, CJP-2, CJP-3, CSD-4 and CLD-5, displayed varying degrees of resistance to beta-cypermethrin compared to a susceptible field population (CBP-5). Mutations of VGSC gene in populations of PRMs associated with pyrethroid resistance were identified through sequencing its fragments IIS4–IIS5 and IIIS6. The mutations I917V, M918T/L, A924G and L925V were present in multiple populations, while no mutations were found at positions T929, I936, F1534 and F1538. CONCLUSION The present study confirmed the presence of extremely high levels of pyrethroid resistance in PRM populations in China, and for the first time detected four pyrethroid resistance mutations in the VGSC gene. Identifying pyrethroid resistance in the field population of PRM in China can be achieved through screening for VGSC gene mutations as an early detection method. Our findings underscore the importance of implementing chemical PRM control strategies based on resistance evidence, while also considering the management of acaricide resistance in the control of PRMs. © 2024 Society of Chemical Industry.

The relationship between the Huanglongbing pathogen and Diaphorina citri with body color polyphenism: field dynamics, acquisition efficiency, and immunity response

Jiayao Fan,Huimin Pan,Chenyang Yuan,Tianyuan Liu,Feng Shang,Yuxi Zhou,Yifan Gao,Long Yi,Jinjun Wang,Wei Dou

BACKGROUND Phenotypic plasticity is generally regarded as a crucial adaptation that enables organisms to survive in the face of environmental changes. Diaphorina citri , a vector of Candidatus Liberibacter asiaticus ( C Las) that causes citrus Huanglongbing, displays three distinct body color morphotypes: blue/green, gray/brown, and orange/yellow morphs. Here, dynamics monitoring of D. citri population in the field was established to identify the occurrence peak. The orange/yellow is a shared phenotype, and hence we focused on the two most prevalent color morphs blue/green and gray/brown. We evaluated the life-history characteristics and susceptibility to insecticides of the two color morphs. For the association between the body color polyphenism and C Las, we compared the C Las acquisition efficiency of field and laboratory populations, and investigated immunological competence of the two color morphs. RESULTS Diaphorina citri adults were present in all seasons in year one, with higher incidences recorded in July, August, and September. During these periods, the blue/green morph predominated in the field, while during the overwintering period, the proportion of gray/brown morph increased. Population parameters showed no significant differences between the two color morphs. Additionally, gray/brown morphs were more susceptible to insecticides and C Las acquisition. Furthermore, the two color morphs displayed different immune responses to pathogen infection. CONCLUSION The C Las infection rate was closely associated with color morphology in D. citri . Different field distribution and trait combinations of the two color morphs represent their responses to variable environments. This study not only broadens the understanding of color polyphenism, but it may also provide potential implication for controlling strategies in D. citri . © 2025 Society of Chemical Industry.

Synthesis of paeonol hydrazone derivatives and their anti-oomycete, anti-fungal, and nematicidal activities

Mei Hu,Xiaolong Guo,Yuee Tian,Yan Li,Yibo Liu,Xiaobo Huang,Genqiang Chen,Zhiping Che

BACKGROUND The natural product paeonol is a rich and sustainable natural bioresource, and its derivatives have various unique biological efficacy. As is well known, Schiff bases are a class of organic compounds with a wide range of biological activities, including anti-fungal, insecticidal, anti-viral, and nematicidal. RESULTS To discover biorational natural product-based pesticides, nine intermediates (2–10), 12 sulfonylhydrazones (11a–11c, 12a–12c, 13a–13c, and 14a–14c) and 20 benzylidene hydrazones (18a–18r, 19a, and 20a) were synthesized by structural modification of paeonol, and their structures were characterized by proton nuclear magnetic resonance ( 1 H NMR), carbon-13 ( 13 C) NMR, and high-resolution mass spectrometry (HRMS). The stereochemical configurations of compounds 14a, 18d, and 18r were unambiguously confirmed by single-crystal X-ray diffraction. Furthermore, bioactivities of these compounds as anti-oomycete, anti-fungal, and nematicidal agents against three serious agricultural pests, Phytophthora capsici , Fusarium graminearum , and Heterodera glycines were evaluated. Among all tested compounds: (i) compound 7 exhibited promising anti-oomycete against Phytophthora capsici , with a half maximal effective concentration (EC 50 ) value of 15.81 mg L −1 ; (ii) compounds 2, 7, 10, and 19a displayed promising anti-fungal against F. graminearum , with EC 50 values of 12.22, 14.72, 23.39, and 33.10 mg L −1 , respectively; (iii) ten compounds (12a–12c, 14c, 18g–18j, 18m, and 19a) showed significant nematicidal activity against H. glycines , with median lethal concentration (LC 50 ) values all less than 30.00 mg L −1 . Especially for compound 18g, its LC 50 value is the smallest, at 12.65 mg L −1 . CONCLUSION The research results indicate that introducing nitro groups at the C5 position of paeonol, or introducing halogens at both C5 and C3 positions, can significantly enhance its biological activity against Phytophthora capsici , F. graminearum , and H. glycines . © 2024 Society of Chemical Industry.

