Fabrication of chitosan microspheres for efficient adsorption of methyl orange

Linlin Zhai,Zhishan Bai,Yong Zhu,Bingjie Wang,Wenqiang Luo

In this article, morphology, structure and size controllable chitosan microspheres with high mechanical strength were synthesized by microfluidic technology combining chemical crosslinking and used as an adsorbent for methyl orange. The synthesized adsorbents were characterized using scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), and an Energy Dispersive Spectrometer (EDS). The effect of pH revealed that the adsorption process depended on pH and the pH variation of methyl orange solution after adsorption indicated that adsorption capacity was affected through the associated role of chitosan nature and pH variation. Experimental results suggested that the as-prepared chitosan microspheres were controlled within a narrow size distribution (coefficients of variation is 1.81%), whose adsorption capacity reached to 207   mg·g −   1 and mechanical strength was suitable to resist forces. In addition, the adsorption isotherm was well fitted with the Langmuir model, and the adsorption kinetic was best described by the pseudo-second-order kinetic model. The high performance microfluidic-synthesized chitosan microspheres have promising potentials in the applications of removing dyes from wastewater.

PEGylated chitosan microspheres as mucoadhesive drug-delivery carriers for puerarin

Xiaofen Hu,Yuyan Zhang,Huifen Zhou,Haitong Wan

PEGylated chitosans with different degrees of grafting were synthesized, and the application potential of microspheres based on PEGylated chitosan as mucoadhesive drug-delivery carriers for puerarin was investigated. Compared with chitosan microspheres, PEGylated chitosan microspheres (PCMs) exhibited better physical stability and higher swelling capacity, and the amount of water uptake increased as the content of poly(ethylene glycol) methyl ether in the microspheres increased. PCMs showed obviously improved mucoadhesive behavior on a mucosa-like surface. Puerarin was incorporated into the microspheres, and the release experiments in vitro showed that the PEGylation of chitosan accelerated puerarin release from the particles and decreased the retention of the drug. The abilities of all of the tested microspheres to open tight junctions and improve the permeability of puerarin were demonstrated with a Caco-2 cell monolayer as an in vitro model. The amount of puerarin permeating across the Caco-2 cell monolayer was significantly increased by the incorporation of puerarin into the PCMs. © 2015 Wiley Periodicals, Inc. J. Appl. Polym. Degradation and compatibility behaviors of poly(glycolic acid) grafted chitosan

Luzhong Zhang,Sufeng Dou,Yan Li,Ying Yuan,Yawei Ji,Yaling Wang,Yumin Yang

The films of poly(glycolic acid) grafted chitosan were prepared without using a catalyst to improve the degradable property of chitosan. The films were characterized by Fourier transform-infrared spectroscopy and X-ray photoelectron spectroscopy (XPS). The degradation of the poly(glycolic acid) grafted chitosan films were investigated in the lysozyme solution. In vitro degradation tests revealed that the degradation rate of poly(glycolic acid) grafted chitosan films increased dramatically compared with chitosan. The degradation rate of poly(glycolic acid) grafted chitosan films gradually increased with the increasing of the molar ratio of glycolic acid to chitosan. Additionally, the poly(glycolic acid) grafted chitosan films have good biocompatibility, as demonstrated by in vitro cytotoxicity of the extraction fluids. The biocompatible and biodegradable poly(glycolic acid) grafted chitosan would be an effective material with controllable degradation rate to meet the diverse needs in biomedical fields.

