Fabrication of a pH-responsive carrier for antifoulant delivery and investigation of its controlled release properties

Chuanjun Gu,Zhanping Zhang,Yuhong Qi,Miaomiao Ma,Chen Zhang

A pH-responsive nanocomposite enables on-demand antifoulant release. • Sharp release triggered by biofilm acidification minimizes environmental leakage. • The smart coating retains PDMS properties with enhanced pH-switchable antifouling. In this work, a pH-responsive antifouling system was developed to address the problem of inefficient use of antifoulants in antifouling coatings. A nanocomposite (MSN-MOF-CA@BIT) was fabricated via a layer-by-layer (LBL) assembly strategy, integrating mesoporous silica with a metal-organic framework for controlled antifoulant (BIT) release. This composite was then incorporated into a polydimethylsiloxane (PDMS) coating matrix. The composite powder exhibited clear pH-dependent release kinetics, with minimal release at seawater pH and accelerated release under acidic conditions. Correspondingly, it demonstrated potent, pH-enhanced antibacterial activity, achieving rapid bacterial eradication in acidic environments. Within the PDMS coating, the composite showed excellent dispersibility, as confirmed by laser confocal microscopy. The coating’s surface roughness increased moderately with filler loading, while its low surface free energy was preserved (<20 mJ/m²). Mechanically, the composite acted as a reinforcing filler, increasing the fracture elongation to 345.6% and the tensile strength to 0.628 MPa at a 10 wt.% loading. Most importantly, the composite coating retained its pH-responsive functionality. Immersion tests confirmed the stability of the encapsulated BIT during coating preparation and its on-demand release in service. Bacterial adhesion assays directly verified the coating's pH-responsive antifouling efficacy: significantly less biofilm formed under acidic conditions triggered by bacterial metabolism, demonstrating the system's ability to prevent fouling expansion proactively. In summary, this study successfully fabricates and validates a novel nanocomposite-based pH-responsive controlled-release coating. It achieves efficient, environmentally triggered antifoulant utilization while maintaining and even enhancing the key physical properties of the PDMS coating, showing great promise for sustainable antifouling technologies. Preparation of hydrophobically modified sodium alginate-chitosan hydrogel and its characteristics in urea controlled-release fertilizers

Feifei Qu,Xueyi Zhai,Hongyao Bai,Jianwei Tang,Mengxiao Zheng,Peng Wang,Yong Liu,Baoming Wang,Pengfei Liu

Urea is widely used in agriculture because of its high nitrogen content and low cost, but its utilization efficiency is often limited by volatilization, leaching, and nitrification-denitrification losses. In this study, sodium alginate (SA) was hydrophobically modified by free-radical grafting with methyl methacrylate (MMA) to obtain SA-PMMA, which was subsequently ionically cross-linked with chitosan (CS) to construct hydrogel carriers for controlled-release urea fertilizers. The resulting materials were characterized by FTIR, XRD, TG-DTG, XPS, and SEM-EDS, confirming successful grafting and indicating interactions between urea and the SA-PMMA/CS matrix. The effects of grafting ratio on water absorption, urea loading, nitrogen release, degradation, and fertilizer performance were systematically investigated. Among the five formulations, sample Z3 exhibited the best overall release behavior, with a cumulative nitrogen release of 77.33% over 56 days, meeting the national standard for slow-release fertilizers. Release-kinetic analysis showed that nitrogen release was best described by the first-order model and was governed by Fickian diffusion. In spinach pot experiments conducted at the same nitrogen input, the hydrogel fertilizers, especially Z3, promoted root development, leaf expansion, chlorophyll a ccumulation, dry matter production, and plant nitrogen uptake compared with conventional urea. These results demonstrate that hydrophobically modified SA/CS hydrogels are promising carriers for improving urea retention, regulating nitrogen release, and enhancing agricultural nitrogen-use efficiency.

