Tryptophan (Trp) is one of the essential amino acids, which plays important roles in biological systems and the normal growth of human beings, and it is of great significance to be able to detect Trp in a rapid, efficient, and sensitive way. Herein, a 3D network metal–organic framework ([Sm2(BTEC)1.5(H2O)8]·6H2O) with excellent thermal and water stability was synthesized by a hydrothermal method. Interestingly, it could discriminate Trp from other natural amino acids in aqueous solution through a significant fluorescence enhancement effect, and showed high detection sensitivity (LOD = 330 nM) and outstanding anti-interference ability. The sensor system was successfully applied to the detection of Trp in practical samples, so it was expected to be a sensitive and efficient Trp sensor. In addition, the sensing mechanism was explained in detail by a series of characterization methods combined with density functional theory (DFT). There were many coordination water molecules in the crystal structure of the complex. Based on the small steric hindrance and molecular structure of water molecules, it provided the possibility for coordination interaction between Trp and Sm3+. On the other hand, the triplet energy level (T1) of Trp matched with the 4G5/2 vibrational energy level of Sm3+, so Trp could be used as the second “antenna molecule” besides 1,2,4,5-benzenetetracarboxylic acid (H4BTEC). Therefore, it effectively broadened the way for Sm-MOF to absorb excitation light.

Perovskite defect passivation with molecule doping shows great potential in boosting the efficiency and stability of perovskite solar cells (PSCs). Herein, an efficient and low-cost bifunctional Lewis base additive d-tryptophan is introduced to control the crystallization and growth of perovskite grains and passivation defects. It is found that the additive doped in the solution precursors could retard crystal growth by increasing activation energy, resulting in improved crystallization of large grains with reduced grain boundaries, as well as inhibiting ion migration and PbI2 aggregation. As a result, the PSCs incorporated with d-tryptophan additives achieve an improved power conversion efficiency from 18.18 to 21.55%. Moreover, the d-tryptophan passivation agent improves the device stability, which retains 86.85% of its initial efficiency under ambient conditions at room temperature after 500 h. This work provides Lewis base small-molecule d-tryptophan for efficient defect passivation of the grain boundaries toward efficient and stable PSCs.

Dioxygen activation and catalysis around ambient temperature is a long-standing challenge in chemistry. Inspired by the significant roles of the hydrogen bond network in dioxygen activation and catalysis by redox enzymes, this work presents a Lewis acid improved dioxygen activation by an FeII(BPMEN)(OTf)2 complex towards tryptophan 2,3-dioxygenase (TDO) activity for 3-methylindole and common olefinic CC bond oxygenation and cleavage (enzymatic Brønsted acid vs. chemical Lewis acid). It was found that the presence of a Lewis acid such as Sc3+ could substantially improve olefinic CC bond oxygenation and cleavage activity through FeII(BPMEN)(OTf)2 catalyzed dioxygen activation. Notably, a more negative ρ value in the Hammett plot of para-substituted styrene oxygenations was observed in the presence of a stronger Lewis acid, disclosing the enhanced electrophilic oxygenation capability of the putative iron(III) superoxo species through its electrostatic interaction with a stronger Lewis acid. Thereof, this work has demonstrated a new strategy in catalyst design for dioxygen activation and catalysis for olefin oxygenation, a significant process in the chemical industry.