Photothermal controlled-release microcapsule pesticide delivery systems constructed with sodium lignosulfonate and transition metal ions: construction, efficacy and on-demand pesticide delivery

Lingling Huang,Guangqi Jiang

BACKGROUND Widespread application of controlled-release pesticide delivery systems is a feasible and effective method to improve the utilization efficiency of pesticides. However, owing to the high cost and complicated preparation technologies of controlled-release pesticide delivery systems, their applications in agricultural production have been seriously hindered. RESULTS This study aimed to construct inexpensive photothermally controlled-release pesticide delivery systems using chitosan (CS) and sodium lignosulfonate (LS) as the wall materials, and a coordination assembly strategy of LS with transition metal ions to encapsulate a model pesticide, avermectin (AVM). The resulting complex or nanoparticle photothermal layers in these systems effectively achieved photothermal conversions, and replaced the use of common photothermal agents. In the prepared pesticide-delivery systems, two systems had remarkable photothermal conversion performance and photothermal stabilities with a photothermal conversion efficiency ( η ) of 24.03% and 28.82%, respectively, under 808 nm, 2 W near-infrared irradiation. The slow-release and ultraviolet-shielding performance of these two systems were markedly enhanced compared with other formulations. The insecticidal activities of these two systems against Plutella xylostella under irradiation with light-emitting diode (LED)-simulated sunlight were also enhanced by 5.20- and 5.06-fold, respectively, compared with that without irradiation of LED-simulated sunlight. CONCLUSION Because of their convenient preparations, inexpensive and renewable raw materials, and excellent photothermally controlled-release performance, these on-demand pesticide delivery systems might have significant potential in improving the utilization efficiency of pesticides in modern agriculture. © 2024 Society of Chemical Industry.

Pesticide loaded nanohydrogel based on attapulgite and gelatin with improved foliar adhesion

Sijia Zhang,Zhenggang Xiao

BACKGROUNDS Pesticides have been widely applied to promote plant growth due to their high insecticidal efficiency. However, most pesticides easily slip from the leaf surface due to the low foliar adhesion, leading to significant damage to the environment and human health. Increasing foliar adhesion of pesticides has been a major challenge. Nanomaterial technology, which can deliver pesticide active ingredients, offers a new opportunity to increase pesticides efficacy with higher foliar adhesion. RESULTS In this study, we prepared the pesticide loaded nanohydrogel ALP@Ge-g-P(AA-co-AM)/ATP by grafting attapulgite and gelatin onto P(AA-co-AM), and loading the pesticide alphacypermethrin synchronously via the emulsion solvent evaporation method. The size of the new nanohydrogel was 20.5 nm and the pesticide loading capacity was 21.5%. Compared to P(AA-co-AM), the water absorbency of Ge-g-P(AA-co-AM)/ATP increased by 54.3% in 120 min, while the water retention increased from 4.27% to 38.7% after 48 h. Washout experiments and contact angle results suggested that the nanohydrogel, with a contact angle of 44.8° indicating good wettability, can resist rain flush and increase foliar adhesion naturally. CONCLUSIONS Overall, the preparation of pesticide loaded nanohydrogel is simple. Experiments demonstrated the pesticide loaded nanohydrogel has high efficiency and strong foliar adhesion. This makes it a promising candidate for spraying application and reducing pollution levels. © 2024 Society of Chemical Industry.

