Accurate intraoperative delineation of tumor margins remains a major clinical challenge because conventional fluorescence imaging is strongly influenced by probe concentration, tissue heterogeneity, and imaging conditions. Here, we report a renally clearable, tumor-targeted molecular probe, NY-07, for quantitative tumor-margin delineation using second near-infrared (NIR-II) fluorescence lifetime (FLT) imaging. NY-07 is constructed by conjugating a cyanine-based fluorophore with pemetrexed as an active targeting ligand, yielding a clinically compatible probe with excellent solubility, high optical stability, and efficient renal clearance. NY-07 exhibits tail emission in the NIR-II window with a stable fluorescence lifetime of 480 ± 0.6 ps and NIR-II brightness approximately twice that of indocyanine green (ICG) at the same concentration, while retaining ∼70% of its NIR-II signal after 7 days of storage. In vivo imaging demonstrates rapid renal excretion, with strong kidney and bladder signals within seconds after injection and minimal hepatic accumulation. Using cell models, tumor-bearing mice, and patient-derived tissues, we show that NY-07 enables target-specific NIR-II FLT imaging and achieves quantitative discrimination between tumor, inflammation, and normal tissues based on distinct lifetime signatures, enabling precise visualization of tumor margins independent of probe concentration and imaging parameters. Compared with conventional NIR-II fluorescence intensity (FLI) imaging, FLT imaging significantly reduces false-positive signals and improves boundary definition in complex biological environments. This work establishes a clinically translatable strategy that integrates renal clearance, active tumor targeting, and lifetime-resolved imaging for functional tumor-margin delineation, providing a robust platform for improving surgical precision and advancing next-generation image-guided oncologic surgery.

Photothermal therapy (PTT) has gained significant attention as a promising cancer treatment due to its low systemic toxicity, minimal invasiveness, and other advantages. Aggregation-induced emission luminogens (AIEgens) with second near-infrared (NIR-II) fluorescence have emerged as ideal photothermal agents (PTAs), offering key benefits such as high biosafety, excellent biocompatibility, and precise tumor theranostics. Nevertheless, achieving an optimal balance between radiation-mediated NIR-II fluorescence imaging and non-radiation NIR-PTT poses a considerable challenge in designing efficient NIR-II AIEgens. In this work, we employed side chain engineering by strategically grafting n -hexyl onto the π-bridge of a conjugated D-π-A-π-D structure, leading to the development of four new AIEgens ( BQ1 ∼ BQ4 ). These compounds feature 6,7-diphenyl- [1,2,5] thiadiazolo [3,4- g ] quinoxaline as the acceptor, thiophene as the π-bridge, and phenothiazine as the donor. This molecular design enables precise regulation of backbone distortion, effectively balancing fluorescence emission and photothermal conversion. Among them, BQ1 , BQ3 , and BQ4 exhibited strong fluorescence, longer emission wavelengths, and superior photothermal conversion efficiencies (PCEs) (59.2%, 57.8%, 52% respectively), whereas BQ2 , with greater molecular distortion, showed a lower PCE of 33.4%. Notably, BQ1 nanoparticles demonstrated outstanding photothermal conversion, prolonged tumor retention, strong resistance to photobleaching in vitro and in vivo , and NIR-II imaging capability at 1055 nm. This study introduces a novel strategy for optimizing NIR-II/AIE PTAs through precise conformational control via strategic n -hexyl positioning, which remains unreported to date.

Fluorescent ratio-based thermometers, especially the combination of thermal sensitive quantum dots (QDs) with thermal stable lanthanide doped nanoparticles (NPs), have demonstrated great potential for temperature sensing due to their high relative thermal sensitivity. In this work, we synthesized NaYF 4 : 3%Nd@NaYF 4 @SiO 2 @Ag 2 S nanocomposites (NCs), where Ag 2 S QDs were integrated onto the surface of NaYF 4 : 3%Nd@NaYF 4 NPs. Under 808 nm excitation, the luminescence intensity ratio of the NCs in the 1200 nm and 1060 nm exhibits a strong linear relationship with temperature. The NCs show a relative sensitivity of 5.05% °C -1 at 47 °C and maintain photostability through six temperature cycles, under various pH conditions, and during prolonged irradiation. In vitro fluorescence imaging further confirms their potential for temperature measurement in aqueous environments. This approach offers a novel design concept for the creation of a highly sensitive nanothermometer.

