Lactic acid bacteria (LAB) have already been used as fermentation strains to enhance the antioxidant capacity of polyphenols. Antioxidant capacity is one of the most important factors to inhibit advanced glycation end product (AGE) formation and could LAB increase the inhibitory capacity of procyanidins on AGEs formation? It was surprising that opposite results were obtained both in simulated food processing and gastrointestinal digestion systems. After incubation with Lactobacillus plantarum (L. plantarum), litchi pericarp oligomeric procyanidins (LPOPCs) were bioconverted to several phenolic acids, which increased the antioxidant activity as expected. However, antiglycation ability and trapping carbonyl compounds capacity both weakened and it might be the primary reason for decreasing the inhibitory effect on AGE formation. Furthermore, it was found that LPOPCs incubated with L. plantarum inhibited the activity of digestive enzymes and thus decreased the digestibility of glycated protein. Our study systematically proposed for the first time that procyanidins bioconversion is an effective means to improve the antioxidant activity but has no remarkable promoting effect on AGEs inhibition.

SalmonellaTyphimurium (S.Typhimurium) is an aggressive zoonotic pathogen that causes enteritis and diarrhea. Antibiotic therapy is still the primary method at present. However, the increasing emergence of multi-drug resistant bacteria weakens the therapeutic efficacy of antibiotics. Probiotics have been widely studied as an alternative antibiotic therapy. In this study, we established an IPEC-J2 cell model ofS.Typhimurium infection, aiming to determine the protective effect ofLactobacillus johnsoniiL531 (L. johnsoniiL531) onS.Typhimurium infection. As our data showed,S.Typhimurium infection resulted in a robust inflammatory response demonstrated by promoted protein levels of the inflammatory-related pathway (TLR4, MyD88, p-IκBα, and p-p65), increased cytokine levels of IL-6, IL-1β, IL-18, and TNF-α, and activated the NLRP3 inflammasome via promoting its assembly. However,L. johnsoniiL531 pre-incubation inhibited the activation of the above inflammatory signaling pathways and reduced the expression levels of pro-inflammatory cytokines. In addition,L. johnsoniiL531 alleviated the damage ofS.Typhimurium to tight junctions ZO-1, Occludin, and Claudin-1. In summary, our findings suggested thatL. johnsoniiL531 alleviatedS.Typhimurium-induced tight junction injury by inhibiting the TLR4/NF-κB/NLRP3 inflammasome signaling pathway. Keywords:SalmonellaTyphimurium;Lactobacillus johnsonii;TLR4;NF-κB;NLRP3 inflammasome;tight junctions

Lactic acid bacteria immobilization methods have been widely used for lactic acid production. Until now, the most common immobilization matrix used is calcium alginate . However, Ca-alginate gel disintegrated during lactic acid fermentation . To overcome this deficiency, we developed an immobilization method in which Lactobacillus rhamnosus cells were successfully encapsulated into an ordered mesoporous silica-based material under mild conditions with a high immobilization efficiency of 78.77% by using elemental analysis. We also optimized the cultivation conditions of the immobilized L. rhamnosus and obtained a high glucose conversion yield of 92.4%. Furthermore, L. rhamnosus encapsulated in mesoporous silica-based material exhibited operational stability during repeated fermentation processes and no decrease in lactic acid production up to 8 repeated batches.

A functional Chinese fermented glutinous rice has been developed with the supplementation of Fu brick tea (CRW-FBT). In this study, we aimed to evaluate its effect on the growth of potential probiotic strains in the Lactobacillus, Streptococcus, and Weissella genus, compared with traditional Chinese fermented glutinous rice (CRW). The growth profiles of lactic acid bacteria were analyzed based on fermentations in vitro, and the optical densities were recorded at 600 nm during the whole fermentation. Growth curve, maximum OD600 nm, and growth rate were measured and compared among samples with different ratios of CRW-FBT and CRW addition. Through the multiple analysis of growth parameters, we found that all the tested strains obtained better growth results when CRW-FBT was supplemented to the media, compared with the CRW and basic media. The bacterial growth was promoted by exhibiting the shortened lag time, prolonged logarithmic phase and stationary phase, and increased growth rate and cell density, as well as the better performance after 24 h and 48 h fermentation. Besides, short-chain fatty acids and organic acids in CRW-FBT were founded. Our work demonstrated the positive effect of Fu brick tea supplemented in the CRW and illustrated its beneficial role in the food fermentation industry for the purpose of microorganism enrichment and the improvement of microbial metabolism.

