Min Zhang,Baolong Li,Yu Du,Gaojian Zhou,Yanyan Tang,Yanfen Shi,BiYu Zhang,Zhou Xu,Qingli Huang

The preparation and construction of novel nanostructures as targeting theranostic is of great significance for diagnosis and treatment of tumor. Herein, a novel PANI/ [email protected] @MnO 2 (PPAuMns) yolk-shell enzymatic theranostic agent was fabricated firstly via in situ deposition of Au nanoparticles (Au NPs) and reduction of KMnO 4 on the surface of PANI/PPy nanocomposites. In addition to high biocompatible and biodegraded performance, PPAuMns possessed amazing tumor microenvironment (TME)-responsive O 2 generation capacity, magnetic resonance imaging (MRI) property, enzymatic activity and excellent photothermal conversion effect. Chemotherapeutics doxorubicin and folic acid (FA) were further loaded and decorated on it for targeted tumor therapy. Experiments in vitro and in vivo indicated that DOX-loaded [email protected] demonstrated excellent synergistic anti-tumor effect combining chemotherapy (CT), chemo-dynamic therapy (CDT) and photothermal therapy (PTT). This work offers a paradigm to design and construct tumor diagnosis and therapy platforms employing TME-responsive synergistic strategy of photothermal ablation, chemo-dynamic and chemotherapy.

A multifunctional targeted nanoprobe with high NIR-II PAI/MRI performance for precise theranostics of orthotopic early-stage hepatocellular carcinoma

Linyun He,Yachao Zhang,Jiangbo Chen,Gongyuan Liu,Jingyi Zhu,Xiaozhen Li,Dengfeng Li,Yuqi Yang,Chun-Sing Lee,Jiahai Shi,Chao Yin,Puxiang Lai,Lidai Wang,Chihua Fang

Early diagnosis and effective treatment of hepatocellular carcinoma (HCC) is quite critical for improving patients’ prognosis. The combination of second near-infrared window photoacoustic imaging (NIR-II PAI) and T2-magnetic resonance imaging (T2-MRI) is promising for achieving omnibearing information on HCC diagnosis due to the complementary advantages of outstanding optical contrast, high temporospatial resolution and soft-tissue resolution. Thus, the rational design of a multifunctional targeted nanoplatform with outstanding performance in dual-modal NIR-II PAI/T2-MRI is particularly valuable for precise diagnosis and imaging-guided non-invasive photothermal therapy (PTT) of early-stage HCC. Herein, a versatile targeted organic–inorganic hybrid nanoprobe was synthesized as a HCC-specific contrast agent for sensitive and efficient theranostics. The developed multifunctional targeted nanoprobe yielded superior HCC specificity, reliable stability and biocompatibility, high imaging contrast in both NIR-II PAI and T2-MRI, and an excellent photothermal conversion efficiency (74.6%). Furthermore, the theranostic efficiency of the targeted nanoprobe was systematically investigated using the orthotopic early HCC-bearing mice model. The NIR-II PAI exhibited sensitive detection of ultra-small HCCs (diameter less than 1.8 mm) and long-term real-time monitoring of the tumor and nanoprobe targeting process in deep tissues. The T2-MRI demonstrated clear imaging contrast and a spatial relationship between micro-HCC and adjacent structures for a comprehensive description of the tumor. Moreover, when using the targeted nanoprobe, the non-invasively targeted PTT of orthotopic early HCC was carried out under reliable dual-modal imaging guidance with remarkable anti-tumor efficiency and biosafety. This study provides an insight for constructing a multifunctional targeted nanoplatform for precise and comprehensive theranostics of early-stage HCC, which would greatly benefit the patients in the era of precision medicine.