Self-healing bio-based membranes for eco-efficient fertilizers: Abrasion resistance, controlled release and soil biodegradability

Zekun Wang,Ziyao Li,Zhen Yu,Ronglei Li,Wenshuo Liu,Guiwei Wang,Xiaoqi Wang,Bin Gao,Yuanyuan Yao,Yuechao Yang

Bio-based membrane shells with excellent abrasion resistance, high elasticity and self-healing ability hold significant potential for enhancing the quality and utilization of membrane-coated bio-based controlled-release fertilizers (BCRFs). In this study, we developed a simple and practical method for preparing novel bio-based membrane shells that exhibited superior elasticity (>200% water absorption and expansion without rupture), reduced porosity (−13.07%), and enhanced tensile and tensile strength (reaching 1003.8% and 15.704 MPa). The resulting self-healing bio-based controlled-release fertilizer (SBCRF) showed higher compressive strength (withstanding 90 N pressure), extended nitrogen release longevity (+40%) and excellent wear resistance (with overlapped nutrient release curves before and after abrasion). Crucially, the strategic cross-linking oxime-carbamate bonds and hydrogen bonds into the polyurethane network structure conferred SBCRF with self-healing function. Molecular dynamics simulations further confirmed the reversible reorganization of broken chemical bonds with restored energy, facilitating repair of the damaged region. These attributes collectively improved the accuracy and stability of nutrient release from SBCRF, while enabling soil biodegradation of the shells (with 43.77% mass loss after 180 days). Field trials in small cabbage demonstrated an 8.6% increase in nitrogen utilization efficiency compared to conventional BCRFs. This study provides a theoretical and technological foundation for the development of stable and efficient BCRFs, advancing the goals of green and intelligent agriculture.

Integrated platform for EV separation and controlled release based on gelatin microspheres for diabetic wound treatment

Zhang Yiqing,Huang Qilin,Meng Hao,Chen Junli,Hao Yaying,Liu Xi,Xu Yangmengyuan,Ma Liqian,Xu Zhan,Ma Kui,Hu Wenzhi,Pan Xiaohua,Fu Xiaobing,Zhang Cuiping

Diabetic wounds exhibit impaired healing owing to multiple pathological factors and current treatment options remain limited, underscoring the urgent need for novel therapeutic strategies. Extracellular vesicles (EVs) derived from mesenchymal stem cells (MSCs) are emerging as promising candidates for wound healing. However, the process from isolation to application of EVs is complex and time-consuming, which probably influences the activity and the therapeutic effects of EVs. To address these issues, we created the integrated platform for EV separation and controlled release. Gelatin microspheres (GMS) were prepared and functionalized with the variable domain of heavy-chain-only antibodies (GMS-VHH). GMS-VHH enabled the efficient capture of CD9-positive EVs in the culture supernatant of MSCs, forming EVs-loaded gelatin microspheres (GMS@EVs). The characterization of EVs demonstrated that the GMS-VHH isolation method outperformed traditional ultracentrifugation in terms of EV structural integrity. GMS@EVs exhibited excellent biocompatibility and promoted the proliferation of epidermal keratinocytes in vitro. In addition, the controlled release and the therapeutic efficacy of GMS@EVs were also observed in the treatment of diabetic wounds. Transcriptomic analysis illustrated that the wound healing-related genes were significantly upregulated and the pro-inflammatory genes as well as the wound healing-impaired genes were downregulated. Collectively, this study introduces a “ready-to-use” platform for EV isolation and controlled release, providing a promising strategy for diabetic wound repair.

Continuous and scalable preparation of environmentally friendly controlled-release pesticide nanocapsules using low-energy-input microfluidics

Ruixi Xiao,Zejun Cao,Bowen Yuan,Yuxia Chen,Xiaona Li,Jiangtao Dong,Xuezhong Du

Nanopesticides can utilize the unique characteristics of nanoscales to maximize pesticide efficacy. The continuous and scalable preparation of nanopesticides is an industrial demand for their practical application. A universal low-energy-input microfluidic platform was developed for high-throughput continuous preparation of pesticide nanocapsules. The pesticide-loaded 1-butanol/water microemulsions through the co-assembly of sodium lignosulfonate (SL) and cetyltrimethylammonium chloride, were developed and then mineralized through the coordination interactions between Fe 3+ and SL. The prepared pyraclostrobin nanocapsules (Pyr NCs) had a uniform size of approximately 150 nm with a high Pyr loading capacity of 75% and excellent cold/hot storage stability. Encapsulation of Pyr significantly improved its photostability, and the nanocapsules enabled them to better adhere to crop leaves to resist rainwater erosion. Pyr NCs had a photothermal conversion efficiency of up to 42.9% and achieved multiple stimuli-responsive controlled pesticide release owing to physiological changes in infected crops. Both in vitro ( Fusarium graminearum ) and in vivo ( Botrytis cinerea ) evaluations demonstrate that Pyr NCs had better fungicidal efficacy than commercial formulations. Pyr NCs not only demonstrated biosafety to target crops but also enhanced biosafety to non-target organisms. The environmentally friendly controlled-release pesticide nanocapsules using the microfluidic platform have broad application prospects in green and sustainable agriculture.