Melanoma is a highly invasive and metastatic malignant skin tumor. Recently, immunogenic cell death (ICD) has attracted great attention as a promising approach to immunotherapy. However, efficiently and comprehensively activating ICD throughout the dense tumor tissue is a key challenge. Herein, we designed a NIR II light-driven asymmetric nanomotor drug delivery system (Sor@CS-ZIF-8@MO1) to achieve deep penetration into the tumor tissue. By combining photothermal therapy (PTT) and chemodynamic therapy (CDT) to synergistically induce ICD, the immunotherapeutic efficacy against melanoma is enhanced. The research results showed that Sor@CS-ZIF-8@MO1 exhibited good photothermal performance and motor-driven performance, and was able to effectively penetrate 3D tumor cell spheroids deeply. Sor@CS-ZIF-8@MO1 targeted tumor tissues through mannose and controllably released sorafenib under the low pH conditions in tumor tissues and photothermal stimulation, thereby promoting tumor tissue angiogenesis to improve its hypoxic microenvironment and effectively enhancing the CDT effect induced by Cu +/2+ . This could synergistically enhance the ICD of tumor cells with the PTT. Meanwhile, the tumor-associated antigens released by ICD, together with ovalbumin and mannose, stimulated immune response, reshaped the tumor immune microenvironment, enhanced tumor immunity, and ultimately effectively inhibited the growth and metastasis of melanoma tumors. In this work, a nanomotor delivery system that integrates multiple modalities and is capable of deeply penetrating tumor tissues to efficiently and comprehensively induce immunogenic cell death (ICD) has been designed, providing a new strategy to address the problem of insufficient induction of ICD in melanoma immunotherapy.

Rare-earth nanoparticles (RE NPs) with narrow long wavelength emissions have been recently investigated for their potential application for fluorescence imaging in the second near-infrared window (NIR-II). Previously these RE NPs have a very limited application in the diagnosis and treatment of deep-seated tumors such as brain tumors, due to their weak fluorescence in the range of 1300–1700 nm. Herein, we report a significant enhancement of more than 10 times regular emission of NaNdF 4 nanoparticles at 1340 nm wavelength by coating them with an inert layer of NaLuF 4 , followed by sensitizing with a near-infrared dye (IR-808). We deliver these highly bright nanoparticles into the brain by using focused ultrasound to temporarily open the blood-brain barrier (BBB), and then detect the orthotopic glioblastoma by fluorescence imaging at 1340 nm. The images obtained from long wavelength fluorescence ( i.e. 1340 nm) exhibited better resolution and contrast compared to the short wavelength fluorescence ( i.e. 1060 nm). Our study not only provides insights for enhancing often overlooked emissions of rare-earth nanoparticles for NIR-II fluorescence imaging of deep-seated tumors, but also demonstrates great potential of focused ultrasound based technology in delivering nanotheranostic agents.

Immune checkpoint blockade therapy has revolutionized clinical oncotherapy by intercepting immunosuppressive pathways in tumor tissues. Nevertheless, the therapeutic efficacy is limited by a multidimensional immunosuppressive tumor microenvironment, which impedes the activity of tumor-infiltrating immune cells and undermines treatment outcomes. In this study, a zwitterionic polymer is designed for the conjugation and traceless release of a checkpoint inhibition antibody (Atezolizumab), while concurrently enabling the restoration of membrane redox homeostasis on T cell membranes within the oxidative tumor microenvironment. Contrast-enhanced fluorescence imaging in the near-infrared II spectral region with a deep learning network is employed to visualize in vivo dynamics of the antibody conjugates. The antibody conjugates demonstrate enhanced and accelerated tumor accumulation, thereby promoting T cell proliferation and augmenting the antitumor efficacy. Mechanistic investigations reveal that the conjugate-mediated regulation of the T cell surface redox state protects against damage induced by extracellular reactive oxygen species, subsequently mitigating ferroptosis of T cells. In murine models, the antibody conjugates effectively suppressed colorectal tumor growth and recurrence, leading to a significant extension of animal survival. This work thus realizes reversing suppressive immune microenvironment from multiple dimensions with a simple checkpoint inhibitor conjugate, promising for clinical translation and next-generation immune checkpoint blockade therapy.