ZnO nanoparticles delivered by metal–organic framework ZIF-8 are highly effective in controlling tobacco brown spot disease

Xunliang Huang,Weiqiang Tian,Hao Jiang,Yue Xing,Ya Pu,Zhongwei Liu,Song Yang,Zhensheng Kang,Lin Cai

BACKGROUND Fungal phytopathogens pose a significant threat to global crop yields. The overuse of pesticides leads to pesticide-resistant pathogens, environmental contamination and risks to human health. Consequently, there is an urgent need for effective and ecofriendly antifungal strategies to address these challenges. RESULTS Herein, ZnO-ZIF-8 is synthesized via an in situ growth method to integrate the antifungal property of zinc oxide (ZnO) and the control release property of zeolitic imidazolate framework (ZIF)-8. Results suggest that the inhibition rates of ZnO, ZIF-8 and ZnO-ZIF-8 against Alternaria alternata are 44.4%, 74.1% and 94.4%, respectively, indicating that ZnO-ZIF-8 exhibits the most potent antifungal activity. ZnO-ZIF-8 also causes distortion and fracture of mycelia, significantly impairing spore germination and inhibiting germ tube elongation. CONCLUSION The potential antifungal mechanisms of ZnO-ZIF-8 involve the disruption of fungal cell morphology and structure through the release of Zn 2+ , ultimately leading to fungal cell death. Biosafety assessments indicate that the application of ZnO-ZIF-8 exhibits negligible effects on tobacco seeds and plants. This study provides valuable insights for the effective management of plant fungal diseases and supports sustainable agricultural development. © 2025 Society of Chemical Industry.

Antifungal activities and underlying mechanisms of carvacrol against Neopestalotiopsis saprophytica causing leaf brown spot in Rosa roxburghii

Handinghong Zheng,Jiaohong Li,Zhenxiang Guo,Guy Smagghe,Shunhua Gui,Xiaomao Wu,Xiangsheng Chen

BACKGROUND Rosa roxburghii produces a distinctive edible fruit renowned for its high vitamin C content and medicinal properties. Leaf brown spot is a newly identified fungal disease in R. roxburghii , causing considerable production losses in southwest China. RESULTS In this study, the pathogenic fungus causing brown spot in R. roxburghii was identified as Neopestalotiopsis saprophytica (CXCHB-2). Carvacrol, among seven selected phytocompounds, demonstrated strong antifungal activity against N.  saprophytica with a 50% effective concentration (EC 50 ) of 19.72 mg L −1 . The mechanisms underlying the antifungal activity of carvacrol against N. saprophytica were investigated, and it showed that carvacrol disrupted N. saprophytica mycelium growth by inhibiting mycelial matrix, and compromising cell wall and membrane integrity. Moreover, the in vivo antifungal assays confirmed that carvacrol could inhibit the growth of N. saprophytica in R. roxburghii leaves with excellent therapeutic and protective effects (76.61% and 70.63%, respectively). Further investigations indicated that this phytocompound induced the upregulation of 24 differentially expressed genes related to the leaf hormone signal transduction of R. roxburghii , enhancing its resistance to N. saprophytica . CONCLUSION Carvacrol exhibited potent antifungal activity against N. saprophytica , causing leaf brown spot in R. roxburghii by disrupting its cell walls and membranes. Moreover, carvacrol could enhance the resistance of R. roxburghii leaves to N. saprophytica . Findings of this study suggested that carvacrol may serve as a promising plant-derived fungicide for controlling leaf brown spot in R. roxburghii leaves, providing a serviceable reference for its application in the agroforestry system. © 2025 Society of Chemical Industry.

Preparation and insecticidal activities of betulin-cinnamic acid-related hybrid compounds and insights into the stress response of Plutella xylostella L.

Xiaobo Huang,Nan Hao,Lili Shu,Zhenyang Wei,Jiawei Shi,Yuee Tian,Genqiang Chen,Xinling Yang,Zhiping Che

BACKGROUND The emergence and increasing intensification of insect pest resistance greatly shortens the service life of commercial insecticides. Hence, the continuous development of novel insecticides is extremely necessary. Discovering new insecticide molecules based on natural products with insecticidal activity has been a popular technique and has made outstanding contributions in this field. RESULTS A series of betulin-cinnamic acid-related hybrid compounds was prepared by molecular hybridization through the bridge of acyloxy groups and their chemical structures were unambiguously confirmed. Against A. pisum , compounds 2e , 3a-3e , 3g , 3i , 3j , and 3m showed complete antifeedant activity at 2 mg/mL; compound 5b exhibited 2.7 times the aphicidal activity of the precursor betulin. Against P. xylostella , compound 2l displayed 3.9 times the insecticidal activity of betulin. Meanwhile, some interesting structure–activity relationships were obtained. Furthermore, compound 2l could destroy the ultrastructure of midgut cells and significantly inhibit the activity of α-amylase in P. xylostella larvae. Molecular docking indicated that the lupane skeleton and p -nitrocinnamoyloxy groups played a key role in the binding of compound 2l to α-amylase. CONCLUSION Compounds 5b and 2l could be further optimized structurally as the secondary lead compounds for the discovery of novel insecticide molecules. © 2025 Society of Chemical Industry.