The challenge of heavy biofouling in complex sweat environments limits the potential of electrochemical sweat sensors for noninvasive physiological assessment. In this study, a novel semi-interpenetrating hydrogel of PSBMA/PEDOT:PSS was engineered by interlacing PEDOT:PSS conductive polymer with zwitterionic PSBMA network. This versatile hydrogel served as the foundation for developing an anti-fouling wearable molecular imprinting sensor capable of sensitive and robust detection of tryptophan (Trp) in complex sweat. The incorporation of PEDOT:PSS conductive polymer into the semi-interpenetrating hydrogel introduced diverse physical crosslinks, including hydrogen bonding , electrostatic interactions , and chain entanglement. This incorporation considerably boosted the hydrogel's mechanical robustness and imparted commendable self-healing property. At the same time, the synergistic coupling between the well-balanced charge of the zwitterionic network and the high conductivity of the PEDOT:PSS polymer facilitated efficient charge transfer. The formation of the desired molecular imprinting membrane of semi-interpenetrating hydrogel was triggered by self-polymerization of dopamine (DA) in the presence of Trp. The designed biosensor demonstrated good sensitivity, selectivity and stability in detecting the target Trp. Notably, it also exhibited exceptional anti-fouling abilities, allowing for accurate Trp detection in complex real sweat samples, yielding results comparable to commercial enzyme-linked immunoassay (ELISA).

Herein, blue-emitting gold nanoclusters (Au NCs) were carried out through tryptophan as the protecting and reducing agents. In aqueous solution of Au NCs@tryptophan, the addition of furaltadone guaranteed the interaction of furaltadone with tryptophan around Au NCs. The propinquity of furaltadone to Au NCs caused that the fluorescence of Au NCs was weakened by furaltadone based on the inner filter effect (IFE). Under the optimal measurement conditions, the logarithm of relative fluorescence intensity of Au NCs@tryptophan was linearly carried out with the furaltadone amount increasing from 0.5 to 100 μM, the corresponding detection limit was 0.087 μM. The fluorescence change of Au NCs@tryptophan displayed excellent selectivity and sensitivity for furaltadone than other possible substance in the human body. In view of Au NCs@tryptophan, the as-performed fluorescence nanosensor suggested outstanding ability for furaltadone sensing in real samples. Obviously, this nanoprobe of furaltadone could implement the naked-eye visual fluorescence determination of furaltadone.

Amino acids not only play a vital role in the synthesis of biological molecules such as proteins in cancer malignant cells, they are also essential metabolites for immune cell activation and antitumor effects in the tumor microenvironment. The abnormal changes in amino acid metabolism are closely related to the occurrence and development of tumors and immunity. Intestinal microorganisms play an essential role in amino acid metabolism, and tryptophan and its intestinal microbial metabolites are typical representatives. However, it is known that the cyclic amino acid profile is affected by specific cancer types, so relevant studies mainly focus on one type of cancer and rarely study different cancer forms at the same time. The objective of this study was to examine the PFAA profile of five cancer patients and the characteristics of tryptophan intestinal microbial metabolites to determine whether there are general amino acid changes across tumors. Plasma samples were collected from esophageal ( n = 53), lung ( n = 73), colorectal ( n = 94), gastric ( n = 55), breast cancer ( n = 25), and healthy control (HC) ( n = 139) subjects. PFAA profile and tryptophan metabolites were measured, and their perioperative changes were examined using high-performance liquid chromatography. Univariate analysis revealed significant differences between cancer patients and HC. Furthermore, multivariate analysis discriminated cancer patients from HC. Regression diagnosis models were established for each cancer group using differential amino acids from univariate analysis. Receiver-operating characteristic analysis was applied to evaluate these diagnosis models. Finally, GABA, arginine, tryptophan, taurine, glutamic acid, and melatonin showed common alterations across all types of cancer patients. Metabolic pathway analysis shows that the most significant enrichment pathways were tryptophan, arginine, and proline metabolism. This study provides evidence that common alterations of the metabolites mentioned above suggest their role in the pathogenesis of each cancer patient. It was suggested that multivariate models based on PFAA profiles and tryptophan metabolites might be applicable in the screening of cancer patients.

