Fluorescent nanodiamond (FND) has received increasing research interests for bioimaging and drug delivery owing its outstanding physicochemical properties and low toxicity. However, synthesis of FND in large scale with desirable optical property is still a big challenge. In this work, we reported for the first time that non-fluorescent ND can be facilely converted into FND through surface modified with aggregation-induced emission fluorophore containing luminescent polymers through the microwave-assisted typical Diels-Alder (D-A) reaction. The final FND (named as ND-poly(Phe-PEGMA-IA)) displayed extensive fluorescence, high water dispersibility and desirable biocompatibility. Cell imaging results demonstrated ND-poly(Phe-PEGMA-IA) exhibited great stain performance. Furthermore, anticancer drug (cisplatin, DDP) can be effectively loaded onto ND-poly(Phe-PEGMA-IA) and controlled release it under acidic environment. All of these favorable properties endow ND-poly(Phe-PEGMA-IA) great potential for biological tracing and controlled drug delivery. Taken together, we have developed a microwave-assisted D-A reaction for preparation of multifunctional FND with great potential for biological imaging and controlled drug delivery applications.

Aggregation-induced emission (AIE) active fluorescence polymeric nanoparticles (FPNs) have recently received increasing interests for biomedical applications such as cell imaging , drug delivery, disease diagnosis and treatment. Fabricated strategies of AIE-active FPNs with high efficiency, simplification and tenderness are still passionately pursued to promote the development of theranostic systems. In this work, we develop a facile method for the preparation of AIE-active FPNs by adopting Ce(IV)/HNO 3 redox polymerization under near room temperature. Thus-prepared FPNs (named as PEG-PLC-1) possess unique AIE feature, great water dispersity, excellent biocompatibility and biodegradability because of the conjugation of ultra-bright AIE dye (PhE-alc) and biodegradable PEG-PCL linear copolymers . The 1 H nuclear magnetic resonance (NMR) spectroscopy, Fourier transform infrared (FT-IR) spectroscopy, transmission electron microscopy (TEM), UV–Visible and fluorescence spectrometers were used to confirm the successful fabrication of AIE-active FPNs. Cell viability and cellular uptake behavior of PEG-PLC-1 FPNs were further investigated for their potential biomedical applications. Results demonstrated that PEG-PLC-1 FPNs are high water dispersity, intensive luminescence and low cytotoxicity, making them very attractive for biomedical applications. More importantly, the method for the fabrication of AIE-active biodegradable FPNs can be occurred under rather facile conditions (e.g., low temperature, free of metal catalysts, common chain transfer agent and aqueous solution) and are specially used for fabrication of AIE-active polysaccharides with poor organic solubility.

Krebs von den Lungen-6 (KL-6) has been recognized as an effective serum biomarker for interstitial lung disease (ILD). The KL-6 accurate detection is of great significance for evaluating the severity of ILD and the prognosis of patients. In this study, a bright aggregation-induced emission luminogen (AIEgen) N , N '-((1,2-diphenylethene-1,2-diyl)bis(4,1-phenylene))bis( N -phenylnaphthalen-1-amine) (TPETN) with a high quantum yield of 87.57% was designed and synthesized, which was subsequently encapsulated into polystyrene nanoparticles (PSNPs) for construction of ultrahigh fluorescent nanoparticle (TPNPs). The obtained TPNPs was found to be 128 times and 16 times higher sensitivity than traditional gold nanoparticles (AuNPs) and tetramethyl 4ʹ,4ʹʹʹ,4ʹʹʹʹʹ,4ʹʹʹʹʹʹʹ-(ethene-1,1,2,2-tetrayl) tetrakis([1,1ʹ-biphenyl]-4-carboxylate) (TCBPE) embedded nanoparticles (TCNPs), respectively. Then, TPNPs were employed as signal output to construct a lateral flow immunoassay platform (TPNP-LFIA) for KL-6 detection. TPNP-LFIA achieved a limit of detection (LOD) of 0.0534 ng/mL, which was 77.15 times and 7.69 times lower than those of AuNP- and TCNP- based LFIA, respectively. The dynamic linearity of TPNP-LFIA for detecting KL-6 ranged from 0.15 to 333.33 ng/mL. The recovery rates of TPNP-LFIA in serum samples varied from 99.96% to 108.13%, with coefficients of variation ≤6.24%. Hence, TPNP-LFIA has promising potential as a rapid and quantitative method for detecting KL-6.

