Microwave-Actuated Hot-Carrier/Polarization Triggers Catalysis to Coordinate Staphylococcus aureus Ribosome Stalling and Treat Deep-Seated Infections

Yuqian Qiao,Zhiyuan Sang,Ting Zhang,Xiangmei Liu,Qing He,Jiaxiang Qin,Ziting Liu,Xin Qian,Zheng Yang,Yufeng Zheng,Yu Zhang,Zhaoyang Li,Shengli Zhu,Hui Jiang,Zhenduo Cui,Shuilin Wu

Due to the low energy of microwaves (MW), MW dynamic therapy (MWDT) remains inefficient in treating deep-seated infections. Here, an MW-actuated hot-carrier/polarization-triggers catalysis strategy is proposed via two types of tandem reactors to amplify the efficacy of MWDT. By combining MW-thermosensitive carbon nanotube (CNT) with thermal-electricity conversion materials, the Seebeck-type and pyroelectric-type MW-thermal-electricity tandem reactors (CNT-Bi 2 Te 3 and CNT-ZnO) are prepared. The CNT converts MW into a local heat source and rapidly heats Bi 2 Te 3 or ZnO to activate hot-carrier or polarization for high-yield reactive oxygen species production to treat deep-seated infections. Transcriptomic and metabolomic analyses reveal MW stalls the ribosomes of Staphylococcus aureus , thereby hindering its proliferation. Therefore, the biocompatible tandem reactor synergizes with bacterial ribosome stalling to demonstrate a targeted bactericidal effect in vivo and in vitro. Furthermore, a series of antimicrobial products is developed for precise and efficient antimicrobial therapies.

Dual-mode sensing of Staphylococcus aureus in food via fluorescent Apt@Fe/Zr MOF with peroxidase-like activity

Yang Juanli,Chen Xingnu,Ji Yi,Tan Mengyuan,Hu Qin,Gao Lu,Zheng Kaiyi,Yang Zhen-quan,Li Xin,Xu Xuechao

Staphylococcus aureus , a prevalent foodborne pathogenic bacterium, poses a significant threat to human health. Herein, a dual-mode sensing method is proposed for detecting S. aureus in food using aptamer-modified Fe/Zr metal organic framework (Apt@Fe/Zr MOF). The Apt@Fe/Zr MOF was prepared through self-assembly of Fe/Zr MOF followed by surface modification with an aptamer. It exhibits peroxidase-like activity, catalyzing the 3,3′,5,5′-tetramethylbenzidine (TMB) chromogenic reaction. Meanwhile, Apt@Fe/Zr MOF demonstrates fluorescent characteristics, emitting a fluorescence signal at 428 nm upon excitation with light at 350 nm. Based on this feature, dual-mode sensing of S. aureus with comparable specificity and excellent anti-interference was achieved through the integration of colorimetry and fluorometry. The limits of detection (LOD) for both colorimetric and fluorometric analyses were 3.25 × 10 1 and 1.47 × 10 1  CFU mL −1 , respectively. Actual detection of food samples indicates that this method exhibits excellent reliability and accuracy, with recoveries ranging from 91.12 to 107.55% (colorimetry) and 94.40 to 109.47% (fluorometry). The results are highly consistent with those obtained from traditional culture-based methods. Therefore, it is evident that the dual-mode detection of S. aureus using Apt@Fe/Zr MOF offers a rapid and reliable method for ensuring food safety. Graphical

Discovery of natural agents against Staphylococcus aureus based on EIIC by protein modeling, virtual screening and molecular dynamics

Zhiyuan Chen,Miao Li,Yujia Guo,Jianqiang Li,Chi Wei,Jiaying Han,Chunhong Liu,Jingwen Bai,Yu Yang

