Active polylactic acid/tilapia fish gelatin-sodium alginate bilayer films: Application in preservation of Japanese sea bass (Lateolabrax japonicus)

Jingwen Chen,Yinxuan Li,Yifen Wang,Salimata Yakubu,Haibing Tang,Li Li

Most edible films/coatings including those made of gelatin cannot be used independently as packaging films. In this study, active fish gelatin-based bilayer packaging films were developed and applied to preserve Japanese sea bass. The coating method was applied to produce polylactic acid/tilapia fish gelatin-sodium alginate bilayer film (PLA/FGSA). β -cyclodextrin inclusion complexes of cinnamaldehyde ( β -CD-CI) and thymol ( β -CD-Ty) were added into the gelatin-based layer of the PLA/FGSA to obtain active bilayer films (PLA/FGSA-CI and PLA/FGSA-Ty). This combination improved the oxygen and water vapor barrier abilities of the films, compared with PLA and edible gelatin-based monolayer films. Both active bilayer films possessed strong antibacterial and antioxidant activities . PLA/FGSA-Ty film showed the most effective inhibition against the growth of Pseudomonas .spp and H 2 S-producing Shewanella , generation of total volatile basic nitrogen (TVB-N) and thiobarbituric acid reactive substances (TBARS) of fish meat. These results indicate the active bilayer films are promising in the preservation of seafood.

Colloidal toxic trace metals in urban riverine and estuarine waters of Yantai City, southern coast of North Yellow Sea

Yuxi Lu,Xuelu Gao,Jinming Song,Chen-Tung Arthur Chen,Jinling Chu

The environmental characteristics of colloidal toxic trace metals Cd, Cu and Pb in riverine and estuarine waters collected from two urban rivers of Yantai City in eastern China, the Guangdang and Xin'an Rivers, were investigated using a modified centrifugal ultrafiltration (CUF) method in conjunction with acid extraction and inductively coupled plasma mass spectrometry. The target metals in dissolved pool were divided into four CUF fractions, i.e. <1 kDa, 1–3 kDa, 3–10 kDa and 10 kDa–0.2 μm, and the results showed that colloidal Cd, Cu and Pb were dominated by 1–10 kDa (1–3 and 3–10 kDa), 1–3 kDa and 10 kDa–0.2 μm fractions, respectively. The coagulation/flocculation of low-molecular-weight (1–10 kDa) colloidal Cd and Cu in the estuaries was obvious and strong, while the enrichment of dissolved Pb in the 10 kDa–0.2 μm fraction may be mainly related to its biogeochemical interactions with Fe-oxides, which is easy to occur in macromolecular colloids. In addition, the actual molecular weight cutoffs (MWCOs) of the three used CUF units with nominal MWCOs of 1, 3 and 10 kDa were determined to be 4.9, 8.5 and 33.9 kDa, respectively, indicating that membrane calibration is essential for explaining the actual fraction of dissolved trace metals and verifying the integrity of ultrafiltration membrane. Overall, the results in this study provide a further understanding of the heterogeneity in biogeochemical features, migration and fate of toxic trace metals in aquatic ecosystems, especially that of the river-sea mixing zone.

Chemodynamic therapy combined with endogenous ferroptosis based on “sea urchin-like” copper sulfide hydrogel for enhancing anti-tumor efficacy

Shuang Li,Bingjie Wang,Jiaojiao Tao,Yu Dong,Teng Wang,Xia Zhao,Tianze Jiang,Lianxiao Zhang,Hai Yang

Chemodynamic therapy (CDT) is a promising strategy for cancer treatment, however, its application is restricted by low hydrogen peroxide (H 2 O 2 ) concentration, insufficient reactive oxygen species (ROS) generation, and high glutathione (GSH) levels. Here, we developed an injectable thermosensitive hydrogel (DSUC-Gel) based on “sea urchin-like” copper sulfide nanoparticles (UCuS) loaded with dihydroartemisinin (DHA) and sulfasalazine (SAS) to overcome these limitations of CDT. DSUC was cleaved to release DHA, SAS and Cu 2+ under acidic tumor microenvironment to enhance CDT. DHA with peroxide bridge responded to intracellular Fe 2+ to alleviate H 2 O 2 deficiency. SAS prevented GSH synthesis by targeting SLC7A11 and inhibited glutathione peroxidase (GPX4) activity to induce endogenous ferroptosis. ROS produced by Fenton-like reaction of Cu 2+ promoted lipid peroxidation (LPO) accumulation to promote ferroptosis. Enhanced CDT and ferroptosis induced immunogenic cell death (ICD), promoted dendritic cells (DCs) maturation and cytotoxic T lymphocytes (CTLs) infiltration. As a result, DSUC-Gel significantly inhibited tumor growth both in vitro and in vivo . Our study provides a novel approach for enhancing anti-tumor efficacy by combining CDT, endogenous ferroptosis and ICD.