Bacterial diarrhea involves colonic water absorption dysfunction and gut microbiota disruption. Gegen Qinlian decoction (GQD) is clinically used to treat infectious diarrhea, yet its mechanisms remain incompletely understood. This study aimed to investigate how GQD modulates gut microbiota and enhances colonic water absorption in E. coli -induced diarrhea. Using an E. coli -induced piglet diarrhea model together with a rat colonic perfusion model, results show that GQD alleviates diarrhea-associated intestinal injury and inflammation and improves colonic water absorption. Mechanistically, GQD was associated with increased colonic aquaporin-3 (AQP3) expression and attenuation of the cAMP-protein kinase A-cystic fibrosis transmembrane conductance regulator (cAMP-PKA-CFTR) secretory signaling axis. 16S rRNA sequencing indicated enrichment of Lactobacillus amylovorus after GQD treatment, and administration of L. amylovorus partially reproduced the anti-diarrheal phenotype and transport-related signaling changes. In vitro assays further suggested that GQD suppresses E. coli growth while promoting L. amylovorus metabolic activity, and molecular docking provided hypothesis-generating predictions of candidate phytochemicals and bacterial targets. Overall, GQD may improve bacterial dysbiosis by enriching L. amylovorus and inhibiting E. coli , thereby restoring colonic water transport via the cAMP/CFTR/AQP3 pathway and ultimately improving bacterial diarrhea.

This study focuses on developing a water-in-oil-in-water (W 1 /O/W 2 ) double emulsion system using high-intensity ultrasound (HIU)-treated pea protein isolate (HIU-PPI) and pectin to encapsulate Lactobacillus plantarum ( L. plantarum ). The effects of ultrasound treatment on pea protein isolate (PPI) characteristics such as solubility, particle size, emulsification, surface hydrophobicity, and surface free sulfhydryl group were examined, determining optimal HIU processing conditions was 400 W for 10 min. The developed W 1 /O/W 2 double emulsion system based on HIU-PPI demonstrated effective encapsulation and protection of L. plantarum , especially at the HIU-PPI concentration of 4 %, achieving an encapsulation efficiency of 52.65 %. Incorporating both HIU-PPI and pectin as emulsifiers increased the particle size and significantly enhanced the emulsion’s viscosity. The highest bacterial encapsulation efficiency of the emulsion, 59.94 %, was attained at a HIU to pectin concentration ratio of 3:1. These emulsions effectively encapsulate and protect L. plantarum , with the concentration of HIU-PPI being a critical factor in enhancing probiotic survival under simulated gastrointestinal digestion. However, the concurrent utilization of pectin and HIU-PPI as emulsifiers did not provide a notable advantage compared to the exclusive use of HIU-PPI in enhancing probiotic viability during in vitro simulated digestion. This research offers valuable perspectives for the food industry on harnessing environmentally friendly, plant-based proteins as emulsifiers in probiotic delivery systems. It underscores the potential of HIU-modified pea protein and pectin in developing functional food products that promote the health benefits of probiotics.

