It is difficult to effectively treat bacterial osteomyelitis using photothermal therapy or photodynamic therapy due to poor penetration of light. Here, a microwave (MW)-excited magnetic composite of molybdenum disulfide (MoS 2 ) / iron oxide (Fe 3 O 4 ) is reported for the treatment of bacteria-infected osteomyelitis. In in vitro and in vivo experiments, MoS 2 /Fe 3 O 4 is shown to effectively eradicate bacteria-infected mouse tibia osteomyelitis, due to MW thermal enhancement and reactive oxygen species (ROS) ( 1 O 2 and ·O 2 – ) production under MW radiation. In addition, the mechanism of MW heat generation is proposed by MW network vector analysis. By the density functional theory and finite element method, the ROS generation mechanism is proposed. The synergy or conductive network between dielectric MoS 2 and magnetic Fe 3 O 4 can reach both enhancement of the dielectric and magnetic attenuation capability. In addition, abundant interfaces are generated to enhance the attenuation of electromagnetic waves by MoS 2 and Fe 3 O 4, introducing multiple reflections and interfacial polarization. Therefore, MoS 2 /Fe 3 O 4 has excellent MW absorption ability based on the synergy or conductive network between MoS 2 and magnetic Fe 3 O 4 as well as multiple dielectric reflections and interfacial polarization.

Staphylococcus aureus ( S. aureus ) is a common pathogen that is dangerous to humans’ health. Herein, a novel upconversion fluorescent biosensor based on fluorescence resonance energy transfer from aptamer-labeled upconversion nanoparticles (UCNPs-apt) as donor and cobalt oxyhydroxide (CoOOH) nanosheets as acceptor was designed to detect S. aureus in complex matrices . The principle of the work relies on fluorescence resonance energy transfer as UCNPs-apt can self-assemble on CoOOH nanosheet surfaces by van der Waals forces to effectively quench the fluorescence. When S. aureus was added, the aptamer was able to preferentially capture the target, resulting in the dissociation of donor and acceptor and the recovery of fluorescence. The structure and morphology of the nanostructures were assigned in detail by a series of characterizations, and the energy transfer mechanism was evaluated by time-resolved lifetime measurements. Under the optimal conditions, a linear calibration plot was obtained in a concentration range of 45–4.5 × 10 6 CFU/mL with a limit of detection of 15 CFU/mL. In addition, the proposed biosensor was used for S. aureus detection in real samples (e.g., pork, beef), and the detection result showed no significant difference ( p > 0.05) compared with the conventional plate count approach. Hence, the fabricated biosensor holds a potential application for S. aureus in food analysis and public health. Graphical abstract

With the abuse of antibiotics, bacterial antibiotic resistance is becoming a major public healthcare issue. Natural plants, especially traditional Chinese herbal medicines, which have antibacterial activity, are important sources for discovering potential bacteriostatic agents. This study aimed to develop a fast and reliable method for screening out antimicrobial compounds targeting the MRSA membrane fromPsoralea corylifoliaLinn. seed. A UPLC-MS/MS method was applied to identify the prenylated flavonoids in major fractions from the extracts ofPsoralea corylifoliaLinn. seed. The broth microdilution method was used to determine the minimum inhibitory concentrations (MICs) of different fractions and compounds. The morphological and ultrastructural changes of MRSA were determined by scanning electron microscopy (SEM). The membrane-targeting mechanism of the active ingredients was explored by membrane integrity assays, membrane fluidity assays, membrane potential assays, ATP, and ROS determination. We identified eight prenylated flavonoids inPsoralea corylifoliaLinn. seed. The antibacterial activity and mechanism studies showed that this type of compound has a unique destructive effect on MRSA cell membranes and does not result in drug resistance. The results revealed that prenylated flavonoids inPsoralea corylifoliaLinn. seeds are promising candidates for the development of novel antibiotic agents to combat MRSA-associated infections.Keywords:methicillin-resistantStaphylococcus aureus;Psoralea corylifoliaLinn. seed;prenylated flavonoids;UPLC-MS/MS;bacterial membrane

