With increasing age, skeletal muscle gradually loses mass and strength, and the risk of falls and fractures escalates among elderly individuals. Inflammation is closely related to age-related muscle atrophy and is the potential target for treating muscular atrophy. Here, biomimetic curcumin nanoparticles (M12MNCs) are prepared via encapsulating curcumin in the skeletal muscle cell membranes modified via muscle-homing peptides (M12) for the treatment of aging related skeletal muscle atrophy. The M12MNCs have good biocompatibility and can be enriched in aging skeletal muscle. After treatment with the M12MNCs, aging mice present enhanced motor ability and improved skeletal muscle metabolism. The results of in vivo and in vitro experiments confirm that M12MNCs reduce inflammation and decrease the expression of α-synuclein (α-syn). In addition, M12MNCs ameliorate skeletal muscle dysfunction in aging mice via regulating the SphK1/Spns2/S1PR2 axis. This study provides a therapeutic target of inflammatory and myogenic factors to improve the function of aging skeletal muscle, which provides valuable insights for the subsequent treatment of aging-related skeletal muscle function. These findings suggest that M12MNCs can improve age-related skeletal muscle dysfunction by modulating inflammation and cell proliferation, and can be used as a novel drug delivery system for clinical therapeutic regimens for muscle atrophy.

Postmenopausal osteoporosis (PMOP) is a predominant form of clinical osteoporosis. It has led to significant health and social burdens for older patients. Reestablishing the balance between osteogenic and osteoclastic is a crucial strategy for treating PMOP. Curcumin (Cur), a naturally derived polyphenolic substance, has gained recognition as a viable option for treating osteoporosis. Despite its potential, the clinical use of Cur is hindered by its limited bioavailability and the presence of side effects. Nanoparticles modified with aspartic acid octapeptide (ASP8) exhibit a strong affinity for bone tissue, facilitating targeted delivery. This study presents novel acid-responsive zeolite imidazolate framework-8 (ZIF) nanoparticles modified with ASP8 and loaded with Cur (Cur@ZIF@ASP8, CZA). Upon delivery by this nanoparticle drug delivery system, Cur can effectively regulate bone homeostasis, offering a potential therapeutic strategy for osteoporosis. This study demonstrated that CZA nanoparticles could successfully transport Cur to bone tissue without significant toxicity. Furthermore, nanoparticles promote bone formation and inhibit osteoclast activity. They also modify the expression of related genes and proteins, such as OCN, ALP, CTSK, and MMP9. Significant evaluations utilizing microcomputed tomography, Masson’s staining, hematoxylin and eosin staining, and immunofluorescence staining demonstrated that intravenous CZA administration in ovariectomized mice resulted in bone destruction while simultaneously reducing overall bone loss. In conclusion, CZA nanoparticles hold promise as a therapeutic option for osteoporosis.

Current fluorescent polyurethanes face critical challenges, including cytotoxic fluorescent labels, irreversible damage from mechanical stress, and monofunctional behavior lacking stimulus-responsive sensing capability. To address these limitations, we developed a polyurethane elastomer that merges extreme stretchability, autonomous self-healing, and circular recyclability. Engineered via adipic dihydrazide and 4, 4′-diaminodiphenyl disulfides, the network unites reversible disulfide metathesis with hydrogen-bond dissipation, yielding 10 MPa strength, 3200 % elongation, and 90 % toughness recovery within 72 h. Notch tolerance reaches 1200 % strain, while 75 % ethanol depolymerisation at 25 °C enables solvent-cast recycling (>80 % retention in tensile strength). The matrix confines curcumin, suppressing aggregation-caused quenching and furnishing stable solid-state fluorescence under water for 14 days. Emission color switches from green to yellow in response to substrate polarity. Applications include stamp-sealable anti-counterfeiting inks and real-time pork-freshness sensors that visually report spoilage through pH-triggered fluorescence decay. This work overcomes key limitations in natural fluorophore applications by establishing a structure-property optimization paradigm that simultaneously achieves high performance, environmental sustainability, and multifunctionality for safety-sensitive applications.

