A novel ratiometric fluorescent nanohybrid probe was constructed for sensitive and selective determination of folic acid (FA) based on blue D-penicillamine-based carbon dots (CDs) and red glutathione S-transferase-Au nanoclusters (GST-AuNCs). Upon the excitation of 380 nm, the obtained CDs-NCs possessed two distinct emission peaks at 465 and 665 nm. The fluorescence intensity at 465 nm was incrementally enhanced with the addition of FA attributed to the hydrogen bonds formation, while the fluorescence intensity at 665 nm was quenched caused by the electronic interaction and the inner filter effects. The fluorescence intensity ratio ( I 465 /I 665 ) exhibited good linear correlation with FA concentrations in the range 10∼90 μM, and the limit of detection (LOD) was 0.63 μM. Notably, the corresponding fluorescent color changed from red to blue, which could be distinguished by naked eyes. On account of the excellent biocompatibility, the CDs-NCs were further successfully used for bioimaging and intracellular FA detection. Furthermore, the real sample analyses confirmed that the proposed nanoprobe could be expanded as a versatile platform for FA detection in practical applications. Graphical abstract

Optical imaging in vivo holds significant implications for disease diagnosis, and nanoprobes with near-infrared (NIR) emission leverage the deep tissue penetration and high spatiotemporal resolution provided by NIR light, demonstrating considerable application potential. This study presents the design and synthesis of three nitrogen-doped boron–dipyrrin (Aza-BODIPY) molecules: Aza–BDP–OCH 3 , Aza–BDP–OH, and Aza–BDP-I. Leveraging the strong electron-accepting properties of the Aza-BODIPY core, we developed a donor–acceptor–donor (D-A-D) structure for Aza–BDP–OCH 3 through modifications with triphenylamine and methoxy groups, resulting in NIR fluorescence. Aza–BDP–OH was obtained via demethylation using boron tribromide, whereas Aza–BDP-I was synthesized by introducing iodine into Aza–BDP–OCH 3 . These three molecules self-assemble with the amphiphilic polymer PMHC 1 8 -mPEG to form nanoparticles (NPs), yielding optical nanoprobes. The resulting NPs exhibit NIR emission, good water solubility, and biocompatibility. At a concentration of 100 μ g ⋅ mL − 1 , these NPs demonstrate low biological toxicity, highlighting their potential for biological applications. Following tail vein injection, Aza–BDP-I NPs accumulate in tumors and effectively illuminate them via the enhanced permeability and retention (EPR) effect. Furthermore, these organic NPs were metabolized by the liver. Therefore, Aza-BODIPY-based NIR fluorescent NPs offer a promising platform for the development of in vivo optical nanoprobes.

As global milk production continues to rise, the disposal of expired milk contributes to environmental pollution and valuable resource wastage. This study presents the development of a novel L-arabinose isomerase, designatedBmAIase12, and its application in the enzymatic recycling of expired milk.BmAIase12 exhibited a specific activity of 10.7 U/mg and showed optimal performance at 50 °C and pH 7.0. Furthermore, it exhibited higher activity than most other L-arabinose isomerases. It converted D-galactose into D-tagatose with a high conversion ratio of 53.3% after 48 h at 50 °C. The conversion efficiency of expired milk to D-tagatose was recorded at 40.62%, resulting in a maximum tagatose yield of 1.625 g/L. This was accomplished through the incorporation of β-galactosidase (120 U/mL) andSaccharomyces cerevisiae(30 mg/mL) to hydrolyze lactose and metabolize glucose, followed by the addition of 3 U/mL ofBmAIase12. Ultimately, following purification, the purity of tagatose was determined to be 98%, and the final yield was 29.8%. These results suggest thatBmAIase12 may serve as a promising enzyme for D-tagatose production due to its high conversion yield.