Controlled synthesis of up-conversion luminescent Gd/Tm-MOFs for pH-responsive drug delivery and UCL/MRI dual-modal imaging

Yana Liu,Cheng Zhang,Chen Xu,Caixue Lin,Keke Sun,Jie Wang,Xueli Chen,Ling Li,Andrew K. Whittaker,Hai-Bing Xu

Using a facile one-step hydrothermal method, a series of metal–organic frameworks containing Gd/Tm (Gd/Tm-MOFs) were prepared successfully. Through the mutual activation of Gd3+ and Tm3+, Gd/Tm-MOFs showed unexpected excellent up-conversion luminescence (UCL) and a superior positive magnetic resonance image (MRI). Using doxorubicin hydrochloride (DOX) as a drug model, the feasibility of Gd/Tm-MOFs as a multifunctional drug carrier was demonstrated. Through modifying Gd/Tm-MOFs with uniform mesoporous silica (mSiO2) shells and folic acid (FA), the drug loading was improved up to 41.5 mg g−1, and pH responsive drug release increased to 64% from 12% by regulating the pH from 5.8 to 7.4. Also, the particles were tracked by MRI, and the second highest longitudinal relaxivity (r1) among the reported gadolinium complexes of 225.86 mM−1 s−1 was achieved. The cell imaging shows obvious blue and red luminescence under 980 nm laser excitation, and the up-conversion luminescence is unique because there is no autofluorescence from cells under 980 nm excitation. This indicates their promising application in the biological field. All the experiments indicate the promising application of Gd/Tm-MOFs in diagnosis and treatment.

AS1411 aptamer-modified theranostic liposomes co-encapsulating manganese oxide nano-contrast agent and paclitaxel for MRI and therapy of cancer

Jingjing Li,Qing Li,Meijuan He,Fan Ding,Lulu Cai,Mingming Zhao,Lina Dong,Qi Wang,Kai Xu

With the advantages and development of MRI nano-contrast agents (CAs), increasing number of MRI-based theranostic nanoparticles have emerged. Liposome, as a biosafe nanocarrier has been used phase III trial for cancer treatment. In this study, liposome was employed as a nanocarrier to co-encapsulate MRI nano-contrast agent poly(ethylene glycol)-grafted manganese oxide (PEG-MnO) and anticancer drug paclitaxel (PTX) for the fabrication of a novel theranostic nanocomplex. After being further modified with AS1411 aptamer, the obtained nanoprobe AS1411-liposome-PEG-MnO-PTX displayed the potential of simultaneous MRI diagnosis and therapy of renal carcinoma in vitro and in vivo. It was found that compared with PEG-MnO nano-CA, liposome-PEG-MnO and AS1411-liposome-PEG-MnO presented a stronger MR contrast enhancement effect in the tumor and longer retention time in the tumor region. More importantly, the introduction of AS1411 aptamer further enhanced the MRI effect and the tumor growth inhibition effect, showing its potential use as a theranostic nanoprobe for renal carcinoma.

Poly(acrylic acid) modified lanthanide-doped GdVO4 hollow spheres for up-conversion cell imaging, MRI and pH-dependent drug release

Xiaojiao Kang,Dongmei Yang,Yunlu Dai,Mengmeng Shang,Ziyong Cheng,Xiao Zhang,Hongzhou Lian,Ping'an Ma,Jun Lin

In this study, multifunctional poly(acrylic acid) modified lanthanide-doped GdVO4 nanocomposites [PAA@GdVO4: Ln3+ (Ln = Yb/Er, Yb/Ho, Yb/Tm)] were constructed by filling PAA hydrogel into GdVO4 hollow spheres via photoinduced polymerization. The up-conversion (UC) emission colors (green, red and blue) can be tuned by changing the codopant compositions in the matrices. The composites have potential applications as bio-probes for cell imaging. Meanwhile, the hybrid spheres can act as T1 contrast agents for magnetic resonance imaging (MRI) owing to the existence of Gd3+ ions on the surface of composites. Due to the nature of PAA, DOX-loaded PAA@GdVO4:Yb3+/Er3+ system exhibits pH-dependent drug releasing kinetics. A lower pH offers a faster drug release rate. Such character makes the loaded DOX easily released at cancer cells. The cell uptake process of drug-loaded composites was observed by using confocal laser scanning microscopy (CLSM). The results indicate the potential application of the multifunctional composites as theragnostics (effective bimodal imaging probes and pH-responsive drug carriers).