Preparation of controlled-release fresh-keeping cards and application in strawberry treated with simulated transport vibration

Yinghui Ding,Weizhuo Li,Jing Ma,Xizheng Zhang,Zhicheng Zhong,Jumin Hou

Strawberries are highly susceptible to decay because of high moisture, microbial invasion and vibration during transportation and storage. In this study, the hydroxypropyl-β-cyclodextrin (HP-β-CD) was used to encapsulate cinnamaldehyde to form hydroxypropyl-β-cyclodextrin-cinnamaldehyde (CA@HP-β-CD). Afterwards, the CA@HP-β-CD was applied to prepare the controlled-release fresh-keeping cards using food-grade absorbent cardboard and polyvinyl alcohol water-soluble film for strawberry storage after different simulated transport vibrations. During storage, the polyvinyl alcohol water-soluble film exhibited disintegration due to higher humidity environment, releasing cinnamaldehyde and lowering strawberry decay. According to the simulated transportation for 6 h and 48 h, the decay rates of strawberries treated with the cards were (16.67 ± 5.77) % and (23.33 ± 5.77) %, respectively, at the 9th day. It suggested that the cards could mitigate spoilage to extend the strawberry shelf life. It aligned closely with real-world fruit transportation scenarios, providing valuable insights for post-transport preservation strategies.

Multifunctional nanocomposite hydrogel with dual-factor controlled release for stem cell recruitment, immunomodulation and bone remodeling

Yang Jiao,Haixin Wang,Mingyu Zhao,Huiling Liu,Peng Shen,Gang Wu,Tim Forouzanfar,Franklin R. Tay,Li Miao,Hongchen Liu,Qian Liu

Bone regeneration is a dynamic, multistage process requiring the orchestrated interplay of cellular recruitment, immune modulation, and balanced osteoblast-osteoclast activity. To address this clinical challenge, we developed a nanocomposite hydrogel-based drug delivery system integrating gelatin methacrylate (GelMA) and silk fibroin methacrylate (SFMA) scaffolds containing mesoporous silica nanoparticles (MSNs) for spatiotemporally controlled co-delivery of stromal cell-derived factor-1α (SDF-1α) and osteoprotegerin (OPG). This dual-delivery platform enabled sustained and localized release of immunoregulatory and osteoprotective agents, achieving a synchronized modulation of the regenerative microenvironment. The MSNs functioned as an effective nanocarrier to preserve bioactivity and prolong release kinetics of bioactive factors, while the hydrogel matrix provided structural integrity and protease-responsive degradation matching bone repair dynamics. The system potentiated bone marrow mesenchymal stem cell (BMSC) recruitment, proliferation, and osteogenic differentiation, reprogrammed macrophage polarization toward pro-regenerative M2 phenotypes, and suppressed osteoclastogenesis. In vivo validation using a rat critical-sized femoral defect model revealed accelerated osseous regeneration. Mechanistically, transcriptomic profiling identified concerted PI3K/Akt-MAPK pathway activation. The immunomodulatory hydrogel reduced pro-inflammatory cytokines while elevating osteogenic factors. Crucially, the hydrogel restored osteoblast-osteoclast crosstalk balance, suppressing TRAP + osteoclast formation through OPG-mediated RANKL inhibition. Overall, this hydrogel-based delivery platform demonstrates a clinically translatable strategy for drug-enabled bone regeneration, with precise control over therapeutic release, spatial localization, and immuno-osteogenic coupling.