D-amino acid oxidase (DAAO)-catalyzed selective oxidative deamination is a very promising process for synthesizing l -amino acids including l -phosphinothricin ( l -PPT, a high-efficiency and broad-spectrum herbicide). However, the wild-type DAAO's low activity toward unnatural substrates like d -phosphinothricin ( d -PPT) hampers its application. Herein, a DAAO from Caenorhabditis elegans ( Ce DAAO) was screened and engineered to improve the catalytic potential on d -PPT. First, we designed a novel growth selection system, taking into account the intricate relationship between the growth of Escherichia coli ( E. coli ) and the catalytic mechanism of DAAO. The developed system was used for high-throughput screening of gene libraries, resulting in the discovery of a variant (M6) with significantly increased catalytic activity against d -PPT. The variant displays different catalytic properties on substrates with varying hydrophobicity and hydrophilicity. Analysis using Alphafold2 modeling and molecular dynamic simulations showed that the reason for the enhanced activity was the substrate-binding pocket with enlarged size and suitable charge distribution. Further QM/MM calculations revealed that the crucial factor for enhancing activity lies in reducing the initial energy barrier of the reductive half reaction. Finally, a comprehensive binding-model index to predict the enhanced activity of DAAO toward d -PPT, and an enzymatic deracemization approach was developed, enabling the efficient synthesis of l -PPT with remarkable efficiency.

Effective cancer immunotherapy is often hindered by V-domain Ig suppressor of T cell activation (VISTA) signaling and myeloid-derived suppressor cells (MDSCs) within tumor microenvironment (TME). Here, we report D-peptide-nanodrug conjugates that synergize VISTA blockade and innate activation to overcome these constraints. We developed Reorpn7, a proteolytically stable D-peptide VISTA inhibitor with high binding affinity and in vitro serum stability. This inhibitor was co-loaded with a toll-like receptor 7/8 (TLR7/8) agonist (R848) into an matrix metalloproteinase-2 (MMP-2) responsive poly lactic-co-glycolic acid (PLGA) nanoplatform (r7PL-PLGA@R848). Upon enrichment in the TME, MMP-2 responsive cleavage enables the precise release of the VISTA-targeting peptide Reorpn7 and the TLR7/8 agonist R848, thereby achieving synergistic therapeutic effect of VISTA blockade and innate immune activation. In MC38 tumor models, this nanosystem significantly increased intratumoral CD8 + T cell infiltration and achieved robust antitumor efficacy without obvious systemic toxicity. Mechanistically, the platform reshaped the TME from an inhibitory milieu into an immunosupportive one by coordinating checkpoint blockade, myeloid reprogramming, and T cell recruitment. This stimuli-responsive paradigm offers a translatable strategy to overcome immune checkpoint blockade resistance in challenging malignancies.

The anti-glycation effects and mechanisms of eight quinone-related compounds, primarily sourced from medicinal and edible plants, were evaluated in this study. In vitro glycation experiments were conducted to study the structural perturbation induced by d -ribose on human myoglobin (HMb) in the presence of the tested compounds. Spectroscopic and electrophoretic analyses indicated that among the four benzoquinone-related substances, thymoquinone and its structurally closest analogue 2,5-dimethyl-1,4-benzoquinone, exhibited significant inhibitory effects on the d -ribose-induced glycation, thereby maintaining protein structural stability, reducing advanced glycation end products (AGEs) formation, and attenuating the covalent protein modification by d -ribose. In contrast, 1,4-benzoquinone and hydroquinone (a reduction product of 1,4-benzoquinone) promoted HMb denaturation through the Michael addition mechanism. Of the two naphthoquinones, juglone and plumbagin, showed anti-glycation effects, whereas the anthraquinones emodin and chrysophanol showed no significant effects. Nano-HPLC-MS/MS analysis further showed that thymoquinone was more effective in preventing glycation than juglone and plumbagin. This study investigates the mechanisms underlying glycation inhibition by quinones, revealing that the presence of dual β -substituted carbons on the benzoquinone ring is critical for stabilizing protein conformations and suppressing glycation. It provides a preliminary mechanistic basis and identifies candidate molecules for potential applications of quinones in retarding glycation, such as in managing diabetic complications or inhibiting glycation reactions during food processing.

