Colorimetric method for Salmonella spp. detection based on peroxidase-like activity of Cu(II)-rGO nanoparticles and PCR

Lijun Wang,Tianyu Liao,Hong Zhou,Yukun Huang,Pengfei Chen,Xiao Yang,Xianggui Chen

Development of a rapid and sensitive method for Salmonella spp. detection is of great importance for ensuring food product safety due to its low infective dose. In this study, a colorimetric method based on the peroxidase-like activity of Cu(II)-modified reduced graphene oxide nanoparticles (Cu 2+ -rGO NPs) and PCR was successfully developed to detect Salmonella spp. in milk. Under optimal conditions, the developed colorimetric method exhibited high sensitivity and strong specificity for Salmonella spp . detection. The limit of detection was 0.51 CFU/mL with a linear range from 1.93 × 10 1 to 1.93 × 10 5  CFU/mL. A specificity study demonstrated that this method can specifically distinguish Salmonella typhimurium and Salmonella enteritidis from other foodborne pathogens. The application of the proposed method for milk sample detection was also validated, and the recovery rates of S . typhimurium in spiked milk sample ranged from 102.84% to 112.25%. This colorimetric sensor exhibits enormous potential for highly sensitive detection of bacteria in milk sample.

Cathodic electrodeposited Cu-BTC MOFs assembled from Cu(II) and trimesic acid for electrochemical determination of bisphenol A

Hu Peng,Zhu Xiaoming,Luo Xiaohui,Hu Xiaofeng,Ji Liudi

The authors describe a novel electrochemical determination method for bisphenol A (BPA) based on the electrosynthesised Cu-BTC (H 3 BTC: trimesic acid) films. Using H 3 BTC as the ligand, Cu(NO 3 ) 2 as the precursor of copper ions, and triethylamine hydrochloride (Et 3 NHCl) as the probase source, Cu-BTC films were directly deposited on glassy carbon electrode (GCE) surface via cathodic electrochemical reduction under −1.30 V. Considering the electrocatalytic activity of metal center Cu 2+ , Cu-BTC films were applied to construct the electrochemical determination platform for BPA. Chronocoulometry and electrochemical impedance spectroscopy were used to study the signal enhancement mechanism. The determination conditions were optimized. As a result, a sensitive electrochemical method was constructed for BPA. The peak currents, best measured at voltage of 0.496 V vs. SCE (KCl saturated calomel reference electrode), increase linearly in the range from 5.0 to 2000 nM. The value of determination limit is 0.72 nM. The proposed method was successfully applied to determine BPA in spiked urine, spiked waste water samples and plastic products. The results were in good agreement with those obtained for the same samples by high-performance liquid chromatography (HPLC). Schematics for the construction of electrochemical determination for bisphenol A.

Dense and robust aminopolycarboxylic acid-decorated porous monoliths for eliminating trace Cu(II) or Zn(II) from water

Ziyun Xu,Shiqi Weng,Chenhui Li,Ming Jin,Decheng Wan

Aminopolycarboxylic acids (APAs) show a strong affinity toward heavy metal species, but straightforward and robust studies on the surface of porous adsorbents with APAs are rarely available. In this study, branched polyethyleneimine (PEI) is directly used to investigate the surface of a microcrystalline cellulose (MC)-supported porous monolith (MC@PEI) through an oil-in-water high internal phase emulsion technique. The residual amines of the covalently immobilized PEI are transformed into dense APAs (MC@PEI-APA). The open-cellular MC@PEI-APA monolith can effectively eliminate trace Cu(II) or Zn(II) from water relative to MC@PEI. Moreover, its adsorption complies with the second-order kinetics and follows the Langmuir adsorption model. MC@PEI-APA shows high adsorption capacities (e.g., 96 mg g −1 for Cu(II)). It also presents stronger metal affinities than the commercial Amberlite IRC-748 adsorbent possibly because of the cooperative ligation of metal species by the relatively dense APA groups of MC@PEI-APA. The proposed adsorbent is durable and has good recyclability.