The healing of chronic diabetic wounds is a common and significant challenge in the medical field. Despite extensive efforts, the development of hydrogel dressings with satisfactory functionality remains an ongoing concern. In this study, a multifunctional hydrogel wound dressing (PAN/Ag-PLG) with adhesion, antibacterial, hemostatic, and other properties, which can effectively repair diabetic wounds infected with methicillin-resistant Staphylococcus aureus (MRSA), is presented. The hydrogel dressing is composed of gallic acid (GA)-functionalized polylysine (PL)-reduced silver nanoparticles (Ag-PLG), oxidized hyaluronic acid (OHA), and cross-linked polyacrylic acid grafted with N -hydrosuccinimide ester. Notably, compared to most conventional wound dressing that lack adhesion or are difficult to remove, the prepared hydrogels exhibit excellent adhesion and mild stimulation-triggered detachment. In vitro and in vivo experiments reveal that the PAN/Ag-PLG hydrogel exhibits outstanding biocompatibility and antibacterial properties and promotes diabetic wound repair by reducing oxidative damage and promoting cell migration and angiogenesis. The smart PAN/Ag-PLG hydrogel reported in this study provides an approach for the potential clinical development of painless antibacterial dressings.

A ratiometric fluorescence and colorimetry dual-mode nano-biosensor has been established for Staphylococcus aureus ( S. aureus ) determination. The prepared approaches of Manganese dioxide nanosheets (MnO 2 NSs) and carbon dots (BCDs) were facile, efficient and labor-saving and MnO 2 NSs-mediated fluorescence quenching and oxidation could amplify detection signals. The dual-mode determination had a broad linear range of 37 ∼ 3.7 × 10 6 CFU/mL and low detection limits of 9 CFU/mL (ratiometric fluorescence) and 22 CFU/mL (colorimetry). Meanwhile, the method was applied in real samples with recovery ranging of 90 ∼ 102% and RSD < 4.44%, which was an insignificant difference with standard plate counting. The new dual-mode approach of S. aureus possesses the advantages of superior sensitivity, precision, accuracy and specificity. Moreover, the dual-mode nano-biosensor can be adopted in other foodborne pathogens determination by changing corresponding aptamers and provide an enlightenment in monitoring food safety.

The purpose of the present study was to utilize solution blow spinning to fabricate curcumin (CUR)-loaded polycaprolactone (PCL)/carboxymethyl chitosan (CMCH) nanofibers with photodynamic inactivation activity for inactivation of S. aureus . The results of ATR-FTIR and XRD spectra demonstrated the successful encapsulation of CUR without disrupting the structure of the nanofibers and increasing the crystallinity of the nanofibers. After the encapsulation of CUR, the thermal stability, tensile strength, and hydrophobicity of the PCL/CMCH nanofibers were improved. Besides, the CUR-loaded PCL/CMCH nanofibers possessed good biocompatibility and low toxicity, and could release CUR sustainably over 120 h. Moreover, the obtained nanofibers showed better inhibition of S. aureus under blue light irradiation at a wavelength of 450 nm compared to the no-light condition. The above results suggested that the developed nanofibers with photodynamic inactivation can be applied as food packaging materials to reduce the impact of S. aureus on human wellness.

To reduce the harmful effects of Staphylococcus aureus ( S. aureus ) infection during wound healing, the exploitation of novel hydrogel-based wound dressings with antibacterial and anti-inflammatory abilities is necessary. Herein, we proposed a novel alginate-based antibacterial hydrogel (SPT) consisting of boronate ester bonds formed between tannic acid (TA) and phenylboronic acid (PBA). TA not only functions as a crosslinker of the hydrogel, but also as an antibacterial agent with antioxidant and anti-inflammatory capacities, which achieved continuous release in response to the mildly-acidic bacterial infection microenvironment. 1 H-nuclear magnetic resonance ( 1 H-NMR), Fourier transformed infrared spectroscopy (FTIR), ultraviolet-visible (UV-vis) spectroscopy, rheometer and scanning electron microscope (SEM) verified the successful production of hydrogel and pH-dependent release of TA. Furthermore, its bactericidal and antioxidant activities were intensively assessed in vitro and in vivo . The experimental results illustrated that the SPT hydrogel possessed good antimicrobial ability against S. aureus and anti-inflammatory capacity due to the pH-responsive release of TA. In addition, the SPT hydrogel exhibited satisfying antioxidant capacities in the free radical assays and ROS fluorescence staining. To sum up, the prepared SPT hydrogel showed good antibacterial effect against S. aureus and anti-inflammatory capacity. Therefore, it has great potential to be employed as wound dressing in clinical applications.

