( R )-Pantolactone (PL) is a key chiral intermediate for the synthesis of calcium ( R )-pantothenate and ( R )-panthenol used as food additives. The commercial production of ( R )-pantothenate is performed by the resolution of racemic pantothenate, which is synthesized through an aldol condensation and a cyanation reaction. In this study, we investigated another synthetic method of ( R )-pantothenate through the stereoselective reduction of ketopantoyl lactone (KPL) by aldo-keto reductase (AKR). A series of conjugated polyketone reductases (CPRs) were discovered from GenBank database by genome mining approach. The putative CPR gene from Candida orthopsilosis Co 90–125 ( Cor CPR) was cloned and functionally expressed in Escherichia coli BL21 (DE3). The optimum pH and temperature of recombinant Cor CPR were 6.0–7.0 and 40 ℃, respectively. The K m and v max toward KPL were1.3 mM and 227.3 μ mol/min/mg protein, respectively. The conserved sequences suggest that Cor CPR belongs to AKR3C family of AKR superfamily. Furthermore, a catalytic tetrad was proposed, and the detailed mechanism was clarified by molecular docking. In a batch reaction, 50 mM KPL was reduced to ( R )-PL with 99% conversion and > 99% enantiomeric excess within 5 h. The recombinant Cor CPR from C. orthopsilosis shows potential application in the asymmetric synthesis of ( R )-pantothenate preparation.

Dye wastewater is a type of wastewater that is difficult to treat, and the degradation of dye using microalgae is both effective and profitable. This study cultivated microalgae Asterarcys quadricellulare R-56 under anaerobic condition for the removal of Congo red dye and explored the growth and lipid accumulation of microalgae with only Congo red dye as the carbon source, and a pathway for the degradation of Congo red by microalgae was proposed. The removal efficiency of Congo red (60 mg/L) by microalgae under light anaerobic culture mode reached 91.6 %, which was higher than that of light aerobic condition (60.8 %). The highest microalgae biomass was 406.4 ± 18.3 mg/L with a concentration of Congo red of 100 mg/L, which was 1.6 times of the control group. Moreover, the highest lipid production and lipid content were 187 ± 3.2 mg/L and 61.1 ± 3.1 %, respectively. In addition, the total saturated and monounsaturated fatty acid content of microalgae lipids reached 96.7 % at 100 mg/L Congo red, which was higher than that of the control group (85.5 %), more favorable for the production of high-quality biodiesel. The mechanism of removing Congo red by microalgae was mainly the biosorption and biodegradation. This study shows that the use of microalgae for dye purification and resource recovery is a promising strategy.

Hyperuricemia is associated with dietary purine intake, dysregulated purine metabolism, and impaired uric acid excretion. This study aimed to isolate multifunctional LAB from fermented foods and apply the selected strain to enhance the functional potential of Smilax glabra Roxb. fermentation. Among 79 isolates, 46 were identified as LAB. Limosilactobacillus fermentum R-9 showed efficient degradation of inosine, guanosine, and adenosine, while exhibiting favorable gastrointestinal tolerance and safety-related probiotic properties. Its survival rates after simulated gastric and intestinal digestion were 82.28% and 67.18%, respectively. Genomics revealed genes related to nucleoside hydrolysis, purine/pyrimidine transport, nucleotide salvage, starch utilization, α-glucan metabolism, and β-glucosidase-related glycoside transformation, supporting the dual potential of R-9 for purine metabolism and adaptation to starch-rich S. glabra . L. fermentum R-9 was further used to ferment Smilax glabra Roxb., a starch-rich medicinal and edible plant material containing flavonoid glycosides. After 4 days of fermentation, total phenolics increased from 0.384 to 0.572 mg GAE/mL, and total flavonoids increased from 2.11 to 3.02 mg RE/mL. Xanthine oxidase (XOD) inhibitory activity increased from 16.58% to 70.81%, accompanied by enhanced DPPH and ABTS radical-scavenging activities. Astilbin reached 59.75 μg/mL, increasing by 33.79%. Mechanistically, L. fermentum R-9 fermentation promoted acidification, amylase and β-glucosidase activities, starch matrix disruption, and flavonoid release. These findings highlight L. fermentum R-9 as a promising starter for developing fermented Smilax glabra Roxb. products with enhanced antioxidant and xanthine oxidase inhibitory potential.

