The development of a multifunctional wound dressing that can adapt to the shape of wounds and provide controlled drug release is crucial for diabetic patients. This study developed a carboxymethyl chitosan-based hydrogel dressing with enhanced mechanical properties and tissue adherence that were achieved by incorporating pectin (PE) and polydopamine (PDA) and loading the hydrogel with recombinant human epidermal growth factor (rhEGF). This EGF@PDA-CMCS-PE hydrogel demonstrated robust tissue adhesion, enhanced mechanical properties, and superior water retention and vapor permeability. It also exhibited significant antioxidant capacity. The results showed that EGF@PDA-CMCS-PE could effectively scavenge 2,2′-Azinobis-(3-ethylbenzthiazoline-6-sulphonate), (1,1-diphenyl-2-picrylhydrazyl), and superoxide anions and increase superoxide dismutase and catalase levels in vivo . In vitro cytotoxicity and antibacterial assays showed good biocompatibility and antimicrobial properties. The sustained release of EGF by the hydrogel was confirmed, with a gradual release profile over 120 h. In vivo studies in diabetic mice showed that the hydrogel significantly accelerated wound healing, with a wound contraction rate of 97.84 % by day 14. Histopathological analysis revealed that the hydrogel promoted fibroblast proliferation, neovascularization, and orderly connective tissue formation, leading to a more uniform and compact wound-healing process. Thus, EGF@PDA-CMCS-PE hydrogel presents a promising tool for managing chronic diabetic wounds, offering a valuable strategy for future clinical applications.

Photoactivated pesticides have many advantages, such as high activity, low toxicity, and no drug resistance. However, poor photostability and a low utilization rate limit their practical application. Herein, the photosensitizer hematoporphyrin (HP) was used as a photoactivated pesticide, covalently linked with pectin (PEC) via ester bonds, to prepare an amphiphilic polymer pro-bactericide, and subsequently self-assembled in aqueous solutions to obtain an esterase-triggered nanobactericide delivery system. The fluorescence quenching effect due to the aggregation of HP in nanoparticles (NPs) enabled the inhibition of photodegradation of HP in this system. Esterase stimulation could trigger HP release and increase its photodynamic activity. Antibacterial assays have shown that the NPs had potent antibacterial capacity, almost completely inactivating bacteria after 60 min of exposure to light. The NPs had good adherence to the leaves. Safety assessment indicated that the NPs have no obvious toxic effects on plants. Antibacterial studies on plants have shown that the NPs have excellent antibacterial effects on infected plants. These results provide a new strategy for obtaining a photoactivated bactericide nanosystem with a high utilization rate and good photostability and targeting ability.

Quercetin (Que) is a hydrophobic flavanol that has the potential to prevent colon diseases. This study aimed to design hordein/pectin nanoparticle as a colon-specific delivery system for quercetin. The encapsulation efficiency, physicochemical stability and release properties of the nanoparticles were estimated. The FTIR and secondary structure analysis indicated that hydrogen bonds, hydrophobic interactions and electrostatic attractions were formed in the quercetin-loaded hordein/pectin nanoparticles (Que-hordein/pectin NPs). In comparison to Que-hordein NPs, Que-hordein/pectin NPs exhibited better colloidal stability (physical, UV light, heating and salt). Furthermore, the release properties studies showed that pectin coating restrained the premature release of Que from hordein NPs in gastric fluid and intestinal fluid. In-vitro release, when the Que-hordein/pectin NPs were exposed to simulated colonic fluid (SCF) for 6 h, quercetin was greatly released from the hordein/pectin NPs (15.29 ± 1.17% – 80.60 ± 1.78%). In-vivo release, the concentration of Que (μg/g) in Que-hordein/pectin NPs was 2.18 times higher than Que-hordein NPs in colon tissue after 6 h of oral administration. This study suggests that Que-hordein/pectin NPs have promising applications in the specific delivery and release of quercetin to the colon.

