Ionic liquids (ILs) which synthesized from bio-renewable materials have recently attracted much attention for their applications in biocatalysis. Ethyl ( R )-3-hydroxybutyrate (( R )-EHB) as a versatile chiral intermediate is of great interest in pharmaceutical synthesis. This study focuses on evaluating the performances of choline chloride (ChCl)-based and tetramethylammonium (TMA)-based neoteric ILs in the efficient synthesis of ( R )-EHB via the bioreduction of ethyl acetoacetate (EAA) at high substrate loading by recombinant Escherichia coli cells. It was found that choline chloride/glutathione (ChCl/GSH, molar ratio 1:1) and tetramethylammonium/cysteine ([TMA][Cys], molar ratio 1:1) as eco-friendly ILs not only enhanced the solubility of water-insoluble EAA in the aqueous buffer system, but also appropriately improved the membrane permeability of recombinant E. coli cells, thus boosting catalytic reduction efficiency of EAA to ( R )-EHB. In the developed ChCl/GSH- or [TMA][Cys]-buffer systems, the space-time yields of ( R )-EHB achieved 754.9 g/L/d and 726.3 g/L/d, respectively, which are much higher than neat aqueous buffer system (537.2 g/L/d space-time yield). Meanwhile, positive results have also been demonstrated in the bioreduction of other prochiral ketones in the established IL-buffer systems. This work exhibits an efficient bioprocess for ( R )-EHB synthesis under 325 g/L (2.5 M) substrate loading, and provides promising ChCl/GSH- and [TMA][Cys]-buffer systems employed in the biocatalysis for hydrophobic substrate.

Using natural-based lipids to construct biocompatible, controllable and efficient nanocarriers and elucidating their structure–function relationships, was regarded as an important area for creating sustainable biomaterials. Herein, we utilized two natural steroids: cholesterol and diosgenin (bearing different hydrophobic tails) as the building blocks, to synthesize a series of natural steroid-based cationic random copolymers PMA6Chol-r-PDMAEMA and PMA6Dios-r-PDMAEMA via RAFT polymerization. The results demonstrated that the steroid-r-PDMAEMA copolymers could efficiently bind pDNA (N/P < 3.0) and then form near-spherical shape (142–449 nm) and positively-charged (+11.5 to +19.6 mV) nanoparticles. The in vitro cytotoxicity and gene transfection efficiency greatly depend on the steroid hydrophobic tail structures and steroid/PDMAEMA block ratios. Optimum transfection efficiency of the (Chol-P1/pDNA and Dios-P3/pDNA) nanoplexes could reach to 18.1–31.2% of the PEI-25K/pDNA complex. Moreover, all of the steroid-r-PDMAEMA/Cy3-pDNA nanoplexes have an obvious “lysosome localization” effect, indicating the steroid structures do not remarkably influence the intracellular localization behaviors of these nanoplexes.

Blood–brain barrier (BBB) dysfunction is a well-established pathological phenotype of ischemic stroke, and targeting BBB integrity has emerged as a promising therapeutic strategy. Danshen-Chuanxiong formula (DS-CX), an effective herbal combination against ischemic stroke, has demonstrated regulatory effects on the BBB at various stages of ischemic stroke. However, its specific BBB-protective components and underlying molecular mechanisms remain unclear. Recent advances in multicellular self-assembled BBB spheroids have shown distinct advantages in disease modeling and drug discovery, offering a novel approach to address these questions. To simulate ischemic stroke-induced BBB dysfunction, we developed an oxygen–glucose deprivation/reoxygenation (OGD/R)-induced BBB disruption model using multicellular spheroids. To identify the effective substances of DS-CX responsible for BBB protection, we conducted a multi-parametric evaluation to assess BBB permeability, tight junctions, cell viability, reactive oxygen species (ROS) levels, inflammatory markers, and apoptotic phenotypes using high-content imaging. Further immunofluorescence and transcription analyses were performed to elucidate the BBB-protective mechanisms of DS-CX and its active components. Similar to the overall effects of DS-CX on BBB protection, preliminary screening fortunately found that both protocatechuic acid, ferulic acid, and senkyunolide I significantly reduced OGD/R-induced leakage, and upregulated the protein and mRNA levels of ZO-1 and Claudin-5 in BBB spheroids. Further multi-phenotypic assessments manifested that DS-CX and its active compounds effectively improved cell survival, reduced ROS production, inhibited inflammation, and decreased apoptosis, compared to the damaged BBB spheroids without drug intervention. Molecular experiments showed that DS-CX and its active constituents not only rescued the abnormal protein levels of pivotal targets related to oxidative stress (HO-1), inflammation (MMP-9, TLR-4), and apoptosis (Caspase-3, Bax, Bcl-2) in OGD/R-treated BBB spheroids, but also normalized the dysregulated mRNA levels of vWF, HO-1, MMP-9, TLR-4, TNF-α, IL-6, IL-1β, and IL-18 caused by OGD/R stimulation. Collectively, the present work successfully identified protocatechuic acid, ferulic acid, and senkyunolide I as key BBB-protective components of DS-CX against ischemic stroke. These compounds likely exert their therapeutic effects through multi-target regulation of oxidative stress, inflammation, and apoptosis. Our findings provide a novel spheroid-based multi-parametric screening approach for discovering BBB-targeted therapies in ischemic stroke. Graphical abstract

