Tuning complexation of carboxymethyl cellulose/ cationic chitosan to stabilize Pickering emulsion for curcumin encapsulation

Xiangwei Zhu,Jianjun Chen,Yuxi Hu,Ni Zhang,Yuqing Fu,Xiaoqiang Chen

Carboxymethyl cellulose (CMC) is an economical and easily available cellulose derivative that ionizes in water and is used in several charge-related interfacial applications. However, compared with other chemically functionalized cellulose derivatives, such as cellulose nanocrystals, the development of CMC-based (CMC as the major ingredient) O/W emulsions remains unexplored. In this study, we synthesized a CMC and quaternized chitosan (QCS) complex for the preparation of Pickering emulsions . Partially protonated CMC was complexed with QCS, followed by neutralization at pH 7 to obtain the CMC/QCS (CQ) particles. The prepared CQ-stabilized emulsion showed a gel-like rheological behavior and exhibited emulsion stability against several environmental stresses, including pH, salt, temperature, and long-term storage. The curcumin was then encapsulated in the CQ-stabilized emulsion and displayed a reduced degradation rate. This study provides a new route for the development of a Pickering emulsion delivery system with improved stability, sustainability and reduced complexity.

Stabilization of zein nanoparticles with k-carrageenan and tween 80 for encapsulation of curcumin

Xiaoxiao Sun,Chuqiao Pan,Zhuoyang Ying,Dongyan Yu,Xitai Duan,Fangfang Huang,Junhong Ling,Xiao-kun Ouyang

We aimed to develop a stable nanosystem with k -carrageenan (Kc), an anionic polysaccharide , combined with tween 80 to stabilize zein nanoparticles. The experimental results showed that the particle size, zeta-potential, and stability of the system depend on the Kc amount when the zein amount was unchanged. We found that the nanosystem with 5.0 and 10.0 mg Kc had a smaller particle size (250.1 ± 0.9 and 287.6 ± 1.4 nm, respectively), low polydispersity index (PDI, ≤ 0.20), and high stability when the external conditions changed. No obvious flocculation or deposition was observed at pH of 3–9, salt ion concentrations of 0–500 mmol/L, temperature of 30 °C and 50 °C–90 °C, and storage at 4 °C for 30 days. When the Kc-ZNP system was loaded with curcumin (Cur), there was a good encapsulation effect (5.0 mg Kc with 1.00 mg Cur (EE (%) = 34.69% ± 2.02%, size = 380.23 ± 5.80 nm, PDI = 0.252 ± 0.018) and 10.0 mg Kc with 2.00 mg Cur (EE (%) = 20.13% ± 1.05%, size = 431.27 ± 0.78 nm, PDI = 0.269 ± 0.020), ensuring that the particle size was not too large. Therefore, Kc–ZNPs could be a potential delivery system for fat-soluble drugs.

Effect of curcumin on the formation of polycyclic aromatic hydrocarbons in grilled chicken wings

Huixin Tian,Jing Yu,Min Li,Jing Li,Yifeng Lu,Xiaobo Yu,Shaoyan Lin,Xianming Zeng,Xinglian Xu,Minyi Han

Organic macromolecules form carcinogenic and toxic substances such as polycyclic aromatic hydrocarbons (PAHs) under high temperature baking. Thus, this study investigated the effects and inhibition pathways of different curcumin concentrations (0.01, 0.05, 0.25, 0.3 mg/g) on seven PAHs in grilled chicken wings. The results demonstrated that curcumin concentrations displayed positive effects in inhibiting the formation of PAHs (16%–72%), increasing the total phenolic content (397.5–1934.4 mg/g) and free radical scavenging activity, and reducing TBARS values (31.15%–47.76%) and fatty acid content. Additionally, PCA and Pearson correlation analyses indicated that lipid oxidation (r = 0.42) and unsaturated fatty acids (r = 0.55) could promote the production of PAHs, while DPPH, ABTS and TPC could counteract their facilitation of PAHs. In conclusion, the addition of appropriate amounts of curcumin before grilling is a feasible strategy to reduce fat oxidation levels and the number of free radicals for the purpose of limiting PAHs content.