Drug-resistant bacterial biofilm-associated infections pose a significant threat to human health, as the physical barrier and protective matrix of biofilms enable bacterial evasion of antibiotic effects, leading to refractory infections. To address this challenge, we develop a novel photosensitizer PY and its sulfonated zwitterionic derivative PYSO 3 . Both photosensitizers exhibit aggregation-induced emission (AIE) properties coupled with exceptional photodynamic and photothermal performance in aggregates. Sulfonated zwitterionic modification significantly enhance the biological activity of PYSO 3 , demonstrating potent bactericidal efficacy against methicillin-resistant Staphylococcus aureus (MRSA) and mature biofilms under laser irradiation. Mechanistically, PYSO 3 -mediated combinational photodynamic therapy and photothermal therapy disrupt critical bacterial survival pathways, including quorum sensing systems, glycolysis/gluconeogenesis pathways, and oxidative stress homeostasis, thereby inhibiting normal bacterial metabolism and biofilm formation. In vivo experiments reveal remarkable antibacterial performance with accelerated wound healing rates and favorable biosafety profiles. This AIE-active zwitterionic photosensitizer not only holds great promise for combating MRSA infections but also provides new design strategies for developing multifunctional antimicrobial agents targeting drug-resistant pathogens.

Staphylococcus aureus (SA) is a predominant foodborne pathogen, frequently associated with nutrient-rich matrices. However, rapid and ultrasensitive detection of SA in complex food samples remains a formidable challenge. In this work, we developed a relay biosensing system for detecting SA-16S rDNA that combines a magnetic bead-anchored APE1 DNA walker with an asymmetric CRISPR/Cas12a system. The magnetic bead-anchored DNA walker enables efficient separation of SA-targets from complex food matrices, while generating abundant activators through APE1-mediated catalytic cleavage. Employing spatially decoupled reactions with differential crRNAs, the asymmetric Cas12a system achieves programmable cascade amplification. Dual-mode fluorescence/electrochemical readout supports both laboratory analysis and on-site detection. The system demonstrates exceptional sensitivity, capable of detecting trace amounts of SA-16S rDNA (7.5 aM by fluorescence, 1.0 fM by electrochemistry) and SA (4 CFU/mL by fluorescence, 34 CFU/mL by electrochemistry). Overall, this work provides a modular and programmable biosensing strategy for bacterial identification, thereby facilitating advanced nucleic acid analysis within complex biological matrices.

