Organic macromolecules form carcinogenic and toxic substances such as polycyclic aromatic hydrocarbons (PAHs) under high temperature baking. Thus, this study investigated the effects and inhibition pathways of different curcumin concentrations (0.01, 0.05, 0.25, 0.3 mg/g) on seven PAHs in grilled chicken wings. The results demonstrated that curcumin concentrations displayed positive effects in inhibiting the formation of PAHs (16%–72%), increasing the total phenolic content (397.5–1934.4 mg/g) and free radical scavenging activity, and reducing TBARS values (31.15%–47.76%) and fatty acid content. Additionally, PCA and Pearson correlation analyses indicated that lipid oxidation (r = 0.42) and unsaturated fatty acids (r = 0.55) could promote the production of PAHs, while DPPH, ABTS and TPC could counteract their facilitation of PAHs. In conclusion, the addition of appropriate amounts of curcumin before grilling is a feasible strategy to reduce fat oxidation levels and the number of free radicals for the purpose of limiting PAHs content.

Curcumin is a polyphenolic compound derived from the plant turmeric and the structural instability of which limits its further clinical applications. In this study, 11 curcumin analogs with more stable scaffold were prepared and evaluated. The results indicated that the optimal compound Y-11 exhibited the strongest antiproliferative activities against lung cancer cells including H460 and H1650. Further studies showed that Y-11 potentially inhibited hDHODH, induced cell cycle arrest and apoptosis as well as down-regulated crucial signal pathway protein expression in H1650 cells. In the conclusion, the newly designed curcumin analog Y-11 may be suitable for further development in lung cancer treatment.

Metal–organic frameworks (MOFs) with tunable structures, high porosity, and abundant active sites are considered to be promising drug delivery systems (DDSs). In this study, novel MOFs-based nanoparticles ( [email protected] ) were successfully prepared using Zn-based IRMOF-16 as a carrier and curcumin (CUR) as the model drug. This nanocargo delivery platform has dual capabilities, enabling simultaneous drug delivery and fluorescence imaging. In vitro release experiments showed that nanoparticles were released more quickly under mildly acidic conditions, which proved their potential for tumor microenvironment-responsive drug release. Through in vitro cell experiments, it was found that the nano-drug platform had high antitumor cytotoxicity, which may be related to the mechanism of increasing intracellular reactive oxygen species (ROS), reducing intracellular mitochondrial membrane potential (MMP), and inducing apoptosis. In addition, confocal laser microscopy showed that [email protected] could localize to the nucleus to exert an antitumor effect. Meanwhile, [email protected] exhibited superior antitumor activity compared with pure CUR in vitro experiments. The biocompatibility test of IRMOF-16 showed reasonable biosafety, and no evidence of obvious toxicity was observed in vitro. [email protected] has unique advantages with regard to pH response, biocompatibility, and antitumor efficacy, indicating that the nano-drug platform is a promising drug delivery carrier with an antitumor effect.

Drug resistance, perplexing a lot of doctors and patients, was an inevitable challenge in the field of effective cancer therapy. Poor water solubility, inefficient aggregation at tumor site and killing normal cells were also the great obstacles, which gave rise to failure in the tumor
clinical treatment. In order to overcome these difficulties, modified targeting nano-micelles (MT-NMs) loading Curcumin (Cur) and Chrysin (Chr) were designed. In this work, dithiodipropionic acid (DA), a reduction-sensitive material, acted as a "messenger" linker connecting Chr and oligomeric
hyaluronic acid (oHA). Then, the amphiphilic polymer, oligomeric hyaluronic acid-dithiodipropionic acid-Chrysin (oHA-DA-Chr), was synthesized. What's more, combinational therapy is a better way to cripple cancer drug-resistant and reinforce the therapeutic efficacy. In this study, 1H-NMR
was used to authenticate the structure of oHA-DA-Chr. The amphiphilic oHA-DA-Chr could self-assemble into nano-micelles (NMs) in water. The NMs with small size (223.2±2.4 nm) could concentrate well at tumor sites by enhancing permeability and retention effect (EPR) and CD44 receptor
targeting property of oHA. The Cur could release well from NMs when disulfide bond (S–S) of DA was exposed to tumor environment containing high concentration of glutathione (GSH).