Acute kidney injury (AKI) causes renal tubular damage, driven primarily by mitochondrial dysfunction and reactive oxygen species (ROS)-mediated oxidative stress, leading to a cellular energy crisis. The physiological architecture of the kidney hampers targeted drug delivery, rendering metabolic restoration a therapeutic challenge. To address this, we developed a second near-infrared (NIR-II) light-driven, bioenergetic nanoplatform (UCTR) that leverages upconversion nanoparticle (UCNPs)-enhanced photosynthesis for energy replenishment and mitochondrial repair in AKI. The UCTR consists of thylakoid membrane (TM)-encapsulated UCNPs cloaked with activated renal tubular epithelial cell membranes (RECM), enabling targeted accumulation in injured tubules. The UCNPs convert deep-tissue-penetrating NIR-II light into visible wavelengths, activating photosynthetic adenosine triphosphate (ATP) and nicotinamide adenine dinucleotide phosphate (NADPH) synthesis within the TM component under irradiation. This exogenous bioenergetic supply mitigates hypoxia-induced mitochondrial energy deficits, while the AMPK/PGC-1α pathway is simultaneously activated to restore the mitochondrial membrane potential. Moreover, UCTR synergizes the antioxidative and anti-inflammatory effects of plantain, accelerating tubular repair.

Studies have shown that emissions in the NIR-II and NIR-III regions exhibit high tissue penetration, along with superior performance in biological and medical imaging applications. Therefore, developing NIR phosphors capable of simultaneously show emissions in both the NIR-II and NIR-III regions holds significant value. Herein, we developed a novel Cr 3+ /Er 3+ co-doped NaBaScSi 2 O 7 phosphor that demonstrates efficient dual-band emission in both NIR-II and NIR-III regions under visible light excitation, enabled by highly effective Cr 3+ →Er 3+ energy transfer. The energy transfer efficiencies calculated by the emission intensity and fluorescence lifetime are 65.3% and 47.4%, respectively, with the energy transfer mechanism dominated by dipole-dipole interaction. The prepared phosphor can be effectively excited by multiple excitation sources (378 nm, 492 nm, 808 nm, and 980 nm), generating distinct emission responses, highlighting its potential for anti-counterfeiting applications. Additionally, the distinct temperature-dependent optical properties of Cr 3+ and Er 3+ offer a viable approach for non-contact temperature detection, achieving a relative sensitivity of 0.8% K -1 and a temperature resolution of 0.126 K at 498 K. The NIR-II/III pc-LED fabricated by NaBaScSi 2 O 7 : 1%Cr 3+ , 0.2%Er 3+ demonstrates outstanding non-invasive imaging capability and accurate predictions for organic solvents. This study presents a strategy for developing phosphors capable of simultaneous NIR-II/III emission, and significantly advances their multifunctional applications in NIR optical thermometry and NIR pc-LED technologies.

The development of organic molecules activated by near-infrared II (NIR-II) light holds considerable potential for deep image-guided disease diagnosis and therapeutic applications. However, the design and synthesis of such materials remain a significant challenge due to limitations in controllable molecular architecture. In this study, two small molecules, TPEPyC2 and DTPEPyC5 , were firstly synthesized. Subsequently, their respective assemblies, namely TPEPyC2@HSA and DTPEPyC5@HSA , were obtained through co-assembled with the hydrophobic human serum albumin (HSA). Comprehensive analyses demonstrated that only DTPEPyC5@HSA exhibited superior photostability, biosafety, and a large three-photon absorption cross-section (3.85 × 10 −80 cm 6 s 2 ). Notably, this probe enabled precise mitochondrial targeting in live HepG2 cells, as indicated by a co-localization coefficient of Pr = 0.88, and achieved tumor tissue penetration of 143 μm using three-photon fluorescence imaging. This bio-protein encapsulation strategy provides a novel molecular design framework for developing NIR–II–excitable fluorescent probes, thereby significantly enhancing their imaging performance.