In this work, sequential electrospinning was utilized to fabricate a multilayer film with ethylcellulose nanofibers as the outer layer and curcumin-loaded gelatin nanofibers as the inner layer. Field-emission scanning electronic microscopy observations showed that the outer and inner layers had a smooth surface and clear boundary. The hydrophobic outer layers decreased the water vapor permeability and improved the water contact resistance of the hydrophilic inner layer, and the intimate interactions of hydrogen bonds between two adjacent layers enhanced the thermal stability . The multilayer film exhibited a sustained release manner of the encapsulated curcumin for 96 h, compared to the burst release within 30 min from the gelatin film. In addition, the antioxidant activities of the released curcumin from the multilayer film were well retained within 96 h. These results suggested that the multilayer nanofibrous film fabricated by sequential electrospinning has potential applications in bioactive encapsulation and controlled release.

Novel curcumin (Cur) cellular delivery system based on assembly of clover shaped trityl polyethylene glycol (TrPEG) with heat denatured ovalbumin (OVA) through hydrophobic interactions was developed in this study. TrPEGs of different degree polymerization (n = 3, 6, 9) were prepared by the coupling of trityl chloride and PEGs under the optimized condition, and the structures were verified by NMR and MS measurements. The excellent amphiphilic and biocompatible TrPEG-9 demonstrated marvelous selective binding capability with the denatured OVA and stabilization of the assembled particles through the clover shaped head. Introduction of TrPEG-9 (EG) with OVA significantly enhanced encapsulation efficiency (83 %) of Cur and afforded uniform compact nanoparticles (63 nm). Evaluation by Saccharomyces cerevisiae model revealed effectively promoted Cur cellular delivery and significantly reactive oxygen species (ROS) reduction capacity of EG-OVA-Cur, and clathrin-mediated endocytosis and macropinocytosis cellular uptake followed by intracellular vacuolar trafficking. This assembled system has great potential for cellular delivery of nutraceutical and biomedical bioactives.

BackgroundAdding a suitable surfactant can enhance the transdermal permeability of transethosomes while also leveraging its functionality as a functional material. In this study, transethosomes were prepared using D-α-tocopherol acid polyethylene glycol succinate (TPGS) as edge activators for transdermal delivery of curcumin (Cur).MethodsThe TPGS-mediated curcumin-loaded transethosomes (Cur@TES) were prepared and formulated optimally, and the optimized formulations were characterized for their morphology, particle size, entrapment efficiency (EE) and drug loading (DL). The stability and deformability of Cur@TES were investigated, while the transdermal delivery of Cur@TES was investigated through in vitro transdermal assays and fluorescence imaging. A mouse ear swelling model was performed to determine the anti-inflammatory effect of Cur@TES.ResultsCur@TES appeared round or elliptical in shape. The particle size, EE and DL for the optimized formulation were observed as 131.2 ± 7.2 nm, 97.68 ± 2.26%, and 6.58 ± 0.62%, respectively. X-ray diffraction analysis confirmed the formation of disordered structures in the inner core of the vesicles. Moreover, Cur@TES system demonstrated better stability and deformability compared to the curcumin-loaded ethosomes (Cur@ES). In vitro transdermal experiments demonstrated that Cur@TES significantly increased the amount of drug retained in the skin (P<0.05). Fluorescence imaging confirmed that the skin distribution was distinctly enhanced with the delivery by TPGS mediated transethosomes. In addition, Cur@TES showed a significant inhibitory effect on Inflammatory swelling in the mouse ear-swelling model.ConclusionTPGS-mediated transethosomes exhibit significant transdermal advantages and enhanced anti-inflammatory effects, providing a new perspective for the transdermal delivery of curcumin.

