Egg yolk proteins have garnered significant attention from researchers due to their outstanding functional properties, and physical modification methods serve as a key approach to enhancing the characteristics of these proteins. In this study, phosphorylated egg yolk high-density lipoprotein (HDL) was prepared using sodium tripolyphosphate (STP) to investigate the properties of the high-internal-phase emulsion formed after cross-linking with phosvitin (Pv), as well as its protective effect on curcumin. Phosphorylation significantly enhanced the functional properties of HDL, with the 6% STP treatment yielding the highest phosphorus content (7.32 mg/g) in the resulting phosphorylated HDL (P-HDL). Structural analyses confirmed covalent phosphate binding, which reduced HDL particle size, induced protein unfolding, and exposed hydrophobic regions, thereby improving its emulsifying capacity and thermal stability. A composite stabilizer composed of phosphorylated HDL (P-HDL) and Pv was developed to construct high internal phase emulsions (HIPEs). These HIPEs also exhibited superior performance in curcumin delivery, increasing its bioaccessibility to 38.10% compared with 15.40% in the control, while effectively inhibiting lipid oxidation during storage. This study provides a promising strategy for designing high-performance emulsion delivery systems.

Scavenging excess reactive oxygen species (ROS) and positively improving intestinal dysbiosis is a promising therapeutic strategy for alleviating inflammatory bowel disease (IBD). However, conventional clinical drugs and probiotic-based adjuvant therapies often fail to achieve satisfactory results due to systemic side effects of drugs, low bioactivity of probiotics, short intestinal retention time, and poor targeting ability. To address these challenges, we developed an antioxidant-functionalized curcumin loaded nanocomposite inulin hydrogel for targeted IBD therapy. In this system, curcumin was encapsulated within chitosan-coated Poly (lactic-co-glycolic acid) PLGA nanoparticles, which not only exhibited excellent ROS-scavenging capacity but also demonstrated the enhanced cellular uptake behavior. In a dextran sulfate sodium induced ulcerative colitis mouse model, the nanocomposite hydrogel significantly prolonged the intestinal retention time of curcumin, thereby suppressing the expression of pro-inflammatory factors, alleviating intestinal inflammation, and promoting the recovery of intestinal barrier and microbial diversity. This study has developed a synergistic therapeutic strategy via combining anti-inflammatory effects with gut microbiota regulation, offering a novel therapeutic approach for the clinical management of colitis.

Objective Autophagy dysfunction results in the accumulation of pro-inflammatory components and drives inflammatory injury. Transcription factor EB (TFEB) is a master regulator of autophagy and a novel target for controlling inflammatory injury. Curcumin analog C1 directly binds to and activates TFEB, but its anti-inflammatory effects remain unclear. Methods The anti-inflammatory and protective effects of curcumin analog C1 were investigated in lipopolysaccharide (LPS)-induced endotoxemia mice. Results Curcumin analog C1 reversed LPS-induced autophagy dysfunction, downregulated pyruvate kinase M2 (PKM2), and reduced pro-inflammatory cytokines including TNF–α, IL–6, and MCP–1. It also attenuated systemic abnormalities, alleviated lung injury, and decreased blood urea nitrogen (BUN), brain-type natriuretic peptide (BNP), and extracellular DNA levels. Conclusions Curcumin analog C1 exhibits potential pharmacological value in the control of inflammatory injury.Thank you for your assistance.

An RF spacer creates a void between NaA and a mesoporous silica shell. • Shell thickness tunes curcumin loading in yolk–shell NaA@HMSNs. • NaA@HMSN-2 loads 124.0 mg/g curcumin and adsorbs Cd 2+ in vitro. • Curcumin release and Cd 2+ adsorption occur in the same buffer system. • HepG2 viability remains above 90% up to 64 μg/mL after 48 h. Cadmium contamination is an environmental and health concern. Here, a yolk–shell composite was constructed using sodium A-type zeolite (NaA) as the microporous yolk and a hollow mesoporous silica nanoparticle (HMSN) layer as the separated shell. Removal of a resorcinol–formaldehyde spacer generated an internal void, and three shell thicknesses were prepared. The NaA component provided Cd 2+ adsorption sites, whereas the amino-functionalized HMSN compartment was used for curcumin loading. N-NaA@HMSN-2, with an approximately 100 nm shell, loaded 124.0 mg/g curcumin. In buffer containing 100 mg/L Cd 2+ , Cur/N-NaA@HMSN-2 released 34.8% and 39.5% curcumin and adsorbed 73.8% and 67.8% Cd 2+ at pH 7.4 and 1.2, respectively. In HepG2 assays, all three NaA@HMSN carriers maintained more than 90% relative cell viability at concentrations up to 64 μg/mL after 48 h. These findings demonstrate coupled Cd 2+ adsorption and curcumin release under in vitro conditions and support further evaluation of this bifunctional material platform. With increasing age, skeletal muscle gradually loses mass and strength, and the risk of falls and fractures escalates among elderly individuals. Inflammation is closely related to age-related muscle atrophy and is the potential target for treating muscular atrophy. Here, biomimetic curcumin nanoparticles (M12MNCs) are prepared via encapsulating curcumin in the skeletal muscle cell membranes modified via muscle-homing peptides (M12) for the treatment of aging related skeletal muscle atrophy. The M12MNCs have good biocompatibility and can be enriched in aging skeletal muscle. After treatment with the M12MNCs, aging mice present enhanced motor ability and improved skeletal muscle metabolism. The results of in vivo and in vitro experiments confirm that M12MNCs reduce inflammation and decrease the expression of α-synuclein (α-syn). In addition, M12MNCs ameliorate skeletal muscle dysfunction in aging mice via regulating the SphK1/Spns2/S1PR2 axis. This study provides a therapeutic target of inflammatory and myogenic factors to improve the function of aging skeletal muscle, which provides valuable insights for the subsequent treatment of aging-related skeletal muscle function. These findings suggest that M12MNCs can improve age-related skeletal muscle dysfunction by modulating inflammation and cell proliferation, and can be used as a novel drug delivery system for clinical therapeutic regimens for muscle atrophy.

