At present, glutathione (GSH) is a very important biomarker in vivo. However, the sensitive and selective assay for the detection of GSH in biological matrix still keeps challenging due to the high complexity of samples. Lanthanide-doped upconverting phosphors (UCPs) which exhibit unique near-infrared (NIR) excitation nature are able to overcome interferences from complex samples. In this study, a novel probe based on the linkage of UCPs and dopamine-quinone through hydrogen bonding and electrostatic interaction has been designed for rapid, economic, sensitive and selective detection of GSH in aqueous solution and human blood serum . Here, dopamine-quinone served as an efficient quencher for upconverting fluorescence, while GSH as a strong reducing agent chemically reduced the dopamine-quinone turning on the fluorescence. The fluorescence recovery was found to be proportional to GSH concentration within the range from 1 to 75 μM. The detection limit of this sensor was 0.29 μM, which was quite competitive for GSH detection. The simple, sensitive and selective fluorescence method took full advantages of UCPs properties of low interferences and broadened the application scope of UCPs in complex biological detection.

Targeted nanodrug delivery systems are highly anticipated for the treatment of malaria. It is known that Plasmodium can induce new permeability pathways (NPPs) on the membrane of infected red blood cells (iRBCs) for their nutrient uptake. The NPPs also enable the uptake of nanoparticles (NPs) smaller than 80 nm. Additionally, Plasmodium maintains a stable, slightly acidic, and reductive internal environment with higher glutathione (GSH) levels. Based on this knowledge, methyl artelinate (MA, a prodrug-like derivative of dihydroartemisinin) nanoparticles (MA-PCL-NPs) were developed using poly(ethylene glycol)-b-poly(ε-caprolactone) (mPEG-PCL) by a thin-film dispersion method and were further coated with polydopamine (PDA) to obtain MA-PCL@PDA-NPs with a particle size of ∼30 nm. The biomaterial PDA can be degraded in slightly acidic and reductive environments, thereby serving as triggers for drug release. MA could generate reactive oxygen species and decrease GSH levels, consequently causing parasite damage. The in vitro release experiment results indicated that the cumulative release percentage of MA from MA-PCL@PDA-NPs was considerably higher in phosphate buffer with 10 mM GSH at pH 5.5 (88.10%) than in phosphate buffer without GSH at pH 7.4 (16.98%). The green fluorescence within iRBCs of coumarin 6, the probe of NPs (C6-PCL@PDA-NPs), could be reduced significantly after adding the NPP inhibitor furosemide (p < 0.001), which demonstrated that MA-PCL@PDA-NPs could be ingested into iRBCs through NPPs. In vivo antimalarial pharmacodynamics in Plasmodium berghei K173-bearing mice showed that the inhibition ratio of MA-PCL@PDA-NPs (93.96%) was significantly higher than that of commercial artesunate injection (AS-Inj, 63.33%). The above results showed that the developed MA-PCL@PDA-NPs possessed pH-GSH dual-responsive drug release characteristics and targeting efficacy for iRBCs, leading to higher antimalarial efficacy against Plasmodium.

Cysteine (Cys), homocysteine (Hcy), and glutathione (GSH) are of great importance in many physiological and pathological processes. Due to their similar structure and reactivity of these three biothiols, it is a great challenge to simultaneously distinguish one from another. By virtue of two different fluorescence signals, nitrobenzofurazan ( NBD ) has been proved to be an effective sensing functional group as well as a green fluorophore precursor to construct fluorescent probes for simultaneous discrimination of Cys/Hcy and GSH. However, most of the reported NBD-based fluorescent probes suffered from a poor water-solubility, a slow response, double-wavelength excitation or two interfering fluorescence signals (narrow wavelength separation in λ max Em and/or big difference in Φ Fl ). In this study, we developed a fluorescent probe, MCQ-O-NBD , for simultaneous discrimination of Cys/Hcy and GSH using two matched fluorescent signals (well separated red and green fluorescence and comparable Φ Fl ), which could function in a pure aqueous media with a rapid response under a single-wavelength excitation. This probe exhibited high sensitivity and good selectivity as well as low detection limit, and had been successfully applied to visualize and differentiate Cys/Hcy and GSH in living cells.