New Family of Multistimuli-Responsive Acrylamide-Based Homopolymers: Synthesis, Responsive Behavior, and Application in Controlled Release

Ke Wang,Xiaofang Liu,Ziyan Li,Fan Zhao,Guiyan Liu,Yongfei Zeng

A new family of multistimuli-responsive acrylamide-based homopolymers with different end pendant substituents were prepared by reversible addition–fragmentation chain transfer (RAFT) polymerization. They include three types of multistimuli-response: temperature/pH/CO2-response, temperature/pH/redox-response, and temperature/pH/light-response. When the terminal substituent was ethyl and tetrahydropyran, P(Et-N-AM) and P(THP-N-AM) exhibited lower critical solution temperature (LCST) behavior in water, which can be adjusted by degree of polymerization (DP) and concentration of polymer, additives, pH, and the injection of CO2/N2 to the solution. When the terminal substituents were phenyl, ferrocene, and azobenzene, P(Ph-N-AM), P(Fc-N-AM), and P(Azo-N-AM) were insoluble in water due to the increase of hydrophobicity, but the upper critical solution temperature (UCST) was exhibited in the mixed solution of nBuOH/water and can be adjusted by the addition of acid or base. P(Fc-N-AM) was also redox-responsive, and its UV/vis absorption peaks changed with the addition of FeCl3/VC. P(Azo-N-AM) was also light-responsive, and under UV/blue light radiation, the characteristic peaks of its UV/vis absorption spectrum changed reversibly. In addition, the three homopolymers were amphiphilic, which can self-assemble in solution, and the obtained micelles were also responsive. Finally, nile red (NR) was used as a hydrophobic drug model, and the controlled release behavior of NR-loaded micelles of P(Fc-N-AM) and P(Azo-N-AM) under different stimuli was investigated. This work has greatly enriched the family of multistimuli-responsive poly(N-substituted acrylamide)s, and the synthetic steps of homopolymers were simple; the responsive behavior was sensitive, and the species were rich. It provides a reference for the design of novel stimuli-responsive polymers and has a good application prospect in self-assembly, drug release, and other fields.

Controlled release of vitamin A palmitate from crosslinked cyclodextrin organic framework for dry eye disease therapy

Xinyue Ye,Falan Li,Mingwei Li,Guoqing Zhang,Weixing Wang,Zhigang Wang,Hui Zhang,Liyun Dong,Xueyuan Lin,Li Wu,Can Peng,Lifeng Wang,Weidong Chen,Jiwen Zhang

Controlled release drug delivery systems of eye drops are a promising ophthalmic therapy with advantages of good patient compliance and low irritation. However, the lack of a suitable drug carrier for ophthalmic use limits the development of the aforementioned system. Herein, the crosslinked cyclodextrin organic framework (COF) with a cubic porous structure and a uniform particle size was synthesized and applied to solidify vitamin A palmitate (VAP) by using the solvent-free method. The VAP@COF suspension eye drops were formulated by screening co-solvents, suspending agents, and stabilizing agents to achieve a homogeneous state and improve stability. According to the in vitro release study, the VAP@COF suspension exhibited a controlled release of VAP within 12 h. Both the ex vivo corneal contact angle and in vivo fluorescence tracking indicated that the VAP@COF suspension prolonged the VAP residence time on the ocular surface. This suspension accelerated the recovery of the dry eye disease (DED) model in New Zealand rabbits. Furthermore, the suspension was non-cytotoxic to human corneal epithelial cells and non-irritation to rabbit eyes. In summary, the particulate COF is an eye-acceptable novel carrier that sustains release and prolongs the VAP residence time on the ocular surface for DED treatment.

Preparation and characterization of glutamine/chito-oligosaccharide modified zein nanoparticles: Controlled release naringin and anti-diabetes

Yiyan Guo,Mingze Xu,Jiapeng Chen,Jingfeng Che,Wenqing Zhu,Yuxin Zheng,Wanting Huang,Wenjun Dai,Hong Chen,Limin Zhai,Li Zhang,Yan-Qing Guan

In this study, we employed FDA-approved food-grade zein to address the issue of poor water solubility of Naringin (Nar) through self-assembly into pH-responsive oral zein nanoparticles with active intestinal absorption. Subsequently, a detailed analysis of the nanoparticles was conducted using a series of characterization tools, as well as in vivo and in vitro experiments. The research results show that the diameter of the nanoparticles is about 116 nm, which is in line with the theory of oral drugs. The loading efficiency of nanoparticles reached 88.25 %, and the release of Naringin in simulated intestinal fluid environment was as high as 94.75 %, indicating that nanoparticles have certain intestinal permeability and significantly improve the oral bioavailability of Naringin. This eco-friendly synthesis process yields food-grade nanoparticles that have the potential to be used as food additives or pharmaceuticals, and may even pave the way for treatments targeting type 2 diabetes and hepatic steatosis.