Background Danshen Ligustrazine Injection (DSLI), composed of the extract of Salvia miltiorrhiza Bunge and Conioselinum anthriscoides 'Chuanxiong', is commonly used in the clinical treatment of ischemic stroke, which is the most common type of cerebrovascular disease, affected by multiple factors. The brain-gut axis is a new type of influencing factor for ischemic stroke, however, there is a lack of systematic research on the relationship between microbiota and host metabolic phenotype. This study aimed to investigate the host metabolic phenotype-microbiome interface in ischemic stroke and the immunomodulatory effects of DSLI. Methods In the study, liquid chromatography-mass spectrometry was used to primarily analyze the quality markers (Q-markers) and the polyphenols of DSLI. Based on 16S rRNA gene sequencing and metabolomics, the variations of microbial composition in different groups were characterized. In addition, a network diagram was constructed to reveal the interrelationship between DSLI-regulated gut microbiota changes and DSLI-regulated brain metabolites in MCAO/R rats. The expression of the aryl hydrocarbon receptor (AhR) in the colon and brain by immunofluorescence and suppressors of cytokine signaling 2 (SOCS2) by western blot were further detected. Results Diminished metabolic diversity and alterations in the tryptophan metabolism pathway were a feature of IS in the study. The results revealed that Clostri_UCG-014 was positively correlated with indoleacrylic acid, indole-3-methylacetate, and Ruminococcaceae was strongly related with L-tryptophan, 3-methylindole, and indole. The results of immunofluorescence showed that the expression of AhR in the colon and brain tissues of rats in the model group was greatly reduced compared with that in the sham-operated group, and was heavily activated with the treatment of DSLI. The levels of aryl hydrocarbon nuclear receptor ARNT2 and SOCS2 were down-regulated in the brains of Model rats, whereas increased by DSLI, further confirming that the brain-gut microbiota-Trp metabolism exists in the pathological changes of stroke and the intervention mechanism of DSLI. Conclusions The present study findings suggest Clostri_UCG-014, Ruminococcaceae, and Firmicute_RF39 play an important role in improving the regulation of immune function in stroke through tryptophan metabolites by acting on the AhR-SOCS2 axis.

A simple chiral electrochemical sensor based on N-carboxymethyl chitosan (NCCS) covalently bound with ethylenediamine-carboxylic multiwalled carbon nanotubes (MWCNTs-EA) was developed for enantioselective recognition of tryptophan (Trp) enantiomers; NCCS preferably combined with L-Trp than with D-Trp. The recognition efficiency prominently increased after the introduction of N-carboxymethyl into chitosan (CS). The carboxymethyl and imino groups of NCCS play an important role in enantioselective discrimination system. Moreover, the parameters of enantioselective recognition such as pH, temperature and response time were investigated. The electrochemical recognition of amino acid enantiomers with different functional groups was carried out. The chiral recognition mechanism was investigated by quantum computing. The results were confirmed by quantum chemical calculations.

In this work, a glassy carbon electrode modified with poly-sulfosalicylic acid (PSA/GCE) was prepared using electropolymerization method and applied for the determination of l -tryptophan ( l -Trp) in the presence of ascorbic acid and dopamine. The morphologies and interface properties of PSA film were examined by scanning electron microscopy and electrochemical impedance spectroscopy . The electrocatalytic oxidation of l -Trp on the PSA/GCE from potentially interfering species, e.g. ascorbic acid (AA) and dopamine (DA) was investigated using cyclic voltammetry (CV) and differential pulse voltammetry (DPV). Under optimum conditions, the proposed method exhibited wide linear dynamic range of 5 × 10 −8 to 1 × 10 −5 M with a detection limit (S/N = 3) of 6.8 × 10 −9 M. Moreover, the modified electrode exhibited good reproducibility and high selectivity, demonstrating its feasibility for the analytical purpose.