Breast cancer, as the common malignant carcinoma in women, seriously reduces the life quality and increases mortality of female worldwide. Fortunately, insufficient chemotherapy with external photothermal therapy could remarkably improve the therapeutic efficiency of breast cancer. In this work, we proposed a polydopamine (PDA) and tetrasulfide bond co-doped hollowed mesoporous silica nanospheres with Dox loading and surface hyaluronic acid coating for drug leakage avoidance (PhMSON@Dox-HA). After intravenously injected into tumor bearing-mice, this therapeutic agent could effectively target to the tumor site owing to the HA modification. The biocompatible property and GSH-induced decomposition capability endows the precise release of drugs in the cytoplasm of tumor cells. Meanwhile, PDA facilitated the efficient photothermal therapy under 808 nm laser irradiation. Therefore, the synergistic effect of chemo-photothermal therapy possessed most effective tumor eradication ability. Our work exploits the design approach of biocompatible and degradable drug delivery nanocarriers and sheds bright light on the chemotherapy and photothermal therapy combination for the malignant tumor elimination.

Ferroptosis is an iron-dependent programmed cell death modality characterized by the massive accumulation of lipid peroxides (LPO). However, the efficiency of ferroptosis is commonly limited by insufficient supply of iron ions and endogenous hydrogen peroxide (H 2 O 2 ) in the tumor cells. To overcome these drawbacks, MIL-100 NPs were synthesized via a microwave-assisted method to encapsulate β-lapachone (Lap), followed with the hyaluronic acid (HA) surface modification (designated as LMH NPs). LMH NPs exhibited GSH-responsive degradation in the tumor cells to release Lap and Fe 3+ . The released of Lap could generate H 2 O 2 via catalysis by tumor-overexpressed quinone oxidoreductase 1 (NQO1), leading to increased intracellular H 2 O 2 levels. The abundant production of H 2 O 2 could supply substrates for Fe 2+ -mediated Fenton reactions to produce toxic hydroxyl radicals (·OH), thereby promoting LPO accumulation and enhancing ferroptosis. Meanwhile, Fe 3 ⁺ released from LMH NPs was reduced to Fe 2+ through GSH consumption, which could diminish glutathione peroxidase 4 (GPX4) activity. In vitro and in vivo experiments confirmed that LMH NPs possessed effective targeted uptake and exhibited optimal tumor inhibition without significant toxic side effects. Overall, the integrated strategy presents a novel ferroptosis-based clinical approach for enhanced breast cancer therapy. Scheme 1 Preparation process and antitumor mechanisms of LMH NPs. Covalent organic frameworks (COFs), as one of the most significant members of the porous organic frameworks, have been well used in the photocatalysis owing to their outspread π-conjugated framework, high crystallinity and regular pore structure. Herein, after reducing the labile imine-linked COF-300 to the more stable amine-linked COF-300-AR, we for the first time demonstrated that COF-300-AR was the light-responsive oxidase mimic. COF-300-AR exhibited excellent oxidase-mimicking activity under purple light stimulation (λ = 400 nm), which can catalyze the oxidation of classical substrates such as 3,3′,5,5′-tetramethylbenzydine (TMB) and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) by the formation of •OH and O2•– free radicals in the presence of dissolved oxygen. The COF-300-AR oxidase mimic has outstanding advantages of easy light control, high stability, good reusability, and highly catalytic oxidation capacity and has been applied to detect glutathione (GSH) levels in HL60 cells with good selectivity and high sensitivity. This study will broaden the sensing applications of COFs and offer a promising build block for the construction of artificial enzymes.

In this work, a unique ratiometric and NIR fluorescent probe , Hecy-NBD, was designed and synthesized to detect cysteine/homocysteine/glutathione in aqueous solution. Hemicyanine skeleton was chosen as the major fluorophore and nitro benzoxadiazole (NBD) chromophore was selected as the recognition unit to construct a ratiometric fluorescent probe. The fluorescence of probe Hecy-NBD was very weak due to the introduction of quenching group NBD . Upon the addition of 100 μM GSH to 10 μM Hecy-NBD, the fluorescence intensity at 702 nm was very strong with the releasing of compound Hecy-OH. While upon the addition of 100 μM Cys or 100 μM Hcy, probe exhibited distinct fluorescence enhancement at 540 nm and 702 nm, which resulted from the formation of compound NBD-Cys and NBD-Hcy through the 5/6-membered cyclic transition and the fluorescence releasing of compound Hecy-OH. The proposed mechanism based on S N Ar reaction and molecular rearrangement, which were confirmed by HRMS spectra. The detection limit of these three biothiols was also calculated (GSH, 50 nM; Cys, 81.0 nM; Hcy, 130 nM). Moreover, probe Hecy-NBD could function well at wide pH conditions from 4 to 10. Besides that, probe Hecy-NBD with low cell toxicity had been successfully applied to dual-channel fluorescence imaging in living cells. Considering the good characteristics, Hecy-NBD had great potential to detect biothiols-associated diseases in the future.

