Double-stranded RNA (dsRNA) and reactive oxygen species (ROS) contribute to the exacerbation of skin inflammatory diseases such as psoriasis and atopic dermatitis. While antioxidant strategies have been extensively explored, approaches to neutralize pathogenic dsRNA remain limited. Here, we show that Mo 90 Ce 10 , a cerium-doped polyoxometalate, concurrently targets dsRNA and ROS to mitigate cutaneous inflammation. In vitro, Mo 90 Ce 10 scavenges ROS and suppresses type I interferon responses by directly binding dsRNA through its oxygen-rich, hydrogen bond-forming surface. Its structural template, {Mo 154 }, exhibits weaker activity. In vivo, Mo 90 Ce 10 reduces disease severity, dsRNA-induced inflammation, ROS burden, and neutrophil infiltration. Leveraging its negative charge and self-assembly properties, we further develop a transdermal platform by complexing Mo 90 Ce 10 with the cationic peptide TD-1 and co-loading methotrexate, thereby enhancing drug penetration andtherapeutic efficacy while reducing recurrence. Together, these findings establish Mo 90 Ce 10 as a multifunctional modulator of dsRNA- and ROS-driven inflammation and a promising transdermal delivery platform for inflammatory diseases. Oxidative stress and dsRNA are key drivers of skin inflammation, while therapies targeting oxidative stress are common, dsRNA targeting is not. Here, the authors report on a cerium-doped polyoxometalate with anti-oxidative and dsRNA binding properties showing potential for a transdermal strategy to reduce skin inflammation.

Osteoarthritis (OA) is a prevalent degenerative joint disease with limited effective treatment options. Joint inflammatory pain is a primary reason patients seek care, but systemic drug administration often causes severe side effects, while intra‐articular injection suffers from rapid clearance and poor tissue penetration. Herein, we develop nanozyme‐based Janus nanomotors loaded with metformin (MET) for reactive oxygen species (ROS)‐targeted therapy in OA. Leveraging intrinsic superoxide dismutase (SOD) and catalase (CAT) enzymatic activity, these nanomotors harness pathologically elevated ROS within the articular microenvironment as chemical fuel, achieving self‐propelled deep penetration into both cartilage and synovial tissue. This mechanism facilitates simultaneous on‐the‐move ROS scavenging and sustained deep‐tissue MET release, which further restores redox homeostasis and protects chondrocytes by regulating the NRF2/KEAP1 signaling pathway. Furthermore, the nanomotors can suppress nociceptive signaling in the dorsal root ganglia (DRG), thereby alleviating joint pain and improving mobility. This research offers a novel and efficient approach for cartilage protection and arthritis pain management. ROS‐responsive NM@MET nanomotors utilize elevated ROS in osteoarthritic joints to promote deep penetration into cartilage and synovium and enable sustained metformin release. By scavenging ROS, regulating NRF2/KEAP1 signaling, protecting chondrocytes, suppressing synovial inflammation, and attenuating nociceptive signaling, NM@MET alleviates pain and improves joint function.

Reactive oxygen species (ROS) play a critical role in
tumor progression
and therapeutic response, yet real-time monitoring of transmembrane
hydrogen peroxide (H 2 O 2 ) gradients─a
key indicator of metabolic activity and antioxidant capacity─remains
challenging due to the lack of integrated platforms combining active
access and sensitive detection. Here, we present a biogenic microrobotic
surface-enhanced Raman scattering (SERS) sensor fabricated from sunflower
pollen grains. Leveraging the natural spiky morphology and magnetic
actuation, the microrobot achieves controlled membrane penetration
to access both intra- and extracellular compartments. The plasmonic
surface coating enables high-sensitivity SERS detection with excellent
reproducibility, allowing quantitative monitoring of hydrogen peroxide
dynamics across the cell membrane. In addition, a responsive polymer
coating confers on-demand release capability upon exposure to elevated
reactive oxygen species levels. By integrating navigation, transcellular
sensing, and triggered release within a single biocompatible platform,
this pollen-based microrobot offers a potential strategy for precision
monitoring and targeted intervention in the tumor microenvironment.

Skin injury triggers the production of reactive oxygen species (ROS) such as H2O2 in the wound microenvironment. In addition to ROS produced by the wound itself, ROS produced by bacterial infections likewise cause severe damage to blood vessels and endothelial cells, leading to chronic or unhealed wounds. Hyaluronic acid (HA) is widely used in hydrogel construction due to its rich bioactivity. In this study, an antibacterial hydrogel with ROS responsiveness was synthesized by grafting HA through phenylboronic acid, cross-linking it with PVA in a ROS-sensitive phenylboron ester bond, and loading it with AMP for the treatment of bacterial-infected wounds. In vitro experiments proved that the hydrogel has good biocompatibility, antioxidant ability, and significant inhibition of E. coli and S. aureus. In vivo experiments proved that the hydrogel promotes the healing of infected wounds in mice, raises the level of IL-6/TGF-β at the wounds, reduces the inflammation of the wounds, raises the level of expression of CD31/VEGF, and promotes the angiogenesis of wounds.