I n this work, an electrochemical chiral sensor of a nanowire-like chiral metal-organic framework/multiwalled carbon nanotube-chitosan/glassy carbon electrode( D -MOF/MWCNTs-CS/GCE) was proposed for the enantiorecognition of L / D -tryptophan( L / D -Trp). The asymmetrical spatial structure of D -MOF provides the feasibility for the enantiorecognition of Trp enantiomers. Moreover, differential pulse voltammetry(DPV) was carried out to be the detection method and the DPV peak potential difference(Δ E p ) between L -Trp and D -Trp was referred as the index of the enantiorecognition performance. Several parameters, such as mass ratios and drop-coated volume of MWCNTs-CS, drop-coated volume and concentration of D -MOF, pH and detection temperature of D -MOF/MWCNTs-CS/GCE were optimized for the largest Δ E p value. And the molecular dynamics(MD) simulation was used to elucidate the enantiorecognition mechanism. Furthermore, the proportions of D -Trp( D -Trp%) in Trp mixtures were detected in a good linear relationship with the DPV peak potentials( E p ), and the proposed electrochemical chiral sensor exhibited good reproducibility, stability and enantiorecognition ability. Additionally, the proposed electrochemical chiral sensor( D -MOF/MWCNTs-CS/GCE) has a good application prospect in the fields of biomedicine, clinical diagnosis, chemical production, pharmaceuticals safety and so on.

Monosaccharides from biomass can be used as cost-effective raw materials to produce rare sugars which are valuable sweeteners. Enzymes play a pivotal role in such biomass biorefinery processes to develop novel synthetic routes, and improve the environmental friendliness of current routes. Here we propose a proof of concept biorefinery process to prepare useful rare sugars of D-xylulose and D-ribulose from D-xylose which was abundant in biomass. Ribose-5-phosphate isomerase B from Ochrobactrum sp. (OsRpiB) with a unique substrate scope was engineered to intensify the activity of transforming D-xylose to D-xylulose. A double mutant R94N/N137Q with 80% higher catalytic efficiency was identified, and kinetic and molecular dynamic studies were conducted to explore the mechanism underneath. The results revealed a correlation between enhancing stability and activity simultaneously. Then a one-pot enzyme cascade was constructed with OsRpiB and D-tagatose-3-epimerase from Pseudomonas cichorii and transformed D-xylulose further to D-ribulose. The conversions of D-xylose and D-xylulose in cascade reaction were 1.80-fold and 1.83-fold of that in separate reactions with the aid of process parameter adjustment. The presented work is the first reported enzyme cascade converting D-xylose to D-ribulose, and proves the promising role of monosaccharide isomerase platforms in the refinery of biomass.

Elimination of specific enteropathogenic microorganisms is critical to gut health. However, the complexity of the gut community makes it challenging to target specific bacterial organisms. Accumulating evidence suggests that various foods can change the abundance of intestinal bacteria by modulating prophage induction. By using pathogenic Escherichia coli ( E. coli ) ATCC 25922 as a model in this research, we explored the potential of dietary modulation of prophage induction and subsequent bacterial survival. Among a panel of sugars tested in vitro, D -xylose was shown to efficiently induce prophages in E. coli ATCC 25922, which depends, in part, upon the production of D -lactic acid. In an enteric mouse model, prophage induction was found to be further enhanced in response to propionic acid. Dietary D -xylose increased the proportion of Clostridia which converted D -lactic acid to propionic acid. Intestinal propionic acid levels were diminished, following either oral gavage with the dehydrogenase gene ( ldhA )-deficient E. coli ATCC 25922 or depletion of intestinal Clostridia by administration of streptomycin. D -Xylose metabolism and exposure to propionic acid triggered E. coli ATCC 25922 SOS response that promoted prophage induction. E. coli ATCC 25922 mutant of RecA, a key component of SOS system, exhibited decreased phage production. These findings suggest the potential of using dietary components that can induce prophages as antimicrobial alternatives for disease control and prevention by targeted elimination of harmful gut bacteria.