Comprehensive Summary Due to the immunosuppressive tumor microenvironment (TME), T cells are usually inactivated and tend to differentiate into regulatory T cells (Tregs) in the tumor tissues, which seriously hinders the anti-tumor efficiency of immunotherapy. In this study, an immune-stimulating polypeptide hydrogel conjugated with an indoleamine-2,3-dioxygenase (IDO) inhibitor, 1-methyl- D -tryptophan (D1MT), through a glutathione (GSH)-responsive spacer was developed as an immunotherapy platform capable of regulating the TME. A combined immunotherapy system was further constructed through encapsulating doxorubicin (Dox) and immune checkpoint blocking antibody targeting programmed cell death protein 1 (aPD-1) in the hydrogel. Dox released from the hydrogel could cause the immunogenic cell death (ICD) of tumor cells and induce the maturation of antigen presenting cells (APCs). After intratumoral injection, the multiple agent-loaded hydrogel elicited effective anti-tumor immunity in mice bearing B16F10 melanoma. Moreover, compared with the control hydrogel without D1MT, the immune-stimulating hydrogel showed better efficiency in improving the immunosuppressive TME with increased number of activated T cells and reduced number of Tregs. Therefore, the immune-stimulating hydrogel has great potential as a stimuli-responsive platform for regulating suppressive TME and enhanced anti-tumor chemo-immunotherapy.

Herein, we engineered a novel pH-responsive ratiometric assay by virtue of the composite system of Ce 3+ -enhanced glutathione-encapsulated copper nanoclusters (GSH-CuNCs-Ce 3+ ) and N-doped carbon quantum dots (N-CQDs). The GSH-CuNCs-Ce 3+ and N-CQDs were fabricated using one-pot chemical reduction and hydrothermal approaches, respectively, and their morphologies as well as physical and chemical properties were testified in detail by a series of characterization techniques. At 350 nm excitation wavelength, the GSH-CuNCs-Ce 3+ /N-CQDs system (GCC-NC) featured the 440/650 nm dual-emitting property. The ratios of fluorescence intensities (F 650 /F 440 ) demonstrated a strong pH-dependence from 3.0 to 5.0 with a coefficient of determination of 0.9982. Consequently, the GCC-NC system was feasible for constructing a pH-responsive dual-emitting fluoroprobe. Based on the catalytic effect of acetylcholinesterase on acetylcholine to yield acetic acid and further trigger varying solution pH, the as-constructed GCC-NC fluoroprobe was satisfactorily applied for detecting malathion . Under optimized conditions, the newly developed GCC-NC fluoroprobe supplied a wider linear range (0.25–200 μM), lower detection limit (0.075 μM), satisfactory recoveries (96.3–106.2%) and higher precision for malathion in several kinds of fruit juice samples. The fortified experiments by malathion’s structural analogues and metal ions provided compelling evidence that this fluoroprobe had strong anti-interference capacity and high specificity for malathion. These findings render us to believe that the as-constructed pH-responsive dual-emitting fluoroprobe holds great promise in trace malathion assay in food matrices.