The coronavirus disease-2019 pandemic reflects the underdevelopment of point-of-care diagnostic technology. Nuclei acid (NA) detection is the “gold standard” method for the early diagnosis of the B.1.1.529 (Omicron) variant of severe acute respiratory syndrome-coronavirus disease-2. Polymerase chain reaction is the main method for NA detection but requires considerable manpower and sample processing taking ≥ 3 h. To simplify the operation processes and reduce the detection time, exonuclease III (Exo III)-aided MoS 2 /AIE nanoprobes were developed for rapid and sensitive detection of the oligonucleotides of Omicron. Molybdenum disulfide (MoS 2 ) nanosheets with excellent optical absorbance and distinguishable affinity to single-strand and duplex DNAs were applied as quenchers, and aggregation-induced emission (AIE) molecules with high luminous efficiency were designed as donor in fluorescence resonance energy transfer-based nanoprobes. Exo III with catalytic capability was used for signal amplification to increase the sensitivity of detection. The composite nanoprobes detected the mutated nucleocapsid (N)-gene and spike (S)-gene oligonucleotides of Omicron within 40 min with a limit of detection of 4.7 pM, and showed great potential for application in community medicine.

Heavy metal pollution is life-threatening, the detection of heavy metals is crucial to human health and environment. In this work, AIE featured LA-stabilized gold nanoclusters (NL-AuNCs) were designed and fabricated by modulating precursors in various reaction conditions, and utilized as an efficient fluorescent sensor for portable detecting Ag + with high sensitivity and selectivity. The NL-AuNCs exhibited intense red fluorescence at 625 nm via the secondary reducing agent N-acetyl-L-cysteine (NAC), achieving a quantum yield (QY) of 15.7 %. Notably, the introduction of Ag + enhanced the red fluorescence intensity of NL-AuNCs on account of the aggregation-induced emission (AIE) process, rendering NL-AuNCs uniquely capable of detecting Ag + with the ultralow detection limit of 1.3 nM within a wide concentration range of 0.002–200 μM. Furthermore, the handheld intelligent sensing strategy was constructed by integrating the smartphone App, which enabled swift portable monitoring and convenient readout by visual assessment of color evolution. The acceptable recoveries of 94.6 % to 114.9 % were attained from testing real water, indicating the excellent environmental tolerance of NL-AuNCs, which allowed the possibility for practical in-situ detection.

R-BTOBD exhibited significant aggregation-induced Emission (AIE) characteristics, including high brightness (αAIE ≈ 40), robust light stability, a substantial Stokes shift (128 nm), and a high signal-to-noise ratio, effectively overcoming aggregation-caused quenching (ACQ). Derived from the axially chiral R-H8–BINOL, R-BTOBD was synthesized via nucleophilic cyclization and exhibited pronounced self-assembly properties. Through robust intra- and intermolecular hydrogen bonding interactions, R-BTOBD formed diverse supramolecular structures, including spherical flower-like aggregates, hollow-core triangular tubules, hexagonal tubules, and irregular white block-like stacks. The specific morphology was influenced by factors such as solvent and pH. For fluorescence applications, R-BTOBD can be employed as an effective visual pH detector through an AIE-enhanced excited-state intramolecular proton transfer (AIE-ESIPT) mechanism. When incorporated into pH paper, it exhibited a bright yellow fluorescence across a wide pH range (pH 1–11), transitioning to an intense bright blue fluorescence under strongly basic conditions (pH 12–14). In chiral applications, R-BTOBD demonstrated potential as a chiral switching agent, leveraging the pronounced changes and inversions in its circular dichroism (CD) signals under varying aggregation states and pH conditions. Additionally, it serves as a specific chiral enantioselective fluorescence probe for l-lysine, demonstrating high efficiency and a low detection limit. Given its exceptional fluorescence and chiral properties, this fluorescent material has significant potential for diverse applications in fluorescence, chirality, and visual inspection.