Postmenopausal osteoporosis (PMOP) is a predominant form of clinical osteoporosis. It has led to significant health and social burdens for older patients. Reestablishing the balance between osteogenic and osteoclastic is a crucial strategy for treating PMOP. Curcumin (Cur), a naturally derived polyphenolic substance, has gained recognition as a viable option for treating osteoporosis. Despite its potential, the clinical use of Cur is hindered by its limited bioavailability and the presence of side effects. Nanoparticles modified with aspartic acid octapeptide (ASP8) exhibit a strong affinity for bone tissue, facilitating targeted delivery. This study presents novel acid-responsive zeolite imidazolate framework-8 (ZIF) nanoparticles modified with ASP8 and loaded with Cur (Cur@ZIF@ASP8, CZA). Upon delivery by this nanoparticle drug delivery system, Cur can effectively regulate bone homeostasis, offering a potential therapeutic strategy for osteoporosis. This study demonstrated that CZA nanoparticles could successfully transport Cur to bone tissue without significant toxicity. Furthermore, nanoparticles promote bone formation and inhibit osteoclast activity. They also modify the expression of related genes and proteins, such as OCN, ALP, CTSK, and MMP9. Significant evaluations utilizing microcomputed tomography, Masson’s staining, hematoxylin and eosin staining, and immunofluorescence staining demonstrated that intravenous CZA administration in ovariectomized mice resulted in bone destruction while simultaneously reducing overall bone loss. In conclusion, CZA nanoparticles hold promise as a therapeutic option for osteoporosis.

Hepatocellular carcinoma (HCC) is a highly prevalent cancer with a significant impact on human health. Curcumin, a natural compound, induces cytoskeletal changes in liver cancer cells and modifies the distribution of lipids, proteins, and polysaccharides on plasma membranes, affecting their mechanical and electrical properties. In this study, we used nanomechanical indentation techniques and Kelvin probe force microscopy (KPFM) based on atomic force microscopy (AFM) to investigate the changes in surface nanomechanical and electrical properties of nuclear and cytoplasmic regions of HepG2 cells in response to increasing curcumin concentrations. CCK-8 assays and flow cytometry results demonstrated time- and concentration-dependent inhibition of HepG2 cell proliferation by curcumin. Increasing curcumin concentration led to an initial increase and then decrease in the mechanical properties of nuclear and cytoplasmic regions of HepG2 cells, represented by the Young's modulus (E), as observed through nanoindentation. KPFM measurements indicated decreasing trends in both cell surface potential and height. Fluorescence microscopy results indicated a positive correlation between curcumin concentration and phosphatidylserine translocation from the inner to the outer membrane, which influenced the electrical properties of HepG2 cells. This study provides valuable insights into curcumin's mechanisms against cancer cells and aids nanoscale evaluation of therapeutic efficacy and drug screening.

Hepatic macrophages have been considered as a therapeutic target for liver fibrosis treatment, and phosphatidylserine (PS)-containing nanoparticles are commonly used to mimic apoptotic cells that can specifically regulate macrophage functions, resulting in anti-inflammatory effects. This study was designed to test the efficacy of PS-modified nanostructured lipid carriers (mNLCs) containing curcumin (Cur) (Cur-mNLCs) in the treatment of liver fibrosis in a rat model. Carbon tetrachloride-induced liver fibrosis in rats was used as an experimental model, and the severity of the disease was examined by both biochemical and histological methods. Here, we showed that mNLCs were spherical nanoparticles with decreased negative zeta potentials due to PS decoration, and significantly increased both mean residence time and area under the curve of Cur. In the rats with liver fibrosis, PS-modification of NLCs enhanced the nanoparticles targeting to the diseased liver, which was evidenced by their highest accumulation in the liver. As compared to all the controls, Cur-mNLCs were significantly more effective at reducing the liver damage and fibrosis, which were indicated by in Cur-mNLCs-treated rats the least increase in liver enzymes and pro-inflammatory cytokines in the circulation, along with the least increase in collagen fibers and alpha smooth muscle actin and the most increased hepatocyte growth factors (HGF) and matrix metalloprotease (MMP) two in the livers. In conclusion, PS-modified NLCs nanoparticles prolonged the retention time of Cur, and enhanced its bioavailability and delivery efficiency to the livers, resulting in reduced liver fibrosis and up-regulating hepatic expression of HGF and MMP-2.

