In this study, we synthesized an amphiphilic α-cyanostilbene derivative ( E -form)-1-(12-(2-(2-(benzo[d]thiazol-2-yl)-2-cyanovinyl)-5-(diethylamino)phenoxy)dodecyl)pyridin-1-ium bromide (denoted by BCPB). As being dissolved in aqueous solution, BCPB shows strong emission of excimers at around 660 nm due to the formation of aggregates. Addition of different amount of CB[7] into the solution leads to the emission shift from 660 nm, firstly to 540 nm and then to 588 nm. The emission shift is attributed to the two-step assembly driven by the concentration of CB[7], and the molar ratios of CB[7]/BCPB (denoted by R m ) play a critical role for the shift. As R m ≤ 4, CB[7] mainly interacts with the cationic pyridinium group of BCPB to form BCPB@CB[7] complex. As R m > 4, another CB[7] binds to the diethylamino group at one end of α-cyanostilbene to form BCPB@CB[7] 2 complex. The two-step assembly well explains the emission shift of BCPB.

Pr-doped CeO2with different doping levels was prepared from Ce(NO3)3∙6H2O and Pr(NO3)3∙6H2O by solvothermal method without any additional reagents, in which the mixed solution of ethylene glycol and distilled water was employed as a solvent. The influences of Pr-doping on phase composition, crystal structure and morphology were investigated, as well as Pr valence and oxygen vacancy defects. The Pr cations entered into the CeO2crystal lattice with normal trivalence and formed a Pr-CeO2solid solution based on the fluorite structure. The larger trivalent Pr was substituted for tetravalent Ce in the CeO2crystal and compensated by oxygen vacancy defects, which caused the local lattice expansion of the crystal lattice. Moreover, the Pr-doped CeO2solid solutions exhibited visible color variation from bright cream via brick red to dark brown with the increasing of Pr contents. The degradation of AO7 dye was also investigated using a domestic medical ultraviolet lamp; the removal efficiency of AO7 by 1% and 2% Pr-doped CeO2approached 100%, much higher than 66.2% for undoped CeO2.Keywords:CeO2;Pr-doping;photocatalysis;pollutant degradation;mesoporous structure

Background:
Micro- and nanoplastics (MNPs) and homosalate (HMS) are ubiquitous emerging environmental contaminants detected in human samples. Despite the well-established endocrine-disrupting effects (EDEs) of HMS, the interaction between MNPs and HMS and its impact on HMS-induced EDEs remain unclear.
Objectives:
This study aimed to investigate the influence of MNPs on HMS-induced estrogenic effects and elucidate the underlying mechanisms in vitro and in vivo.
Methods:
We assessed the impact of polystyrene nanospheres (PNSs; 50 nm, 1.0mg/L) on HMS-induced MCF-7 cell proliferation (HMS: 0.01–1μM, equivalent to 2.62–262μg/L) using the E-SCREEN assay and explored potential mechanisms through transcriptomics. Adult zebrafish were exposed to HMS (0.0262–262μg/L) with or without PNSs (50 nm, 1.0mg/L) for 21 d. EDEs were evaluated through gonadal histopathology, fertility tests, steroid hormone synthesis, and gene expression changes in the hypothalamus–pituitary–gonad–liver (HPGL) axis.
Results:
Coexposure of HMS and PNSs resulted in higher expression of estrogen receptor α (ESR1) and the mRNAs of target genes (pS2, AREG, and PGR), a greater estrogen-responsive element transactivation activity, and synergistic stimulation on MCF-7 cell proliferation. Knockdown of serum and glucocorticoid-regulated kinase 1 (SGK1) rescued the MCF-7 cell proliferation induced by PNSs alone or in combination with HMS. In zebrafish, coexposure showed higher expression of SGK1 and promoted ovary development but inhibited spermatogenesis. In addition, coexposure led to lower egg hatchability, higher embryonic mortality, and greater larval malformation. Coexposure also modulated steroid hormone synthesis genes (cyp17a2, hsd17β1, esr2b, vtg1, and vtg2), and resulted in higher 17β-estradiol (E2) release in females. Conversely, males showed lower testosterone, E2, and gene expressions of cyp11a1, cyp11a2, cyp17a1, cyp17a2, and hsd17β1.

