Reduction of Nε-(carboxymethyl) lysine by (−)-epicatechin and (−)-epigallocatechin gallate: The involvement of a possible trapping mechanism by catechin quinones

Yuting Li,Lin Li,Marianne N. Lund,Bing Li,Yi Hu,Xia Zhang

(−)-Epicatechin (EC) and (−)-epigallocatechin gallate (EGCG) (0.001–5%) were found to reduce N ε -(carboxymethyl) lysine (CML) concentrations by up to 0.80 ± 0.11 mM and 0.85 ± 0.07 mM, respectively, as quantified by HPLC-MS during heating at 80 °C and 100 °C in a glucose-lysine model system at pH 5, 7 and 8, which mimic different food processing conditions. With the addition of 1 mM EC, the free CML concentration in coconut milk was reduced to 5.7 ± 0.1 μg/mL (∼2.8 × 10 −2  mM), which was significantly lower than that in unheated coconut milk (6.1 ± 0.2 μg/mL (∼3.0 × 10 −2  mM), p < 0.05) indicating that the free CML may be eliminated by the addition of EC in coconut milk. Products of EC/EGCG quinones reacted with CML by a Michael addition or Schiff base addition in model systems were tentatively identified by UPLC-ESI-MS/MS.

Calcium carbonate/ε-poly(lysine) hybrid microparticles for uranium seawater extraction: an eco-compatible emergency response to marine nuclear disasters

Jérémie Courtois,Chunyan Li,Chen Wang,Hongping Zhang,Heping Jin,Wei Feng

During the past decade, a large variety of materials dedicated to uranium seawater extraction has been developed for harvesting purposes. Yet, materials especially designed for decontamination following nuclear accidents are scarce and eco-friendly ones even rarer. In this study, hybrid microparticles made of biocompatible calcium carbonate and ε-poly(lysine) are used as efficient U sorbent in seawater. Their eco-friendly nature makes them interesting for both harvesting and on-site immobilization of radionuclides, also accounting their ability to settle in the seabed with limited impact on eco-systems. The material shows excellent U extraction capability in contaminated seawater ([U] = 10 ppm) with removal rate R of 87.7 % and extraction capacity of 513 mg per g of ε-poly(lysine) and remains efficient in natural condition ([U] = 2.8 ppb) with R = 39.8 %. In addition, the sorbent has a remarkable selectivity toward U versus V in U contaminated seawater as well as a significant affinity for Sr 2+ . DFT and XPS analyses revealed that U is adsorbed to ε-poly(lysine) via a chelation mechanism that involves amide and amine groups of lysine. Lability of the formed coordination complex allows easy recovery of captured U by contact with NaHCO 3 desorbing solution, ensuring recyclability of the material.

Dendron-like poly(ε-benzyloxycarbonyl-L-lysine)/linear PEO block copolymers: Synthesis, physical characterization, self-assembly, and drug-release behavior

You-Cui Xu,Chang-Ming Dong

Dendron-like poly(ε-benzyloxycarbonyl- L -lysine)/linear poly(ethylene oxide) block copolymers (i.e., Dm-PZLys- b -PEO, m = 0 and 3; Dm are the propargyl focal point poly(amido amine) dendrons having 2 m primary amine groups) were for the first time synthesized by combining ring-opening polymerization (ROP) of ε-benzyloxycarbonyl- L -lysine N -carboxyanhydride (Z-Lys-NCA) and click chemistry, where Dm-PZLys homopolypeptides were click conjugated with azide-terminated PEO. Their molecular structures and physical properties were characterized in detail by FTIR, 1 H NMR, gel permeation chromatography, differential scanning calorimetry, polarized optical microscopy, and wide angle X-ray diffraction. Both homopolypeptides and copolymers presented a liquid crystalline phase transition for PZLys block, and the transition was irreversible. Moreover, the degree of crystallinity of PEO block within linear copolymers decreased from 96.2% to 20.4% with increasing PZLys composition, whereas that within dendritic copolymers decreased to zero. The secondary conformation of PZLys progressively changed from β-sheet to α-helix with increasing the chain length. These copolymers self-assembled into spherical nanoparticles in aqueous solution, and the anticancer drug doxorubicin-loaded nanoparticles gave a similar morphology compared with their blank counterparts. The drug-loaded nanoparticles showed a triphasic drug-release profile at aqueous pH 7.4 or 5.5 and 37 °C and sustained a longer drug-release period for about 2 months. © 2012 Wiley Periodicals, Inc. Effects of maltose and lysine treatment on coffee aroma by flash gas chromatography electronic nose and gas chromatography–mass spectrometry