A “turn-on” ESIPT fluorescence probe of 2-(aminocarbonyl)phenylboronic acid for the selective detection of Cu(II)

Dandan Jiang,Minghao Zheng,Xiaoyang Yan,Bin Huang,Hui Huang,Tianhao Gong,Kunming Liu,Jinbiao Liu

Herein, we report a highly selective fluorescent probe for the detection of Cu(II). The detection mechanism relies on the Cu(II)-catalyzed oxidative hydroxylation of 2-(aminocarbonyl)phenylboronic acid into salicylamide, thus recovering the excited-state intramolecular proton transfer (ESIPT) effect and inducing more than 35-fold fluorescence enhancement. The simple structure and readily available fluorescent probe give a novel method for quantitatively detecting Cu(II) in the linear range of 0–22 μM, with a limit of detection down to 68 nM, and exhibiting high selectivity for Cu(II) over 16 other metal ions.

An eco-friendly wheat straw adsorbent functionalized with mercaptopropionyl for the removal of Cu(II) from aqueous solution

Yong-peng Sun,Gang Wang,Ya-ling Guo,Xiao-yan Zhu

A novel absorbent called mercaptopropionyl wheat straw (MPWS) was prepared through alkali pretreatment and mercaptopropionyl modification for efficient removal of Cu(II) from aqueous solutions. Single factor experiments were conducted to investigate various parameters affecting the adsorption of Cu(II) with MPWS. Adsorption experiments including kinetics, isotherms, thermodynamics, as well as various characterization methods were employed to discuss the mechanism of Cu(II) uptake by MPWS in this study. The results showed that MPWS exhibited high removal capacity for Cu(II), achieving a maximum removal efficiency of 97.28 % under optimal conditions: oscillation rate at 150 rpm, temperature at 303 K, duration at 2 h, pH value at 6.0, and initial concentration of Cu(II) at 100 mg/L. The fitting results indicated that pseudo-second-order kinetic model better described the experimental data while Freundlich model provided a better fit for isothermal data. Thermodynamic calculations confirmed that the adsorptive process was spontaneous and endothermic. Characterization techniques such as FTIR spectroscopy SEM imaging, and EDS analysis confirmed chemisorption between Cu(II) ions and functional groups present on MPWS including sulfhydryl groups, amine groups, hydroxyl groups, and carboxyl groups. Overall, the main mechanisms governing Cu(II) uptake onto MPWS included coordination, ion exchange, electrostatic adsorption, and physical adsorption.

Dithiocarbamate functionalized Al(OH)3-polyacrylamide adsorbent for rapid and efficient removal of Cu(II) and Pb(II)

Ya Liu,Peng Qian,Yang Yu,Li Xiao,Yongliang Wang,Shufeng Ye,Yunfa Chen

Heavy metal ions such as Cu 2+ and Pb 2+ impose a significant risk to the environment and human health due to their high toxicity and non-degradable characteristics. Herein, Al(OH) 3 -polyacrylamide chemically modified with dithiocarbamates (Al-PAM-DTCs) was synthesized using formaldehyde, diethylenetriamine, carbon disulfide, and sodium hydroxide for rapid and efficient removal of Cu 2+ and Pb 2+ . The synthesized adsorbent was characterized by Fourier transform infrared (FTIR) spectroscopy, thermogravimetric analysis, scanning electron microscopy–energy dispersive X-ray spectroscopy analysis, and transmission electron microscope measurements. Al-PAM-DTCs showed rapid removal of Cu 2+ (<30 min) and Pb 2+ (<15 min) with high adsorption capacities of 416.959 mg/g and 892.505 mg/g for Cu 2+ and Pb 2+ respectively. Al-PAM-DTCs also had high capacities in removing suspended solids and metal ions simultaneously in turbid bauxite suspensions. FTIR, thermodynamic study, and elemental mapping were used to determine the adsorption mechanism. The rapid, convenient, and effective adsorption of Cu 2+ and Pb 2+ indicated that Al-PAM-DTCs has great potential for practical applications in purification of other heavy metal ions from aquatic systems. © 2017 Wiley Periodicals, Inc. Application of EDTA-functionalized bamboo activated carbon (BAC) for Pb(II) and Cu(II) removal from aqueous solutions

