Surface-enhanced Raman spectroscopy (SERS) relies on the formation of precise and stable plasmonic nanogaps to achieve high sensitivity and reproducibility. However, the dynamic nature of plasmonic nanoparticles often leads to uncontrolled nanogap fluctuations, limiting SERS signal stability. Here, we present a novel strategy for stable plasmonic nanogap engineering by employing covalent organic frameworks (COFs) to confine the motion of Au nanoparticles at the nanoscale. The porous and rigid structure of COFs effectively restricts nanoparticle mobility, ensuring the formation of well-defined and fixed plasmonic nanogaps. Time-resolved studies demonstrate that COFs-modified Au nanoparticles exhibit a highly stable SERS signal, with only 37.5 % intensity fluctuations, whereas bare Au nanoparticles undergo significant blinking, leading to an 82.8 % signal loss over 300 s. This confinement strategy not only contributes to enhancing SERS signal stability but also holds promise as a platform for ultrasensitive molecular detection, biosensing, and in situ chemical analysis, offering potential insights for the development of precisely engineered nano-plasmonic sensing systems.

Salmonellais a rapidly spreading and widespread zoonotic infectious disease that poses a serious threat to the safety of both poultry and human lives. Therefore, the timely detection ofSalmonellain foods and animals has become an urgent need for food safety. This work describes the construction of an aptamer-based sensor forSalmonelladetection, using Fe3O4magnetic beads and Ag@Au core–shell nanoparticles-embedded 4-mercaptobenzoic acid (4MBA). Leveraging the high affinity between biotin and streptavidin, aptamers were conjugated to Fe3O4magnetic beads. These beads were then combined with Ag@4MBA@Au nanoparticles functionalized with complementary aptamers through hydrogen bonding and π-π stacking interactions, yielding a SERS-based aptamer sensor with optimized Raman signals from 4MBA. When target bacteria are present, aptamer-conjugated magnetic beads exhibit preferential binding to the bacteria, leading to a decrease in the surface-enhanced Raman scattering (SERS) signal. And it was used for the detection of five different serotypes ofSalmonella, respectively, and the results showed that the aptamer sensor exhibited a good linear relationship between the concentration range of 102–108CFU/mL and LOD is 35.51 CFU/mL. The SERS aptasensor was utilized for the detection of spiked authentic samples with recoveries between 94.0 and 100.4%, which proved the usability of the method and helped to achieve food safety detection.

Surface-enhanced Raman scattering (SERS), as one of the most powerful analytical methods, undertakes important inspection tasks in various fields. Generally, the performance of an SERS-active substrate relies heavily on its structure, which makes it difficult to integrate multiple-functional detectability on the same substrate. To address this problem, here we designed and constructed a film of graphene/Au nanoparticles (G/Au film) through a simple method, which can be conveniently transferred to different substrates to form various composite SERS substrates subsequently. By means of the combination of the electromagnetic enhancement mechanism (EM) and the chemical enhancement mechanism (CM) of this structure, the film realized good SERS performance experimentally, with the enhancement factor (EF) approaching ca. 1.40 × 105. In addition, the G/Au film had high mechanical strength and had large specific surface area and good biocompatibility that is beneficial for Raman detection. By further transferring the film to an Ag/Si composite substrate and PDMS flexible film, it showed enhanced sensitivity and in situ detectability, respectively, indicating high compatibility and promising prospect in Raman detection.

A facile strategy has been reported to obtain on-line fresh Ag as surface-enhanced Raman scattering (SERS) active substrates by making AgCl nanoparticles exposed to the laser beam of Raman spectrometer. The composition and morphology of AgCl nanoparticles were characterized by X-ray diffraction (XRD), UV–visible (UV–vis) spectroscopy and scanning electron microscopy (SEM). The laser-driven evolvement and possible formation mechanism of cubic AgCl nanoparticles to Ag/AgCl composites were also investigated by transmission electron microscopy (TEM), energy dispersive X-ray spectroscopy (EDS) and high-resolution transmission electron microscopy (HRTEM). Raman measurements demonstrate that the fresh Ag nanoparticles with a few defects have a prominent SERS sensitivity to probe molecules, such as the 4-mercaptopyridine (4-Mpy) and 4-aminothiophenol (PATP) molecules. The SERS intensity of 4-Mpy and PATP increases up to the maximum when the laser irradiation time is prolonged to 50 s, which corresponds to the defect extent and the proportion of fresh Ag in the Ag/AgCl composites. This work provides a simple, efficient and feasible approach for obtaining on-line fresh Ag as SERS substrates.