The most common in vitro cell culture platform, standard two-dimensional (2D) monolayer cell culture, often fails to mimic the tumor microenvironment, while animal models complicate research on the effect of individual factors on cell behaviors. Both are unsatisfactory in the research of molecular mechanisms of tumor development and progression and the discovery and development of anticancer drugs. In vitro three-dimensional (3D) cell culture can partially simulate in vivo conditions and 3D-cultured cancer cells can recapture many essential features of native tumor tissues. In this study, to mimic the in vivo breast tumor microenvironment, novel reduction-responsive poly(Nε-acryloyl L-lysine) (pLysAAm) hydrogels were synthesized by rapid photo-polymerization of Nε-acryloyl L-lysine and using N,N′-bis(acryloyl)-(L)-cystine as a crosslinker, and their physicochemical properties were characterized systemically. The results showed that the pLysAAm hydrogels were formed within 93 s under UV irradiation and exhibited almost total elastic recovery from compressions as high as 75%. The lyophilized hydrogel samples displayed a highly porous structure with interconnected pores, had an equilibrium swelling ratio of about 20, and were degraded faster in a glutathione-containing solution than in PBS solution. The biological versatility of the pLysAAm hydrogels was demonstrated by both in vitro MCF-7 cell culture and in vivo tumor formation. Compared to cells cultured as 2D monolayers, the 3D-cultured cells presented 3D cell morphology, exhibited better cell viability, expressed higher levels of pro-angiogenic factors, and showed significantly greater migration and invasion abilities. The results from assay of tumorigenicity in nude mice and histologic analysis demonstrated the enhanced tumorigenic and angiogenic capabilities of the MCF-7 cells pre-cultured in pLysAAm hydrogels. These findings suggest that pLysAAm hydrogels may be used to bridge the gap between standard in vitro cell cultures and living tissues, aid breast cancer research, and help researchers to develop novel anticancer therapeutics.

Enzymatic approaches have become promising alternatives to chemical methods for the production of semi-synthetic β-lactam antibiotics. In this work, enzymatic synthesis of N-bromoacetyl-7-aminocephalosporanic acid (N-bromoacetyl-7-ACA), the key intermediate for the production of cefathiamidine, was reported for the first time.Of the immobilized penicillin acylases (PAs) tested, PGA-750 was the best biocatalyst. Optimization of the biocatalytic process was conducted. The optimal acyl donor, molar ratio of acyl donor to 7-ACA, pH, temperature, 7-ACA concentration, and enzyme dosage were methyl bromoacetate, 3, 7.5, 20 °C, 50 mmol/L and 4 U/mL, respectively. Under the optimal conditions, enzymatic N-acylation of 7-ACA with methyl bromoacetate afforded the desired product with the yield of 85% in 2 h, where the synthesis/hydrolysis (S/H) ratio was approximately 1.5. The immobilized enzyme PGA-750 exhibited good operational stability, and the relative yields of approximately 90% and 63% were achieved, respectively, when it was reused in 7th and 11th batch. An enzymatic approach to N-bromoacetyl-7-ACA, the key intermediate for the industrial production of cefathiamidine, has been developed successfully in a fully aqueous medium. The present work may open up a novel opportunity for the production of cefathiamidine through a simple and green process.