In this study, a reasonable 3D structure of enzyme IIC (EIIC) was established using ab initio methods. Curcumin, baicalein and phloretin were screened out from 127,695 natural products based on EIIC protein in Staphylococcus aureus ( S. aureus ) by using the virtual screening technology. The antibacterial assays results confirmed that these three compounds could inhibit S. aureus cell growth. The results of molecular docking, molecular dynamics and molecular mechanics/generalized born surface area calculation indicated that curcumin, baicalein and phloretin mainly bound to Glu275, His231, and Phe359 of EIIC through hydrogen bonds and van der Waals forces to obstruct the transport of glucose-based carbohydrates, and thereby inhibited the carbohydrate metabolism of S. aureus . The steered molecular dynamics analysis revealed the existence of transfer channel in EIIC and illustrated that curcumin, baicalein and phloretin could dissociate from EIIC when pulling force on them. Furthermore, alanine scanning and hydrogen bond occupancy analyses found that Glu275 played an important role in stabilizing EIIC-baicalein/curcumin/phloretin complexes, and 275Glu@OE1 was a stable hydrogen bond acceptor. Therefore, Glu275 may be a hot spot residue that can maintain the stable binding of agents to EIIC, which may be a direction for the development of new drugs against S. aureus .

Bacterial cell wall-specific nanomedicine for the elimination of Staphylococcus aureus and Pseudomonas aeruginosa through electron-mechanical intervention

You Yanling,Yu Xu,Jiang Junjie,Chen Zhixin,Zhu Ya-Xuan,Chen Yihan,Lin Han,Shi Jianlin

Personalized synergistic antibacterial agents against diverse bacterial strains are receiving increasing attention in combating antimicrobial resistance. However, the current research has been struggling to strike a balance between strain specificity and broad-spectrum bactericidal activity. Here, we propose a bacterial cell wall-specific antibacterial strategy based on an in situ engineered nanocomposite consisting of carbon substrate and decorated TiO x dots, termed TiO x @C. The fiber-like carbon substrate of TiO x @C is able to penetrate the bacterial membrane of Pseudomonas aeruginosa ( P. aeruginosa ), but not that of Staphylococcus aureus ( S. aureu s) due to its thicker bacterial wall, thus achieving bacterial wall specificity. Furthermore, a series of experiments demonstrate the specific electro-mechanical co-sterilization effect of TiO x @C. On the one hand, TiO x @C can disrupt the electron transport chain and block the energy supply of S. aureus . On the other hand, TiO x @C capable of destroying the membrane structure of P. aeruginosa could cause severe mechanical damage to P. aeruginosa as well as inducing oxidative stress and protein leakage. In vivo experiments demonstrate the efficacy of TiO x @C in eliminating 97% of bacteria in wounds and promoting wound healing in wound-infected female mice. Overall, such a bacterial cell wall-specific nanomedicine presents a promising strategy for non-antibiotic treatments for bacterial diseases.

Sea hedgehog-inspired surface-enhanced Raman scattering biosensor probe for ultrasensitive determination of Staphylococcus aureus in food supplements

Yi Tao,Qing Liu,Ningtao Cheng

Staphylococcus aureus contamination in food supplements poses substantial challenges to public health and large-scale production but the sensitive detection in a timely manner remains a bottleneck. Drawing inspiration from the sea hedgehog, gold nanostars (AuNSs) were leveraged to design an ultrasensitive surface-enhanced Raman scattering (SERS) biosensor for the determination of Staphylococcus aureus in food supplements. Besides the surface enhancement furnished by the AuNSs, Raman reporter molecules and specific aptamers sequentially self-assembled onto these AuNSs to construct the “three-in-one” SERS biosensor probe for label-based quantitation of Staphylococcus aureus . Following incubation with contaminated health product samples, the gold nanostars@Raman reporter-aptamer specifically recognize and assemble around Staphylococcus aureus cells, forming a distinctive sea hedgehog structure. This unique configuration results in an amplified Raman signal at 1338 cm −1 and an enhancement factor of up to 6.71 × 10 7 . The entire quantitative detection process can be completed within 30 min, boasting an exceptional limit of detection as low as 1.0 CFU mL −1 . The method exhibits a broad working range for the determination of Staphylococcus aureus , with concentrations spanning 2.15 CFU mL −1 to 2.15 × 10 5  CFU mL −1 . Furthermore, it demonstrates outstanding precision, with relative standard deviation values consistently below 5.0%. As a showcase to validate the practicality of the SERS method, we conducted tests on determining Staphylococcus aureus in a herbal food supplement, i.e., Ginkgo Biloba extract (GBE); the results align closely with those obtained through the conventional lysogeny broth agar plate method, pointing to the potential applicability in real-world scenarios.