A surface-enhanced Raman scattering (SERS)–fluorescence dual-mode pH-sensing method based on Janus microgels was developed, which combined the advantages of high specificity offered by SERS and fast imaging afforded by fluorescence. Dual-mode probes, pH-dependent 4-mercaptobenzoic acid, and carbon dots were individually encapsulated in the independent hemispheres of Janus microparticles fabricated via a centrifugal microfluidic chip. On the basis of the obvious volumetric change of hydrogels in different pHs, the Janus microparticles were successfully applied for sensitive and reliable pH measurement from 1.0 to 8.0, and the two hemispheres showed no obvious interference. The proposed method addressed the limitation that sole use of the SERS-based pH sensing usually failed in strong acidic media. The gastric juice pH and extracellular pH change were measured separately in vitro using the Janus microparticles, which confirmed the validity of microgels for pH sensing. The microparticles exhibited good stability, reversibility, biocompatibility, and ideal semipermeability for avoiding protein contamination, and they have the potential to be implantable sensors to continuously monitor pH in vivo.

A monolayer-MoS 2 /ZnO-microtube heterostructure is fabricated for SERS detection. • Deep defects and optical resonances promote SERS performance in monolayer-MoS 2. • A limit of detection of 10 −12 M with an enhancement factor of 2.4 × 10 7 is realized. • Monolayer-MoS 2 /ZnO-microtube heterostructure is reusable by photodegradation. Two-dimensional transition metal dichalcogenides (2D-TMDCs) have attracted considerable attention as substrate enhancers for surface-enhanced Raman spectroscopy (SERS) due to their extraordinary chemical stability and spatial homogeneity. Unfortunately, the enhancement factors of 2D-TMDCs are limited by weak optical confinement within the atomically-thick layer. Construction of 2D-TMDCs/semiconductor-microcavity heterostructures significantly boosts the light confinement by optical resonances as well as charge-transfer (CT) resonances by energy-level alignment, by which the SERS performance is improved. Here we fabricate a monolayer-MoS 2 /ZnO-microtube (ML-MoS 2 /ZnO-MT) heterostructures, of which the ML-MoS 2 was capped onto the ZnO-MT via van der Waals forces. The deep-defect level in ZnO-MT grown by optical vapor supersaturated precipitation provide an extra CT channel transferring carriers to ML-MoS 2 for chemical enhancement. Meanwhile, the Fabry-Perot resonances supported in the ZnO-MT promote exciton, molecule and CT resonances as well as fluorescence resonance energy transfer between ML-MoS 2 and analyte molecules, by which a SERS enhancement factor of 2.4 × 10 7 with the limit of detection down to 10 −12 M for methylene blue is achieved. The ML-MoS 2 /ZnO-MT also exhibits the pronounced catalytic behavior, enabling the SERS heterostructure reusable by photodegradation of analyte molecules under 5-min ultraviolet irradiation. This work provides a new approach for highly-sensitive and recyclable plasmon-free SERS substrates for trace detection. Download: Download high-res image (69KB) Download: Download full-size image

Surface-enhanced Raman scattering (SERS) is a highly sensitive and molecule-specific analytical technique with broad applications in biomedicine and chemistry. However, the poor reproducibility caused by the non-uniform distribution of “hot spots” in conventional substrates has hindered its further development and practical use. In this work, stainless-steel carriers with different pore sizes were employed to construct free-standing membranes (FSMs) by combining them with nanoparticle suspensions. The influence of pore size on FSM formation was systematically investigated. Specially, SERS detection of 10 −5 M rhodamine 6G as a model analyte, relative standard deviations was as low as combined 1.7% demonstrating excellent signal reproducibility. When applied to embryo culture media, relative standard deviations values of 7.76% and 7.49% were obtained for FSMs derived from high-quality and low-quality embryos, respectively. The proposed FSM fabrication method is highly reproducible, simple, and cost-effective, offering a promising strategy for optimizing and extending the practical applications of SERS.

The early detection of esophageal cancer is crucial for enhancing patient survival. Surface-enhanced Raman scattering (SERS), renowned for its precise detection capabilities of low-concentration molecules and organisms, holds immense promise for early diagnosis. However, the use of silver nanoparticles to enhance the Raman signal can disrupt the detection process. To address this problem in this study, we synthesized polyethylene glycol (PEG)-coated silver nanocubes (PEG-AgNCs) as stable, label-free, and low-toxicity nanoprobes for detecting esophageal cancer. AgNCs with sharp corners and edges are selected as the SERS substrate to increase the strength of the Raman signal. To minimize the toxicity of AgNCs, the surface is coated with PEG, which exhibits outstanding biocompatibility. The SERS effectiveness of PEG-AgNCs was evaluated using 4-mercaptobenzoic acid as a Raman probe molecule, and the toxicity of PEG-AgNCs at different concentrations was investigated using human keratinocytes (HaCaT) and human esophageal carcinoma cells (TE-1). Results showed that PEG-AgNCs can effectively reduce the toxicity of AgNCs while maintaining a strong SERS signal. Remarkable differences in Raman spectra were observed between HaCat and TE-1 cells within the spectral range of 588–768 cm −1 , indicating the potential of PEG-AgNCs in the early detection of esophageal cancer using Raman detection. Our study highlights the potential of developing low-toxicity SERS biosensors for biological and chemical analyses.