Nutrients co-encapsulation is a significant research focus in functional foods. This study developed a method to co-encapsulate fish oil and curcumin (Cur) using bovine bone gelatin (BBG)-chitosan (CS) complex coacervation, followed by freeze-drying with maltodextrin (MD) as a drying aid. The results confirmed that the successful encapsulation of fish oil/Cur droplets in the powder particles. As the MD/BBG mass ratios increased (0–8), several trends were observed for the powders: the oil loading capacity decreased progressively (17.7%–2.7%), oil encapsulation yields increased and then decreased (54.1%–27.9%), oil encapsulation efficiencies exponentially increased (75.6%–92.9%), Cur loading capacities exponentially decreased (0.82%–0.24%), Cur encapsulation yields exponentially increased (63.7%–85.9%), Cur encapsulation efficiencies linearly decreased (79.8%–73.4%), and peroxide values during the oxidative stability process logistically increased (19.5–44.5 mmol/kg oil at 1 h). During simulated small intestinal digestion, an MD/BBG mass ratio of 5 resulted in the highest percentage (92.8%) among all ratios tested (0–8). These results suggested fish oil can act as an effective carrier for Cur in functional powders, and that miscible nutrients can be co-encapsulated within complex coacervates. This work provided an efficient strategy for developing raw materials with co-encapsulated nutrients for functional foods and contributed to understanding the underlying mechanism of the multi-nutrient co-encapsulation.

Osteoarthritis (OA) is a persistent inflammatory joint disorder for which there are currently no effective therapeutic interventions. The pathogenesis of OA is intimately associated with oxidative stress and an inflammatory environment. Therefore, this study developed curcumin-loaded mesoporous catechin nanoparticles (Cur@MCN) for enhanced treatment of OA. MCN could effectively encapsulate curcumin due to their mesoporous features, thereby significantly enhancing curcumin's dispersibility and bioavailability. In vitro studies revealed that the Cur@MCN surpassed either free curcumin or MCN alone in scavenging reactive oxygen species, attenuating inflammatory responses, and protecting chondrocytes. Mechanistically, the Cur@MCN drive macrophage repolarization from the M1 (pro-inflammatory) type toward the M2 (anti-inflammatory) type via the targeted modulation of the NLR family pyrin domain-containing 3 (NLRP3) inflammasome signaling axis. In a validated Sprague-Dawley rat model of OA, intra-articular administration of Cur@MCN significantly alleviated cartilage erosion, reduced synovial inflammation, and improved OARSI histological scores, confirming its potent chondroprotective and anti-inflammatory effects. These findings indicate that Cur@MCN may be an effective therapeutic agent for treating OA and other diseases related to oxidative stress.

Under pH 4.0 HAc–NaAc buffer medium, curcumin alone possesses extraordinary weak fluorescence emission . Nevertheless, the introduction of Triton X–100 micelles can largely enhance the fluorescence intensity of curcumin . Uranyl ions can complex with micelles–capped curcumin , along with the slight red shift of curcumin fluorescence (about 1–7 nm), a clear decrement of absorbance (424 nm) and fluorescence (507 nm) intensities, and a distinct color change from bright yellow to orange. The fluorescence decrements ( Δ F, 507 nm) are positively correlated to the amount of uranyl ions in the concentration range of 3.7 × 10 − 6 –1.4 × 10 − 5 mol L − 1 . The detection limit of this fluorescence quenching methods is 3.7 × 10 − 6 mol L − 1 , which is nearly 9000 times lower than the maximum allowable level in drinking water proposed by World Health Organization. Good selectivity is achieved because of a majority of co–existing substances (such as Ce 4 + , La 3 + , and Th 4 + ) do not affect the detection. The content of uranyl ions in tap water samples was determined by the proposed method with satisfactory results.