Shear induced self-thickening in chitosan-grafted polyacrylamide aqueous solution

Lei Jin,Yonggang Shangguan,Tao Ye,Hu Yang,Quanfu An,Qiang Zheng

A remarkable shear induced self-thickening of chitosan-graft-polyacrylamide aqueous solution was observed. After the polyelectrolyte solution presenting shear thinning was subjected to a high-rate shear for several minutes, their viscosities recovered and then a much higher zero shear viscosity than the original one appeared. Obviously, the self-thickening differs from conventional shear thickening or viscous recovery, as reported previously. The mechanism of self-thickening was investigated by rheological methods together with TEM, 1H NMR and DLS, etc. It was found that some aggregates exist in original chitosan-graft-polyacrylamide aqueous solution and the scale of such aggregations would become larger within several minutes after a strong shear. The thickening was proven to be the result of an enhanced scale of GPAM aggregation in aqueous solution, and the mechanism of aggregation was proven to be intermolecular hydrogen bonding effects. Besides, the shear-induced self-thickening appears to be facile, maintainable and easily controllable by changing the shear conditions.

Preparation of Chitosan-Based Hemostatic Sponges by Supercritical Fluid Technology

Hu-Fan Song,Ai-Zheng Chen,Shi-Bin Wang,Yong-Qiang Kang,Shi-Fu Ye,Yuan-Gang Liu,Wen-Guo Wu

Using ammonium bicarbonate (AB) particles as a porogen, chitosan (CS)-based hemostatic porous sponges were prepared in supercritical carbon dioxide due to its low viscosity, small surface tension, and good compatibility with organic solvent. Fourier transform infrared spectroscopy (FTIR) spectra demonstrated that the chemical compositions of CS and poly-(methyl vinyl ether-co-maleic anhydride) (PVM/MA) were not altered during the phase inversion process. The morphology and structure of the sponge after the supercritical fluid (SCF) process were observed by scanning electron microscopy (SEM). The resulting hemostatic sponges showed a relatively high porosity (about 80%) with a controllable pore size ranging from 0.1 to 200 μm. The concentration of PVM/MA had no significant influence on the porosity of the sponges. Comparative experiments on biological assessment and hemostatic effect between the resulting sponges and Avitene®were also carried out. With the incorporation of PVM/MA into the CS-based sponges, the water absorption rate of the sponges increased significantly, and the CS-PVM/MA sponges showed a similar water absorption rate (about 90%) to that of Avitene®. The results of the whole blood clotting experiment and animal experiment also demonstrated that the clotting ability of the CS-PVM/MA sponges was similar to that of Avitene®. All these results elementarily verified that the sponges prepared in this study were suitable for hemostasis and demonstrated the feasibility of using SCF-assisted phase inversion technology to produce hemostatic porous sponges. Keywords:chitosan;hemostatic sponge;porous structure;supercritical fluids

Electrospun nanofibrous chitosan membranes modified with polyethyleneimine for formaldehyde detection

Na Wang,Xianfeng Wang,Yongtang Jia,Xiaoqi Li,Jianyong Yu,Bin Ding

Here we describe a formaldehyde sensor fabricated by coating polyethyleneimine (PEI) functionalized chitosan nanofiber-net-binary structured layer on quartz crystal microbalance (QCM). The chitosan fibrous substrate comprising nanofibers and spider-web-like nano-nets constructed by a facile electro-spinning/netting process provided an ideal structure for the uniform PEI modification and sensing performance enhancement. Benefiting from the fascinating nanostructure, abundant primary amine groups of PEI, and strong adhesive force to the QCM electrode of PEI–chitosan membranes, the developed formaldehyde sensor presented rapid response and low detection limit (5   ppm) at room temperature. These findings have important implications in fabricating multi-dimensional nanostructures on QCM for gas sensing and chemical analysis.