BackgroundThe emergence of linezolid−resistant methicillin−susceptible Staphylococcus aureus (MSSA) is a growing clinical threat, but the underlying mechanisms are unclear. Here, we generated stable resistant derivatives and used a multi−omics strategy to investigate the transcriptional and metabolic regulation underlying linezolid resistance in MSSA.MethodsA clinical isolate of linezolid-susceptible S. aureus (strain S. aureus_97) was used as the parental strain. A stable linezolid-resistant derivative (designated LZR) was generated through stepwise induction with subinhibitory concentrations of linezolid. The genetic stability of the resistant phenotype was subsequently confirmed via phenotypic stability assays. To elucidate the mechanisms underlying resistance, comparative transcriptomic (RNA-seq) and untargeted metabolomic (LC-MS/MS) analyses were performed to identify differential gene expression and metabolic profiles between the resistant (LZR) and susceptible (LSS) strains.ResultsThe successfully induced linezolid-resistant derivative (LZR) exhibited a 16-fold increase in minimum inhibitory concentration (MIC) against linezolid (32 μg/mL) compared with the parental strain (2 μg/mL), and this resistant phenotype remained stable after 50 serial passages in antibiotic-free medium. CCCP was observed to reduce the MIC of LZR in a concentration-dependent manner. Transcriptomic and metabolomic analyses revealed widespread transcriptional reprogramming and metabolic perturbations in the resistant strain. Differentially expressed genes were mainly enriched in pathways associated with oxidative phosphorylation, ribosome function, and glycerolipid metabolism, while differential metabolites were primarily involved in the biosynthesis of secondary metabolites and amino acid metabolism. Integrated analysis identified key co-enriched pathways with distinct regulatory patterns, which collectively remodeled bacterial material and energy metabolism and were closely associated with the emergence of linezolid resistance.ConclusionA stable linezolid-resistant MSSA strain was developed in this work, revealing an intricate network of regulatory interactions that drive resistance at both the transcriptional and metabolic levels. These findings provide an experimental foundation and theoretical basis for further understanding the mechanisms of linezolid resistance in MSSA and for developing strategies to combat resistance.

Hydrogel dressings that can simultaneously curb infection and dampen inflammation are particularly attractive for accelerating multidrug-resistant bacteria-infected wound healing. However, clinical translation of many reported hydrogels is often constrained by multi-component formulations, complicated preparation procedures, and insufficient sprayability, which limit conformal coverage on irregular wound beds. Polyitaconate possesses intrinsic antibacterial and anti-inflammatory activities, yet its weak antioxidative properties and lack of thermogelling properties limit its utility in infected wound healing. In this study, we develop a curcumin-modified polyitaconate-based hydrogel (FPIC) that integrates antibacterial, anti-inflammatory, and antioxidant properties. FPIC hydrogel exhibits thermoresponsive gelation, injectability, sprayability, and shear-thinning properties, allowing convenient close contact with complex wound geometries. FPIC hydrogel inhibits bacterial growth, moderates excessive inflammatory responses, and eliminates reactive oxygen species while promoting angiogenesis, thereby facilitating the shift from the inflammatory phase to the proliferative phase of healing. Moreover, FPIC hydrogel enhances collagen deposition, speeds re-epithelialization, and supports balanced tissue remodeling, enabling improved skin regeneration with a lower propensity for fibrosis. These findings establish FPIC hydrogel as a multifunctional and highly bioactive polyitaconate-based hydrogel dressing with strong promise for treating infection- and inflammation-associated wounds and advancing toward clinical use.

Rapid and accurate pathogen detection is critical for optimizing clinical decision-making within precision medicine frameworks, specifically for identifying Staphylococcus aureus (S. aureus) in wound infections. Most of conventional detection methods suffer from procedural complexity, protracted analysis times, and insufficient sensitivity, thereby constraining their clinical deployment. In this work, an optical fiber surface plasmon resonance (SPR) sensor enhanced by Ti 3 C 2 T x MXene was developed for detecting S. aureus , biofunctionalized with goat anti-human immunoglobulin G (IgG). The optimal thickness of Ti 3 C 2 T x MXene layer, determined to be approximately 94 nm based on maximizing figure of merit (FOM) of 17.88 RIU −1 , yielded high sensitivity and low full width at half maximum (FWHM). By monitoring SPR wavelength shifts in the transmission spectra, the sensor was employed to detect S. aureus with a low limit of detection (LOD) of 1.14 CFU/mL and a high sensitivity of 3.063 nm/log (CFU/mL). It exhibited excellent linearity across the concentration range of 10 2 –10 8 CFU/mL, alongside high stability and selectivity. Validation using S. aureus -infected wound tissue from diabetic murine models exhibited a precision exceeding 97 %. This technology represents a promising avenue for developing next-generation, high-performance biosensing platforms with significant translational potential in precision medicine diagnostics.