Efficient extraction of uranium from seawater is of great significance to the stable development of nuclear energy. The graft modification of pectin is expected to realize the efficient extraction of uranium through the dual action of aminoxime and carboxyl groups. In this work, novel biomimetic pectin adsorbents were developed and their performance on the adsorption performance of uranium from seawater was investigated. The physical structure and morphology were characterized by employing FT-IR, BET, SEM and elemental analysis. The skeleton structure and surface functional groups of the pectin-derived adsorbents, which were modified to improve the adsorption selectivity, favored the adsorption of uranium in simulated seawater. The modified apple pectin (MPA) adsorbents exhibited the highest adsorption capacity of 212.27 mg/g at optimal conditions. The thermodynamic results showed that the adsorption of uranium on MPA surface followed the Langmuir single-layer model, and the theoretical maximum adsorption capacity could reach up to 296.25 mg/g. The adsorption process conformed to the pseudo-second-order model. Furthermore, the synergistic mechanism between aminoxime and carboxyl groups of the pectin-based adsorbents was proposed. In conclusion, this work provides a new bio-based adsorbent with high efficiency for uranium extraction from concentrated seawater.

Encapsulation is usually adapted to improve probiotic viability under adverse environmental conditions, and used to ensure adequate amount of probiotics reach the distal gastrointestinal tract and achieve probiotic action. In the present study, sub-species of Bifidobacterium were encapsulated with pectin or alginate by an emulsification/internal gelation method. Bovine milk osteopontin (OPN) was used as an excipient with potential to improve the protection of the probiotics during processing and/or gastrointestinal digestion. Here, we reported that the use of OPN significantly improved probiotic viability during freeze drying and simulated infant gastrointestinal digestion. The viability of Bifidobacterium bifidum R0071 and Bifidobacterium breve M-16V loaded in pectin-based freeze-dried microcapsules with 0.5% OPN addition remained at over 10 6 CFU·g −1 after simulated infant digestion. In comparison, the microcapsules without OPN had a probiotic viability of only 10 3 CFU·g −1 . The protective effect of OPN was proven to be dose dependent. For example, the B. bifidum R0071 viability was 6.1 log CFU·g −1 after the gastrointestinal digestion for the pectin-based microencapsulation without OPN addition, while the value remained at 8.4 log CFU·g −1 when OPN concentration reached 5%. In this study, we also demonstrated that extracellular calcium ions could chelate with OPN molecules which could be attributed to preventing damage of the probiotic cell wall and the improved protective effective of microencapsulation. Overall, these results provide new insight into the use of OPN as a microencapsulation excipient to promote probiotic viability and its potential future application in functional foods such as infant formula with enhanced gut health.

Hesperitin (Ht) and hesperidin (Hd) have a variety of biological activities, but their applications are limited due to the poor aqueous dispersion, weak stability and low bioavailability. In this study, a complex coacervate, formed by glutenin heated at pH 2, 80 °C for 8 h and orange peel pectin (P), was used as a carrier to enhance the water dispersibility of Ht/Hd, and their release behavior in vitro were investigated. The main driving forces for the complexes of Ht/Hd with glutenin (Glu) or fibrillated glutenin (Glu-F) were hydrogen bonding and hydrophobic forces. However, the enthalpy change of the interaction of the fibrillated glutenin with Ht/Hd was reduced, implying the formation of a more stable Ht/Hd-Glu-F complex. Furthermore, the optimum mass parameter ratio of Ht/Hd:P:Glu-F obtained from orthogonal experiments was 5:4:40 at pH 6. Both Ht and Hd were almost insoluble in water (pH 6), while the formation of the Ht-Glu-F-P and Hd-Glu-F-P complexes increased the solubility of the almost insoluble Ht and Hd to 102.16 ± 4.82 and 220.57 ± 10.82 μg/mL, respectively. After 30 days of storage, the particle size of the emulsions formed by Ht-Glu-F-P and Hd-Glu-F-P complexes increased from 171.90 to 174.84 nm to 231.82 and 219.81 nm, respectively, which had better storage stability than those formed from Ht/Hd-Glu-P complexes. The increased intermolecular repulsion caused by fibrillation modification improved the stability of the delivery system. These results provide a unique method for improving the aqueous dispersion and storage stability of Ht/Hd.