ABSTRACT A nano-drug delivery system that is responsive to glutathione (GSH) and combines chemotherapy (CT) and photodynamic therapy (PDT) was designed precisely for treating melanoma. The carboxymethyl chitosan (CMC), a water-soluble and biocompatible nanocarrier, was used to load the chemotherapeutic small-molecule drug camptothecin (CPT) and the photosensitizer Rhodamine-B (RhB). The development of dual drug combo (CCR NPs) nanoparticles with a mean particle diameter of 170 ± 2.2 nm in aqueous solution has been achieved using ultrasonic self-assembly. Nanoparticle disintegration and camptothecin (CPT) release occur as CPT, sensitive to glutathione (GSH), breaks in cancer cells with excellent GSH levels. The RhB encapsulated in the CCR NPs was released quickly to attain successive CPT release. The CPT dramatically lowers tumor cell GSH levels, inhibiting the antioxidant effect of GSH on photosensitizers' ROS production capabilities and improving PDT synergistically. Nanoparticle diameter, zeta potential, and outstanding stability were all demonstrated by the CCR NPs. In vitro toxicity study on A375 human melanoma cells showed that CPT and RhB were highly selective (SI=4.25) and had a synergic effect (CI=0.24). Biochemical staining methods confirmed the morphological changes and nuclear damages. Apoptosis was confirmed by FITC-Annuxin V and PI staining by flow cytometer. Investigating drug delivery systems integrating RhB and CPT represents a valuable novel strategy for treating melanoma cancer.

Reactive oxygen species (ROS) are widely considered to the effective therapeutics for fighting bacterial infections especially those associated with biofilm. However, biofilm microenvironments including hypoxia, limited H 2 O 2 , and high glutathione (GSH) level seriously limit the therapeutic efficacy of ROS-based strategies. Herein, we have developed an acidic biofilm microenvironment-responsive antibacterial nanoplatform consisting of copper-dopped bovine serum albumin (CBSA) loaded with copper peroxide (CuO 2 ) synthesized in situ and indocyanine green (ICG). The three-in-one nanotherapeutics (CuO 2 /ICG@CBSA) are capable of releasing Cu 2+ and H 2 O 2 in a slightly acidic environment, where Cu 2+ catalyzes the conversion of H 2 O 2 into hydroxyl radical (•OH) and consumes the highly expressed GSH to disrupt the redox homeostasis. With the assistance of an 808 nm laser, the loaded ICG not only triggers the production of singlet oxygen ( 1 O 2 ) by a photodynamic process, but also provides photonic hyperpyrexia that further promotes the Fenton-like reaction for enhancing •OH production and induces thermal decomposition of CuO 2 for the O 2 -self-supplying 1 O 2 generation. The CuO 2 /ICG@CBSA with laser irradiation demonstrates photothermal-augmented multi-mode synergistic bactericidal effect and is capable of inhibiting biofilm formation and eradicating the biofilm bacteria. Further in vivo experiments suggest that the CuO 2 /ICG@CBSA can effectively eliminate wound infections and accelerate wound healing. The proposed three-in-one nanotherapeutics with O 2 /H 2 O 2 -self-supplied ROS generating capability show great potential in treating biofilm-associated bacterial infections. Ligand-receptor recognition-based nanodrug delivery systems incorporate stimulus-responsive functionality to proactively transport chemotherapeutics to lesional tissues and intelligently release drugs in response to stimulation, thereby augmenting drug accumulation at target sites, boosting drug bioavailability, and avoiding systemic toxicity, which reveals a promising potential in malignant tumor therapy. Herein, we designed and prepared a dual-targeted nanomedicine, FA-HA-SS-PPT, utilizing microwave-assisted quantitative conjugating of podophyllotoxin (PPT) to folic acid (FA) and hyaluronic acid (HA) carriers via short-chain PEG bearing the tumor microenvironment-sensitive disulfide bond. The FA-HA-SS-PPT nanoparticles developed by self-assembly exploited the peculiarity of specifically binding to the folate receptor (FR) and CD44 receptor, initiated targeting to translocate the drug into the tumor cells and released the PPT by self-cyclization in high intracellular GSH concentration. Furthermore, this nanomedicine demonstrated superior tumor cytotoxicity and proliferation inhibition in vitro by triggering the mitochondrial apoptotic pathway and suppressing the microtubule protein formation. Notably, quantitative disulfide bond conjugation and nanomedicine formation endowed FA-HA-SS-PPT nanoparticles with astonishing in vivo circulatory stability and tumor microenvironmental responsiveness, further reinforcing drug security and anti-tumor capability. Therefore, quantitatively conjugating drugs provide a promising safe and efficient means to treat multi-receptor overexpressing tumors with multi-targeted nanomedicines.