Hydrophobic and nanoporous chitosan–silica composite aerogels for oil absorption

Qian Ma,Yanfei Liu,Zhe Dong,Jianglang Wang,Xin Hou

Hydrophobic and nanoporous chitosan–silica composite aerogels with low density, high porosity, and superior oil absorbency were successfully prepared by a typical sol–gel method and a two-step hydrophobic treatment. The morphologies, porosity characteristics, mechanical properties, thermal stability, hydrophobicity, and oil absorbencies of the composite aerogels were systematically investigated. The nitrogen physisorption analysis showed that composite aerogels had large specific surface areas and uniform nanoporous structures. In addition, the composite aerogels could support 7000 times its own weight; this indicated the role of the supporting skeleton played by chitosan. The hydrophobicity and lipophilicity was demonstrated with a water contact angle of 137° and an oil contact angle of 0°. Importantly, the composite aerogel with 20 wt % chitosan had a relatively high oil absorbency of 30 g/g and could be reused up to 10 times. Therefore, the chitosan–silica composite aerogels in this study had a broad prospect to be used as efficient and recyclable oil absorbents. © 2014 Wiley Periodicals, Inc. J. Appl. Polym. Preparation and characterization of vanillin-crosslinked chitosan therapeutic bioactive microcarriers

Qin Zou,Junfeng Li,Yubao Li

Chitosan microspheres with diameter of 14.3–48.5   μm were prepared by emulsion method and using natural vanillin as cross-linking agent. The surface morphology and microstructure of the microspheres were characterized by scanning electron microscopy, X-ray diffraction and Fourier-transform infrared spectroscopy, etc. The hollow microspheres showed a well-defined spherical shape with median diameter of 30.3   μm and possessed a uniform surface with micro-wrinkles, which is in favor of the drug release. Interpenetrating network cross-linking mechanism might result from the Schiff base reaction and the acetalization of hydroxyl and carbonyl between chitosan and vanillin. Berberine, as a model drug, was loaded in the microspheres and released in a sustainable manner. The drug loading ratio could change from 9.16% to 29.70% corresponding to the entrapment efficiency of 91.61% to 74.25%. In vitro cell culture study using MG63 cells and in vivo implantation clearly showed that the microspheres could provide favorable cell attachment and biocompatibility. The results confirm that the drug-loaded vanillin-crosslinked chitosan microspheres could be a worthy candidate either as carriers of drugs and cells, or as therapeutic matrix for bone repair and regeneration.

Novel antimicrobial chitosan–cellulose composite films bioconjugated with silver nanoparticles

Shan Lin,Lihui Chen,Liulian Huang,Shilin Cao,Xiaolin Luo,Kai Liu

Cellulose-based membranes have emerged as an attractive alternative to non-biodegradable petrochemical materials. An important drawback, however, is that cellulose-based membranes are prone to biofouling. Silver nanoparticles (AgNPs) encapped with polyacrylic acid were conjugated with the chitosan/cellulose composite films to enhance the antimicrobial activities. Using the 1-ethyl-3-(3-dimethylaminopropyl) carbodii-mide hydrochloride and N -hydroxysuccinimide as biocoupling agents, AgNPs with an average size of 9   nm were distributed evenly in the film without agglomeration. The presence of AgNPs in the chitosan/cellulose–AgNPs composite films was further confirmed by X-ray diffraction measurements. Fourier transform infrared spectroscopy analysis supported the presence of amide bonds between the primary amino groups of chitosan and the carboxylic residues of coordination to silver nanoparticles. The antimicrobial properties of the chitosan/cellulose and chitosan/cellulose–AgNPs composite films were determined using the disk diffusion tests with Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ). As compared to the chitosan/cellulose composite films, the chitosan/cellulose–AgNPs composite films showed significantly improved antimicrobial activities.

Zinc ion-induced formation of hierarchical N-succinyl chitosan film

Hongjiao Wang,Shunli Liu,Aokai Zhang,Kewen Li,Olayinka Oderinde,Fang Yao,Guodong Fu

A series of N-succinyl Chitosan (NSCS) hydrogel derived from different substitution degree of NSCS were fabricated through the zinc ions crosslinking. Subsequently, a light-weight, transparent, and hierarchical NSCS-Zn(II) film was obtained by drying the hydrogel. The hierarchical structure of NSCS-Zn(II) film was confirmed by scanning electron microscopy. The NSCS-Zn(II) film exhibited excellent mechanical strength because of the preferential formation of the alignment structure. The tensile strength and Young's modulus reaches 119.3 MPa and 2.67 GPa when the NSCS substitution degree was 91.95%. Furthermore, a remarkable and enhanced broad-spectrum bactericidal properties of the NSCS-Zn(II) film was revealed through E. coli and S. aureus exposure, respectively. These features indicated that NSCS-Zn(II) film is a promising material that can be used in biomedical engineering. © 2017 Wiley Periodicals, Inc. J. Appl. Polym. Sci. 2017 , 134 , 44664.