Host–guest interactions by artificially synthesized macrocyclic molecules, such as cucurbit[n]uril (CB[n], n refers to the number of glycoluril units), exhibited comparable affinity to natural biological interactions, offering orthogonal selectivities. These unique high-affinity interactions are crucial in bioseparation, particularly in the separation of complex biomacromolecules. However, to date, the application of supramolecular systems in bioseparation remains insufficient and challenging. In this work, a site-specific host–guest interaction-based method by CB[6] and CB[7] was developed for the selective enrichment of basic peptides, and their recognition for basic lysine/arginine (K/R) residues exhibited distinct selectivity. Although they both exhibited affinity to basic peptides, CB[6] tended to bind to peptides containing multiple K/R residues, while CB[7] preferred to adsorb peptides containing both phenylalanine residues and basic K/R residues. This difference was attributed to their distinct cavity sizes. Furthermore, CB[6]-containing monolith was employed for the enrichment of basic peptides from mouse liver sample digested with Staphylococcus aureus protease V8 (Glu-C). It is noteworthy that basic peptides containing 3–4 K/R residues can be successfully enriched, whereas those with more than 5 K/R residues cannot be effectively eluted. It was also observed that 1043 unique peptides were newly identified from 2.5 μg of mouse liver after the capture of basic peptides, with a markable increase in the identification coverage (31%). These newly identified peptides are generally more acidic and shorter than those depleted by CB[6], with median isoelectric point (pI) values of 5.8 and 7.0 and median lengths of 10 and 16 amino acids, respectively.

Background. Oxymatrine (OMT) is the primary pharmacological component of Sophora flavescens Aiton., which has been shown to possess potent antifibrotic, antioxidant, and anti-inflammatory activities. The aim of the present study was to clarify the protective mechanism of OMT on acute lung injury (ALI) subjected to myocardial ischemia/reperfusion (I/R). Methods. A myocardial I/R-induced ALI model was achieved in diabetic rats by occluding the left anterior descending coronary artery for 1 h, followed by reperfusion for 1 h. The levels of inflammatory factors (tumor necrosis factor-α, interleukin- (IL-) 6, and IL-17) in bronchoalveolar lavage fluid were assessed using commercially available kits. The index of myocardial injury, including the detection of cardiac troponin I (cTnI), cardiac troponin T (cTnT), lactate dehydrogenase (LDH), and creatine kinase-MB (CK-MB), was also determined using commercially available kits. Hematoxylin and eosin staining and terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling were used to identify histological changes. The expression levels of endoplasmic reticulum chaperone BiP (GRP78), DNA damage-inducible transcript 3 protein (CHOP), eukaryotic translation initiation factor 2-alpha kinase 3 (PERK), inositol dependent enzyme 1α (IRE1α), ATF6, caspase-3, -9, and-12, Bcl-2, and Bax were determined by Western blotting. The mRNA expression levels of GRP78 and CHOP were detected by reverse transcription-quantitative PCR. Results. Myocardial I/R increased the levels of cTnI, cTnT, LDH, and CK-MB in diabetic rats. Damaged and irregularly arranged myocardial cells were also observed, as well as more serious ALI with higher lung injury scores and WET/DRY ratios and lower PaO2. Moreover, the expression of key proteins of endoplasmic reticulum stress (ERS) was increased by I/R injury, including phosphorylated- (p-) PERK, p-IRE1ɑ, and ATF6, as well as decreased levels of apoptosis. These effects were all significantly reversed by OMT treatment. Conclusions. OMT protects against ALI subjected to myocardial I/R by inhibiting ERS in diabetic rats.

Although ginsenoside Rg3 was isolated as a major component of Korea red ginseng and confirmed to exert potential hepatoprotective effect on acetaminophen (APAP)-induced liver injury via induction of glutathione S-transferase (GST) in vitro, the in vivo hepatoprotective effect of Rg3 and the underlying molecular mechanism of action remain unclear. The current study was aimed to explore whether 20( R )-Ginsenoside Rg3 (20( R )-Rg3) could alleviate acetaminophen-induced liver injury in mice and to determine the involvement of PI3K/AKT signaling pathway . Our findings demonstrated that a single injection of APAP (250 mg/kg) increased the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), tumor necrosis factor-α (TNF-α), and interleukin-1β (IL-1β); such increases were attenuated by pretreatment of mice with 20( R )-Rg3 for seven days. The depletion of glutathione (GSH), generation of malondialdehyde (MDA) and the over expression of cytochrome P450 E1 (CYP2E1) and 4-hydroxynonenal (4-HNE) caused by APAP exposure were also inhibited by 20( R )-Rg3 pretreatment. Moreover, 20( R )-Rg3 pretreatment significantly alleviated APAP-induced apoptosis , necrosis, and inflammatory infiltration in liver tissues. Importantly, 20( R )-Rg3 effectively attenuated APAP-induced liver injury in part via activating PI3K/AKT signaling pathway. In summary, 20( R )-Rg3 exerted liver protection against APAP-caused hepatotoxicity evidenced by inhibition of oxidative stress and inflammatory response, alleviation of hepatocellular necrosis and apoptosis via activation of PI3K/AKT signaling pathway, showing potential as a novel therapeutic agent to prevent liver damage.