Presently, a combination of chemotherapy, radiotherapy, thermotherapy, and other treatments has become a hot topic of research for the treatment of cancer, especially lung cancer. In this study, novel hollow gold nanoparticles (HGNPs) were used as drug carriers, and in order to improve the targeting ability of HGNPs to a lung tumor site, polyoxyethylene sorbitol oleate (PSO) was chosen here as a target ligand since it can be specifically recognized by the low-density lipoprotein (LDL) receptor which is usually over expressed on A549 lung cancer cells. In this way, a PSO-modified doxorubicin-loaded HGNP drug delivery system (PSO-HGNPs-DOX) was constructed and its physicochemical properties, photothermal conversion ability, and drug release of PSO-HGNPs-DOX was investigated. Further, the effects of triple combination therapy, the intracellular uptake, and the ability to escape macrophage phagocytosis of PSO-HGNPs-DOX were also studied using A549 cells in vitro. In addition, an in vivo mouse model was also used to study the targeting of PSO-HGNPs-DOX to lung cancer. PSO-HGNPs-DOX demonstrated a good triple therapeutic effect for lung cancer (A549 cell viability was only 10% at 500 μM) by LDL receptor mediated endocytosis and was able to escape macrophage phagocytosis to enhance its accumulation at the target site. Therefore, PSO-HGNPs-DOX is a novel, safe, promising, and targeted drug carrier designed for triple combination lung cancer therapy which should be further studied for such applications.

R -/ S -2-(2-hydroxypropanamido) benzoic acid ( R -/ S -HPABA), marine-derived anti-inflammatory antiplatelet drugs, were initially synthesised in our group. However, preliminary research showed that R -/ S -HPABA were eliminated rapidly because of extensive hydroxylation metabolism of phenyl ring in vivo. In order to reduce significant hydroxylation metabolism to improve pharmacological activity and bioavailability, trifluoromethyl group was incorporated into R -/ S -HPABA to synthesise R -/ S -2-(2-hydroxypropanamido)-5-trifluoromethyl benzoic acid ( R -/ S -HFBA), respectively. The purposes of this study were to report the synthesis of R -/ S -HFBA and compare the anti-inflammatory antiplatelet effect and pharmacokinetic properties of R -/ S -HFBA with those of R -/ S -HPABA. Carrageenan-induced rat paw edema assay was used for the evaluation of the anti-inflammatory activity. R -/ S -HFBA showed better results in inhibiting edema and were able to prolong the anti-inflammatory effect after carrageenan injection. The antiplatelet aggregation activity of R -/ S -HFBA and R -/ S -HPABA was studied on arachidonic acid-induced platelet aggregation of rabbit platelet-rich plasma. The aggregation inhibition rate of R- / S -HFBA was significantly ( p < 0.05) higher than that of R- / S -HPABA, respectively. Molecular docking study revealed that R -/ S -HFBA possess more potent binding affinity with COX-1/COX-2 than R -/ S -HPABA, respectively, and that the presence of trifluoromethyl group leads to increase in activity of R -/ S -HFBA. R -/ S -HFBA also afford more favorable pharmacokinetic properties than R- / S -HPABA, respectively, such as higher C max , larger AUC 0-∞ , and longer t 1/2 , which, as expected, are more metabolically stable.