Objective Epithelial mesenchymal transition (EMT) and inflammation have been identified as carcinogenic agents. This study aims to investigate whether inhibition of trichloroethylene (TCE) associated hepatocellular carcinoma (HCC) by curcumin is associated with inflammation and EMT.Methods In the current study, TCE sub-chronic cell model was induced in vitro, and the effects of TCE on cell proliferation, migration, invasion, and expression of functional proteins were verified by Western blot, MTT, clone formation, wound healing, Transwell. The detoxification effect of curcumin on TCE was explored by a mouse tumor-bearing experiment. Results TCE induces hepatocyte migration, colony formation, and EMT in vitro. In vivo studies have shown that curcumin significantly reduces the mortality of mice and control the occurrence and size of liver tumors by inhibiting the IL-6/STAT3 signaling pathway. In vitro, curcumin inhibits the proliferation of HepG2 cells as determined by MTT assay. In addition, curcumin significantly inhibited the protein expression of IL-6R, STAT3, snail, survivin, and cyclin D1 in THLE-2 and HepG2 cells induced by IL-6.Conclusion Curcumin has anti-inflammatory and anti-proliferative effects, and inhibits the development of HCC induced by TCE by reversing IL-6/STAT3 mediated EMT.

Intracerebral hemorrhage (ICH) is a kind of fatal stroke with the highest mortality and morbidity in the world. To date, there is no effective treatment strategy for ICH. Curcumin, a major active ingredient of Curcuma longa L., possesses a potential anti-inflammatory activity in many types of disease. In the current study, the mechanism underlying curcumin attenuated ICH-induced neuronal apoptosis and neuroinflammation was explored. Herein, we studied that curcumin decreased brain edema and improved neurological function by using brain edema measurement, assessment of neurological-deficient score, immunofluorescence, and Western blotting analyses after ICH. The results showed that curcumin improved ICH-induced neuronal apoptosis and neuroinflammation. Functionally, the polarization of microglia was assessed by immunofluorescence and Western blotting analyses after ICH in the absence or presence of curcumin. The results suggested that the M1-type microglia were activated after ICH, while the effect was blocked by curcumin treatment, suggesting that curcumin alleviates the neuroinflammation and apoptosis of neurons by suppressing the M1-type polarization of microglia. Mechanically, M1 polarization of microglia was regulated by JAK1/STAT1, and the activation of JAK1/STAT1 was blocked by curcumin. Meanwhile, the protective function of curcumin can be blocked by RO8191, an activator of JAK1. Taken together, our study suggested that curcumin improved the ICH-induced brain injury through alleviating M1 polarization of microglia/macrophage and neuroinflammation via suppressing the JAK1/STAT1 pathway.

Guanidine-nanocellulose encapsulates curcumin forming a stable Pickering emulsion. • Emulsion encapsulation ensures curcumin stability and pH-responsive color change. • The film enables simultaneous food freshness monitoring and shelf-life extension. • The film offers an antibacterial coating for fruit surfaces that is easy to clean. Traditional food packaging suffers from drawbacks such as environmental pollution and limited functionality. Herein, we fabricated a multifunctional film based on a curcumin Pickering emulsion stabilized by cellulose nanofibrils grafted with polyhexamethylene guanidine (PCNF). PCNF acts as both a Pickering emulsifier and an antibacterial agent. The PCNF-stabilized emulsion not only protects curcumin from degradation but also forms a composite film after incorporation of PVA and glycerol. This film exhibits good mechanical strength, color stability and pH-responsive color changes. In shrimp freshness monitoring tests, the film showed a distinct color transition from pale yellow to brown. Moreover, the tunable rheology of the emulsion allows adaptable coating on fruits. After 7 days of storage, the composite coating significantly reduced banana browning, owing to its exceptional antibacterial performance. Notably, the combination of PCNF and glycerol enables rapid water disintegration. The film completely disperses within 30 s of immersion, imparting outstanding easy-cleaning properties. These synergistic features provide a robust foundation for practical applications of this material in the field of food packaging and preservation. Traditional food packaging materials are characterized by short preservation periods and inadequate mechanical properties. In this study, curcumin-loaded halloysite nanotubes (HNTs) were incorporated into polyvinyl alcohol (PVA), and active nanofiber films were fabricated via electrospinning. The encapsulation efficiency of curcumin-HNT composite at a 1:1 mass ratio was 13.8 ± 0.2%, with a drug loading capacity of 14.5 ± 0.2%. XRD analysis revealed a characteristic halloysite nanotube peak at 24.55°, while FTIR spectra confirmed successful incorporation through weakened bands in the 1510–1282 cm −1 region. HNT incorporation enhanced curcumin thermal stability, increasing its decomposition temperature from 730 °C to 800 °C due to nanotubular confinement and structural interactions. The nanofiber films exhibited improved tensile strength, high moisture absorption (∼181%), and effective moisture-barrier properties. Further, the films demonstrated antioxidant, antibacterial, and pH-responsive colorimetric functions. Application studies on baked goods and fresh fruits showed improved water retention and frost resistance, reducing storage-related quality deterioration.