Poly (hydroxyethyl methacrylate - acrylic acid) microspheres loaded with magnetically responsive Fe3O4 nanoparticles for arterial embolization, drug loading and MRI detection

Xin Li,Wenxia Lv,Weiqi Yang,Yinfeng Guo,Jianwen Huang,Wenyi Liang,Yugang Huang,Aiping Qin,Xingmei Deng,Xufeng Li,Mianrong Chen,Hainan Yang,Lu Liang,Lingran Du

In recent decades, transcatheter arterial embolization (TAE) has been attached great importance in the noninvasive vascular interventional therapy for treating various diseases. In this study, a novel magnetic embolic agent of Fe 3 O 4 -loaded hydrophilic poly (hydroxyethyl methacrylate - acrylic acid) microspheres (FPHA-Ms) was constructed using inverse suspension polymerization and sequential in situ co-precipitation method. The loading of Fe 3 O 4 nanoparticles in FPHA-Ms was verified by infrared spectrum, vibrating sample magnetometer, X-ray diffraction and X-ray photoelectron spectroscopy, respectively. The weight percentage of iron content in lyophilized FPHA-Ms was measured by inductive coupled plasma emission spectrometer. The morphology, particle size distribution , equilibrium water content, elasticity, drug loading & eluting capacity, biosafety, magnetic resonance imaging (MRI) detectability and embolic efficacy of FPHA-Ms were systematically evaluated, respectively. FPHA-Ms possessed many merits including spherical shape, uniform and wide size range, considerable water content, high drug loading, sustained drug release and good elasticity and biocompatibility . The in vivo experiments showed FPHA-Ms could effectively block the blood supply and could be detected by MRI device. All the results proved that FPHA-Ms could serve as a novel MRI-visible multi-functional embolic agent for clinical vessel embolization, drug loading and MRI tracing in the near future.

Highly sensitive T 1–T 2 dual-mode MRI probe based on ultra-small gadolinium oxide-decorated iron oxide nanocrystals

Yashuang Miao,Peirong Chen,Miao Yan,Jianmin Xiao,Biao Hong,Ke Zhou,Guilong Zhang,Junchao Qian,Zhengyan Wu

Single-mode magnetic resonance imaging (MRI) contrast agents (CAs) in clinical settings are easily disturbed by calcification, bleeding, and adipose signals, which result in inaccurate diagnoses. In this study, we developed a highly efficient T1–T2 dual-mode MRI CA using an ultra-small gadolinium oxide-decorated magnetic iron oxide nanocrystal (GMIO). The gadolinium element could effectively alter the magnetic properties of the GMIO from soft-ferromagnetism to superparamagnetism. In addition, when the Gd/Fe ratio was 15% (designated as GMIO-2), the GMIO-2 possessed the best superparamagnetism and highest magnetism. Subsequently, T1 and T2 values of GMIO-2 were measured through a series of turbo spin-echo images and then multi-spin echo sequence, respectively. Based on this, T1 and T2 relaxivities of GMIO-2 were calculated and were the highest (r1: 1.306 m M−1 s−1 and r2: 234.5 m M−1 s−1) when compared to other groups. The cytotoxicity of GMIO-2 was negligible under a wide range of dosages, thus exhibiting excellent cell biocompatibility. Moreover, GMIO-2 could quickly diffuse into cells, leading to its effective accumulation. The systemic delivery of GMIO-2 resulted in an excellent T1–T2 dual-mode MRI contrast effect in kidneys, which is expected to improve the diagnosis of kidney lesions. Therefore, this work provides a promising candidate for the development of a T1–T2 dual-mode MRI CA.