Preparation of Porous Ga-Doped TiO2 Composite Aerogel and Its Bactericidal Activity against Escherichia coli and Staphylococcus aureus

Zihao Song,Longjin Huang,Sheng Cui

In this work, a novel strategy for preparing Ga-doped TiO 2 composite aerogel antibacterial agent through the sol–gel method and supercritical technology is demonstrated. The specific surface area of the samples is as high as 175 m 2 g −1 and exhibits a 3D network structure with a multistage pore distribution. The obtained antibacterial agent combines the phototoxicity of TiO 2 , photo-independent toxicity of Ga 3+ , and structural properties of aerogels, showing a synergistic antibacterial effect. On the one hand, TiO 2 produces reactive oxygen species in the presence of light, thus destroying the structure of bacteria. On the other hand, Ga 3+ competitively replaces Fe 3+ and hinders the metabolic process of bacteria. Moreover, the structural properties of aerogels promote superior antimicrobial performance of antibacterial agents. Their antibacterial properties against Escherichia coli and Staphylococcus aureus bacteria are mainly investigated through the inhibition zone and natural colony counting method. In the results, it is shown that the structure and bactericidal activity of TiO 2 aerogel is effectively optimized with Ga 3+ doping. The maximum zone of inhibition of the composite aerogel against E. coli and S. aureus is 17.5 and 16 mm, and the bactericidal rate is as high as 98.57% and 96.60%, which exhibit outstanding bactericidal performance.

Polyvinyl alcohol/chitosan hydrogel based on deep eutectic solvent for promoting methicillin-resistant Staphylococcus aureus-infected wound healing

Ruigang Zhou,Wenhai Zhang,Jiahao Wu,Junjie Huang,Weimei Wang,Weilong Peng,Jun Chen,Ruonan Bo,Mingjiang Liu,Jingui Li

Bacterial-infected wounds usually lead to slow wound healing due to increased inflammation, especially wounds infected by drug-resistant bacteria, which is a serious challenge in the biomedical field. Traditional antimicrobial strategies such as antibiotics lead to a significant increase in drug-resistant strains and have limited efficacy. Therefore, there is an urgent need to develop multifunctional dressings with excellent antibacterial activity and promotion of wound healing. In this study, the hydrogel is cross-linked by natural deep eutectic solvent (DES), polyvinyl alcohol (PVA), chitosan (CS), tannic acid (TA), and Cu 2+ through intermolecular interactions (hydrogen bonds and metal coordination bonds). The hydrogel was prepared using a simple one-step mixing method without toxic chemical cross-linkers, which conformed to environmentally friendly chemistry. The prepared hydrogel had excellent injectability and self-healing properties and could fill irregular wound surfaces. In addition, in vitro studies have shown that the hydrogel has excellent hemostatic properties, good biocompatibility, and broad-spectrum antibacterial properties. Most importantly, in the methicillin-resistant Staphylococcus aureus- infected skin wound model, the hydrogel could significantly accelerate the healing of infected wounds by inhibiting the intensification of inflammation, increasing collagen deposition, accelerating re-epithelialization, and promoting the formation of hair follicles and new blood vessels.

The anti-biofilm mechanism of reuterin against Staphylococcus aureus and its potential application on milk-contact material surfaces