Background Spinal cord injury (SCI) induces gut microbiota dysbiosis, which significantly affects recovery. Buyang Huanwu Decoction (BHD), a traditional Chinese medicine formula, has shown therapeutic effects on SCI. Although BHD is known to modulate gut microbiota, whether its benefits are mediated through the gut–spinal cord axis remains unclear. Methods A rat SCI model was established. BHD was administered orally, and fecal microbiota transplantation (FMT) from BHD-treated rats (BHD-FMT) was performed to assess neuroprotective and gut-protective effects. Behavioral testing, histology, and immunofluorescence evaluated motor recovery, inflammation, and neuroregeneration. Gut microbiota profiling was performed using 16S rDNA sequencing and metagenomics, while targeted metabolomics quantified tryptophan metabolites. Transcriptomics validated key pathways, and a microbiota–metabolite–signaling network was constructed. Results BHD significantly improved motor function, reduced spinal inflammation, and promoted neuronal survival and axonal regeneration. It restored gut function, reduced colonic inflammation, and enhanced ZO-1 and Occludin expression, which were further confirmed by FMT. BHD-FMT reshaped the gut microbiota and enriched Lactobacillus johnsonii , which correlated positively with recovery. Metabolomics showed increased tryptophan metabolites, including indole-3-lactic acid (ILA) and indole-3-propionic acid (IPA), with ILA strongly associated with functional improvement. Transcriptomic analysis and Western blot validation demonstrated that BHD-FMT activated the AhR–PI3K/Akt pathway, which was suppressed by an AhR antagonist. Conclusion BHD promotes neuroregeneration after SCI by reshaping gut microbiota and enhancing tryptophan metabolism, potentially exerting its effects through the L. johnsonii –ILA–AhR–PI3K/Akt network. These findings reveal a gut–spinal cord axis–mediated mechanism of BHD and highlight microecological targets for SCI therapy. Graphical

The protective effects ofLactobacillus murinuson chemotherapy-related intestinal injury and immune imbalances were explored by establishing a cyclophosphamide (CTX)-induced mouse model of immunosuppression. CTX treatment led to intestinal barrier destruction, exacerbated local inflammation, and significantly reduced short-chain fatty acid levels (especially butyrate), accompanied by systemic immune suppression.Lactobacillus murinusintervention, especially at medium and high doses, dose-dependently repaired the intestinal barrier, inhibited inflammatory responses, restored levels of metabolites such as butyrate, and systematically regulated splenic immune cell proportions, restoring the CD4+/CD8+balance. Metabolomic analysis further revealed that, at different doses, this regulation affected distinct metabolic pathways: low doses enhanced glutathione and purine metabolism, medium doses restored folate and steroid hormone metabolism, and high doses promoted fatty acid β-oxidation and galactose metabolism, forming a multi-level metabolic protective network. This suggests thatL. murinuscan alleviate chemotherapy-induced intestinal mucositis and mitigate systemic immune suppression through a dual local anti-inflammatory and systemic immune-regulatory effect, with potential mechanisms related to butyrate-mediated regulation of the “metabolism–immune axis,” providing evidence for probiotic-assisted chemotherapy.

Fatty liver hemorrhagic syndrome (FLHS) has become a common and serious disease in intensive laying hen farming, seriously endangering the productivity and health of laying hens. Although probiotics have been recognized as an emerging candidate therapeutic strategy for liver disease, their effective substances and mechanisms for alleviating FLHS remain to be elucidated. In this study, corticosterone (Cort) was applied to induce FLHS in laying hens, and Lactobacillus plantarum ( L. plantarum 1–2-3) was used for the intervention. The results showed that L. plantarum 1–2-3 treatment significantly ameliorated Cort-induced hepatic steatosis and liver damage in laying hens, as evidenced by decreased levels of triglyceride (TG), total cholesterol (TC), aspartate aminotransferase, and alanine aminotransferase. Importantly, we found that L. plantarum 1–2-3 could biosynthesize gamma-aminobutyric acid (GABA) by expressing glutamic acid decarboxylase. Surprisingly, feeding GABA to laying hens replicated the effect of L. plantarum 1–2-3 in alleviating Cort-induced FLHS. GABA significantly down-regulated the mRNA expression of Acc , Fasn , Cd36 , and Gpam while upregulating the levels of Ppara and Cpt1a . An in vitro verification study showed that GABA decreased TG and TC content in Leghorn male hepatoma cells. Moreover, GABA significantly enhanced antioxidant enzyme activity, inhibited oxidative stress in hepatocytes, regulated the AMPK/NLRP3 signaling pathway, and reduced the protein expression of NF-κB, TNFα, and IL-1β. In conclusion, our study demonstrated that L. plantarum 1–2-3 could effectively alleviate FLHS in laying hens, while GABA was an important postbiotic for L. plantarum 1–2-3 to exert lipid-lowering and anti-inflammatory effects.