Widespread occurrence of cyhalofop-resistant Leptochloa chinensis in eastern China was mainly propelled by target-site mutations

Guoqi Chen,Kai An,Jiahao Xue,Wei Deng,Zeyue Huang,Yang Xu

BACKGROUND Various mechanisms or factors may cause the wide infestation of herbicide-resistant weeds with different contributions, such as target-site resistance (TSR), non-target-site resistance (NTSR), hormesis, and dissemination of herbicide-resistant weeds. Here we tested this hypothesis in eastern China in Leptochloa chinensis , for which cyhalofop resistance is a major challenge to rice planting. RESULTS Our results revealed that 30% of the total 242 populations held high (10 < RI < 100) or very high resistance (RI > 100) to cyhalofop. Resistance increased from north to south, and from transplanted rice fields to direct-seeded rice fields. Six mutant types conferring TSR to cyhalofop were found in 89% sampled populations, with 18% carrying two types, 2% carrying three types, and another 2% carrying four types of mutations. W2027C was the most frequent TSR mutation, present in 47.3% of sampled populations, followed by I2041N (21.8%), W1999C (20%), L1818F (20%), W1999G (5.5%) and C2088R (1.8%). Non-target-site resistance linked with P450 monooxygenases (P450) and glutathione S-transferases (GST) were observed in 7% and 10% cyhalofop-resistant populations sampled. No significant correlation between genetic and geographic distances were observed among cyhalofop-resistant populations. Hormetic effects of cyhalofop to L. chinensis were evident in 13% of overall populations. CONCLUSIONS This study compared the contribution of different mechanisms towards herbicide resistance, revealed herbicide hormesis and revealed the distribution of resistance. Insights from this study could be important for weed management practices in other areas of the world. © 2025 Society of Chemical Industry.

Genome identification of C-type lectins in Bactrocera dorsalis and functional characterization of BdCTL-S12 with a broad-spectrum pathogen recognition

Wei Zhao,Di Zou,ZhengYang Wang,GuiQiang Wang,HongBo Jiang,Guy Smagghe,JinJun Wang,DanDan Wei

BACKGROUND Bactrocera dorsalis (Diptera: Tephritidae) is a highly adaptable invasive pest that causes significant economic losses. Long-term chemical control strategies have led to high levels of insecticide resistance and severe environmental risks. C-type lectins (CTLs), key pattern recognition receptors in insect innate immunity, are named for their calcium-dependent carbohydrate recognition domains (CRDs) and they play a critical role in detecting and binding to foreign pathogens. This study aims to identify B. dorsalis CTLs (BdCTLs) and investigate their roles in pathogen defense. RESULTS A total of 42 BdCTL genes were identified. Based on the number of CRDs, 36 genes were classified into the S subfamily, while the remaining six were assigned to the X subfamily. Spatio-temporal expression analysis by qPCR revealed that BdCTL genes were expressed throughout all developmental stages with a higher expression observed in the midgut, Malpighian tubules, and fat body. The expression levels of BdCTL-S4 , BdCTL-S12 , and BdCTL-S31 were significantly upregulated under the infection of the entomopathogenic fungus Beauveria bassiana . Notably, BdCTL-S12 also exhibited upregulated expression patterns in response to four different pathogens bacteria with sustained expression over time. Using RNA interference (RNAi), we successfully knocked down BdCTL-S12 expression, achieving a silencing efficiency of 68%. Survival assays demonstrated a 10–30% reduction in the survival rate of B. dorsalis after pathogen infection following dsRNA injection compared to the control group. Further in vitro experiments confirmed that BdCTL-S12 can agglutinate and bind to pathogens in a calcium-dependent manner. This binding was competitively inhibited by polysaccharides, underscoring a key functional mechanism of BdCTL-S12 in pathogen recognition. CONCLUSION In summary, we identified 42 BdCTLs and demonstrated that BdCTL-S12 is critical for pathogen recognition and immune defense in B. dorsalis . These results advance our understanding of the molecular mechanisms underlying B. dorsalis immunity. Specifically, elucidating the role of CTLs may facilitate the development of microbial-based biological control strategies (e.g., entomopathogenic fungi or bacteria) against this invasive pest. Additionally, RNAi-mediated knockdown of CTL-encoding genes could weaken the immune system of Bactrocera flies, increasing their susceptibility to microbial infections and thereby enhancing the effectiveness of biological control. © 2025 Society of Chemical Industry.