Purpose DOX is one of the most potent anticancer drugs. But its short half-life and the occurrence of multi-drug resistance (MDR) markedly limit its clinical application. To solve these problems, we develop DOX loaded polymersomes (DOX polymersomes). Methods An methoxy poly(ethylene glycol)-b-poly(epsilon-caprolactone) (mPEG-b-PCL) copolymer was synthesized and used to prepare DOX polymersomes. The pharmaceutical properties of DOX polymersomes were characterized. The in vitro release profile of DOX from polymersomes was investigated. The in vitro cytotoxicity and cell uptake studies were performed on MCF-7 and MCF-7/ADR cells. The in vivo pharmacokinetic profiles were investigated on Sprague–Dawley rats. Results DOX polymersomes had a nano-scale particle size of about 60 nm with a hydrophobic membrane about 10 nm in thickness. Release of DOX from the polymersomes took place in a sustained manner. Cell experiments showed DOX polymersomes enhanced the cytotoxicity and the intracellular accumulation of DOX in MCF-7/ADR cells, compared with free DOX. In vivo pharmacokinetic study showed the DOX polymersomes increased the bioavailability and prolonged the circulation time in rats. Conclusions The entrapment of DOX in biodegradable polymersomes could enhance cytotoxicity in MCF-7/ADR cells and improve its in vivo pharmacokinetic profile.

A facile one-pot 1,8-diazacyclo-[5,4,0]undec-7-ene (DBU) catalyzed transesterification/atom transfer radical polymerization (ATRP) strategy has been successfully developed through the combination of copper/DBU-catalyzed ATRP and DBU-catalyzed transesterification reactions. Well-defined poly(methacrylate)s with various side ester groups have been synthesized by ATRP and transesterification of acyl donor 2,2,2-trifluoethyl methacrylate with various alcohols, such as benzyl alcohol, n -butanol, iso -propyl alcohol, methanol, triethylene glycol monomethyl ether, propargyl alcohol, and 6-azido-1-hexanol by the one-pot process. Kinetic studies indicate that the ATRP process proceeded in a controllable manner without the interference of the transesterification reactions. Expansion of the binary system to a higher level ternary system has been successfully achieved by the combination of copper(I)-catalyzed azide–alkyne cycloaddition, transesterification, and ATRP reactions. © 2014 Wiley Periodicals, Inc. Multidrug resistance (MDR) has been a major obstacle to tumor chemotherapy. Pluronic unimers have been reported to be promising copolymers to reverse MDR, and the intracellular delivery of Pluronic unimers is a problem worth thinking. To exert the excellent reversal effect of Pluronic unimers, DOX-loaded G4.0 PAMAM was modified with PluronicL64 via cis-aconitic acid as a pH-sensitive linkage (PCPAMAM/DOX), which could release DOX and Pluronic unimers into cytoplasm. The Pluronic-modified PAMAM (PCPAMAM) exhibited favorable biocompatibility and pH-sensitivity. PCPAMAM/DOX showed a nano-scale size and a sustained in vitro release profile. Compared with a control formulation, PCPAMAM/DOX showed a higher reversal effect on MCF-7/ADR cells and enhanced intracellular drug accumulation. The results of P-gp activity, subcellular distribution of PluronicL64, the ATP level and mitochondrial transmembrane potential all illustrated that free Pluronic unimers could be released by PCPAMAM functioning as reversal agents. In conclusion, PCPAMAM could be a promising vehicle to enhance DOX accumulation by overcoming MDR in MCF-7/ADR cells. This work also provided an effective method to deliver Pluronic unimers into MDR cells.

A series of organic–inorganic hybrid materials were prepared by linking lanthanide (Eu 3+ , Sm 3+ ) complexes to mesoporous SBA-15/SBA-16 through 8-hydroxybenz[de]anthracen-7-one modified silane (HBA-Si) as linker. The physical characterizations of these hybrids revealed that they all have high surface area, uniformity in mesostructure. The luminescence properties of these covalently bonded materials (denoted as Ln(HBA-SBA-15) 3 phen and Ln(HBA-SBA-16) 3 phen) were compared with ternary complexes (Ln(HBA) 3 phen) (Ln = Eu, Sm). Eu(HBA-SBA-15(16)) 3 phen hybrids display better thermal stability, whose luminescent lifetimes and quantum efficiencies were matchable with those of Eu(HBA) 3 phen complex in spite of its much lower effective condensation of Eu 3+ species. In addition, the luminescent performance of functionalized SBA-15 hybrids was more favorable than that of functionalized SBA-16 hybrids, revealing that SBA-15 was a better host material for lanthanide complex than mesoporous silica SBA-16.