IntroductionOsteoarthritis (OA) is a common degenerative joint disease for which no definitive effective treatment currently exists. Therapeutic hydrogen (H2) exhibits antioxidant and anti-inflammatory properties and holds promise for alleviating OA; however, targeted delivery and sustained release of H2 remain challenging.MethodsIn this study, we developed an upconversion nanocomposite for integrated diagnosis and therapy of OA. Upconversion nanoparticles (UCNP) were first synthesized and coated with hydrogen-producing TiO2 to form UCNP@TiO2. A H2O2-sensitive thioketal (TK) linker was then conjugated onto the surface of UCNP@TiO2 to attach a black hole quencher (BHQ), yielding the functionalized nanocomposite UCNP@TiO2-TK-BHQ.ResultsIn the presence of hydrogen peroxide (H2O2), cleavage of the TK linker triggers the release of BHQ, thereby restoring the fluorescence of UCNP for real-time reactive oxygen species (ROS) detection. Simultaneously, near-infrared (NIR) light activates UCNP@TiO2 to generate hydrogen, which scavenges ROS and promotes the restoration of chondrocyte health.Discussion/ConclusionThe UCNP@TiO2-TK-BHQ nanocomposite thus represents a promising theranostic platform that combines ROS sensing with on-demand H2 therapy, offering an integrated approach for both diagnosis and treatment of OA.

The accumulation of reactive oxygen species (ROS) leads to enhanced osteoclast activity, causing severe bone destruction in postmenopausal osteoporosis. Immunity-related GTPase family M member 1 (Irgm1) plays an essential role in affecting the production of intracellular ROS. To detect whether deletion of Irgm1 could suppress osteoclastogenesis through cellular redox regulation, we first evaluated whether the Irgm1 level was significantly elevated in mice bone marrow-derived monocytes/macrophages (BMDMs) from ovariectomy (OVX)-induced osteoporosis mice. Moreover, bioinformatics network analysis was performed to identify Irgm1 as a key upregulated gene during osteoclast differentiation. Next, we found that macrophage-specific Irgm1 knockout (Irgm1-cKO, Lyz2-Cre; Irgm1 flox/flox ) in OVX mice resulted in slower bone loss compared with OVX mice from the control group (Irgm1 flox/flox ). We then demonstrated that loss of Irgm1 inhibited osteoclast differentiation and bone resorption function via suppressing ROS accumulation. Further mechanism revealed that Irgm1 could endogenously bind to kelch-like ECH-associated protein 1 (Keap1) and keep Keap1 from ubiquitination and degradation. In the absence of Irgm1, Keap1 was downregulated and causing nuclear factor erythroid 2-related factor 2 (Nrf2) to translocate to the nucleus, thereby activating the level of the antioxidant system to combat oxidative stress. Moreover, Irgm1 deficiency in RAW264.7 promoted osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs) through inhibiting the M1 phenotype polarization. Taken together, our results revealed that loss of Irgm1 significantly alleviates OVX-induced bone loss, thus laying the foundation for exploring Irgm1 as a novel targeting approach for the treatment of osteoporosis.

Oxidative stress is closely linked to androgenetic alopecia (AGA). However, research on antioxidants specifically targeting hair follicle tissue has been limited. The utilization of nanomaterials that possess superior reactive oxygen species (ROS)-scavenging capabilities and biocompatibility is a promising approach for the treatment of AGA. The microneedle-mediated transdermal delivery system can overcome the limitations of the stratum corneum and effectively deliver functional nanomaterials to the vicinity of hair follicles. In this study, we report a degradable microneedle patch loaded with ROS-scavenging CuxO nanozyme (CuxO-MNs) for targeted treatment of AGA. Tiny CuxO nanozymes were synthesized using a robust and eco-friendly method, demonstrating effective ROS-scavenging ability at both in vitro and cellular levels. With the penetration and dissolution of sodium hyaluronate microneedles, CuxO can be rapidly released and effectively reach the area around the hair follicle, enabling targeted removal of ROS. In the AGA model, CuxO-MNs effectively alleviate oxidative stress, restore the hair follicle microenvironment, and stimulate the transition of hair follicles from the telogen to the anagen. These combined effects significantly enhance hair regeneration and exhibit therapeutic efficacy comparable to the clinical drug minoxidil, suggesting the safety and translational potential of this strategy for AGA treatment.

Multimodal treatment of cancer is an unstoppable revolution in clinical application. However, designing a platform that integrates therapeutic modalities with different pharmacokinetic characteristics remains a great challenge. Herein, we designed a universal lipid nanoplatform equipping a ROS-cleavable docetaxel prodrug (DTX-L-DTX) and an NF-E2-related factor 2 (NRF2) inhibitor (clobetasol propionate, CP). This simply fabricated nanomedicine enables superior synergistic molecularly targeted/chemo/radio therapy for lung cancer cascade by a transcription factor-driven ROS self-sustainable motion. Chemotherapy is launched via ROS-triggered DTX release. Subsequently, CP inhibits the expression of NRF2 target genes, resulting in efficient targeted therapy, meanwhile inducing sustained ROS generation which in turn facilitates chemotherapy by overcoming ROS consumption during the DTX release process. Finally, the introduction of radiotherapy further amplifies ROS, offering continuous mutual feedback to amplify the ultimate treatment performance. This strategy is conceptually and operationally simple, providing solutions to challenges in clinical cancer treatment and beyond.

