Food safety is critically linked to human health, necessitating advanced monitoring of biogenic amines (BAs), key biomarkers of food freshness. Here, a supramolecular fluorescence probe was developed using sulfobutyl ether-β-cyclodextrin (SBE-β-CD) encapsulated nitro-spiropyran (SP), for ultra-sensitive sensing of histamine. Hydrophobic interactions drive the closed-loop SP into the SBE-β-CD cavity, forming a non-fluorescent complex (SBE-β-CD@SP). Upon histamine introduction, competitive displacement ejects SP from the cavity. The liberated SP undergoes rapid ring-opening under 365-nm UV excitation to form fluorescent merocyanine (MC), emitting the fluorescence signal. This turn-on response enables quantitative detection of histamine, exhibiting a linear range of 10–70 nM (R 2 = 0.996) with a low detection limit of 0.069 nM. The probe demonstrated high selectivity over interfering amines and achieved 94.2–97.9% recovery in spiked fish samples, validating real-world utility. This work establishes a host–guest displacement-based sensing strategy via histamine-triggered MC activation, which exhibits potential applicability to fish freshness assessment. By integrating supramolecular chemistry with nanomaterial design, this probe offers a novel approach for food safety monitoring.

Oral delivery is the most convenient drug administration route. However, oral delivery of peptides is extremely challenging due to the physical and chemical barriers within the gastrointestinal tract. Polysaccharides are often utilized as polymeric biomaterials in drug delivery. Among these, dietary polysaccharides extracted from okra, yam, and spirulina have been reported to stimulate innate immunity with well-known nutritional benefits. In this study, we developed a dietary-polysaccharide-modified fish-oil-based emulsion for oral co-delivery of a hydrophilic PD-L1 blocking peptide and the hydrophobic small molecule simvastatin. The optimal emulsion was nano-sized and exhibited a negative surface charge, high drug encapsulation efficiency of over 97%, low viscosity, and sustained drug release manner. The formulation could significantly increase the uptake of peptides by intestinal Caco-2 cells, which demonstrated the great potential of the formulation for promoting the oral absorption of peptides. Additionally, these dietary polysaccharides could promote dendritic cell maturation and cytokine expression in macrophages, demonstrating that these nutraceutical polysaccharides had dual roles of functioning as promising colloidal delivery systems and as potential immune modulators or adjuvants. Thus, this food-based colloidal delivery system shows promise for the oral delivery of peptide drugs and lays a great platform for future applications in immunotherapy. Keywords:dietary polysaccharide;water-in-oil-in-water emulsion;colloidal delivery system;immune modulator;PD-L1 blocking peptide;oral drug delivery

Due to many special characteristics, zinc oxide nanoparticles (ZnO NPs) are widely used all over the world, leading to their wide distribution in the environment. However, the toxicities and mechanisms of environmental ZnO NP-induced changes of physiological processes and metabolism remain largely unknown. Here, we found that addition of dietary ZnO NPs disturbed hepatic Zn metabolism, increased hepatic Zn and lipid accumulation, downregulated lipolysis, induced oxidative stress, and activated mitophagy; N,N,N′,N′-tetrakis (2-pyridylmethyl) ethylenediamine (TPEN, Zn2+ ions chelator) alleviated high ZnO NP-induced Zn and lipid accumulation, oxidative stress, and mitophagy. Mechanistically, the suppression of mitochondrial oxidative stress attenuated ZnO NP-activated mitophagy and ZnO NP-induced lipotoxicity. Taken together, our study elucidated that mitochondrial oxidative stress mediated ZnO NP-induced mitophagy and lipotoxicity; ZnO NPs could be dissociated to free Zn2+ ions, which partially contributed to ZnO NP-induced changes in oxidative stress, mitophagy, and lipid metabolism. Our study provides novel insights into the impacts and mechanism of ZnO NPs as harmful substances inducing lipotoxicity of aquatic organisms, and accordingly, metabolism-relevant parameters will be useful for the risk assessment of nanoparticle materials in the environment.

Zinc-based flow batteries receive widespread attention due to their advantages of low cost and high energy density. However, zinc dendrites are easy to appear during the charge process, pierce the membrane and thus destroy the battery, which seriously restrict its further development. In this article, MFI-type zeolite nanosheets ( ns -MFIs) with high mechanical strength and hydrophobicity are in situ introduced to porous polymer membranes, which spontaneously form turnup fish-scale-like structure through the one-step phase inversion/surface segregation process. This special structure well disperses mechanical energy to provide effective protection characteristics to resist the penetration of zinc dendrites, and meanwhile promotes the uniform zinc depositions on the electrode by alleviating the water migration and accelerating zincate ion diffusion, so as to prolong the cycle life of the battery for more than 600 cycles, which is 4 times and 2.5 times longer than the commercial Nafion 212 and pristine porous polymer membrane, respectively. Moreover, the sub-nano size pores and high-aspect-ratio of ns -MFIs afford membranes extra ion sieving ability and transport area for the charging-balancing ions OH − to ensure superior battery performance, delivering an average coulombic efficiency (CE) of ~98.5%, voltage efficiency (VE) of ~83.2%, and energy efficiency (EE) of ~81.9% at 80 mA/cm 2 .