Surface-enhanced Raman Spectroscopy (SERS) has emerged as a powerful and sensitive technique for the detection of various molecules, including gaseous analytes. However, the development of efficient, stable, and reusable SERS substrates for gaseous molecule remains a challenge. This work introduces a novel Cu-Ag@ZIF-8 film for SERS detection of gaseous molecule, which consists of Ag nanoparticles (AgNPs) core and ZIF-8 porous shell on Cu foils. With strong enhancement of electromagnetic field by AgNPs core and effective caption of gaseous molecule by ZIF-8 porous shell, this substrate exhibits good SERS performance of p-aminothiophenol (PATP) and 4-mercaptophenol (DP) gaseous molecules, whose limit of detection (LOD) could reach 9.12 × 10 −10 M and 9.91 × 10 −9 M, respectively. Furthermore, the detection of gaseous benzaldehyde diffused by oil paint is explored by this developed SERS substrate for application. To achieve accurate identification of harmful gaseous molecule in complex volatile organic compounds (VOC), the deep learning (DL) approach is performed based on SERS spectra obtained from this Cu-Ag@ZIF-8 film. Our results demonstrate that the identification of gaseous benzaldehyde in VOC diffused from commercial oil paint has a high accuracy at 97.7% with the help of ANN algorithms, which points out the Cu-Ag@ZIF-8 film has good performance and huge potential for SERS detection of gaseous molecule.

Thoracolumbar fascia (TLF) plays an important role in the biomechanics of the spine. TLF often causes low back pain (LBP), leading to limb movement disorders and numbness. At present, the clinical diagnosis mainly depends on doctors' clinical experience and magnetic resonance imaging. In order to make the diagnosis more convenient and accurate, we designed a "sandwich" biosensor based on surface-enhanced Raman scattering (SERS) to detect C-reactive protein (CRP) and serum amyloid A (SAA) in fascia. In this study, Ag nanocubes (AgNCs) were used as the substrate, so that the antibodies of CRP and SAA were modified on the substrate surface. Then substrate combined with antigen to form the bottom layer of the sandwich. 4-Mercaptobenzoic acid (4-MBA) and Nairn blue chloride (NBA) were used as signalling molecule probes. Hollow gold nanoparticles (HGNPs) modified with (EDC / NHS) enabled the attachment of antibodies to CRP and SAA to the surface of HGNPs. Eventually, the complex formed by the antigen-antibody reaction reacted with the sandwich substrate after the modification reaction to form the final "sandwich" structure. We successfully applied it to the specific detection of inflammatory markers in fascial tissue of TLF. The results showed that it had high stability and repeatability, which can bring some help for clinical work.

Excessive thiram residues in food have the potential to negatively impact human health. Hence, the development of a convenient and fast detection method is highly desirable. In this study, an efficient, repeatable, and sensitive surface-enhanced Raman scattering (SERS) active chip was manufactured via a low-cost colloidal lithography technique. The plasmonic structure was composed of a series of silver nanospheres and nanowires. Interestingly, this type structure creates a nanocavity space with a characteristic geometry generating a strong electromagnetic field coupling. The finite-different time-domain software was employed to simulate the electromagnetic field distribute on the nanocavity. Accordingly, SERS active chip that displays ultra-low concentration detection of thiram (10−11 M) was realized. Moreover, the excellent reproducibility of thiram (10−6 M) practical detection on an apple pericarp has great potential for application in food safety.

The variation of tumor-associated metabolites in extracellular microenvironment timely reflects the development, the progression and the treatment of cancers. Conventional methods for metabolite detection lack the efficiency to grasp the dynamic metabolic alterations. Herein, we developed a SERS bionic taster which enabled real-time analysis of extracellular metabolites. The instant information of cell metabolism was provided by the responsive Raman reporters, which experienced SERS spectral changes upon metabolite activation. Such a SERS sensor was integrated into a 3D-printed fixture which fits the commercial-standard cell culture dishes, allowing in-situ acquisition of the vibrational spectrum . The SERS taster can not only accomplish simultaneous and quantitative analysis of multiple tumor-associated metabolites, but also fulfill the dynamic monitoring of cellular metabolic reprogramming, which is expected to become a promising tool for investigating cancer biology and therapeutics.