Clinically frequently-used glutaraldehyde (GA)-crosslinked bioprosthetic valve leaflets (BVLs) are still curbed by acute thrombosis, malignant immunoreaction, calcification, and poor durability. In this study, an anticoagulant heparin-like biomacromolecule, sulfonated, oxidized pectin (SAP) with a dialdehyde structure was first obtained by modifying citrus pectin with sulfonation of 3-amino-1-propane sulfonic acid and then oxidating with periodate. Notably, a novel crosslinking approach was established by doubly crosslinking BVLs with SAP and the nature-derived crosslinking agent quercetin (Que), which play a synergistic role in both crosslinking and bioactivity. The double crosslinked BVLs also presented enhanced mechanical properties and enzymatic degradation resistance owing to the double crosslinking networks formed via CN bonds and hydrogen bonds, respectively, and good HUVEC-cytocompatibility. The in vitro and ex vivo assay manifested that the double-crosslinked BVLs had excellent anticoagulant and antithrombotic properties, owing to the introduction of SAP. The subcutaneous implantation also demonstrated that the obtained BVLs showed a reduced inflammatory response and great resistance to calcification, which is attributed to quercetin with multiple physiological activities and depletion of aldehyde groups by hydroxyl aldehyde reaction. With excellent stability, hemocompatibility, anti-inflammatory, anti-calcification, and pro-endothelialization properties, the obtained double-crosslinked BVLs, SAP + Que-PP, would have great potential to substitute the current clinical GA-crosslinked BVLs.

“High-pressure processing (HPP) plus” combined technologies are applied to overcome the limitation of single HPP and to produce juices with more stable quality during storage. This research explored the potential of HPP in combination with centrifugation to produce cloud stable orange juice during refrigerated storage. The results indicated that HPP combined processing technology significantly improved the cloud stability of orange juice, which was related to removed large particles, reduced viscosity, decreased protein contents, and inactivated pectin methylesterase activity induced by centrifugation ( P < 0.05). Besides, chelator solubilized pectin (CSP) decreased but water solubilized pectin (WSP) maintained in the juice after centrifugation. During storage, the conversion of pectin fraction from WSP to CSP, resulting in sedimentation appeared in centrifugation treated orange juice when stored for 28 days. In general, pectin characteristics changes and pectin fractions conversion were the main driving forces affecting cloud stability of orange juice pasteurized by HPP in combination with centrifugation and during chilled storage.

In this study, pectin (Pe) with different esterification degrees was used as raw materials to prepared hydrogel adsorbents via free radical polymerization. The effect of Pe esterification degree on hydrogel structure and adsorption performance was studied by FTIR , SEM and XPS characterization and copper ion adsorption experiment. The results demonstrated that the carboxyl group in the hydrogels was bonded to Cu 2+ through electrostatic force and coordination, which was an important factor in its adsorption capacity. The hydrogels prepared from Pe with low esterification degree had finer pores and higher carboxyl content, so the adsorption capacity on both water and Cu 2+ was stronger. The preparation of hydrogels from low-ester Pe was more conducive to the adsorption of copper ions. Besides, the adsorption behavior of the hydrogels on Cu 2+ was investigated through the adsorption thermodynamics and kinetics. The results indicated that the adsorption kinetics of the hydrogels was in accordance with the quasi-second-order model. The adsorption of Cu 2+ by hydrogels was the result of physical and chemical adsorption, which was endothermic under natural condition, and a higher temperature will result in more favorable spontaneous adsorption.