Gadolinium-Doped BTO-Functionalized Nanocomposites with Enhanced MRI and X-ray Dual Imaging to Simulate the Electrical Properties of Bone

Peng Wang,Lili Hao,Zongliang Wang,Yu Wang,Min Guo,Peibiao Zhang

Physicochemical properties of biomaterials play a regulatory role in osteoblast proliferation and differentiation. Inspired by the electrical properties of natural bone, the electroactive composites applied to osteogenesis have gradually become the hotspot of research. In this work, an electroactive biocomposite of poly(lactic-co-glycolic acid) mixed with gadolinium-doped barium titanate nanoparticles (Gd-BTO NPs) was investigated to establish the structure–activity relationship between electrical property, especially surface potential, and osteogenic activity. Furthermore, the potential mechanism was also explored. The results showed that the introduction of Gd-BTO NPs was more conducive to improve the elastic modulus and beneficial to utilize MRI and X-ray dual imaging. The electrical characteristics of composites indicate that the introduction of Gd-BTO NPs can effectively improve the electrical properties of materials including dielectricity, piezoelectricity, and surface potential. Moreover, adjusting the amount of gadolinium doping could optimize electrical activity and enhance MRI compatibility. The surface potential of 0.2Gd-BTO/PLGA could reach −58.2 to −60.9 mV at pH values from 7 to 9. Functional studies on cells revealed that the negative surface potential of poled Gd-BTO/PLGA enhanced cell attachment and osteogenic differentiation significantly. Furthermore, the negative surface potential could induce intracellular Ca2+ ion concentration oscillation and improve osteogenic differentiation via the calcineurin/NFAT signal pathway. These findings suggest that electroactive Gd-BTO/PLGA nanocomposites may have huge potential for bone regeneration and be expected to have wide applications in the field of bone tissue engineering.

Magnetic drug carrier with a smart pH-responsive polymer network shell for controlled delivery of doxorubicin

Wan-Fu Ma,Ke-Yi Wu,Jing Tang,Dian Li,Chuan Wei,Jia Guo,Shi-Long Wang,Chang-Chun Wang

A smart magnetic targeting drug carrier (MCNC/PAA) comprising an approximately 100 nm sized magnetic colloid nanocrystal cluster (MCNC) core and a pH-responsive cross-linked poly(acrylic acid) (PAA) shell is reported. The abundant carboxyl groups in the shell enable the resultant MCNC/PAA to easily load a large amount of doxorubicin (DOX) (up to 44.6%) via the strong interaction between the DOX and the carboxyl group in a neutral solution. Interestingly, a synergistic pH-responsive effect derived from the entrapped DOX and PAA network was found to effectively manipulate the drug releasing behavior at 37 °C. It was found that the premature release was highly restricted at a pH of 7.4, and upon reduction in pH from 7.4 to 5.0 or 4.0, a large amount of drug was rapidly released. Compared with the synthesized MCNC/PNIPAM, MCNC/PHEMA and MCNC/PDMAPMA nanocarriers, the MCNC/PAA was preferably suited to drug delivery. In addition, the composite nanocarriers could be tracked by magnetic resonance imaging (MRI). The cytotoxicity assay of MCNC/PAA to normal cells indicated that the composite nanospheres were biocompatible and suitable as drug carriers. Meanwhile, the DOX-loaded composite nanospheres had more potent cytotoxicity than free DOX to HeLa cells. These results clearly imply that the MCNC/PAA nanocarrier is a promising platform that can be applied to construct a smart drug delivery system with magnetic targeting and pH-stimulation, as well as tracking by MRI.