Characterization of a novel deep-sea microbial esterase EstC10 and its use in the generation of (R)-methyl2-chloropropionate

Gong Yanhui,Ma Sanmei,Wang Yongfei,Xu Yongkai,Sun Aijun,Zhang Yun,Hu Yunfeng

A novel esterase EstC10 from Bacillus sp. CX01 isolated from the deep sea of the Western Pacific Ocean and the functionalities of EstC10 was characterized. At present, the reports about the kinetic resolution of racemic methyl 2-chloropropionate were quite rare. So we developed deep-sea microbial esterase EstC10 as a novel biocatalyst in the kinetic resolution of racemic methyl 2-chloropropionate and generate ( R )-methyl 2-chloropropionate with high enantiomeric excess (>99%) after the optimization of process parameters such as pH, temperature, organic co-solvents, surfactants, substrate concentration and reaction time. Notably, the optimal substrate concentration (80 mmol/L) of esterase EstC10 was higher than the kinetic resolution of another esterase, Est12-7 (50 mmol/L). The novel microbial esterase EstC10 identified from the deep sea was a promising green biocatalyst in the generation of ( R )-methyl 2-chloropropionate as well of many other valuable chiral chemicals in industry.

Environmentally friendly and temperature resistant water-based drilling fluids with highly inhibitory synthetic nanocellulose polymers for deep sea drilling

Yikang Guo,Xianyu Yang,Ren Wang,Meng Zhao,Jianlong Wang,Jingyu Xie,Zhaokai Dai,Quanwei Xu,Zichen Han,Guosheng Jiang,Jihua Cai

The ocean is rich in sustainable hydrocarbon. The process of extracting these resources poses challenges such as coexistence of low temperature and high temperature, high pressure of deepwater and unpredictable shale formations. A kind of ionic crosslinking polymer consisting of N, N-dimethylacrylamide (DMA), 2-acrylamido-2-methylpropane sulfonic acid (AMPS), and Cellulose Nanofibers (CNF) (PADC), was synthesized to address these challenges. Cellulose was first oxidized to nanocellulose (CNF) through 2,2,6,6-tetramethylpiperidinyl-1-oxide (TEMPO) oxidation. Subsequently, the ionic crosslinking polymer, consisting of DMA, AMPS, and CNF, was synthesized using ionic crosslinking polymerization. The surface characteristics and morphology were characterized by transmission electron microscopy (TEM), nanoparticle size analysis, and zeta potential analysis. The structural composition was analyzed by Fourier Transform Infrared Spectroscopy (FT-IR). The shale inhibition mechanism was investigated through contact angle and pressure transmission experiments. The results indicated that the average length of PADC is approximately 1450 nm, with an average diameter of less than 100 nm and contains sulfonic acid groups. The shale contact angle increased from 16.75° to 82°, indicating that PADC increased the hydrophobicity of the shale by 390%. Pressure transfer experiments show that PADC can reduce shale permeability by 37%, which means PADC has good hydration inhibition performance. In addition, a drilling fluid system suitable for deep sea drilling was proposed. The drilling fluid system exhibits good rheological properties at 160 °C and 0 °C. The fluid loss after hot rolling at 160 °C is less than 12 mL. The linear expansion of shale core after 16 h is only 0.7%, and the rolling recovery rate at 160 °C is as high as 99.48%. Luminescent bacteria EC 50 is 2.0 × 10 5  mg/L, which means it is non-toxic and environmentally friendly. And drilling fluid conforms to the power law flow model. Therefore, the drilling fluids proposed in this paper can meet the requirements of deep sea drilling in terms of rheological properties and filtration loss properties at 0–160 °C, and can seal shale pores and inhibit shale hydration through the ionic cross-linking PADC. This research offers insights into the utilization of cellulose in well drilling processes and provides references for designing deep sea drilling fluids.

Effects of cholesterol-lowering probiotic fermentation on the active components and in vitro hypolipidemic activity of sea buckthorn juice

Ying Fan,Yueling Shang,Fengwei Li,Jinbin Liu,Dujun Wang,Ya Zhang,Xiaohong Yu,Wei Wang

Sea buckthorn has lipid-lowering properties and is widely used in the development of functional foods. In this study, a probiotic ( Lactobacillus plantarum , Lp10211) with cholesterol-lowering potential and acid and bile salt resistant was screened for the fermentation of sea buckthorn juice. Changes in the active ingredients, such as sugars and phenolics, before and after fermentation, as well as their in vitro lipid-lowering activities, were compared. The contents of reducing and total sugars decreased substantially after fermentation. Lp10211 primarily utilized fructose for growth and reproduction, with a utilization rate of 76.9%. The phenolic compound content of sea buckthorn juice increased by 37.06% after fermentation and protected the phenolic components from degradation (protocatechuic and p -coumaric acids) and produced new polyphenol (shikimic acid). Enhanced inhibition of pancreatic lipase activity (95.42%) and cholesterol micellar solubility (59.15%) was evident. The antioxidant properties of the fermentation broth were improved. Notably, Lp10211 preserved the color and reversed browning in sea buckthorn juice. The collective findings indicate that fermentation of sea buckthorn juice by Lp10211 may enhance the functional components and lipid-lowering activity of sea buckthorn, which may provide a new approach for the development of lipid-lowering foods.