Genomic mining has identified a novel microbial alkaline esterase from the Indian Ocean. This esterase was overexpressed in E. coli BL21 (DE3) and further functionally characterized. Under optimal conditions (10 mmol/L substrate, pH 6.0, 2 h at 40 °C), this esterase can hydrolyze racemic methyl mandelate to ( R )-methyl mandelate with very high optical purity ( e.e. >99%) and yield (nearly 90%). Interestingly, the stereoselectivity of this esterase is opposite to that of two previously reported lipases that can generate ( S )-methyl mandelate through the hydrolysis of racemic methyl mandelate. No organic solvents or other additives were required to optimize the optical purity and production of the final chiral product ( R )-methyl mandelate, which can potentially simplify the production procedure of ( R )-methyl mandelate catalyzed by esterase.

Optically pure 1-phenyl-1,2-ethanediol is a very important chiral building block and intermediate in fine chemical and pharmaceutical industries. Reduction of 2-hydroxyacetophenone provides a straightforward approach to access these important compounds. In this study, two enantiocomplementary carbonyl reductases, BDHA (2,3-butanediol dehydrogenase from Bacillus subtilis ) and GoSCR (polyol dehydrogenase from Gluconobacter oxydans ) were discovered for the first time to convert 2-hydroxyacetophenone (2-HAP) to ( R )-1-phenyl-1,2-ethanediol (( R )-PED) and ( S )-1-phenyl-1,2-ethanediol (( S )-PED) with excellent stereochemical selectivity, respectively. The two enzymes were purified and characterized. In vitro bioreduction of 2-HAP catalyzed by BDHA and GoSCR coupled with glucose dehydrogenase (GDH) from Bacillus subtilis for cofactor regeneration were demonstrated, affording both ( R )-PED and ( S )-PED in >99% ee and 99% conversion. Recombinant Escherichia coli whole cells co-expressing both GDH and BDHA or GoSCR genes were used to asymmetric reduction of 2-HAP to ( R )-PED or ( S )-PED. Under the optimized conditions, the bioreduction of 400 mM (54 g/L) substrate was proceeded smoothly without the external addition of cofactor, and the product ( R )-PED and ( S )-PED were obtained with 99% yield, >99% ee and 18.0 g/L/h volumetric productivity. These results offer a practical biocatalytic method for the preparation of both ( R )-PED and ( S )-PED with high volumetric productivity.

Astrocytes may fail to perform their neuroprotective roles against cerebral ischemia/reperfusion (I/R) injury due to phenotypic transformation. We aimed to demonstrate the distinct roles of A1 astrocytes and A2 astrocytes on the blood-brain barrier (BBB) injury following cerebral I/R, and explore whether H 2 S-mediated A2 astrocytes polarization protects against BBB injury via inhibiting ROCK/NF-κB pathway. The mice cerebral I/R model and the oxygen-glucose deprivation/re‑oxygenation (OGD/R) model of astrocytes were used in present study. Cerebral I/R-induced BBB injury is evidenced by increased EB dye leakage and reduced expressions of ZO-1 and occludin in mice hippocampal tissues. We found that H 2 S-mediated A2 polarization and lipopolysaccharide (LPS)-induced polarization of A1 astrocytes respectively display beneficial and harmful role in BBB injury. Besides, harmful roles of A1 astrocytes in BBB injury can be reduced by H 2 S. Additionally, A1 astrocytes exhibit excessive activation of NF-κB and enhanced expressions of MMP9 and AQP4, which can be inhibited by H 2 S. Moreover, H 2 S-mediated polarization of A2 astrocyte displays enhanced phosphorylation and nuclear translocation of STAT3 and reduced expressions of MMP9 and AQP4. Importantly, ROCK inhibitor Fasudil likewise inhibits the activation of NF-κB and promotes STAT3 activation in OGD/R astrocytes, and NF-κB inhibitor BAY11–7082 reduces the expressions of MMP9 and AQP4 in OGD/R astrocytes. In conclusion, H 2 S-mediated polarization of A2 astrocyte protects against BBB injury following cerebral I/R, the mechanisms may be related to inhibition of ROCK/NF-κB pathway, and activation of STAT3.