Fluorescent material-based lateral flow immunochromatographic assay (LFIA) has been widespread applied for the point-of-care test of mycotoxins. However, aggregation-caused quenching (ACQ) phenomenon of fluorescent labels seriously affects the detection performance of the methods. Herein, a red-emitted aggregation-induced emission fluorescent microspheres (AIEFMs) based LFIA was developed and firstly applied for the visual and quantitative detection of aflatoxin M1 (AFM1) in milk. The fluorescent probe was constructed by directly coupling the carboxylated AIEPMs with anti -AFM1-mAb, which demonstrated a strong AFM1-specific affinity and exceptional fluorescent characteristics. Under the optimal conditions, this AIEFMs-LFIA could achieve a rapid detection of AFM1 within 15 min, with a visual limit of detection (vLOD) of 0.1 ng/mL and a quantitative limit of detection (qLOD) of 0.01 ng/mL. The accuracy and precision of the established AIEFMs-LFIA method were assessed though the spiked milk samples, and the recoveries were in the range of 86.81%-106.37% with coefficients of variation below 15%. Furthermore, the test strip exhibited a wide detection range for AFM1 (0.01-1 ng/mL) and a remarkable stability for 180 days when stored at 4 °C and 25 °C. Importantly, the results for the spiked milk samples were consistent with those of standard UPLC-MS/MS method, with a correlation coefficient of 0.9911. Overall, the newly developed AIEFMs-LFIA demonstrates significant potential for the point-of-care test of AFM1 in milk.

Pseudomonas aeruginosa , a hazardous pathogen, poses a significant public health risk through its potential contamination of drinking water. Current culture-based detection methods remain limited by their prolonged duration and operational complexity. In this study, ultrabright orange-emitting aggregation-induced emission nanodots (AIENDs) were successfully prepared through a self-assembly approach constructing a metal-polyphenol network formed by coordination between tannic acid and Zr 4+ . The synthesized ultrabright orange-emitting AIENDs demonstrated excellent biocompatibility and enabled direct conjugation with anti- P. aeruginosa monoclonal antibodies (mAbs) without the need for coupling reagents, enabling rapid antibody immobilization with a coupling efficiency exceeding 94 %. The resulting fluorescent probe (AIEND@mAbs) was employed as an amplified signal reporter in a lateral flow immunoassay (LFIA), establishing a sensitive and rapid quantitative detection platform for P. aeruginosa . This AIENDs-LFIA achieved a linear detection range of 1.0 × 10 3 to 5.0 × 10 4 CFU/mL, with a detection limit of 9.74× 10 2 CFU/mL within 15 min. Comparative analysis revealed that the AIENDs-LFIA exhibits 2.2- to 5.7-fold higher sensitivity than conventional LFIAs that use fluorescent microspheres or gold nanoparticles. Furthermore, spiked-recovery experiments in tap water, mineral water, and purified water yielded acceptable recoveries and satisfactory precision. These results demonstrate the AIENDs-LFIA as a promising, efficient, and practical tool for monitoring pathogenic bacteria in drinking water.

The macrocycles with the features of large cavity, optical property and CPL response to guest simultaneously are rarely reported but very attractive based on their potential application. Herein, a macrocycle ( BiCDB-MC ) with dynamically racemic feature, inwardly directed functional groups, a large hollow with size of 1.3 nm and fluorescent property was designed and synthesized. BiCDB-MC was found to have two equally chiral conformations and was dynamically racemic in solution based on their rapidly interconvertion via macrocycle flipping. It was interesting that BiCDB-MC exhibited a selective turn-on CPL response with g lum = 0.00248 toward ursolic acid since one of the stable chiral conformations could form a more stable inclusion complex. This kind of turn-on CPL emission for dynamical-racemic macrocycle with CPL recognition for complicated natural chiral compound was described for the first time.