Yuqin He,Haide Zhang,Nana Wen,Rongsuo Hu,Guiping Wu,Ying Zeng,Xiong Li,Xiaodan Miao

BACKGROUND Arabica coffee is a sub-tropical agricultural product in China. Coffee undergoes a series of thermal reactions to form abundant volatile profiles after roasting, so it loses a lot of reducing sugars and amino acids. Adding carbonyl compounds with amino acids before roasting could ensure the nutrition and flavour of coffee. The technology is versatile for the development of coffee roasting process. This investigation evaluates the effects of combining maltose and lysine (Lys) to modify coffee aroma and the possibly related mechanisms. Arabica coffee was pretreated with a series of solvent ratios of maltose and Lys with an identical concentration (0.25 mol L −1 ) before microwave heating. RESULTS It was found that the combination of maltose and Lys significantly ( P ≤ 0.05) influenced quality indices of coffee (pH and browning degree). Ninety-six aromatic volatiles have been isolated and identified. Twelve volatile profiles revealed the relationship between fragrance difference and compound content in coffee. Moreover, coffee aroma was modified by a large number of volatiles with different chemical classes and character. CONCLUSION Thus, our results suggest that the combination of reagents changed overall aroma quality through a series of complex thermal reactions, especially the ratio of Lys/maltose over 2:1. © 2017 Society of Chemical Industry

Development of nonfouling polypeptides with uniform alternating charges by polycondensation of the covalently bonded dimer of glutamic acid and lysine

Qinghua Yang,Longgang Wang,Weifeng Lin,Guanglong Ma,Jiang Yuan,Shengfu Chen

In this work, nonfouling polypeptides with homogenous alternating charges were synthesized by polycondensation of the covalently bonded dimer of glutamic acid (E) and lysine (K) (EK dimer) with benzyloxycarbonyl (Z)-protected side chains. This facile method successfully solved the uniformity problem of nonfouling peptides caused by the copolymerization of two different monomers and enabled the incorporation of various terminal functional groups for future applications. The molecular weights (MWs) of the nonfouling peptides can be easily controlled by the ratio of the terminal group, lipoic acid, to the EK dimer. The nonfouling peptides can form self-assembling monolayers (SAMs) on a gold surface through two terminal thiol groups, which were characterized by attenuated total reflection Fourier transform infrared (ATR-FTIR), X-ray photoelectron spectroscopy (XPS) and ellipsometry (ELL). The resistance to nonspecific protein adsorption, cell attachment and bacterial adhesion of these nonfouling peptide SAMs and the in vitro cytotoxicity and haemolytic activity of these peptides were also evaluated. The results show that the lowest relative protein adsorptions of antibody (anti-IgG) and fibrinogen (Fg) on the SAMs are 5.1 ± 1.6% and 7.3 ± 1.8%, respectively, determined by enzyme-linked immunosorbent assay (ELISA), where the protein adsorption on a tissue culture polystyrene (TCPS) surface was set to 100%. Almost no obvious cell attachment and bacterial adhesion were observed, and no cytotoxicity and no haemolytic activity in vitro were detected. With the advantages of biocompatibility, biodegradability and the abundance of moieties for ligand immobilization, these nonfouling peptides developed by the facile method can be used in a wide range of biomedical applications.

Hydroxypropyl chitosan/ε-poly-l-lysine based injectable and self-healing hydrogels with antimicrobial and hemostatic activity for wound repair

Shuting Peng,Sen Niu,Qin Gao,Ruiyuan Song,Zhengxiao Wang,Ziyun Luo,Xi Zhang,Xiaofei Qin

The biggest obstacle to treating wound healing continues to be the production of simple, inexpensive wound dressings that satisfy the demands associated with full process of repair at the same time. Herein, a series of injectable composite hydrogels were successfully prepared by a one-pot method by utilizing the Schiff base reaction as well as hydrogen bonding forces between hydroxypropyl chitosan (HCS), ε-poly- l -lysine (EPL), and 2,3,4-trihydroxybenzaldehyde (TBA), and multiple cross-links formed by the reversible coordination between iron (III) and pyrogallol moieties. Notably, hydrogel exhibits excellent physicochemical properties, including injectability, self-healing, water retention, and adhesion, which enable to fill irregular wounds for a long period, providing a suitable moist environment for wound healing. Interestingly, the excellent hemostatic properties of the hydrogel can quickly stop bleeding and avoid the serious sequelae of massive blood loss in acute trauma. Moreover, the powerful antimicrobial and antioxidant properties also protect against bacterial infections and reduce inflammation at the wound site, thus promoting healing at all stages of the wound. The study of biohydrogel with multifunctional integration of wound treatment and smart medical treatment is clarified by this line of research.