Celery ( Apium graveolens L . ) is rich in phenolic compounds, yet their transformation mechanisms during fermentation remain unclear. This study investigated the metabolic remodeling of phenolics in celery pulp fermented by Lactiplantibacillus plantarum NCU116 ( L. plantarum NCU116) using an integrated omics approach. Untargeted metabolomics, transcriptomics, targeted quantification, enzymatic assays, and structural imaging were employed. A total of 63 free and 29 bound phenolic compounds were identified. Notably, the levels of representative flavonoid glycosides, including apiin and graveobioside A, significantly decreased to 0.76 and 0.73 times their original levels, respectively, while 3′-methoxy apiin showed a slight decrease to 0.90 times. Transcriptomic analysis revealed upregulation of β-glucosidase genes ( bglB , bglH ), consistent with elevated enzyme activity. Confocal laser scanning microscope (CLSM) confirmed progressive cell wall depolymerization and reduced lignin autofluorescence, coinciding with the release of bound phenolics. These findings demonstrate that phenolic conversion in fermented celery is co-regulated by microbial enzymatic hydrolysis and structural remodeling of the plant matrix.

Nano-sized magnetic photocatalysts (nano TiO2-SiO2/Fe3O4) with a particle size of approximately 20 nm were developed for efficient water treatment. Their photocatalytic performance was comparable to the nano particle photocatalysts, P25. Furthermore, the photocatalysts could be conveniently separated from treated water using a magnet. We then applied the photocatalysts to photokilling of Lactobacillus casei and degradation of persistent herbicides (simetryn, prometryn, metolachlor, and sulfometuron methyl). Lactobacillus casei, a Gram-positive bacterium lacking an outer membrane, was introduced and validated as a reliable model bacterium for photocatalytic antibacterial assessments. Complete photokilling of Lactobacillus casei occurred within 50 minutes of UV irradiation, demonstrating simpler kinetic behavior compared to traditionally used Gram-negative model organisms. The photocatalysts also exhibited effective degradation toward four persistent herbicides. Their concentrations decreased from 10 ppm to below 1 ppm within 120 minutes.

The study investigates the impacts of indigenous bacterial strains inoculation specifically L. pentosus , on the flavor characteristics, microbial composition, and metabolite profiles of fermented oyster hydrolysates. This research aimed to elucidate potential mechanisms underlying the reduction of off-flavors in fermented hydrolysates. A total of 46 and 57 volatile compounds were detected by GC–MS and GC-IMS in hydrolysates inoculated with different core microbes, respectively. The 9 key volatile compounds detected by GC–MS analysis. ( E , E )-2,4-heptadienal, heptanal, octanal, pentanal, and ( E )-2-octenal reduced the off-flavor of the fermented oyster hydrolysate. Meanwhile 1-octen-3-ol, 3-octanone, 4-octanone, and ( E , Z )-2,6-nonadienal enhanced the direct contribution of desirable flavors. Variation in 16 amino acids, 10 organic acids and 3 nucleotides were monitored to further understand the metabolic changes affecting flavor quality. Moreover, pyruvate decarboxylase [EC 4.1.1.1], phosphomannanase [EC 3.2.1.109], lipoyl-CoA synthetase [EC 6.3.2.4], and arginine kinase [EC 2.7.3.3] were the main microbiologically active enzymes. An increase in the content of aromatic compounds and a decrease in the content of C6-C9 unsaturated aldehydes through Lys, Phe, Asp, Glu, phosphoenolpyruvate, oleic acid, and linoleic acid metabolism pathways improved the flavor of oyster hydrolysates fermented by L. pentosus . This research provides a theoretical basis for leveraging autochthonous microbial fermentation to systematically improve flavor characteristics in fermented products.