A novel photoelectrochemical (PEC) aptasensor coupled with a Z‑scheme was developed to realize quantitative determination of 3,3′,4,4′-tetrachlorobiphenyl (PCB77) at a concentration of sub-picogram per liter. The PEC aptasensor was fabricated by immobilizing ZnSe/Ag 2 Se on indium-tin oxide (ITO) that could covalently bind sulfurized aptamer. Meanwhile, complementary DNA-functionalized CdTe units were used as the sensitizers, and these were immobilized on the aptamer to form Z‑scheme dual-photosystem (PS). The Z‑scheme PS based on both ZnSe/Ag 2 Se and CdTe would be provided with better photocatalytic activity than conventional heterojunctions , and improving the photo-to-current conversion efficiency without biased voltage. Following the process of hatching, PCB77 was captured via the aptamer on the PEC aptasensor, producing aptamer–PCB77 complexes with weak conductive performance. Using the photocurrent change as a response signal for the quantitative detection of PCB77, the detection limits of the proposed method were 0.05 pg L –1 and 0.22 ng L –1 for the linear ranges of 0.01–500 pg L –1 and 0.5–10 5 ng L –1 , respectively. Compared with other technologies, the PEC aptasensor is more flexible, sensitive, specific and rapid in the absence of biased voltage under visble light. The results presented herein can potentially help build a model for the detection of various polychlorinated biphenyls (PCBs) with stable chemical properties and insulation performances that can be used to study persistent organic pollutants (POPs) in marine organisms and in the analysis of environmental conditions.

In this work, konjac glucomannan (KGM)-based film reinforced with pullulan (PL) and acai berry extract (ABE) was developed by solvent casting method. The as-prepared films performed pH-sensitive properties, which can be potentially applied for fish freshness detection. Rheology , Fourier transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD), scanning electron microscopy (SEM) were used to characterize chemical structure and morphology of ABE-loaded KGM/PL (KP) films (KP-ABE). FT-IR spectrum indicated that hydrogen bond dominated the formation of KP-ABE films. Adding PL contributed to enhanced mechanical properties of KGM film with increased tensile strength (TS) from 21.25 to 50.27 MPa and elongation at break (EAB) from 10.64 to 19.19 %. Incorporating ABE upgraded flexibility, UV-shielding, thermostability , water barrier (decreased Water vapor permeability (WVP) from 2.07 to 1.67 g·mm/m 2 ·day kPa), antioxidant, and antibacterial ability of KP films, but weakened TS. In addition, KP-ABE films can reflect fish freshness in real time through color variability. Therefore, KP-ABE films exhibited potential applications in intelligent food packaging materials.

The development of an efficient pretreatment, prior to enzymatic hydrolysis, is a good strategy for the sustainable use of refractory fish byproducts. This study compared hydrothermal pretreatments at 159 °C for 2 min, followed by water extraction (steam explosion-assisted extraction, SE) and 121 °C for 70 min (hot-pressure extraction, HPE), for the recovery of proteins from fish backbones. The effect of enzymatic hydrolysis on the properties of the obtained fish bone protein (FBP) was also evaluated. The results demonstrated that FBP had high contents of protein (81.09–84.88 g/100 g) and hydroxyproline (70–82 residues/1000 residues). After hydrolysis with Flavourzyme, for 3 h, the FBP hydrolysates that were pretreated with SE (SFBP-H) exhibited a better degree of hydrolysis (DH) and nitrogen recovery (NR), and a higher level of umami taste free amino acids (151.50 mg/100 mL), compared with the HPE-treated samples. The obtained SFBP-H mainly distributed below 3000 Da and had strong scavenging effects on 1,1-diphenyl-2-picrylhydrazy (DPPH) (IC50= 4.24 mg/mL) and 2,2-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) (IC50= 1.93 mg/mL) radicals. Steam explosion-assisted extraction is a promising route for recovering proteins from native fish bone materials, and improving the flavor and antioxidant activity of the hydrolysates. Keywords:fish backbone;protein;steam explosion;hot-pressure extraction;enzymatic hydrolysis

Bone hydrolysates from bighead carp (Aristichthys nobilis) were prepared using Protamex and Alcalase with degrees of hydrolysis (DH) of 5%, 10% and 15%. The antioxidant activity of bone hydrolysates was evaluated in vitro and then the hydrolysates with better antioxidant activity were used to immerse bighead carp fillets through a vacuum impregnation process at concentrations of 1% and 2%. Among the six hydrolysates, fish bone hydrolyzed with Protamex at DH 10% exhibited the highest ability to scavenge 1, 1-diphenyl-2-picrylhydrazyl (DPPH) (88.79%), 2, 2′-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid (ABTS) (57.76%) and hydroxyl radicals (62.72%), as well as to chelate ferrous ions (91.46%). The hydrolysates effectively postponed freezing- and thawing-induced protein/lipid oxidation. Compared with the fillets without treatment, the impregnated fillets had higher sulfhydryl contents, greater Ca2+-ATPase activity, lower carbonyls and lower thiobarbituric acid-reactive substances (TBARS). Bone hydrolysates also have a positive effect on the texture and water-holding ability of freeze-thawed fish fillets. Fish bone hydrolysates of Protamex could serve as potential antioxidants to preserve fish fillets. Keywords:bone hydrolysates;antioxidant activity;fillet;vacuum impregnation;freeze–thaw cycles