Injectable nanoparticle-crosslinked xyloglucan/ε-poly-l-lysine composite hydrogel with hemostatic, antimicrobial, and angiogenic properties for infected wound healing

Jingmei Teng,Wei Zhao,Shengyu Zhang,Dan Yang,Yu Liu,Rongjian Huang,Yuxi Ma,Lei Jiang,Hua Wei,Jiantao Zhang,Jing Chen

Skin wounds are susceptible to infection, leading to severe inflammatory reactions that can progress to chronic wounds, ultimately causing significant physical and mental distress to the patient. In this study, we propose an injectable composite hydrogel achieved through one-pot gelation of oxidized xyloglucan (OXG), cationic polyamide ε-Poly- l -lysine (EPL), and surface amino-rich silicon nanoparticles (SiNPs). OXG exhibits commendable anti-inflammatory properties and provides crosslinking sites. SiNPs serve as mechanically reinforced crosslinkers, facilitating the construction of a dynamic Schiff base network. SiNPs significantly reduced the gelation time to 3 s and tripled the storage modulus of the hydrogels. Additionally, the combination of EPL and SiNPs demonstrated synergistic antimicrobial activity against both S. aureus and E. coli . Notably, the hydrogel effectively halted liver bleeding within 30 s. The hydrogel demonstrated outstanding shear-thinning and self-healing properties, crucial considerations for the design of injectable hydrogels. Furthermore, its efficacy was evaluated as a wound dressing in a mouse model with S. aureus infection. The results indicated that, compared to commercial products, the hydrogel exhibited a shorter wound healing time, decreased inflammation, thinner epithelium, increased hair follicles, enhanced neovascularization, and more substantial collagen deposition. These findings strongly suggest the promising potential of the proposed hydrogel as an effective wound dressing for the treatment of infected wounds.

The L-lysine-assisted synthesis of a nano-ZSM-5 zeolite as a catalyst for oleic acid pyrolysis to BTX

Sen Fu,Hanxu Guo,Hong Yuan

A nano-ZSM-5 zeolite having a hierarchical pore structure was synthesized using L-lysine as a mesopore template and growth inhibitor. The effects of the L-lysine/Si and Si/Al molar ratios on the morphology and pore structure of the zeolite were studied. The products were characterized by scanning electron microscopy, transmission electron microscopy and N2-Brunauer–Emmett–Teller surface area analysis. These assessments showed that the particle size of the ZSM-5 zeolite could be reduced to 60 nm with a surface area as high as 395 m2 g−1 and a high density of mesopores. These zeolites were applied to the catalytic pyrolysis of oleic acid in a fixed-bed reactor to generate benzene, toluene and xylenes (BTX). At a reaction temperature of 500 °C, the catalyst having L-lysine/Si and Si/Al ratios of 0.4 and 30, respectively, gave a BTX yield of 27.4 wt%. This yield decreased with increases in the Si/Al ratio whereas varying the L-lysine/Si ratio did not produce a specific trend. The spent catalyst was characterized by thermogravimetric analysis and exhibited superior anti-coking ability and longer catalytic lifetime compared with a standard ZSM-5 zeolite.

Preparation and characterization of carboxymethyl glucuronoxylan/polyvinyl alcohol/ε-poly-(L-lysine) carbon dots/ZnCl2 composite films with antibacterial properties

Liu Yushen,Fan Xiaotong,Zhang Xiaoran,He Chao,Xiang Zhouyang,Hu Zhenhua

Polyvinyl alcohol (PVA)-based films are widely used due to their mechanical properties and biodegradability. However, their limitations in water resistance and oxygen barrier properties hindered their further application in packaging industry. In this work, carboxymethyl glucuronoxylan/polyvinyl alcohol/ε-poly-(L-lysine) carbon dots/ZnCl 2 composite films with high mechanical properties and antibacterial properties were developed. First, carboxymethyl glucuronoxylan (CMX) and ε-poly-(L-lysine) carbon dots (PL-CD) were synthesized. The structural groups of CMX and PL-CD were confirmed by the FTIR. Then, CMX/PVA/ZnCl 2 /PL-CD composite films were prepared by casting method. With the addition of PVA, ZnCl 2 and PL-CD, the composite films demonstrated a notable improvement in mechanical properties, contact angle (tensile strength: 8.65 MPa ~ 22.4 MPa, elongation at break: 1.1% ~ 4.2%, Young’s modulus: 549.1 MPa ~ 1114.7 MPa, contact angel: 74.7° ~ 105.3°, OTR: 2.04 cm 3 /(m 2 ·day) ~ 3.67 cm 3 /(m 2 ·day)). In addition, all the composite films showed excellent antibacterial properties with the addition of PL-CD. Hence, the carboxymethyl glucuronoxylan/polyvinyl alcohol/ε-poly-(L-lysine) carbon dots/ZnCl 2 composite films holds significant promise for diverse applications in the packaging industry.