A Dual-Synergy strategy integrates supramolecular co-assembly with heterogeneous protein networks for efficient delivery. • DFT and IGMH analyses reveal π–π and C–H···π interactions drive Cur–Que co-assemblies’ synergistic antioxidant activity. • Cur/Que. co-assemblies reinforce the OVA/PPI network via extensive non-covalent interactions, elevating T peak to 91.21°C. • The composite gel restricts lipid digestion and boosts Cur and Que in vitro bioavailability to 58.96% and 63.17%. Hydrophobic phytochemicals often show limited solubility and low bioaccessibility during digestion, which restricts their use in functional food systems. In this study, a protein-based emulsion gel was developed by embedding a curcumin–quercetin supramolecular co-assembly within a composite network formed from ovalbumin and peanut protein isolate. The curcumin–quercetin co-assembly at a 5:5 ratio showed the strongest synergistic antioxidant activity, with a combination index of 0.598. Density functional theory calculations and independent gradient model analysis based on Hirshfeld partition indicated that π–π stacking and C–H···π interactions stabilized the co-assembled structure, modulated frontier orbital energy levels, reduced the energy gap, and increased electrophilicity, thereby enhancing radical-scavenging activity. The ovalbumin–peanut protein isolate composite at a 5:5 ratio formed a well-developed gel network and generated nanoscale embedded structures of approximately 779 nm through electrostatic and hydrophobic interactions. Incorporation of 1% curcumin–quercetin co-assembly resulted in encapsulation efficiencies above 97% for both phytochemicals. It also increased the storage modulus by 77.5% and improved water-holding capacity to 82.15%. Low-field nuclear magnetic resonance analysis showed a higher proportion of bound water, while the thermal transition temperature increased from 81.83 to 91.21 °C. These results indicate that the co-assemblies served as active crosslinking domains within the protein network through hydrogen bonding and hydrophobic interactions. During simulated digestion, the integrated co-assembly gel system delayed lipid digestion, with a free fatty acid release of approximately 58.55%, and increased the bioaccessibility of curcumin and quercetin to 58.96% and 63.17%, respectively. This study reveals how molecular co-assembly and protein gel structuring jointly improve the stability and delivery performance of hydrophobic phytochemicals, providing a basis for designing nutrient-dense functional food delivery systems.

Natural biopolymer-based nanoparticles are promising candidates for emulsion stabilization and delivery of hydrophobic compounds. In this study, tea protein (TP) and chitosan (CS) were employed to fabricate complex nanoparticles (NPs) using a pH-dependent method. At a mass ratio of 2:1 (TP:CS), the NPs exhibited favorable properties (206.62 nm, −30.86 mV, 85.41° contact angle) owing to electrostatic interactions, hydrogen bonding, and hydrophobic interactions (binding energy of the complex was −5.40 kcal/mol). At a particle concentration of 2 % and an oil-phase volume fraction of 60 %, the Pickering emulsion stabilized by TP-CS 2:1 NPs remained physically stable. Compared with TP alone, TP-CS 2:1 NPs significantly enhanced the encapsulation efficiency and antioxidant capacity (DPPH/ABTS) of curcumin (Cur). In vitro digestion confirmed that the emulsions reduced gastric degradation of Cur, increasing its bioaccessibility by 17.4 %. Accordingly, this study provides a strategy for designing natural protein-polysaccharide NPs for functional emulsions and sustained delivery systems.

Camptothecin (CPT) is a potent broad-spectrum antitumor agent with efficient therapeutic effect for various cancers. However, its application in glioma therapy has been impeded by the tumor immunosuppressive environment and blood–brain barrier (BBB)-associated drug delivery challenges. Herein, neurotransmitter analogs-modified liposomes (NTs-LIP) were prepared by doping lipidized tryptamine (Tryp) to co-deliver CPT and curcumin (CUR) for improved chemo-immunotherapy in glioma. The introduction of Tryp promotes the delivery efficiency of CPT and CUR across the BBB. CPT inhibits cell proliferation after cellular uptake of NTs-LIP, the combination of which with CUR downregulates the elevated expression of the programmed cell death 1 ligand 1 (PD-L1) caused by CPT to prevent the inactivation of T-cells and synergistically enhance chemo-immunotherapy efficacy. Furthermore, both Tryp and CUR interfere with the indoleamine 2,3-dioxygenase (IDO) pathway to reduce regulatory T cell (Treg)-mediated immunosuppression, exhibiting the potential to combine with PD-L1 inhibition for synergistic antitumor immunity. Taken together, this platform contributes towards targeted delivery and alleviation of the immunosuppressive environment in glioma therapy.