The development of effective vaccines and delivery systems in aquaculture is a long-term challenge for controlling emerging and reemerging infections. Cost-efficient and advanced nanoparticle vaccines are of tremendous applicability in prevention of infectious diseases of fish. In this study, dihydrolipoamide dehydrogenase (DLDH) antigens of Vibrio alginolyticus were loaded into mesoporous silica nanoparticles (MSN) to compose the vaccine delivery system. Hydroxypropyl methylcellulose phthalate (HP55) was coated to provide protection of immunogen. The morphology, loading capacity, acid-base triggered release were characterized and the toxicity of nanoparticle vaccine was determined in vitro. Further, the vaccine immune effects were evaluated in large yellow croaker via oral administration. In vitro studies confirmed that the antigen could be stable in enzymes-rich artificial gastric fluid and released under artificial intestinal fluid environment. In vitro cytotoxicity assessment demonstrated the vaccines within 120 μg/ml have good biocompatibility for large yellow croaker kidney cells. Our data confirmed that the nanoparticle vaccine in vivo could elicit innate and adaptive immune response, and provide good protection against Vibrio alginolyticus challenge. The MSN delivery system prepared may be a potential candidate carrier for fish vaccine via oral administration feeding. Further, we provide theoretical basis for developing convenient, high-performance, and cost-efficient vaccine against infectious diseases in aquaculture.

In this study, a novel signal amplification strategy on photoelectrochemical (PEC) aptasensor was designed for high-sensitivity and -selectivity detection of 3,3′,4,4′-tetrachlorobiphenyl (PCB77) on the basis of Schottky junction and sensitization. First, the Schottky barrier not only provided an electron-transfer irreversible passage from CuO to Au Nanoparticles (NPs) but also generated excellent local surface plasmon resonance between CuO and Au NPs, thus improving the efficiency of charge separation and light absorption. Second, to further improve the response of the PEC aptasensor under the action of the sensitization, the complementary-DNA-functionalized CdS quantum dots were introduced onto the surface of CuO/Au NPs via hybridization of the target aptamer. The PEC aptasensor exhibited a low detection limit of 17.3 pg L −1 , and a wide linear response was shown at a range of 0.2 – 220 ng L −1 depending on the variation of photocurrent before and after incubation.

The appeal for environment friendly films with bioactive components to replace synthetic plastic packaging has been increasing in recent years. In this study, ginger essential oil (GEO) nanoemulsion at different levels (0.5%, 1.0%, 1.5%, 2.0%, and 3.0%) were incorporated into fish sarcoplasmic protein (FSP) and chitosan (CH) blend films to evaluate the physical, mechanical, antioxidant, and thermal properties. The water distribution state of films was also determined by NMR transverse relaxation (T2) measurements. The tensile strength (TS) of samples was significantly reduced with increasing GEO nanoemulsion contents ( p < 0.05), while the value of elongation at break (EAB) was the highest when the film with 1.0% GEO nanoemulsion. The addition of GEO nanoemulsion enhanced the water barrier properties and affected the water distributions of films, but had negative effects on thermal degradation of films. Scanning electric microscope (SEM) demonstrated the different microstructure at varied levels of GEO nanoemulsion and Fourier transform infrared (FTIR) spectroscopy gave the detailed information about the interactions at molecular level. These results revealed that FSP blended with CH was excellent film-forming material and mixing defined amount of GEO nanoemulsion into film-forming solution enhanced the physical and antioxidant properties of films.

Sialic acid (Sia) is widespread in vertebrates and humans as well as in some microbes and invertebrates. It is well known that O-acetylation is a common modification for Sia. Also, highly O-acetylated Sias have been reported in humans, bovines, and fish. Derivatization based on 1,2-diamino-4,5-methylenedioxybenzene (DMB) coupled with LC-fluorescence detection or LC-MS analysis has been the routine analytical strategy for O-acetylation profiling of Sias. Unfortunately, DMB offers poor chromatographic resolution for some O-acetylated species. Herein, we report another derivatization reagent, 4,5-dimethylbenzene-1,2-diamine (DMBA), for the LC-MS analysis of Sia isomers. The DMBA method was first evaluated using Sia reference standard, horse serum and carp blood. The results demonstrate that DMBA provides superior chromatographic separation efficiency and comparable MS/MS spectra to DMB. The proposed method was then applied to the analysis of relative abundance of each Sia species in fish tissues, including liver, intestine, muscle and brain. It was found that the relative amounts of Sia species were significantly different among the four tissues. Thus, DMBA derivatization-based LC-MS methods have great potential in investigating the O-acetylation level of Sias in various samples, such as human food and bio-pharmaceutical products.