An investigation of IRMOF-16 as a pH-responsive drug delivery carrier of curcumin

Mengru Cai,Boran Ni,Xueling Hu,Kaixin Wang,Dongge Yin,Gongsen Chen,Tingting Fu,Rongyue Zhu,Xiaoxv Dong,Changhai Qu,Xingbin Yin

Metal–organic frameworks (MOFs) with tunable structures, high porosity, and abundant active sites are considered to be promising drug delivery systems (DDSs). In this study, novel MOFs-based nanoparticles ( [email protected] ) were successfully prepared using Zn-based IRMOF-16 as a carrier and curcumin (CUR) as the model drug. This nanocargo delivery platform has dual capabilities, enabling simultaneous drug delivery and fluorescence imaging. In vitro release experiments showed that nanoparticles were released more quickly under mildly acidic conditions, which proved their potential for tumor microenvironment-responsive drug release. Through in vitro cell experiments, it was found that the nano-drug platform had high antitumor cytotoxicity, which may be related to the mechanism of increasing intracellular reactive oxygen species (ROS), reducing intracellular mitochondrial membrane potential (MMP), and inducing apoptosis. In addition, confocal laser microscopy showed that [email protected] could localize to the nucleus to exert an antitumor effect. Meanwhile, [email protected] exhibited superior antitumor activity compared with pure CUR in vitro experiments. The biocompatibility test of IRMOF-16 showed reasonable biosafety, and no evidence of obvious toxicity was observed in vitro. [email protected] has unique advantages with regard to pH response, biocompatibility, and antitumor efficacy, indicating that the nano-drug platform is a promising drug delivery carrier with an antitumor effect.

Novel Reductive Responsive Chrysin-Oligomeric Hyaluronic Acid Nanomaterials to Curcumin Delivery for Cancer Therapy

Wang Bingjie,Ma Chuanjiang,Guo Chunjing,Wang Kaili,Yuan Dehao,Cheng Haoran,Liu Mengna,Fang Lei,Fan Xinxin,Chen Daquan

Drug resistance, perplexing a lot of doctors and patients, was an inevitable challenge in the field of effective cancer therapy. Poor water solubility, inefficient aggregation at tumor site and killing normal cells were also the great obstacles, which gave rise to failure in the tumor clinical treatment. In order to overcome these difficulties, modified targeting nano-micelles (MT-NMs) loading Curcumin (Cur) and Chrysin (Chr) were designed. In this work, dithiodipropionic acid (DA), a reduction-sensitive material, acted as a "messenger" linker connecting Chr and oligomeric hyaluronic acid (oHA). Then, the amphiphilic polymer, oligomeric hyaluronic acid-dithiodipropionic acid-Chrysin (oHA-DA-Chr), was synthesized. What's more, combinational therapy is a better way to cripple cancer drug-resistant and reinforce the therapeutic efficacy. In this study, 1H-NMR was used to authenticate the structure of oHA-DA-Chr. The amphiphilic oHA-DA-Chr could self-assemble into nano-micelles (NMs) in water. The NMs with small size (223.2±2.4 nm) could concentrate well at tumor sites by enhancing permeability and retention effect (EPR) and CD44 receptor targeting property of oHA. The Cur could release well from NMs when disulfide bond (S–S) of DA was exposed to tumor environment containing high concentration of glutathione (GSH).

Weiya Cao,Yinci Zhang,Amin Li,Pan Yu,Li Song,Jiaojiao Liang,Niandie Cao,Jiafeng Gao,Ruyue Xu,Yongfang Ma,Xiaolong Tang

Objective Epithelial mesenchymal transition (EMT) and inflammation have been identified as carcinogenic agents. This study aims to investigate whether inhibition of trichloroethylene (TCE) associated hepatocellular carcinoma (HCC) by curcumin is associated with inflammation and EMT.Methods In the current study, TCE sub-chronic cell model was induced in vitro, and the effects of TCE on cell proliferation, migration, invasion, and expression of functional proteins were verified by Western blot, MTT, clone formation, wound healing, Transwell. The detoxification effect of curcumin on TCE was explored by a mouse tumor-bearing experiment. Results TCE induces hepatocyte migration, colony formation, and EMT in vitro. In vivo studies have shown that curcumin significantly reduces the mortality of mice and control the occurrence and size of liver tumors by inhibiting the IL-6/STAT3 signaling pathway. In vitro, curcumin inhibits the proliferation of HepG2 cells as determined by MTT assay. In addition, curcumin significantly inhibited the protein expression of IL-6R, STAT3, snail, survivin, and cyclin D1 in THLE-2 and HepG2 cells induced by IL-6.Conclusion Curcumin has anti-inflammatory and anti-proliferative effects, and inhibits the development of HCC induced by TCE by reversing IL-6/STAT3 mediated EMT.