Nowadays, the rational design of a mechanochromic luminescent (ML) material with ultra-sensitivity toward external stimuli is a greatly challenging task. In this contribution, TPA–Pyr–BP is readily synthesized and verified as a typical AIE luminogen (AIEgen), which consists of a twisted electron donor of triphenylamine (TPA) and a strong electron acceptor of pyridinium (Pyr), and this structural fabrication method shows an elevated electron D–A interaction and intra-molecular charge transfer (ICT) behavior. Specifically, due to the presence of a benzophenone (BP) derivative within the molecular structure, TPA–Pyr–BP exhibits a strong UV absorption ability (ε = 42 360 L mol−1 cm−1). In contrast to a variety of analogues, TPA–Pyr–BP shows ultra-sensitivity with regard to the stimuli of mechanical force, and a sharp signal contrast is expressed with not only a large fluorescence intensity enhancement, but also an obvious emission wavelength shift of 102 nm. Then, the corresponding sensitive performance is extended to demonstrations of different applications, such as being used as a mechanosensor, solid material surface damage reporter, and fingerprint information extractor, as well as in the preparation of security papers. It is also found that the different aggregation states of TPA–Pyr–BP are responsible for this smart behavior.

Control of stereochemistry plays a key role in medicinal chemistry, material and life science. As a prominent AIE luminogen, tetraphenylethene (TPE) derivatives have E/Z isomers which are challenging to separate even by HPLC. Herein, we designed oxetane-substituted TPE (TPE-2OXE) and separated pure isomers by simple column chromatography with high yields, as confirmed by mass spectrometry, IR and NMR spectroscopy. The isomerization of the two isomers can occur by photo- and thermo-activation. Importantly, (Z)-TPE-2OXE isomer solid shows bathochromic emission with a quantum yield 5 times higher than that of (E)-TPE-2OXE. The differences in emission wavelength and quantum yield are derived from distinct emission mechanisms of locally excited (LE) state emission of (E)-TPE-2OXE and charge transfer (CT) state emission of (Z)-TPE-2OXE. The two isomers are also good piezochromic luminescent materials, which have not only an obvious emission color shift but also significantly enhanced luminescence brightness by external force. In addition, (E)-TPE-2OXE solids show self-healing ability, which can crystallize spontaneously from ground amorphous state. The higher brightness of (E)-TPE-2OXE can be retained in solution, so fluorescent AIE nanodots are prepared from the two isomers. Cell-labeling experiments also show that (Z)-TPE-2OXE AIE dots have higher labeling brightness as compared to the (E)-TPE-2OXE isomer. The synthesis and distinct properties of E/Z isomers are beneficial to further development of new TPE derivatives for various applications.

A novel AIE-1 probe integrates “ICT + ESIPT+AIE” tri-modal for ratiometric NIR fluorescence imaging of MAO activity. • AIE-1 exhibits wide linear range (0–140 μg/mL), high sensitivity (LOD 0.23 μg/mL), and excellent selectivity. • AIE-1 semi-quantitatively distinguishes MAO activity in normal/cancer cells and evaluates cellular inhibitor efficacy. • Successful real-time visualization of upregulated MAO activity is achieved in a DILI model. Given the crucial role of monoamine oxidase (MAO) in maintaining amine homeostasis and liver pathology, real-time in situ monitoring of its activity is crucial for elucidating the mechanism of drug-induced liver injury (DILI). This study designed and synthesized a ratio type fluorescent probe AIE-1 based on the “ICT + ESIPT” mechanism, which integrates aggregation induced luminescence (AIE), near-infrared (NIR) emission, and mitochondrial targeting. The probe AIE-1 utilizes MAO catalyzed oxidative deamination reaction to trigger intramolecular charge transfer and excited state proton transfer processes, producing significant ratiometric fluorescence signals at 518 nm and 712 nm. The sensing mechanism was thoroughly validated by 1 H NMR titration, DFT calculations of HOMO-LUMO energy gaps, and solvent-polarity-dependent experiments, collectively confirming the occurrence of both ICT and ESIPT processes. Spectral experiments show that its detection of MAO has a wide linear range (0–140 μg/mL), high sensitivity (0.23 μg/mL), strong selectivity, and anti-interference ability. Cell imaging experiments have confirmed that AIE-1 can successfully semi quantitatively distinguish the differences in MAOs activity between normal liver cells and liver cancer cells, and evaluate the effectiveness of inhibitors. In the drug-induced liver injury model, AIE-1 successfully achieved real-time visualization monitoring of upregulation of MAO activity in vivo with sensitivity and specificity. This probe provides a novel and efficient tool for studying the role of MAO in pathological processes such as DILI.