Dan Lv,Yu Liu,Jiasheng Zhou,Kunlun Yang,Zimo Lou,Shams Ali Baig,Xinhua Xu

In this study, a novel bamboo activated carbon (BAC) with ethylene diamine tetraacetic acid (EDTA) functionality was prepared by direct grafting in the presence of tetraethyl orthosilicate (TEOS) as a crosslinking agent. The BAC@SiO 2 -EDTA was characterized by SEM, TEM, TGA, FTIR, XPS and its adsorption property for removal of Pb(II) and Cu(II) under various experimental conditions was also investigated. The characterization results reflected that EDTA was successfully assembled on the surface of the BAC and average pore size increased from 4.10 to 4.83   nm as BAC grafted with EDTA. Adsorption data fitted very well in Langmuir isotherm model and pseudo-second-order kinetic model. As compared with the raw BAC, the maximum adsorption capacities of BAC@SiO 2 -EDTA for the Pb(II) and Cu(II) increased from 45.45 to 123.45   mg   g −1 and from 6.85 to 42.19   mg   g −1 , since the existence of EDTA on modified BAC promoted the formation of chemical complex. The removal of heavy metal ions mainly depended on the complexation with EDTA and the electrostatic attractions with negatively charged surface of BAC@SiO 2 -EDTA. The adsorption of Pb(II)/Cu(II) on the BAC@SiO 2 -EDTA was pH dependent and pH 5–6 was considered an optimum. However, lower temperature favored the adsorption and the maximum adsorption was recorded at 20   °C. In addition, BAC@SiO 2 -EDTA had an excellent reusability with about 40% decline in the adsorption capacity for Pb(II) after fifth reuse. Insignificant influences of co-existing cations and natural organic matter (NOM) were found on the adsorption of Pb(II) and Cu(II). All the results demonstrate that BAC@SiO 2 -EDTA is a potential adsorbent for metal ions in wastewater.

Adsorption of Cu (II), Pb (II) and Cr (VI) from aqueous solutions using black wattle tannin-immobilized nanocellulose

Qinghua Xu,Yulu Wang,Liqiang Jin,Yu Wang,Menghua Qin

A novel nanocomposite based on black wattle (BW) tannin and nanocellulose was prepared and applied in heavy metal ions adsorptive removal from aqueous solutions. Firstly, nanocrystalline cellulose was oxidized by sodium periodate to get dialdehyde nanocellulose (DANC). BW tannin was then covalently immobilized onto DANC, which was used as both the matrix and crosslinker, to obtain tannin-nanocellulose (TNCC) composite. The resulting nanocomposite was characterized using FTIR, AFM, and TG. The successful immobilization was confirmed by the chromogenic reaction between FeCl 3 and TNCC and FT-IR analysis. AFM images revealed that TNCC was ellipsoidal particles with lengths ranging from 100–400   nm. Zeta potential measurement showed that TNCC was negative charged at a pH range from 1–12. Compared to the original tannin, the thermal stability of TNCC was slightly increased by the addition of nanocellulose. TNCC demonstrated the maximum adsorption efficiency at pH   2 for Cr(VI) and pH 6 for Cu(II) and Pb(II), respectively. The adsorption for these three metal ions followed pseudo second-order kinetics, indicating the chemisorption nature. The adsorption isotherms all fitted well with the Sips model, and the calculated maximum adsorption capacities were 51.846   mg   g −1 , 53.371   mg   g −1 and 104.592   mg   g −1 for Cu(II), Pb(II) and Cr (VI), respectively.

NH2-rich polymer/graphene oxide use as a novel adsorbent for removal of Cu(II) from aqueous solution

Hai Tao Xing,Jian Hua Chen,Xue Sun,Yi Hong Huang,Zhen Bo Su,Shi Rong Hu,Wen Weng,Shun Xing Li,Hong Xu Guo,Wen Bing Wu,Ya Shan He,Fei Ming Li,Yang Huang

Graphene oxide nanosheets (GO) were dispersed and crosslinked with a water soluble polymer, namely poly(allylamine hydrochloride) (PAH) with high amine density, to prepared a novel PAH-GO composite, which was used for removing Cu(II) ions from aqueous solutions. The physico-chemical properties of PAH-GO composites were investigated by transmission electron microscopy, thermogravimetric analysis, etc. Adsorption experiments indicated that adsorption process was strongly dependent on pH and temperature, however weakly dependent on ionic strength. The maximum adsorption capacity was about 349.03   mg/g higher than that of currently reported. Competitive adsorption studies for the ternary system suggested that the adsorption of PAH-GO composite for Cu(II) ions was much preferable to Cd(II) and Ni(II) ions. Thermodynamic parameters suggested that adsorptions were endothermic and spontaneous processes. The Langmuir adsorption isotherm was applicable fit adsorption process, and adsorption kinetic process could be described by the pseudo-second-order model. Adsorption–desorption results showed that the reusability of PAH-GO composite was encouraging. Based on the results obtained, we conclude that PAH-GO composite can be an effective and potential adsorbent for removing Cu(II) ions from an aqueous solution.