Curcumin attenuates intracerebral hemorrhage-induced neuronal apoptosis and neuroinflammation by suppressing JAK1/STAT1 pathway

Fei Wang,Jian-jun Xia,Li-juan Shen,Ting-ting Jiang,Wu-lin Li,Da-li You,Qing Chang,Shan-you Hu,Li Wang,Xiao Wu

Intracerebral hemorrhage (ICH) is a kind of fatal stroke with the highest mortality and morbidity in the world. To date, there is no effective treatment strategy for ICH. Curcumin, a major active ingredient of Curcuma longa L., possesses a potential anti-inflammatory activity in many types of disease. In the current study, the mechanism underlying curcumin attenuated ICH-induced neuronal apoptosis and neuroinflammation was explored. Herein, we studied that curcumin decreased brain edema and improved neurological function by using brain edema measurement, assessment of neurological-deficient score, immunofluorescence, and Western blotting analyses after ICH. The results showed that curcumin improved ICH-induced neuronal apoptosis and neuroinflammation. Functionally, the polarization of microglia was assessed by immunofluorescence and Western blotting analyses after ICH in the absence or presence of curcumin. The results suggested that the M1-type microglia were activated after ICH, while the effect was blocked by curcumin treatment, suggesting that curcumin alleviates the neuroinflammation and apoptosis of neurons by suppressing the M1-type polarization of microglia. Mechanically, M1 polarization of microglia was regulated by JAK1/STAT1, and the activation of JAK1/STAT1 was blocked by curcumin. Meanwhile, the protective function of curcumin can be blocked by RO8191, an activator of JAK1. Taken together, our study suggested that curcumin improved the ICH-induced brain injury through alleviating M1 polarization of microglia/macrophage and neuroinflammation via suppressing the JAK1/STAT1 pathway.

Yan Yang,Yiling Wang,Xinran Jiang,Jiahao Mi,Dizhang Ge,Yuna Tong,Yuxuan Zhu

Introduction Alzheimer’s disease (AD), a neurodegenerative condition, stands as the most prevalent form of dementia. Its complex pathological mechanisms and the formidable blood-brain barrier (BBB) pose significant challenges to current treatment approaches. Oxidative stress is recognized as a central factor in AD, underscoring the importance of antioxidative strategies in its treatment. In this study, we developed a novel brain-targeted nanoparticle, Ce/Zr-MOF@Cur-Lf, for AD therapy. Methods Layer-by-layer self-assembly technology was used to prepare Ce/Zr-MOF@Cur-Lf. In addition, the effect on the intracellular reactive oxygen species level, the uptake effect by PC12 and bEnd.3 cells and the in vitro BBB permeation effect were investigated. Finally, the mouse AD model was established by intrahippocampal injection of Aβ1-42, and the in vivo biodistribution, AD therapeutic effect and biosafety of the nanoparticles were researched at the animal level. Results As anticipated, Ce/Zr-MOF@Cur-Lf demonstrated efficient BBB penetration and uptake by PC12 cells, leading to attenuation of H2O2-induced oxidative damage. Moreover, intravenous administration of Ce/Zr-MOF@Cur-Lf resulted in rapid brain access and improvement of various pathological features of AD, including neuronal damage, amyloid-β deposition, dysregulated central cholinergic system, oxidative stress, and neuroinflammation. Conclusion Overall, Ce/Zr-MOF@Cur-Lf represents a promising approach for precise brain targeting and multi-target mechanisms in AD therapy, potentially serving as a viable option for future clinical treatment.