Preparation of Multifunctional Hydrogel with Curcumin-Controlled Release and Photothermal Synergistic Antibacterial Property for Wound Dressing

Yufei Ling,Yuxuan Wang,Yuanpeng Cai,Shizhen Zhao,Hui Liu

Considering the dynamic changes in the wound microenvironment and the risk of bacterial infection during healing, the synergistic design of antibacterial hydrogel dressings has become an effective treatment strategy. A multifunctional hydrogel Cur-F127/CMCs/PDA was prepared through the cross-linking of carboxymethyl chitosan (CMCs) with genipin (GP), followed by the loading of curcumin-F127 (Cur-F127) micelles and polydopamine (PDA) nanoparticles. Successful synthesis of the hydrogel was confirmed by Fourier transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM), and X-ray photoelectron spectroscopy (XPS). Results showed that the introduction of PDA endowed the hydrogel with excellent photothermal capability, achieving a temperature increase to 54.3 °C within 10 min under near-infrared (NIR) irradiation. Meanwhile, rapid Cur release was observed under NIR irradiation, exhibiting distinct “on/off” switching characteristics. The drug-controlled releasing behavior of the hydrogel was evaluated in physiological environments, and relatively high cumulative Cur release was obtained under simulated wound microenvironmental conditions (acid region and high temperature) due to the shrinking of polymer chains and micelle structure. The synergistic effect of Cur release and PDA photothermal therapy enabled the hydrogel to exhibit excellent antibacterial effects, and the survival rates of E. coli and S. aureus were reduced to 6.25% and 1.31%, respectively. Furthermore, in vitro biological studies demonstrated that the hydrogel possessed a good antioxidant property, hemostatic performance, blood compatibility, cell compatibility, and cell migration ability. In summary, the multifunctional wound dressing Cur-F127/CMCs/PDA provided a synergistic antibacterial strategy combining natural active compound release with photothermal effects, which accelerated the wound-healing process and holds significant clinical translational potential.

Construction of a Glutathione-Responsive and Silica-Based Nanocomposite for Controlled Release of Chelator Dimercaptosuccinic Acid

Hongqiang Zhai,Yuli Wang,Menghua Wang,Shuai Liu,Feifei Yu,Chunsheng Gao,Guiling Li,Qiang Wu

Dimercaptosuccinic acid (DMSA) is an oral heavy metal chelator. Although DMSA is the most acceptable chelator in the urinary excretion of toxic elements from children and adults, its defects in plasma binding and the membrane permeability limit its interaction with intracellular elements and affect its efficacy in chelation therapy. Herein, a novel nanocomposite composed of mesoporous silica nanoparticles (MSNs), disulfide bond, and DMSA was synthesized and characterized with a scanning/transmission electron microscope, IR and Raman spectra, and TGA analysis. The in vitro interactions with glutathione (GSH) and cellular uptake assays showed that it was able to be stable in extracellular environments such as in blood, be internalized by cells, and release DMSA inside via GSH-triggered disulfide cleavage reaction. The in vitro adsorption assays showed that MSNs-SH as its intracellular metabolite had strong adsorbability for models of Hg2+or Pb2+. The hemolysis and cell viability assays showed that it was compatible with blood and cells even at a concentration of 1000 μg·mL−1. All above could not only enable it to be a GSH-responsive drug delivery system (DDS) for DMSA delivery but also to be a solution for its defects and efficacy. Thus, introduction of intelligent DDS might open a new avenue for DMSA-based chelation therapy. Keywords:mesoporous silica;disulfide bond;glutathione-responsive;controlled release;dimercaptosuccinic acid