Peirong Ma,Zheng Li,Yuting Jin,Jiale Zuo,Yanling Zhang,Alideertu Dong,Douxin Xiao,Munkhjargal Burenjargal

In recent years, with the advantages of environment friendly, natural deep eutectic solvents (NADES) have been gradually applied for the extracting bioactive compounds from natural products. In this study, NADES-12 (betaine-lactic acid, n / n , 1:2) was selected for the first time to extract flavonoids from sea buckthorn fruit after screening of 23 NADES. The optimal technical conditions of NADES combined with ultrasonic assisted extraction (UAE) were determined by single factor test and response surface optimization test. The results showed that the optimal process for the extraction of antioxidant components by NADES-12 was 37 min extraction time, 40 °C extraction temperature, 17 % (v/v) water content, 40 mL mg −1 liquid–solid ratio, 160 W ultrasonic power, and a maximum extraction rate of 5.248 mg g −1 , and the extraction yield is 2.4 and 3.1 times that of 60 % ethanol and water. The sea buckthorn extract by NADES has antioxidant activity. The study proposes a green and efficient method for the extraction of bioactive components from sea buckthorn fruit and provides a valuable basis for the exploitation of other natural plants.

Xuebin Lu,Zhengya Gong,Zhihao Yu,Jian Xiong,Gaixiu Yang,Mengyan Guo,Yuxin Zhang,Yiming Li

Supercapacitors as one of the new energy storage devices have attracted much attention from researchers. The design and preparation of new electrode materials with high conductivity and energy density has become a frontier and key issue in the research of supercapacitor performance improvement. In addition, flow capacitor deionization (FCDI) relying on the working principle of supercapacitors is a promising technology for desalination and has received crucial attention. NiCo 2 S 4 (NCS) is a promising pseudocapacitive material with high electrical conductivity and ideal stability. In this study, sea urchin-like NiCo 2 S 4 electrode materials (CC@NCS) grown in situ on crab shell carbon (CC) were prepared by a simple molten salt-assisted hydrothermal two-step method. The composite successfully combined the properties of CC and NiCo 2 S 4 . The prepared material exhibited excellent electrochemical properties and energy storage performance. In the three-electrode system, the specific capacitance is up to 692.4 F/g at a current density of 0.5 A/g. Asymmetric supercapacitors assembled with CC as the negative electrode, CC@NCS as the positive electrode, and 3 M KOH as the electrolyte obtained high specific capacitance (0.5 A/g, 163.6 F/g) and high energy density (305.16 W/kg, 8.18 Wh/kg). The device also showed excellent cycling performance in the long cycle test (88.65 % retention after 5000 cycles). In addition, the FCDI device with this material as the flow electrode achieved a final conductivity of 15.16 μS/cm by electrosorption of a 1000 mg/L NaCl solution at a constant pressure of 1.2 V for 60 min along with an impressive average salt removal rate (0.0621 mg/cm 2 ·min −1 ) and charge efficiency value (90.79 %), and a low energy consumption value (0.02 kWh/mol). What's more, it also exhibited excellent desalination properties. This indicates that the composite is a promising active material for energy storage and desalination.

Changes in the Main Physicochemical Properties and Electrochemical Fingerprints in the Production of Sea Buckthorn Juice by Pectinase Treatment

Kaihua Guo

Enzymatic hydrolysis using pectinase is critical for producing high-yield and quality sea buckthorn juice. This study determined the optimal temperature, time, and enzyme dosage combinations to guide manufacturers. A temperature of 60 °C, hydrolysis time of 3 h, and 0.3% enzyme dosage gave 64.1% juice yield—25% higher than without enzymes. Furthermore, monitoring physicochemical properties reveals enzyme impacts on composition. Higher dosages increase soluble solids up to 15% and soluble fiber content by 35% through cell wall breakdown. However, excessive amounts over 0.3% decrease yields. Pectin concentration also declines dose-dependently, falling by 91% at 0.4%, improving juice stability but needing modulation to retain viscosity. Electrochemical fingerprinting successfully differentiates process conditions, offering a rapid quality control tool. Its potential for commercial inline use during enzymatic treatment requires exploration. Overall, connecting optimized parameters to measured effects provides actionable insights for manufacturers to boost yields, determine enzyme impacts on nutrition/functionality, and introduce novel process analytical technology. Further investigations of health properties using these conditions could expand sea buckthorn juice functionality.