Current fluorescent polyurethanes face critical challenges, including cytotoxic fluorescent labels, irreversible damage from mechanical stress, and monofunctional behavior lacking stimulus-responsive sensing capability. To address these limitations, we developed a polyurethane elastomer that merges extreme stretchability, autonomous self-healing, and circular recyclability. Engineered via adipic dihydrazide and 4, 4′-diaminodiphenyl disulfides, the network unites reversible disulfide metathesis with hydrogen-bond dissipation, yielding 10 MPa strength, 3200 % elongation, and 90 % toughness recovery within 72 h. Notch tolerance reaches 1200 % strain, while 75 % ethanol depolymerisation at 25 °C enables solvent-cast recycling (>80 % retention in tensile strength). The matrix confines curcumin, suppressing aggregation-caused quenching and furnishing stable solid-state fluorescence under water for 14 days. Emission color switches from green to yellow in response to substrate polarity. Applications include stamp-sealable anti-counterfeiting inks and real-time pork-freshness sensors that visually report spoilage through pH-triggered fluorescence decay. This work overcomes key limitations in natural fluorophore applications by establishing a structure-property optimization paradigm that simultaneously achieves high performance, environmental sustainability, and multifunctionality for safety-sensitive applications.

In this work, sequential electrospinning was utilized to fabricate a multilayer film with ethylcellulose nanofibers as the outer layer and curcumin-loaded gelatin nanofibers as the inner layer. Field-emission scanning electronic microscopy observations showed that the outer and inner layers had a smooth surface and clear boundary. The hydrophobic outer layers decreased the water vapor permeability and improved the water contact resistance of the hydrophilic inner layer, and the intimate interactions of hydrogen bonds between two adjacent layers enhanced the thermal stability . The multilayer film exhibited a sustained release manner of the encapsulated curcumin for 96 h, compared to the burst release within 30 min from the gelatin film. In addition, the antioxidant activities of the released curcumin from the multilayer film were well retained within 96 h. These results suggested that the multilayer nanofibrous film fabricated by sequential electrospinning has potential applications in bioactive encapsulation and controlled release.

Gelatin possesses excellent film-forming ability, biodegradability, and biocompatibility, which makes it a promising alternative to petroleum-based plastics. However, its application in food packaging is limited by its brittleness, high water sensitivity, and insufficient microbial protection. In this study, curcumin was introduced as a chain extender into castor oil-based waterborne polyurethane (CWU), which was then blended with gelatin as a functional material to create reactive food composite films (CWGL). The effects of varying CWU concentrations on the microstructure and properties of the films were thoroughly investigated. The incorporation of CWU enhanced the mechanical properties, water solubility, water vapor barrier, oxygen barrier, and UV shielding capabilities of the gelatin-based composite films. Additionally, CWU imparted antioxidant, antibacterial, and pH-responsive properties to the CWGL. Packaging application tests demonstrated that CWGL could extend the shelf life of fresh pork up to 10 days under refrigeration. Furthermore, CWGL exhibited a visible color change (∆E > 5) during pork storage, suggesting its potential use as a freshness-indicating innovative packaging. This study presents a novel strategy to advance the application of gelatin in innovative food packaging.

Background:Although electrohydrodynamic atomization (EHDA) consistently provides drug-encapsulation efficiencies (DEE) far above those of conventional bottom-up nanotechnologies, the question of how to systematically push that efficiency even higher remains largely unexplored. Methods:This study introduces a modified triaxial electrospinning protocol tailored to the application and benchmarks it against two conventional techniques: single-fluid blending and coaxial electrospinning. Ethylcellulose (EC) served as the polymeric matrix, while curcumin (Cur) was chosen as the model drug. In the triaxial setup, an electrospinnable, drug-free EC solution was introduced as an intermediate sheath to act as a molecular barrier, preventing Cur diffusion from the core fluid. Ethanol alone was used as the outermost fluid to guarantee a stable and continuous jet. Results: This strategy provided a DEE value of 98.74 ± 6.45%, significantly higher than the 93.74 ± 5.39% achieved by coaxial electrospinning and the 88.63 ± 7.36% obtained with simple blending. Sustained-release testing revealed the same rank order: triaxial fibers released Cur the most slowly and exhibited the smallest initial burst release effect, followed by coaxial and then blended fibers. Mechanistic models for both fiber production and drug release are proposed to clarify how the tri-layer core–shell structure translates into superior performance. Conclusions: The modified triaxial electrospinning was able to open a new practical route to produce core-sheath nanofibers. These nanofibers could provide a higher DEE and a better sustained drug release profile than those from the coaxial and blending processes.