Liposomes are the most valuable nanocarriers in clinical use because of their biocompatibility, biodegradation, and effective encapsulation of hydrophilic or hydrophobic drugs. However, their applications are limited by the structure and functions of the most common phospholipids used as the main component of the liposomes. In this work, novel series of thioether phosphatidylcholines (S-PCs) and S-PC-based liposomes (S-LPs) were developed for reactive oxygen species (ROS)-responsive drug release. First of all, S-PCs with different chain lengths were synthesized by a combination of click reaction and heterogeneous esterification. Differential scanning calorimetry studies indicated that S-PCs had different phase transition temperatures depending on their chain lengths. Their critical aggregation concentrations were measured by the fluorescence probe technique indicating the self-assembly ability. After that, S-PC-based stealth liposomes (S-LPs) containing DSPE-PEG2000 and cholesterol were prepared via a classic thin-film method. Doxorubicin (DOX) as a model drug was loaded in the stealth liposomes (DOX/S-LPs) by using the ammonium sulfate gradient method with high encapsulation efficiency. DOX/S-LPs were characterized by dynamic light scattering (DLS), transmission electron microscope (TEM), and cryogenic TEM, confirming their spherical structure with the bilayer thickness of about 4 nm. The ROS sensitivity of S-PCs and S-LPs was carefully evaluated in the presence of H2O2 by means of mass spectrometry, DLS, TEM, and ultraviolet spectroscopy and release study. The results indicated the significant structural change of S-LPs after H2O2 treatment, which demonstrated that S-LPs possessed an efficient ROS-triggered disintegration because of thioether oxidation of S-PCs. Finally, in vitro and in vivo anticancer efficiency assays revealed the improved drug potency of DOX/S-LPs, which can be attributed to ROS-triggered destruction of S-LPs after the uptake by tumor cells followed by rapid release of DOX. All together, as alternatives of traditional phosphatidylcholines, S-PC-based stealth liposomes are promising ROS-responsive carriers for the controlled delivery of drugs.

Significant embryo loss remains a serious problem in pig production. Reactive oxygen species (ROS) play a critical role in embryonic implantation and placentation. However, the potential mechanism of ROS on porcine trophectoderm (pTr) cell fate during the peri-implantation period has not been investigated. This study aimed to elucidate the effects of ROS on pTr cell phenotypes and the regulatory role in cell attachment and differentiation. Herein, results showed that exogenous H2O2 inhibited pTr cell viability, arrested the cell cycle at S and G2/M phases, and increased cell apoptosis and autophagy protein light chain 3B and Beclin-1, whereas these effects were reversed by different concentrations of N-acetyl-l-cysteine (NAC) posttreatment. In addition, NAC abolished H2O2-induced autophagic flux, inhibited intracellular and mitochondrial ROS, and restored expression of genes important for mitochondrial DNA and biogenesis, cell attachment, and differentiation. NAC reversed H2O2-activated MAPK and Akt/mammalian target of rapamycin pathways in dose-dependent manners. Furthermore, analyses with pharmacological and RNA interference approaches suggested that autophagy regulated cell apoptosis and gene expression of caudal-related homeobox 2 and IL-1β. Collectively, these results provide new insights into the role of the ROS-induced autophagy in pTr cell apoptosis, attachment, and differentiation, indicating a promising target for decreasing porcine conceptus loss during the peri-implantation period.

Nowadays, the stability and on-demand release of drug carriers are still to be solved. To meet the demand of these issues, we developed a reactive oxygen species (ROS) responsive selenium-containing polyphosphoesters nanogel (PSeP) by a facile one-step ring-opening polymerization of the novel monomer 4-selenoctane-1,8-diyl bis(propylphosphatelane) (Se-COP) with polyethylene glycol (mPEG) employed as the macroinitiator. Their structure was confirmed by NMR, FT-IR and GPC. The crosslinked core-shell structure imparted the nanogels with excellent dimensional stability according to the dynamic light scattering (DLS) and transmission electron microscopy (TEM). Moreover, the selenide groups endowed the nanogels with rich ROS responsiveness when subjected to the stimuli of H 2 O 2 , thus the drug-loaded PSeP nanogels displayed swollen behaviors leading to an activated doxorubicin hydrochloride (DOX · HCl) release. The release mechanisms fitted by the Ritger-Peppas power-law model also proved the swollen release process. MTT assays exhibited that the PSeP nanogels were nontoxic up to a tested concentration of 50 μg mL −1 by A549 and HEK293, and the DOX-loaded PSeP had a high anti-cancer behaviour against A549 cancer cells. Additionally, these nanogels possessed enhanced intracellular drug release by CLSM . Therefore, these results highlighted that the selenium-containing polyphosphoesters nanogels could be a potential platform for the ROS-sensitive drug delivery.