Lanthanide NIR-II luminescence free from thermal quenching is attractive for various applications at elevated temperatures. Here, we present a thermal enhancement of Er 3+ NIR-II luminescence based on the synergy of phonon assisted Er 3+ -Er 3+ cross relaxations and controllable Er 3+ -Ho 3+ energy transfer. The lattice expansion of NaYGeO 4 olivine phosphor upon heating (298–823 K) suppresses the Er 3+ -Ho 3+ energy transfer and leads up to 46.1 times enhancement of Er 3+ NIR-II luminescence under 379 nm excitation. The NIR-II luminescence evolution as increasing temperature allows dual-mode ratiometric thermometry, i.e., luminescence intensity ratios (LIRs) based on Er 3+ 4 I 13/2 Stark sublevels and on Er 3+ 4 I 13/2 and Ho 3+ 5 I 6 levels. Obtained relative sensitivities above 0.46% K ‒1 over a broad temperature range of 298–673 K with a maximum of 2.32% K -1 suggest NaYGeO 4 :Er 3+ /Ho 3+ phosphor as a promising NIR-II luminescence thermometer. Moreover, this research offers an available perspective for designing thermal-enhanced NIR luminescence in ordinary thermal expansion materials.

Head and neck squamous cell carcinoma (HNSCC) remains a clinically challenging malignancy due to limited therapeutic options and considerable treatment-related toxicity. In this study, we present an innovative nanotherapeutic platform—monocarboxylate transporter 1 (MCT1) antibody-targeted nanoparticles co-loaded with a second near-infrared window (NIR-II, 1000–1700 nm) absorbing conjugated polymer (PDTP-TBZ-OD, denoted as PTO) and emodin (MCT@PTO-Em NPs) —that synergistically integrates emodin-induced pyroptosis with NIR-II mediated mild photothermal therapy (MPTT) to address critical limitations of current HNSCC treatments strategies. The synthesized NIR-II-absorbing conjugated polymer PTO exhibits strong absorption at 1064 nm and a high photothermal conversion efficiency (39 %), enabling deeptissue penetration and precise hyperthermia control. Accordingly, MCT@PTO-Em NPs implement a dual-modal therapeutic strategy that triggers pyroptosis through NLRP3 inflammasome activation while simultaneously enhancing tumor cell death via mild hyperthermia. Moreover, emodin encapsulated within the nanoparticles (NPs) inhibits heat-shock protein 70 (HSP70), thereby disrupting cellular thermoresistance mechanisms and sensitizing cancer cells to MPTT. Additionally, surface functionalization of the nanoparticles with MCT1-targeting antibodies enhances tumor-specific delivery and intracellular accumulation, thereby minimizing off-target effects. Both in vitro and in vivo studies demonstrate that MCT@PTO-Em NPs achieve superior tumor suppression through the synergistic interplay between pyroptosis and MPTT, while exhibiting excellent biocompatibility. This work presents a precision nanomedicine strategy that integrates molecular targeting, pyroptosis-mediated immune activation, and MPTT, offering a transformative therapeutic paradigm for HNSCC and other solid tumors.

Tumor hypoxia limits the efficacy of photodynamic therapy (PDT), necessitating photosensitizers with hypoxia-adaptive therapy and imaging. Here, we present a fluorinated N -oxide aza-BODIPY ( OFBD ) nanoemulsion ( OFBD-NP ) for hypoxia-responsive, multimodal imaging-guided phototherapy. OFBD generates robust singlet oxygen under normoxia for PDT, but is reduced by CYP450 enzymes in hypoxic cells to photothermal-potent FBD , enabling switchable PDT/PTT. Co-assembly with fluorinated oil enhances oxygen delivery, boosts 19 F MRI sensitivity, and promotes J -aggregation, shifting fluorescence into the NIR-II window for deep-tissue imaging. The redox conversion also activates photoacoustic signals, enabling responsive tri-modal imaging (NIR-II FLI/ 19 F MRI/PAI). OFBD-NP shows potent cytotoxicity in vitro under both normoxic and hypoxic conditions via apoptosis. In vivo, it selectively accumulates in tumors, offers high-contrast imaging, and leads to complete tumor regression in subcutaneous A549 models after laser irradiation, without systemic toxicity. This work demonstrates a smart nanoplatform that integrates deep-tissue imaging and hypoxia-triggered therapeutic switching, addressing major limitations of conventional photosensitizers in cancer imaging and phototherapy.