Mao-Cheng Sun,Yu-Xin Chen,Xiu-Juan Fan,Ying-Di Wang,Tiehua Zhang,Changhui Zhao

Reuterin, a potent antimicrobial compound derived from Limosilactobacillus reuteri , was investigated for its mechanism of action against Staphylococcus aureus biofilm and its efficacy in eliminating biofilms on four distinct milk-contact materials. Results showed that reuterin exhibited a minimum biofilm inhibitory concentration of 0.16 mg/mL against S. aureus ATCC 29213 after treatment for 48 h at 37 °C. Reuterin significantly reduced bacterial hydrophobicity, auto-aggregation and biofilm biomass by 39.36%, 25.49%, and 93.46%, respectively, effectively impeding biofilm formation and bacterial adhesion activity. For the mature biofilm, the minimum biofilm eradication concentration of reuterin incubated for 8 h at 37 °C was 0.31 mg/mL. At this concentration, reuterin significantly decreased the levels of extracellular proteins, polysaccharides and DNA of the biofilm as assayed by the BCA method, the phenol-sulfuric acid test and the spectrophotometric assay. Metabolic activity within the biofilm was reduced by 47.95% determined by the resazurin method, while the viable bacteria was not detected by plate counting. Laser confocal microscopy further confirmed the disruptive effects of reuterin on biofilm architecture. Moreover, RT-qPCR analysis showed that reuterin inhibited the relative expression of the accessory regulator ( sarA ) and bacterial adhesion factors ( clfA, clfB, fnbA, fnbB ). Additionally, reuterin was also found to interfere quorum-sensing genes within S. aureus , particularly targeting the LuxS/AI-2 system ( icaR, icaA, icaB, icaC, icaD ) and the agr system ( agrA, agrB, RNAIII ). Finally, reuterin eliminated biofilms formed on four milk-contact materials, including glass, stainless steel, polyvinyl chloride, and acrylonitrile-butadiene-styrene. Conclusively, reuterin emerges as a highly promising anti-biofilm agent in the food industry.

Ultrasmall high-entropy alloy-nanolabels based immunochromatographic test strip for rapid, ultrasensitive, and catalytic detection of Staphylococcus aureus

Huang Wei,Ren Zhen,Li Xukui,Chen Rong,Fan Dinglin,Da Jingjing,Zha Yan,Xu Yongjie

The use of ultrasmall high-entropy alloys (HEAs, ≈5 nm) with unique catalytic properties, integrating them into lateral flow immunoassay (LFIA) for the detection of Staphylococcus aureus  is proposed. Under optimal conditions, the detection limits of the HEAs-based LFIA were 1.5 × 10 3  CFU/mL via direct coloration and 15 CFU/mL using the DAB-enhanced catalytic signal, representing improvements of 10 3 -fold and 10 5 -fold, respectively. The HEAs demonstrated superior performance compared with traditional AuNPs, including enhanced sensitivity and high peroxidase activity (Km = 0.066 M, Vmax = 1.132 × 10 –6  M min −1 ), as well as excellent specificity and repeatability, with relative standard deviation (RSD) of less than 10%. Moreover, the incubation time for HEAs functionalized with antibodies to bind Staphylococcus aureus was only 5 min, compared with 20 min for functionalized AuNPs. The method also showed high specificity, effectively discriminating Staphylococcus aureus from other foodborne pathogens. The HEAs-based LFIA was successfully applied to food sample analysis, including milk and orange juice, demonstrating its robust applicability for real-world detection scenarios. This approach not only lowered the detection limit but also reduced the total detection time, making it a highly promising candidate for on-site detection of Staphylococcus aureus and other foodborne pathogens by simply changing the recognition element. A novel PEG-mediated boric acid functionalized magnetic nanomaterials based fluorescence biosensor for the detection of Staphylococcus aureus