Photothermal and Immunomodulatory Hydrogels Loaded with Black Phosphorus Nanosheets and Curcumin for Wound Healing

Rong Xiao,Longbao Feng,Xinyue He,Yifan Hao,Zhong Cao,Aliaksandr Astrowski,Yi Zhang,Rui Guo

In the field of skin wound treatment, traditional methods are often limited by the dual challenges of effectively clearing bacterial infections and accelerating wound healing. To address these issues, this study developed an innovative bioactive hydrogel that combines photothermally responsive antimicrobial and immunomodulatory effects. The hydrogel utilized methacrylated gelatin and oxidized dextran, forming a stable network structure through radical and Schiff bonds, loaded with black phosphorus (BP) nanosheets and curcumin (Cur). Characterization of the hydrogel’s physicochemical properties revealed not only its excellent swellability, biodegradability, mechanical strength, thermal stability, and antioxidative properties but also its ability to control the release of Cur through the photothermal effect. Under 808 nm NIR irradiation, the suitable photothermal effect was observed with BP nanosheets at a concentration of 250 μg/mL under a power density of 1.5 W/cm2, and the photothermal conversion efficiency (η) was 41.34%. In vitro experiments showed that the hydrogel exhibited a bactericidal rate of over 90% against E. coli, S. aureus, and P. aeruginosa and more than 80% antibiofilm formation activity. After coculturing with HUVEC cells for 24 and 48 h, cell viability remained above 80%. Cell migration assay confirmed the hydrogel’s effectiveness in promoting cell migration (closure rate reached 97.4% within 24 h). In a rat model with infected wounds, it was shown that on day 14, wounds were nearly healed, with an antibacterial rate of over 85%, the expression levels of proinflammatory factors TNF-α and IFN-γ were reduced, the expression of anti-inflammatory factors IL-4 and TGF-β1 was increased, re-epithelialization was observed, and collagen was deposited, which facilitated the rapid healing of the wounds. The hydrogel in this study with photothermal and immunomodulatory capabilities offers an efficient strategy for treating infected wounds.

Hyaluronic acid-decorated curcumin-based coordination nanomedicine for enhancing the infected diabetic wound healing

Shuai Wu,Li Zhu,Sheng Ni,Yuan Zhong,Kai Qu,Xian Qin,Kun Zhang,Guixue Wang,Da Sun,Wuquan Deng,Wei Wu

Persistent over-oxidation, inflammation and bacterial infection are the primary reasons for impaired wound repairing in diabetic patients. Therefore, crucial strategies to promote diabetic wound repairing involve suppressing the inflammatory response, inhibiting bacterial growth and decreasing reactive oxygen species (ROS) within the wound. In this work, we develop a multifunctional nanomedicine (HA@Cur/Cu) designed to facilitate the repairing process of diabetic wound. The findings demonstrated that the synthesized infinite coordination polymers (ICPs) was effective in enhancing the bioavailability of curcumin and improving the controlled drug release at the site of inflammation. Furthermore, in vitro and in vivo evaluation validate the capacity of HA@Cur/Cu to inhibit bacterial growth and remove excess ROS and inflammatory mediators, thereby significantly promoting the healing of diabetic wound in mice. These compelling findings strongly demonstrate the enormous promise of this multifunctional nanomedicine for the treatment of diabetic wound.

Curcumin Pickering emulsion film stabilized by guanidine-functionalized CNF for food monitoring and preservation

Qianyu Sun,Ye Jiang,Yiyun Jin,Junyi Wu,Tianzhong Yuan,Zhixin Jia,Zhihao Wang,Guihua Yang

Guanidine-nanocellulose encapsulates curcumin forming a stable Pickering emulsion. • Emulsion encapsulation ensures curcumin stability and pH-responsive color change. • The film enables simultaneous food freshness monitoring and shelf-life extension. • The film offers an antibacterial coating for fruit surfaces that is easy to clean. Traditional food packaging suffers from drawbacks such as environmental pollution and limited functionality. Herein, we fabricated a multifunctional film based on a curcumin Pickering emulsion stabilized by cellulose nanofibrils grafted with polyhexamethylene guanidine (PCNF). PCNF acts as both a Pickering emulsifier and an antibacterial agent. The PCNF-stabilized emulsion not only protects curcumin from degradation but also forms a composite film after incorporation of PVA and glycerol. This film exhibits good mechanical strength, color stability and pH-responsive color changes. In shrimp freshness monitoring tests, the film showed a distinct color transition from pale yellow to brown. Moreover, the tunable rheology of the emulsion allows adaptable coating on fruits. After 7 days of storage, the composite coating significantly reduced banana browning, owing to its exceptional antibacterial performance. Notably, the combination of PCNF and glycerol enables rapid water disintegration. The film completely disperses within 30 s of immersion, imparting outstanding easy-cleaning properties. These synergistic features provide a robust foundation for practical applications of this material in the field of food packaging and preservation.