In this study, high-entropy Mg 1.8 R 0.2 Al 4 Si 5 O 18 (R Ni, Ni 1/2 Co 1/2 , Ni 1/3 Co 1/3 Zn 1/3 , Ni 1/4 Co 1/4 Zn 1/4 Cu 1/4 , Ni 1/5 Co 1/5 Zn 1/5 Cu 1/5 Mn 1/5 ) cordierite ceramics based on equimolar ratios were designed and synthesized through the solid-state reaction technique at high temperatures. The effect of various R-ionic substitutions at the octahedral site and their relationship with ionic polarization, chemical bonding, and configurational entropy were explored. These factors have greatly affected the microwave dielectric properties of Mg 1.8 R 0.2 Al 4 Si 5 O 18 ceramics. The presence of cation radius mismatch and electronegativity mismatch at [MgO 6 ]-octahedron lattice sites revealed lattice distortion resulting from multiple cation substitutions. As the entropy values increased, the temperature stability of microwave dielectric properties of Mg 1.8 R 0.2 Al 4 Si 5 O 18 ceramics improved. For R Ni 1/2 Co 1/2 , a higher Q×f of approximately 73,658 GHz @ 13 GHz, with ɛ r ∼4.44 and τ f ∼ −23.9 ppm/°C was obtained. In the case of R Ni 1/5 Co 1/5 Zn 1/5 Cu 1/5 Mn 1/5 , a near zero τ f (−10.7 ppm/°C), ɛ r = 4.68 and Q×f = 35,478 GHz @13 GHz were achieved, making it potentially suitable for practical applications.

Hepatocellular carcinoma (HCC) is a challenging cancer to treat due to its high malignancy, strong invasiveness, and high risk of metastasis. Inhibiting lymphatic metastasis in the early stages is crucial for HCC therapy. Active targeted drug delivery systems, imbued with tumor and lymphatic targeting capability via ligand–receptor mediation, represent a promising therapeutic strategy. Cantharidin (CTD), the primary effective component of poisonous traditional Chinese medicine (PTCM) Mylabris, has unique therapeutic effects on HCC. However, its high toxicity limits its clinical application. In this study, based on the high expression of lymphatic vessel endothelial hyaluronan receptor-1 (LYVE-1) in the peritumoral lymphatic vessels and glycyrrhetinic acid receptor (GA-R) in hepatocellular carcinoma cells, we developed CTD-loaded poly(d,l-lactide-co-glycolide) (PLGA) nanoparticles modified with glycyrrhetinic acid (GA)–hyaluronic acid (HA) copolymers (GA-HA-PLGA/CTD NPs) through electrostatic interactions. This nanoparticle platform could preferentially accumulate cargo into the tumors and peritumoral lymphatic vessels, increasing drug accumulation in tumors and lymph nodes while decreasing drug accumulation in other organs, particularly the heart and kidney. As a result, it could enhance antitumor efficacy and inhibit lymphatic metastasis better than PLGA/CTD NPs while also reducing systemic toxicity. Furthermore, the variation of HA molecular weights and degrees of GA substitution could regulate the distribution of nanoparticles within tumors and lymph nodes. Overall, GA-HA-PLGA/CTD NPs exhibit promising potential as a therapeutic approach for HCC treatment and lymphatic metastasis inhibition, providing a potential strategy for the application of PTCMs in HCC treatment.

Aging is characterized by progressive cellular dysfunction and increased susceptibility to chronic diseases. Enhancing autophagy, a key cellular clearance process, offers a promising strategy to combat aging. In this study, we investigated the autophagy-inducing and anti-aging properties of Salvia plebeia R. Br. ethanol extract (SPE) using in vitro and in vivo models. Our results showed that SPE enhanced autophagosome and autolysosome formation, elevated LC3-II and Beclin-1 levels, and reduced p62 expression in human foreskin fibroblasts (HFFs), confirming its autophagy-inducing activity. SPE also mitigated D-galactose (D-gal)-induced cellular senescence, reducing SA-β-gal staining, senescence markers (p16, p21), SASP factors, and γ-H2AX foci. These effects were Atg7-dependent, as Atg7 knockout abolished the benefits, highlighting autophagy’s role in SPE’s action. In vivo, SPE improved physical function, reversed cellular senescence, enhanced antioxidant defenses, and restored autophagic activity in D-gal-induced prematurely senescent mice. Furthermore, SPE alleviated age-related cognitive decline, reduced neuronal damage, increased synaptic density, and mitigated Tau accumulation. Notably, SPE demonstrated good tolerability over 12 weeks, with stable body weight, no apparent tissue injury, and unchanged serum ALT, AST, and creatinine. Chemical profiling identified rosmarinic acid (RA) as a major bioactive constituent that recapitulated key autophagy- and senescence-related effects in vitro. These findings suggest that SPE is a promising natural autophagy enhancer with potential to promote healthy aging, combat age-related diseases, and serve as a novel therapeutic strategy for cellular dysfunction in aging.