Zhengzheng Wang,Xiaoyan Feng,Fangbin Xiao,Xuekun Bai,Qian Xu,Hengyi Xu

Staphylococcus aureus ( S. aureus ) is one of the pathogens that seriously endanger human health and are widespread in nature. Food poisoning incidents caused by S. aureus frequently break out. Therefore, it is significant that a rapid and accurate method is established for S. aureus detection. In this study, a strategy using magnetic separation combine with fluorescence analysis was proposed for S. aureus detection. Boric acid can selectively enrich compounds with cis -diol groups through a reversible esterification reaction, and has been widely used in the identification of sugars and glycosylated molecules. As a boric acid derivative, 3-aminophenylboronic acid (APBA) has the advantages of stability, low cost, easy synthesis, etc. Therefore, using APBA as a recognition molecule to prepare magnetic nanomaterials can achieve the enrichment of S. aureus . In order to reduce steric hindrance and ensure the dominant orientation of APBA, thus improving the enrichment performance of target bacteria. The polyethylene glycol (PEG) was used as a mediator to prepare PEG-mediated APBA functionalized magnetic nanomaterials (APBA-PEG-MNs). In addition, this study used FITC-pig IgG to label S. aureus because the Fc fragment of IgG to bind protein A on the surface of S. aureus and realized the detection of S. aureus by detecting fluorescence. Under the optimal experimental conditions, the capture efficiency of APBA-PEG-MNs for S. aureus with the concentration of 2.7 × 10 6 CFU/mL in spinach, juice and pool water spiked samples was 92.6%, 91.6%, 91.6%. The limit of detection of the strategy in spiked samples was 2.7 × 10 2 CFU/mL within 80 min.

In situ forming Hydrogel with adding ZnO Nano-particle for effectively methicillin-resistant Staphylococcus aureus infected frostbite injury

Xun Xu,Jun Wang,Tian-Ci Sun,Qing Li,Ren-De Ning

Hydrogel has emerged as a promising wound dressing material, and in situ forming hydrogel has emerged as a promising wound dressing recently. But most in situ forming hydrogel are normally unstable. Herein, we report an in-situ forming hydrogel synthesized from poly(Nisopropylacrylamide166- co - n -butyl acrylate9)-poly(ethyleneglycol)-poly(N-isopropylacrylamide166- co - n -butyl acrylate9) copolymer (P(NIPAM166- co -nBA9)-PEG-P(NIPAM166-conBA9), denoted as PEP) and zinc oxide nano-particle(ZnO nano-particle) in response to skin temperature. This thermoresponsive hydrogel exhibits sol-gel reversibility at high temperatures, which is closed to the temperature of human skin. To investigate its healing effects, we used the Hydrogel dressing® in an SD rat model. The biocompatibility and antibacterial ability against methicillin-resistant Staphylococcus aureus(MRSA) of this PEP-ZnO hydrogel wound dressing are confirmed in vitro and in vivo, which could transparently promote the healing of a MRSA-infected frostbitten skin Injury. Materials and methods Thirty rats were randomly divided into two groups. The treatment group received hydrogel and transparent film dressing 30 min to 1 h post-burn, while the control group received only cotton dressing. The wound area was measured, and the wound closure rate was calculated on days 3, 7, and 14 post-surgery. Tissue samples were collected from each rat on these days and stored at −80 °C for histological analysis using H&E, Masson and immunohistochemical staining. This analysis assessed factors such as granulation tissue length, re-epithelialization, re-angiogenesis, collagen deposition, inflammatory cell infiltration, and collagen production. Clinical and histological assessments at 14 days showed more rapid healing in the hydrogel dressing group compared to the control group. Conclusion Our results indicate that the design of our hydrogel for cooling injury wounds effectively improves healing and mitigates the damage from low temperatures.

One Stone with Two Birds: Functional Gold Nanostar for Targeted Combination Therapy of Drug-Resistant Staphylococcus aureus Infection

Huajuan Wang,Zhiyong Song,Shuojun Li,Yang Wu,Heyou Han

The development of new antibacterial agents to deal with the emergence and spread of antibiotic resistance in Gram-positive bacterial pathogens has become an increasing problem. Here, a new strategy is developed for the effective targeting and killing of Gram-positive bacteria based on vancomycin (Van)-modified gold nanostars (AuNSs). Our work has demonstrated that the Van-modified AuNSs (AuNSs@Van) can not only selectively recognize methicillin-resistant Staphylococcus aureus (MRSA) but also kill MRSA under near-infrared laser irradiation in vitro. Additionally, AuNSs@Van shows satisfactory biocompatibility and antibacterial activity in treating bacterial infection in vivo. The attractive trait of AuNSs@Van is attributed to the physical effect of its antibacterial activity, with less potential for resistance development. The aforementioned advantages indicate the potential of AuNSs@Van as a photothermal antibacterial agent for effectively combating Gram-positive bacteria in the field of health care.