Electrospun collagen/chitosan composite fibrous membranes for accelerating wound healing

Zhan Zhang,Xinzhe Zhao,Ziyu Song,Lu Wang,Jing Gao

The protein-polysaccharide nanofibers have attracted intensive attention in promoting wound healing, due to their components and nanoscale fibrous structure that mimics the native extracellular matrix (ECM). For the full-thickness wounds, in addition to promoting healing, hemostatic property and antibacterial activity are also of critical importance. However, currently, protein-polysaccharide-based nanofiber membranes exhibit poor mechanical properties, lack inherent hemostatic and antibacterial capabilities, as well as the ability to promote tissue repair. In this study, we developed composited membranes, which were composed of collagen (Col) and chitosan (Chs), through solvent alteration and post-processing, the membranes showed enhanced stability under physiological conditions, proper hydrophilic performance and improved mechanical property. Appropriated porosity and water vapor transmission rate, which benefit to wound healing, were detected among all the membranes except for Col membrane. Aimed at wound dressing, hemocompatibility, antibacterial activity and cell proliferation of the electrospun membranes were evaluated. The results indicated that the Col/Chs composited membranes exhibited superior blood clotting capacity, and the membranes with Chs exceeding 60% possessed sufficient antibacterial activity. Moreover, compared with Chs nanofibers, significant increase in cell grow was detected in Col/Chs (1:3) membrane. Taken together, the electrospun membrane with multiple properties favorable to wound healing, superior blood coagulation, sufficient antibacterial performance and promoting cell proliferation property make it favorable candidate for full-thickness skin wound healing.

Developing a multifunctional chitosan composite sponge for managing traumatic injuries

Zhicheng Liu,Hao Zhang,Huining Su,Yang Chen,Xunan Jing,Daquan Wang,Shaohui Li,Hao Guan,Lingjie Meng

Developing porous hemostatic sponges that are both biosafe and multifunctional remains a complex challenge. Conventional hemostatic techniques often fall short in managing bleeding effectively, leading to severe critical cases such as suboptimal hemostasis, increased infection risk, and complications arising from profuse bleeding. To address these deficits, our study introduces a novel multifunctional nanocomposite sponge that synergistically incorporates chitosan (CS), cellulose (Cel), graphene oxide (GO), and silver (Ag) nanoparticles. The resulting CS/Cel/GO/Ag developed demonstrates a swelling rate exceeding 3000 %, an absorption rate of over 2100 %, and the lowest stress surpassing 20 kPa at an initial 80 % strain. In vitro analyses reveal that the CS/Cel/GO/Ag sponge has excellent cytocompatibility, non-hemolytic nature, and competence in blood cell adherence and bacterial inhibition. In vivo evaluations further demonstrate that compared to conventional hemostatic methods, the sponge substantially enhances hemostatic efficacy, as evidenced by the marked reductions in clotting times and diminished blood loss compared to conventional hemostatic methods. Specifically, the test results of the CS/Cel/GO/Ag sponge across three different models are as follows: for the rat tail amputation model, the clotting time was 99 s, while blood loss was 222 mg; for the rat liver injury model, the clotting time was 129 s. while blood loss was 812 mg; for the rat femoral artery laceration model, the clotting time was 96 s, while blood loss was 758 mg. The compelling attributes of the CS/Cel/GO/Ag sponges position them as a promising solution for the acute management of bleeding. Their excellent performance indicates they have potential role to play in trauma care.