Target Ligand Separation and Identification of Isoforsythiaside as a Histone Lysine-Specific Demethylase 1 Covalent Inhibitor Against Breast Cancer Metastasis

Mengzhen Gu,Xiaoqing Xu,Xiaoping Wang,Yun Wang,Yu Zhao,Xiaoxian Hu,Lu Zhu,Zhenzhong Deng,Chao Han

Histone lysine-specific demethylase 1 (LSD1) is hyperactive in breast cancer, which is associated with the metastasis of the tumor. Current irreversible LSD1 inhibitors are all synthesized by covalently binding to the flavin adenine dinucleotide cofactor, which often have side effects due to the high affinity for a variety of targets. Here, we identified isoforsythiaside (IFA), a natural phenylpropanoid glycoside isolated from Forsythia suspensa, as a novel covalent inhibitor of LSD1. The target ligand fishing technique and LC–MS/MS analysis identified that IFA could covalently bind to the Ser817 residue of LSD1 by α,β-unsaturated ketone moiety to block the amine oxidase-like domain of LSD1. Moreover, RBMS3/Twist1/MMP2, the downstream signaling pathway of LSD1, was activated after IFA treatment to inhibit the metastasis of MDA-MB-231 cells in vitro and in vivo. This study provided novel molecular templates for development of LSD1 covalence-binding inhibitor and laid a foundation for developing agents against breast carcinoma metastasis for targeting LSD1.

Poly-L-lysine functionalized silica membrane-enhanced colorimetric loop-mediated isothermal amplification for sensitive and rapid detection of Vibrio parahaemolyticus

Xiujuan Wang,Binghui Shi,Cengceng Xia,Mengnan Hou,Jingying Wang,Anning Tian,Chao Shi,Cuiping Ma

Traditional detection of foodborne pathogen relies on advanced analyzers, which is inadequate for the rapid control of infections, particularly in resource-limited regions, highlighting the necessity of developing detection systems for point-of-care testing (POCT). Herein, taking Vibrio parahaemolyticus as a detecting target, we reported poly- L -lysine functionalized silica membrane (PL-SM) based loop-mediated isothermal amplification (pLAMP) platform for sensitive on-site detection. This platform utilized PL-SM for DNA capture driven by the electrostatic attraction between protonated amine groups of poly-L-lysine and negatively charged phosphate groups of DNA, followed by introducing a colorimetric indicator calcein for LAMP amplification. After optimization, the colorimetric mode of pLAMP allowed the screening of V . parahaemolyticus with the visual limit of detection ( v LOD) of 1 CFU/mL in 50 min, 1000-fold lower than methods based on commercial kits. Validation was performed using 174 seafoods, which was 97 % concordant to those of real-time PCR. Furthermore, an image processing approach was developed based on the analysis of the RGB under UV light. Paired with a smartphone, the objective analytical method could be readily conducted in the field. Thus, we propose a sensitive and visual detection platform, which may play a crucial role in improving testing efficiency and accuracy in food safety, medical diagnostics, and environmental monitoring.

Structural Changes and Antibacterial Activity of Epsilon-poly-l-lysine in Response to pH and Phase Transition and Their Mechanisms

Jia-Ning Liu,Sen-Lin Chang,Peng-Wei Xu,Ming-Hui Tan,Bing Zhao,Xiao-Dong Wang,Qing-Sheng Zhao

ε-Poly-l-lysine (ε-PL) consists of 25–35 lysine residues which are linked by an isopeptide bond formed by dehydration condensation of α-carboxyl and ε-amino groups and has good antibacterial activity and broad-spectrum inhibition range. However, there is no clear conclusion about the structure and antibacterial mechanism of ε-PL in aqueous solution. Herein, a high purity of ε-PL was prepared using Amberlite IRC-50 ion-exchange resin. Membrane filtration and dynamic light scattering were used to study the variations of ε-PL aggregation in aqueous solution with pH value. The conformational changes and antibacterial activities of ε-PL and carbamoylated ε-PL in different water environments were studied with circular dichroism (CD) and inhibition zone. The structural changes during the spray-drying process were determined by Fourier transform infrared spectroscopy. The results indicated that the side chain amino charge played a decisive role in the ε-PL conformation and aggregation. ε-PL exhibited the properties of a β-sheet during spray drying from acidic liquids to solids. The cation enhanced the antibacterial activity of ε-PL but did not play a key role. Instead, the backbone of ε-PL might determine the mechanism of ε-PL antibacterial.