Tryptophan is a potential biomarker for a variety of diseases, so rapid and accurate detection of tryptophan levels in the human body is essential. In this work, surface-enhanced Raman scattering (SERS) detection was combined with acid-catalyzed silicon-based molecularly imprinted polymers to synthesize Cu 2 O@Ag-MIPs (CA-MIPs) for selective detection of L-tryptophan(L-Trp). Compared with traditional detection methods, SERS detection has the advantages of being sensitive, fast and nondestructive. Here, Ag nanoparticles are uniformly loaded on {100} surface-dominated Cu 2 O sub-microcubes to excite strong localized surface plasmon resonance, which exhibits good SERS performance. Considering the complexity of practical detection, an acid-catalyzed method was adopted to load silicon-based imprinted polymers with high biocompatibility on the Cu 2 O@Ag surface, which improved the drawbacks of the common silicon-based imprinting method, such as loose structure and long reaction time. The thickness of the imprinted layer can be controlled to ensure selectivity while ensuring superior SERS sensitivity. The prepared CA-MIPs were shown good linearity with L-Trp concentration in the range of 10 −3 –10 −10 mol L −1 , and the sensor also exhibited excellent selectivity for the determination of L-Trp in the presence of other interfering amino acids. In addition, the method was shown good recoveries in spiked recovery tests of simulated urine samples and was expected to be practically applied to the detection of clinical disease markers.

Background: Curcumin (Cur) and tryptophan (Trp) both show promise for treating ulcerative colitis (UC) alone, but their combination has not been explored. This study investigated the therapeutic advantage of the combination (Cur–Trp) for DSS-induced ulcerative colitis in mice. Methods: We established a mouse model of ulcerative colitis induced by dextran sulfate sodium (DSS). The mice were treated with Cur, Trp, or Cur–Trp, and to evaluate the therapeutic effects, we assessed clinical signs such as body weight, disease activity index (DAI), and colon length. We also examined intestinal barrier function through indicators including histopathological changes, inflammatory factors, oxidative stress levels, mucin secretion, and tight junction protein expression. Additionally, we analyzed the composition of gut microbiota and the content of its metabolites like short-chain fatty acids (SCFAs).Results: The Cur–Trp group produced the most significant improvement, exceeding that of Cur or Trp group. This was evidenced by a significant recovery of this sign, including slower weight loss, reduced colon shortening, and de-creased disease activity index. Compared with the model group, the weight loss of mice in the Cur–Trp group was reduced from 17.15% to 9.73%, which was better than that in the cur group (11.33%) and the Trp group (11.59%). The DAI decreased from the model group (3.6) to the Cur–Trp group (2.4), while the DAI in the Cur group and the Trp group only decreased to 2.9 and 2.8, respectively. The colon length in the Cur–Trp group (6.52 cm) was larger than that in the cur group (6.31 cm), the Trp group (6.23 cm) and the model group (5.5 cm). The Cur–Trp intervention effectively restored intestinal barrier function, as shown by reducing colon tissue dam-age, modulating inflammatory factors, restoring oxidative balance, increasing mucin secretion, and upregulating tight junction protein expression. Further studies showed that the combination uniquely modulated the gut microbiome, increased the Firmicutes/Bacteroidetes (F/B) ratio, decreased the genus of pro-inflammatory bacteria, and in-creased beneficial bacteria, while increasing SCFA levels to alleviate DSS-induced ulcerative colitis. Conclusions: Cur–Trp has shown great potential in alleviating colitis and promoting intestinal barrier function, suggesting that the combination of Cur and Trp has the potential to be developed as a therapeutic functional food or dietary supplement for UC. However, more studies are needed to validate this finding. Future research should focus on elucidating the precise molecular mechanisms, optimizing dosage and clinical trials in chronic models and humans to provide more targeted treatment options.

( 3 ) (C 18 H 16 N 3 O 2 =2-acetylpyridine- d -tryptophan) were synthesized and characterized by elemental analysis, IR, UV, 1 H NMR and X-ray diffraction single crystal analysis. The analyses of the structures indicate that all three materials crystallize in the tetragonal crystal system, space group P 4 1 2 1 2. They have similar structures; i.e., the Schiff base coordinates in its deprotonated form and behaves as a hexadentate (4N+2O) coordinated ligand to form a distorted octahedron geometry. On the other hand, as a result of the alternate arrangement of chains through N–H···O intermolecular hydrogen bonds interactions, 2-D layers are formed for the three complexes. Furthermore, the luminescent properties and thermal stabilities of the three complexes were investigated.