Egg yolk proteins have garnered significant attention from researchers due to their outstanding functional properties, and physical modification methods serve as a key approach to enhancing the characteristics of these proteins. In this study, phosphorylated egg yolk high-density lipoprotein (HDL) was prepared using sodium tripolyphosphate (STP) to investigate the properties of the high-internal-phase emulsion formed after cross-linking with phosvitin (Pv), as well as its protective effect on curcumin. Phosphorylation significantly enhanced the functional properties of HDL, with the 6% STP treatment yielding the highest phosphorus content (7.32 mg/g) in the resulting phosphorylated HDL (P-HDL). Structural analyses confirmed covalent phosphate binding, which reduced HDL particle size, induced protein unfolding, and exposed hydrophobic regions, thereby improving its emulsifying capacity and thermal stability. A composite stabilizer composed of phosphorylated HDL (P-HDL) and Pv was developed to construct high internal phase emulsions (HIPEs). These HIPEs also exhibited superior performance in curcumin delivery, increasing its bioaccessibility to 38.10% compared with 15.40% in the control, while effectively inhibiting lipid oxidation during storage. This study provides a promising strategy for designing high-performance emulsion delivery systems.

Scavenging excess reactive oxygen species (ROS) and positively improving intestinal dysbiosis is a promising therapeutic strategy for alleviating inflammatory bowel disease (IBD). However, conventional clinical drugs and probiotic-based adjuvant therapies often fail to achieve satisfactory results due to systemic side effects of drugs, low bioactivity of probiotics, short intestinal retention time, and poor targeting ability. To address these challenges, we developed an antioxidant-functionalized curcumin loaded nanocomposite inulin hydrogel for targeted IBD therapy. In this system, curcumin was encapsulated within chitosan-coated Poly (lactic-co-glycolic acid) PLGA nanoparticles, which not only exhibited excellent ROS-scavenging capacity but also demonstrated the enhanced cellular uptake behavior. In a dextran sulfate sodium induced ulcerative colitis mouse model, the nanocomposite hydrogel significantly prolonged the intestinal retention time of curcumin, thereby suppressing the expression of pro-inflammatory factors, alleviating intestinal inflammation, and promoting the recovery of intestinal barrier and microbial diversity. This study has developed a synergistic therapeutic strategy via combining anti-inflammatory effects with gut microbiota regulation, offering a novel therapeutic approach for the clinical management of colitis.

Objective Autophagy dysfunction results in the accumulation of pro-inflammatory components and drives inflammatory injury. Transcription factor EB (TFEB) is a master regulator of autophagy and a novel target for controlling inflammatory injury. Curcumin analog C1 directly binds to and activates TFEB, but its anti-inflammatory effects remain unclear. Methods The anti-inflammatory and protective effects of curcumin analog C1 were investigated in lipopolysaccharide (LPS)-induced endotoxemia mice. Results Curcumin analog C1 reversed LPS-induced autophagy dysfunction, downregulated pyruvate kinase M2 (PKM2), and reduced pro-inflammatory cytokines including TNF–α, IL–6, and MCP–1. It also attenuated systemic abnormalities, alleviated lung injury, and decreased blood urea nitrogen (BUN), brain-type natriuretic peptide (BNP), and extracellular DNA levels. Conclusions Curcumin analog C1 exhibits potential pharmacological value in the control of inflammatory injury.Thank you for your assistance.

An RF spacer creates a void between NaA and a mesoporous silica shell. • Shell thickness tunes curcumin loading in yolk–shell NaA@HMSNs. • NaA@HMSN-2 loads 124.0 mg/g curcumin and adsorbs Cd 2+ in vitro. • Curcumin release and Cd 2+ adsorption occur in the same buffer system. • HepG2 viability remains above 90% up to 64 μg/mL after 48 h. Cadmium contamination is an environmental and health concern. Here, a yolk–shell composite was constructed using sodium A-type zeolite (NaA) as the microporous yolk and a hollow mesoporous silica nanoparticle (HMSN) layer as the separated shell. Removal of a resorcinol–formaldehyde spacer generated an internal void, and three shell thicknesses were prepared. The NaA component provided Cd 2+ adsorption sites, whereas the amino-functionalized HMSN compartment was used for curcumin loading. N-NaA@HMSN-2, with an approximately 100 nm shell, loaded 124.0 mg/g curcumin. In buffer containing 100 mg/L Cd 2+ , Cur/N-NaA@HMSN-2 released 34.8% and 39.5% curcumin and adsorbed 73.8% and 67.8% Cd 2+ at pH 7.4 and 1.2, respectively. In HepG2 assays, all three NaA@HMSN carriers maintained more than 90% relative cell viability at concentrations up to 64 μg/mL after 48 h. These findings demonstrate coupled Cd 2+ adsorption and curcumin release under in vitro conditions and support further evaluation of this bifunctional material platform.