The nucleophilic substitution reaction between 4-chloro-7-nitro-2,1,3-benzoxadiazole (NBD-Cl) and –NH2 groups on amino-functionalized carbon dots (CDs) produces a novel carbon dot-based hybrid, CDs@NBD for short. In addition to the characteristic fluorescence emission of carbon dots at λex/λem = 360/443 nm, the CDs@NBD hybrid gives rise to new photoluminescence at λex/λem = 460/544 nm, which is significantly quenched by p-phenylenediamine (PPD) via static quenching. This provides a novel sensing approach for fluorometric detection of PPD with CDs@NBD as a probe. A linear calibration graph is obtained within two concentration ranges of 0.1–1.0 μmol L−1 and 1.0–10.0 μmol L−1, along with a detection limit of 56 nmol L−1 and a RSD of <3% (at 1.0 μmol L−1). The common coexisting species in environmental and biological sample matrices cause no obvious interferences on the detection of PPD. The colour change of the CDs@NBD hybrid solution with the variation in PPD concentration also facilitates visual detection of PPD, which further demonstrates promising applications in environmental and biological sample analysis.

CeO2has a potential application in the purification of organic dye wastewater because of the abundant oxygen vacancy (VO) defects in its crystals. In this study, a cubic CeO2microsphere with layered interleaved symmetrical 3D flower-like morphology was synthesized, and its adsorption capacity for acid orange 7 (AO7) was further enhanced by Y doping. The impact of varying amounts of Y ions on the phase composition, lattice parameters, and morphology of the product was investigated, revealing that 4 mol.% was determined as the doping level limit of Y ions in CeO2crystals. XPS, Raman, and H2−TPR techniques were employed to compare surface species changes before and after 4 mol.% Y doping in the CeO2crystals, including O−Ce(III), O−Ce(IV), O−Y(III), and VOcorrelation, yielding a rough quantitative assessment of these species. The 4 mol.% Y-doped CeO2(2.0 g/L) demonstrated the highest removal rate for 20 mg/L of AO7 dye within just 20 min to reach adsorption–desorption equilibrium, half the time required by undoped CeO2, achieving an impressive adsorption rate of 94.6%, compared to only 69.5% for undoped CeO2at 20 min. The adsorption capacity of undoped CeO2was enhanced by 19.05% through the doping of 4 mol.% Y, achieving a value of 16.56 mg/L. The feasibility of enhancing the adsorption capacity of CeO2by Y doping provides a reference for the application of CeO2and other metal oxides.

Citrus fruits have abundant flavonoid glycosides (FGs), an important class of natural functional and flavor components. However, there have been few reports about the modification of UDP-glycosyltransferases (UGTs) on flavonoids in citrus. Notably, in flavonoid biosynthesis, 7-O-glucosylation is the initial and essential step of glycosylation prior to the synthesis of flavanone disaccharides, the most abundant and iconic FGs in citrus fruits. Here, based on the accumulation of FGs observed at the very early fruit development stage of two pummelo varieties, we screened six novel flavonoid 7-O-glucosyltransferase genes (7GlcTs) via transcriptomic analysis and then characterized them in vitro. The results revealed that four Cg7GlcTs possess wide catalytic activities towards various flavonoid substrates, with CgUGT89AK1 exhibiting the highest catalytic efficiency. Transient overexpression of CgUGT90A31 and CgUGT89AK1 led to increases in FG synthesis in pummelo leaves. Interestingly, these two genes had conserved sequences and consistent functions across different germplasms. Moreover, CitUGT89AK1 was found to play a role in the response of citrus to Huanglongbing infection by promoting FG production. The findings improve our understanding of flavonoid 7-O-glucosylation by identifying the key genes, and may help improve the benefits of flavonoid biosynthesis for plants and humans in the future.