In this study, a novel titanium/zirconium‑tin oxide/alpha‑lead dioxide/beta‑lead dioxide (Ti/Zr-SnO 2 /α-PbO 2 /β-PbO 2 ) electrode was prepared and utilized for degrading alizarin yellow R (AYR) dye. The Integration of impregnation drying, drop coating with a solvent, and high-temperature pyrolysis significantly diminished the characteristic fissure morphology of conventional pyrolytic metal oxide interlayers, thereby modulating the electrodeposition process of alpha‑lead dioxide (α-PbO 2 ) and enhancing electrode stability and electrochemical performance. Electrode characterization revealed that the charge transfer resistance ( R ct ) was 88.0 Ω/cm 2 , and the oxygen evolution potential (OEP, vs. Ag/AgCl) was 2.02 V. The electrochemically active surface area was the largest, with an accelerated lifetime of up to 82.37 h. That was 13.18 times longer than that of the Ti/α-PbO 2 /β-PbO 2 electrode (6.25 h) and 8.83 times longer than that of the Ti/Zr-SnO 2 /β-PbO 2 electrode (9.33 h). Under optimal experimental conditions (400 A/m 2 , 50 mg/L, 20 °C, pH = 8.00), the removal rate of AYR by this electrode was 99.90 %, the chemical oxygen demand (COD) removal rate was 74.64 %, the average current efficiency (ACE) was 50.94 %, and the electric energy per order ( E EO ) was 41.17 kWh/m 3 . The AYR degradation process followed to a quasi-zero-order kinetic model ( k = 0.2745 min −1 ). In the real water treatment, AYR and COD removal rates remained above 90.13 % and 62.57 %, respectively. This study demonstrated that the Ti/Zr-SnO 2 /α-PbO 2 /β-PbO 2 electrode significantly improved electrocatalytic performance and stability through the unique preparation of its interlayer, exhibiting great potential for efficient dye wastewater treatment.

Lipid nanoparticles (LNPs) hold great promise as carriers for developing drug delivery systems (DDSs) aimed at managing ischemic stroke (IS). Previous research has highlighted the vital role played by the lipid composition and biophysical characteristics of LNPs, influencing their interactions with cells and tissues. This understanding presents an opportunity to engineer LNPs tailored specifically for enhanced IS treatment. We previously introduced the innovative concept of reconstituted lipid nanoparticles (rLNPs), which not only retain the advantages of conventional LNPs but also incorporate lipids from the originating cell or tissue. Brain-derived rLNPs (B-rLNPs) exhibit significantly superior accumulation within the cerebral ischemic region when compared to liver-derived rLNPs (L-rLNPs). The homing effect of B-rLNPs was then employed to construct 3-n-butylphthalide (NBP) loaded DDS (B-rLNPs/NBP) for the treatment of IS. Our results demonstrated that compared with free NBP, B-rLNPs/NBP can significantly reduce infarct volume, neurological deficits, blood-brain barrier (BBB) leakage rate, brain water content, neutrophil infiltration, alleviate pathological structures, and improve the motor function in MCAO/R model. We also proved that B-rLNPs/NBP showed further reinforced protective effects on the same model than free NBP through the regulation of TLR4/MyD88/NF-κB (anti-inflammation) and Bax/Bcl-2 (anti-apoptosis) pathways. This study offers a promising tool towards improved IS treatment.

Printed circuit boards (PCBs) serve as fundamental building blocks of modern electronics, and the quality of copper electroplating in microvias is crucial to the reliability and performance of high-density interconnect products. Achieving void-free bottom-up filling of microvias remains a key technical challenge due to the nonuniform current distribution and mass transport constraints inherent in electroplating. In this work, Bismarck Brown R (BBR), an aromatic azo-containing organic molecule, is introduced as a leveler for acidic sulfate copper electroplating. The molecular structure, adsorption mechanism, electrochemical behavior, and synergistic effects of BBR are systematically elucidated using DFT calculations, MD simulations, electrochemical measurements, and in situ FTIR spectroscopy. DFT and MD results reveal that BBR adsorbs strongly onto the copper surface in a planar configuration via combined physical and chemical interactions with an adsorption energy of −693.67 kcal/mol. Electrochemical analysis demonstrates that BBR alone accelerates copper deposition at the bottom and inhibits it at the mouth of the microvias. More importantly, BBR synergistically enhances the inhibiting effect of EO-PO and the accelerating effect of SPS, with such synergy being more pronounced at the microvia mouth. In situ FTIR confirms that both EO-PO and SPS promote BBR adsorption on the copper surface. The ternary additive system (EO-PO + SPS + BBR) achieves a positive relative charge difference (ΔQ = 6.95%), indicative of a superior bottom-up filling performance. Electroplating experiments validate that void-free, dense copper filling of microvias is achieved exclusively in the presence of BBR. Furthermore, BBR modulates the preferred crystallographic orientation of the copper coating from (200) to (111). This work provides a comprehensive molecular-level understanding of BBR as a high-performance additive and offers an effective strategy for void-free microvia filling in advanced PCB manufacturing.