In the present study, nanocomplexes composed of perilla seed protein isolate (PSPI) and pectin (PEC) were assembled for the delivery of curcumin (Cur) based on the pH-driven self-assembly method. The encapsulation efficiency of Cur was 87.77%, and the PSPI-Cur-PEC exhibited controlled release of Cur during the in vitro digestion. Moreover, the PSPI-Cur-PEC possessed efficient antioxidant activity and provided greater defense for Cur against thermal treatment and ultraviolet irradiation. Fourier transform infrared spectroscopy , fluorescence spectroscopy and X-ray diffraction results indicated that entrapped Cur was existed in the form of amorphous state in PSPI-PEC, and the process was driven by hydrophobic effects, hydrogen bonding and electrostatic interactions. Scanning electron microscopy images revealed substantial morphological changes of the nanocomplexes throughout the process, while transmission electron microscopy images observed the core-shell structure of PSPI-Cur-PEC nanocomplexes. Furthermore, PSPI-PEC exhibited excellent biocompatibility by the in vitro cytotoxicity assay on Caco-2 cells, and the intermolecular interactions and possible formation mechanism of PSPI-Cur-PEC were further determined by molecular docking . This work elucidated that PSPI-PEC nanocomplexes assembled by a green and energy-efficient pH-driven method was feasible to protect sensitive hydrophobic bioactive components and provided a possible utilization of perilla seed protein in delivery system for nutraceuticals.

Microcapsule colloidal systems have been widely used as delivery vehicles. Natural biopolymers are highly biocompatible shell materials for the fabrication of microcapsules, but no single substance can fulfill the requirements of aqueous two-phase interfacial assembly. In this study, we fabricated pectin-chitosan-collagen composite microcapsules (PCCMs) in an aqueous two-phase system to improve the robustness and sustained-release capacity of resultant microcapsules. Fourier transform infrared spectra and isothermal titration calorimetry thermodynamics analysis indicated that the PCCM interface can be stabilized by pectin, chitosan, and collagen via intermolecular electrostatic interactions and hydrogen bonds. The swelling experiment and atomic force microscopy nanoindentation measurements demonstrated that the PCCMs had excellent anti-swelling performance and enhanced mechanical strength of the composite interfacial membrane. Contraction-recovery measurements and scanning electron microscopy indicated that the PCCMs exhibited prominent anti-shrinkage capacity and restorable surface smoothness. The release kinetics of encapsulated substances under conditions of different osmotic pressure and salt concentrations revealed that the PCCMs had superior sustained-release capacity. This study yielded natural, biocompatible, and robust aqueous two-phase microcapsules for encapsulation and delivery of active biomacromolecules.

The oral delivery of anti-inflammatory drugs has been a promising strategy for enhancing the clinical efficacy of ulcerative colitis (UC) treatment strategies. However, achieving site specific drug delivery to colon tissues and target cells is a challenging task for formulation scientists. In this study, macrophages-targeted liposome-loaded pectin-chitosan hydrogels were developed for UC treatment via oral administration. Folate-functionalized cholesterol was synthesized as lipid membrane materials for the liposomes containing curcumin (CUR). The incorporation of the liposomal CUR within pectin-chitosan hydrogels resulted in a matrix that exhibited considerable sensitivity to colonic enzymes during in vitro release. The targeted delivery of hybrids was able to effectively reach macrophages. They also showed enhanced capability to downregulate TNF-α, IL-6, and IL-1β in the lipopolysaccharide-induced Raw 264.7 cells model. DSS-induced mice model showed improved anti-UC effects, including accelerated mucosal repair and decreased inflammation and modulate the immune balance in the intestinal tissue of mice with colitis, which may be attributable to increased drug accumulation in the colonic lumen and improved internalization to target cells. Therefore, the incorporation of folate-modified liposomes containing CUR and pectin-chitosan physical hydrogels could potentially serve as a favorable approach for treating UC through an oral colon-targeted drug delivery system.