Magnetization of a Cu(II)-1,3,5-benzenetricarboxylate metal-organic framework for efficient solid-phase extraction of Congo Red

Xu Yan,Jin Jingjie,Li Xianliang,Han Yide,Meng Hao,Song Chaosheng,Zhang Xia

We describe a facile method for the preparation of magnetic microparticles consisting of a metal-organic framework (MOF) of Cu(II)-BTC (BTC = 1,3,5-benzenetricarboxylate) for rapid magnetic solid-phase extraction (MSPE) of the dye Congo Red (CR) from aqueous solution. Magnetization of the MOF and solid-phase extraction of CR was simultaneously accomplished by mixing, under ultrasonication, the MOF and silica-coated magnetite microparticles in the solution to be extracted. Under optimized conditions, the magnetic MOF hybrid exhibits a fast adsorption rate and a high removal efficiency (>97 %) toward CR even over 15-fold reuse, which also features high adsorption capacity of 97.7 and 92.5 % for cationinc Methylene Blue (MB) and Crystal Violet (CV), respectively. The desorption of CR from the magnetic MOF was realized by washing it with ethanol and water. The material is considered to be a promising new adsorbent for use in wastewater treatment and in analytical preconcentration. We report on a method for magnetization of a Cu-BTC metal-organic framework for use in rapid magnetic solid phase extraction of Congo Red from aqueous solutions. The material maintains its high removal efficiency (of >97 %) over 15 recyclable experiments.

From simulation to experiment: A Sagnac interference-based double holes optical fiber sensor for ultrasensitive Cu(II) detection

Qiang Chen,Yi Cai,Tianyu Jiang,Qing Chen,Jianhua Wang,Yong Zhao

Double-hole optical fiber (DHF) exhibits considerable potential for sensing applications due to its distinctive structural characteristics. In this study, a Sagnac interference (SI) optical fiber sensor based on a corroded DHF shows high refractive index (RI) sensitivity, allowing for the detection of copper ions, Cu(II), at very low concentrations. The dependence of DHF corrosion on the birefringence and RI sensitivity are investigated by the optical simulation. Experimental results illustrate that the RI sensitivity reaches 3267 nm/RIU within the range of 1.3333–1.3743, exhibiting good linearity. A chitosan derivative (CSD) with ion-imprinted (IIM) treatment is coated on the surface of the corroded DHF for Cu(II) detection. It is demonstrated that within the concentration range of Cu(II) from 10 −15 to 10 −6 M, both the maximum and average sensitivities can reach 12.08 × 10 9 and 129.96 nm/μM, respectively, without interference from other metal ions or temperature (25.3–44.0 ℃). The proposed corroded DHF sensor is a good candidate for RI and Cu(II) concentration detection due to its simple structure, high sensitivity, and selectivity.

Effect of Cu(II) and Conserved Copper Binding Sites on the Multicopper Oxidase CopA and Characterization of BioMnOx

Wenwei Tang,Zuxun Huang,Yunying Liu,Xinping Zeng

The microbial manganese removal process is believed to consist of the catalytic oxidation of Mn(II) by manganese oxidase. In this study, the multicopper oxidase CopA was purified and exhibited high manganese oxidation activity in vitro, and it was found that Cu(II) can significantly enhance its manganese oxidation activity. Gene site-directed mutagenesis was used to mutate four conserved copper binding sites of CopA to obtain four mutant strains. The manganese removal efficiencies of the four strains were determined, and it was found that H120 is the catalytically active site of CopA. The loss of Cu(II) and the mutation of the conserved copper binding site H120 resulted in the loss of ethoxyformyl and quinone modifications, a reduction in the number of modifications, and a change in the position of modifications, eventually causing a decrease in protein activity from 85.87% to 70.1%. These results reveal that Cu(II) and H120 play an indispensable role in manganese oxidation by the multicopper oxidase CopA. X-ray photoelectron spectroscopy (XPS) analysis indicates that biogenic manganese oxides produced by strains and by CopA were both composed of MnO 2 and Mn 3 O 4 and that the average valence of Mn was 3.2.