Methane valorization is important for climate change mitigation and sustainable carbon utilization, but conventional nonoxidative coupling of methane (NOCM) suffers from the high activation barrier of CH 4 , limited product selectivity, and rapid coking-induced deactivation. Herein, we develop a spark discharge−dielectric barrier discharge (DBD) coupled plasma-catalytic system for the selective conversion of high-concentration methane to ethylene. Spark discharge first activates CH 4 and preferentially converts it to C 2 H 2 , while a downstream catalyst bed, maintained at 100−130 ° C by residual discharge heat, enables acetylene semihydrogenation to C 2 H 4 . A coupled DBD unit is further introduced for in situ carbon removal and catalyst regeneration. Among the tested catalysts, impregnation−reduction-derived 0.1Pd-5Cu/γ-Al 2 O 3 -R with atomically dispersed Pd exhibited the best performance, achieving 72.38% CH 4 conversion, 72.19% C 2 H 4 selectivity, 92.48% acetylene-based C 2 H 4 selectivity, and a C 2 H 4 space-time yield of 69.97 mmol·g· −1 ·h −1 at 36.2 kJ·L −1 . With DBD-assisted regeneration, efficient NOCM operation was maintained for 630 min. Mechanistic analyses associate the enhanced ethylene selectivity with highly dispersed Pd sites and their electronic modulation by predominantly low-valent Cu species, which suppress overhydrogenation and facilitate ethylene desorption.

To simultaneously accelerate both sluggish hydrogen evolution reaction (HER) and urea oxidation reaction (UOR) kinetics, strongly coupling of transition metal hydroxides of high UOR activity with the transition metals of low hydrogen adsorption barrier for highly active HER is a promising strategy. Herein, a tractable hydrothermal protocol and a partial in-situ topological reduction strategy are developed to synthesize hcp Ni/r-Ni(OH) 2 nanosheets containing abundant oxygen defects with an ultrathin carbon layer, which are utilized as the new bifunctional catalysts for both HER and UOR. As for HER, the surface electronic structure of Ni is optimized by introducing the oxygen defects coupling with hcp Ni with excellent electronic structure, which greatly enhances the dissociation ability of H 2 O and reduces the Ni-H bond strength , thus acquiring the optimized H* Gibbs free energy (∆G H *). As for UOR, abundant interfaces and the electron transfer between transition metal hydroxides/transition metal are favorable to regulate the urea adsorption. As expected, the hcp Ni/r-Ni(OH) 2 /C electrocatalyst shows good activity for HER (70.7 mV @ 10 mA cm −2 ), UOR (1.36 V @ 10 mA cm −2 ), and urea electrolysis (1.45 V @ 10 mA cm −2 ). This facile strategy provides a novel way to design high-performance electrocatalysts towards urea splitting, which can utilize to generate hydrogen as well as eliminate urea in water.

In this work, the solubility of unsolvated crystalline (R)-equol in six (alcohols + n -heptane) binary solvents and that of solvated crystalline (R)-equol in methanol along with ethanol were firstly systematically measured and analyzed. The experimental results revealed that the solubility of unsolvated crystalline (R)-equol was positively correlated with temperature and mass fraction of alcohol. And the solubility order was 2-propanol > 1-propanol > 1-butanol > 1-pentanol > 3-methyl-1-butanol > 2-methyl-1-propanol > n -heptane. The transformation temperatures between unsolvated and solvated forms were acquired at 316.28 K and 310.50 K, respectively. Meanwhile, the experimental solubility data of unsolvated crystalline (R)-equol were correlated using modified Apelblat, λh , quartic polynomial and CNIBS/R-K equations, in which the modified Apelblat equation achieved the best regression performance. Besides, the dissolution of unsolvated crystalline (R)-equol was endothermic. Furthermore, solvent parameters, solvation free energy and molecular electrostatic potential surface obtained through molecular simulation were used to explain the experimental phenomena, and found the polarity of solvents, van der Waals and hydrogen bonding interactions made different contributions to various dissolution processes.