Simultaneous Activation of Short-Wave Infrared (SWIR) Light and Paramagnetism by a Functionalized Shell for High Penetration and Spatial Resolution Theranostics

Liyi Ma,Yuxin Liu,Lidong Liu,Anqi Jiang,Fang Mao,Dongdong Liu,Lu Wang,Jing Zhou

Nanoparticle emitting short-wave infrared (SWIR) light has received increased attention in the molecular imaging field due to its deeper tissue penetration, fast imaging, high sensitivity, and resolution. The simultaneously activated SWIR excited directly by an 808 nm laser and T 1 -weighted magnetic resonance imaging (MRI) signal are found in one single-shell nanoparticle NaErF 4 @NaGdF 4 (Er@Gd), which is used as a dual-modality imaging contrast agent in vivo to accurately determine the position of tumors. The conjugated cypate is then aggregated on the surface of Er@Gd@SiO 2 -Cy/bovine serum albumin. With the guidance of dual modality imaging, photothermal therapy is effectively used to ablate tumors in a mouse model. The design of single-shell nanomaterial activation of SWIR imaging and MRI signals is expected to provide a new strategy for high penetration and spatial resolution cancer theranostics.

Jing Wang,Xueao Li,Wenqian Zhao,Hui Tu,Xiangyang Zu,Guangda Li,Jinghua Li,Jing Li,Yuchuan Zhuang,Yanbo Dong,Andrey Tulupov,Fengshou Zhang,Jianfeng Bao

Encapsulated magnetic vortex iron oxide nanorings (MVIONs) within mesoporous silica (MVIONs@SiO 2 ) have been developed to address key challenges in magnetic hyperthermia—specifically, controlled drug release and precise thermal management in tumor therapy. This study showcases the design of these nanoparticles for the targeted delivery of the chemotherapeutic agent doxorubicin (DOX), facilitated by alternating magnetic fields (AMF). The MVIONs@SiO 2 nanoparticles exhibit exceptional specific absorption rates (SAR) of approximately 1021   W∙g -1 , indicating high thermal efficiency that ensures effective tumor heating without harming surrounding tissues. Additionally, these nanoparticles achieve a DOX loading capacity of 23.4% and a release rate of 60% under magnetic heating conditions, optimizing drug delivery to the tumor site. The relaxation rate r 2 of MVIONs@SiO 2 is recorded at 53.32   mM -1 s -1 , demonstrating significant MRI contrast enhancement correlating with nanoparticle concentration within tumors. Experimental results from cell and animal studies confirm the biocompatibility and potent antitumor activity of MVIONs@SiO 2 , highlighting its dual function in improving chemotherapy and magnetic hyperthermia. This innovation in nanoparticle-based theranostics marks a significant advance in integrating diagnostic imaging and therapeutic heating, potentially revolutionizing cancer treatment and diagnostics.

FeIII-Doped Two-Dimensional C3N4 Nanofusiform: A New O2-Evolving and Mitochondria-Targeting Photodynamic Agent for MRI and Enhanced Antitumor Therapy

Zhifang Ma,Mengchao Zhang,Xiaodan Jia,Jing Bai,Yudi Ruan,Chao Wang,Xuping Sun,Xiue Jiang

Local hypoxia in tumors, as well as the short lifetime and limited action region of 1 O 2 , are undesirable impediments for photodynamic therapy (PDT), leading to a greatly reduced effectiveness. To overcome these adversities, a mitochondria-targeting, H 2 O 2 -activatable, and O 2 -evolving PDT nanoplatform is developed based on Fe III -doped two-dimensional C 3 N 4 nanofusiform for highly selective and efficient cancer treatment. The ultrahigh surface area of 2D nanosheets enhances the photosensitizer (PS) loading capacity and the doping of Fe III leads to peroxidase mimetics with excellent catalytic performance towards H 2 O 2 in cancer cells to generate O 2 . As such tumor hypoxia can be overcome and the PDT efficacy is improved, whilst at the same time endowing the PDT theranostic agent with an effective T 1 -weighted in vivo magnetic resonance imaging (MRI) ability. Conjugation with a mitochondria-targeting agent could further increase the sensitivity of cancer cells to 1 O 2 by enhanced mitochondria dysfunction. In vitro and in vivo anticancer studies demonstrate an outstanding therapeutic effectiveness of the developed PDT agent, leading to almost complete destruction of mouse cervical tumor. This development offers an attractive theranostic agent for in vivo MRI and synergistic photodynamic therapy toward clinical applications.