Sea anemone-inspired smart epoxy coating with mechanical energy relieving capability for improved corrosion protection in simulated marine environment

Li Chen,Fan-di Meng,Fu-hui Wang,Li Liu

Organic coatings are one of the most widely used methods for metal protection. However, traditional organic coatings fail to meet the demands for mechanical energy relief under harsh marine conditions characterized by fluid shear and pressure. To address this challenge, a biomimetic strategy is proposed to introduce a slipable functional side chain into the epoxy backbone, mimicking the mechanism of tentacle movement in sea anemones. This innovative design endows the 2-ureido-4-pyrimidone (UPy)-grafted epoxy coating with exceptional mechanical energy dissipation capabilities. An in-depth investigation into the relationship between the molecular structure and properties revealed that the enhancement originates from a unique architecture that mimics the slip mechanism of sea anemones. This structure features chain-sliding motion at the molecular level and the dynamic exchange of hydrogen bond cross-linking sites. This mechanism efficiently dissipates mechanical energy within the three-dimensional crosslinked network of the coating, thereby significantly enhancing its anti-corrosion performance. This research offers a novel strategy for developing mechanical energy relief coatings and shows promise for the practical application of dynamic bond chemistry theory in creating multifunctional coatings for deep-sea equipment.

Boosting tetracycline degradation via synergistic photo-self-Fenton/peroxymonosulfate activation using sea-urchin-like iron-doped nickel phosphonate catalysts

Huage Lin,Mingyang Wei,Haitong Li,Teng Liu,Weinan Xing

Background Transition metal phosphonates have demonstrated significant potential in catalysis due to their versatile coordination configurations and tunable structural frameworks. However, their direct application in photo-self-Fenton and peroxymonosulfate activation systems for organic pollutant degradation has rarely been explored. Methods In this study, a novel three-dimensional (3D) sea-urchin-like iron-doped nickel phosphonate (NiFeP2) was synthesized and employed as a catalyst in a photo-self-Fenton system for the efficient degradation of tetracycline (TC). Characterization revealed that the hierarchical architecture of NiFeP2 not only enhances light absorption and exposes abundant active sites but also facilitates synergistic peroxymonosulfate activation and photo-self-Fenton reactions, thereby improving catalytic performance. Significant findings Under optimized conditions, NiFeP2 achieved an apparent reaction rate constant (k obs ) of 0.0589 min⁻¹ for TC degradation, demonstrating superior pollutant removal efficiency. The synthesized NiFeP2 photocatalyst exhibited excellent recyclability and maintained stable photocatalytic performance over multiple cycles, demonstrating its promising suitability for real-world wastewater purification applications. This work presents an effective photo-self-Fenton system based on NiFeP2, offering a promising strategy for the remediation of refractory organic pollutants in wastewater treatment.

Constructing Ti3C2Tx@PPy-MoO4 hybrid reinforced epoxy coating for achieving active-passive collaborative anti-corrosion under deep sea environment

Guanting Li,Xianze Meng,Yangmin Wu,Wenjie Zhao

Ti 3 C 2 T x nanosheets, which can act as effective physical barrier against corrosion species, have emerged as promising corrosion-resistant reinforcements in organic coatings due to abundant surface functional groups. However, relying solely on Ti 3 C 2 T x for passive protection does not adequately meet higher corrosion protection requirements, especially under high hydrostatic pressure environment in the deep sea. Herein, we synthesized Ti 3 C 2 T x @PPy-MoO 4 (MPMo) hybrid through in-situ oxidative polymerization strategy, which endowed epoxy coating with collaborative protection of passive physical barrier and active corrosion-inhibition. MPMo enhanced epoxy coating (MPMo/EP) exhibited superior anti-corrosion durability in simulated 60 MPa deep sea environment, maintaining a |Z| f=0.01 Hz value above 10 8  Ω·cm 2 and outperforming Ti 3 C 2 T x -based coatings by three orders of magnitude even after six cycles of AHP test. Moreover, polypyrrole (PPy) significantly improved the structural stability of Ti 3 C 2 T x , and successfully loaded with MoO 4 2− through anion doping. Meanwhile, Raman mapping analysis also revealed that MoO 4 2− anions adsorbed onto the Q235 steel surface and formed Fe 2 (MoO 4 ) 3 protective layers, which significantly hindered the generation of corrosion products. This work achieves active-passive synergistic protection of coatings through in-situ oxidation polymerization, providing a new approach for constructing robust anti-corrosion coatings suitable for higher protection demand in deep sea environment.