Polymer solution has extremely extensive applications in many natural and industrial processes, especially in oilfield development field, such as polymer flooding, fracturing and water shut-off. Thus, the study of flow behaviors of polymer in formation pores is significantly important. In this paper, the flow behaviors of the polymer droplets in the 3-D pore throat structure were systematically studied. Additionally, the influencing factors (polymer concentration, molecular weight and pore throat ratio, for instance) were investigated. As the increasing of polymer concentration and molecular weight, the polymer droplets were more difficult to break, which means the critical flow rate decreased and the average sizes of the first daughter droplets (FDD) were longer synchronously. Moreover, with the increase in pore throat ratio, the critical flow rate increased and the length of the FDD decreased. In addition, the prediction models of the length of the FDD with polymer concentration and pore throat ratio were established, respectively. The prediction model revealed that the length of the FDD satisfied an exponential relationship with the polymer concentration and a linear relationship with the pore throat ratio. Finally, the average size of droplets after macroscopic core flooding experiment was 8 μm and 17 μm when the polymer concentration was 0.01% and 0.1%, respectively. The results were consistent with the breakup behaviors of polymer droplets in the microscopic pore throat structure.

Objective Hepatocellular carcinoma (HCC) is one cancer with high death rates. Nowadays, there are no effective drugs to treat it. Cyclovirobuxine D (CVB-D) is the primary ingredient of the traditional Chinese medicine (TCM) Buxus microphylla . Here, we try to explore the impacts of CVB-D on human HCC cells and explain the potential mechanisms. Methods HepG2 and Huh-7 cells were used for our experiments. The cell viability and half inhibitory concentration (IC50) were detected by MTT assays. The apoptosis ratio was examined by Annexin V-FITC/7AAD staining and flow cytometry (FCM). The Fe 2+ content was examined by ferrous ion content assays. The malondialdehyde (MDA) content was evaluated by lipid peroxidation MDA assays. The reactive oxygen species (ROS) level was examined by the DCFH-DA probe. The expression of apoptotic markers (Bax and Bcl-2) and ferroptosis-related proteins (GPX4 and FSP1) was detected by western blotting. The in vivo curative effect of CVB was explored using xenograft models established in C-NKG mice. Results The cell viability could be inhibited by CVB-D in HepG2 and Huh-7 cells. The IC50 value of CVB-D on HepG2 and Huh-7 cells are 91.19 and 96.29 µM at 48 h, and 65.60 and 72.80 µM at 72 h. FCM showed that the apoptosis rate was increased by CVB-D in HepG2 and Huh-7 cells. Next, ferrous ion content assays showed that the level of Fe 2+ was increased by CVB-D in HepG2 and Huh-7 cells. Then, we found the level of MDA and ROS was increased by CVB-D. And the Fe 2+ promotion by CVB-D could be reversed by Fer-1. Additionally, western blotting assays showed that the expression of GPX4 and FSP1 was inhibited by CVB-D in HepG2 and Huh-7 cells. Moreover, in vivo, CVB-D displayed excellent anticancer effects in HCC tumor-bearing C-NKG mice. Conclusion CVB-D suppresses the growth in HCC cells through ferroptosis.

d -Allulose, is an improtant rare sugar, having health-promoting properties and applications in the food, pharmaceutical and nutrition sectors. White the aldol reaction-based method shows promise as an alternative to the “Izumori” strategy for ᴅ-allulose bioproduction, its industrial exploitability is hindered by the low product yield in reaction equilibrium and the high costs associated with the use of purified enzymes . To address these limitations, this study explored cell-based biosynthesis of ᴅ-allulose from low-cost glycerol by overexpression the enzymes involved in the ᴅ-allulose aldol condensation (AC) biosynthesis pathway, namely fructose-1-phosphatase (YqaB), alditol oxidase (AldO), and l -rhamnulose-1-phosphate aldolase (RhaD), in Escherichia coli . Results indicated that RhaD is a rate-limiting step of the ᴅ-allulose biosynthetic pathway. Then, biosensor-based high throughput screening was employed for the directed evolution of RhaD. In this way, two RhaD mutants, namely RhaD N156D and RhaD P184S , were successfully screened from 40,129 individual colonies, which exhibited respectively achieved a ᴅ-allulose titer of 3.1 g/L and 2.9 g/L, respectively in shake flasks, representing a more than 20% improvement compared to the wildtype strain. Overall, the ᴅ-allulose biosensor-assisted key enzyme evolution strategy and the high throughput screening method presented in this work provide a paradigm for improving rare sugars’ high-valued productions from cheap substrates.
