In this study, we engineered pH/glutathione dual-responsive nanoparticles (LP/CC-Cu-Cur NPs) based on a poly-γ-glutamic acid (γ-PGA)/chitosan (CS) core-shell architecture for synergistic chemo/chemodynamic therapy of glioma. The nanoparticles feature a core of CC-Cu-Cur NPs, formed via Cu 2+ -coordinated self-assembly of caffeic acid-grafted CS and curcumin (Cur), encapsulated within a phenylboronic acid-conjugated γ-PGA shell through pH-sensitive borate ester bonds. Surface modification with lactoferrin conferred brain-penetrating and glioma-targeting capabilities. The resulting spherical nanoparticles had a uniform size of 235.89 nm, a zeta potential of −22.66 mV, and high Cur loading (6.02%) and encapsulation efficiency (83.09%). Upon exposure to the acidic tumor microenvironment, the nanoparticle shell detaches, reversing surface charge from negative to positive, thereby enhancing cellular uptake and mitochondrial targeting. Intracellular glutathione then triggers core degradation, releasing Cur and Cu 2+ . Cur induces mitochondrial apoptosis, while Cu 2+ catalyzes a Fenton-like reaction, converting endogenous hydrogen peroxide into highly cytotoxic reactive oxygen species. In vitro, the nanoparticles showed enhanced blood-brain barrier penetration, efficient lysosomal escape, and potent cytotoxicity against GL-261 cells (IC 50 = 18.34 μg/mL) via a synergistic action of Cu 2+ and Cur (CI = 0.28). In vivo, LP/CC-Cu-Cur NPs achieved superior brain accumulation and antitumor efficacy, highlighting their potential as a promising strategy for glioma therapy.

The abnormally high expression of glutathione (GSH) in hepatocellular carcinoma (HCC) cells significantly contributes to chemotherapy resistance by neutralizing reactive oxygen species (ROS). To address this problem, we developed a nanocomposite probe (micelle@PDA@MnO 2 NPs) which was composed of free radical-labeled micelles as the core, polydopamine (PDA) as the inner shell and MnO 2 nanosheets as the outer shell. In the HCC microenvironment, the over-expressed GSH triggered the decomposition of the MnO 2 shell, releasing Mn 2+ ions, and subsequently, the acidic condition and over-expressed carboxylesterase 2 (CES2) worked together to mediate the controlled release of nitroxide free radicals (i.e., TEMPONE) from the micelle core. With the help of dual-signal electron spin resonance (ESR) detection, it was found that much more free radicals and Mn 2+ were released in hepatoma cells than in normal liver cells, rendering the proposed nanotherapeutic platform specific to HCC and a good candidate for free radical therapy and chemodynamic therapy (CDT). Moreover, the therapy featured an intelligent self-amplified property: the abundant GSH in the tumor microenvironment rapidly activated the decomposition of the nanocomposite and triggered the release of free radical TEMPONE and Mn 2+ , and TEMPONE and Mn 2+ further amplified the oxidative stress to induce tumor apoptosis via ROS generation and GSH depletion. Finally, in combination with three therapy modes (Mn 2+ -mediated CDT, the oxidative stress enhancement effect of TEMPONE, and PDA-based photothermal therapy), significant therapeutic effects including facilitation of HepG2 cell apoptosis and inhibition of tumor growth in the HCC mouse model were achieved.

In this study, a highly sensitive surface-enhanced Raman scattering (SERS) microarray chip was developed for the detection of reduced glutathione (GSH) in the serum of colorectal cancer (CRC) patients. Gold-palladium nanorods (Au@Pd NRs) with peroxidase-like activity were synthesized and assembled into holes on the polydimethylsiloxane (PDMS)-indium tin oxide (ITO) glass substrate via electrophoretic deposition (EPD). The Au@Pd NRs catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) to oxidized TMB (oxTMB) in the presence of hydrogen peroxide (H 2 O 2 ), producing a characteristic SERS peak at 1605 cm −1 . Prior to detection, a thiolated GSH aptamer was immobilized on the microarray chip to specifically capture GSH, effectively avoiding interference from other reducing substances in the complex serum matrix. The captured GSH then reduces oxTMB back to TMB, causing signal attenuation proportional to GSH concentration. The limit of detection (LOD) reached 93 pmol L −1 , with a relative standard deviation (RSD) of 6.36% for reproducibility. Clinical serum analysis of 30 healthy individuals and 30 CRC patients demonstrated results consistent with the gold standard testing method of high-performance liquid chromatography (HPLC) (relative error < 8%), confirming the potential of this approach for the effective detection of GSH and early CRC diagnosis. In this study, a highly sensitive surface-enhanced Raman scattering (SERS) microarray chip was developed for the detection of reduced glutathione (GSH) in the serum of colorectal cancer (CRC) patients.

A novel, highly sensitive “on-off-on” fluorescent probe ( E )- N ’-((1 H -imidazol-2-yl) methylene) -4-(5, 5-difluoro-1, 3, 7, 9-tetramethyl-5 H -4 λ 4 ,5 λ 4 -dipyrrolo [1, 2- c :2′, 1′- f ] [1,3,2] diazaborinin-10-yl) benzohydrazide ( BDB ) was developed in this study, and its structural and spectral properties were characterized by 13 C NMR, 1 H NMR, X-ray, ESI-MS, ultraviolet and fluorescence photometry. BDB could specifically and efficiently identify Cu 2+ among many other metal ions with a fluorescence quenching efficiency of 92.7% and a recognition limit of 0.65 μM in CH 3 CN/HEPES (7:3, v/v, pH = 7.23) buffer. The energy difference between BDB and BDB -Cu 2+ complex was calculated by DFT. The BDB -Cu 2+ complex could continuously identify GSH in the same system and show an obvious fluorescence enhancement response with a recovery efficiency of approximately 91% and a low recognition limit of 0.02 μM, which is significantly lower than the physiological concentration of GSH in living cells. The applicability of BDB is verified by the strip test. BDB can also be used to detect GSH in HeLa cells, which expand the application of fluorescent chemical sensors in biology.

A novel tetranuclear copper complex (TCC) has been successfully synthesized and characterized using high resolution mass spectrometry (HRMS), Fourier transform infrared spectroscopy (FTIR) and X-ray single crystal diffraction technique. TCC was capable of triggering by Glutathione (GSH) to produce Cu (Ⅰ), then the resulted Cu (Ⅰ) further converted the overexpression of endogenous H 2 O 2 into •OH, a highly cytotoxic reactive oxygen species (ROS), thereby killing the cancer cells. TCC showed high cytotoxicity to 4T1, MCF7, HepG2 cancer cells. Interestingly, the cytotoxicity of TCC to non-cancerous cells is much lower than that of cancer cells. Cell cycle experiments demonstrated that TCC was capable of arresting the cancer cell cycle in the G2/M phase. The apoptosis experiments shown that TCC could induce apoptosis rather than necrosis.

Herein, a novel and sensitive fluorescence method for cartap determination is established on the basis of the inner filter effect (IFE) of gold nanoparticles (Au NPs) on the fluorescence of glutathione protected Cu NCs (GSH-Cu NCs). In the presence of Au NPs, the fluorescence of GSH-Cu NCs was strongly quenched by the IFE because the absorption spectra of Au NPs overlap well with the emission spectra of GSH-Cu NCs. Upon addition of cartap, cartap could induce the aggregation of Au NPs whose absorption spectrum does not overlap with the emission spectrum of GSH-Cu NCs. Then, with the increase in cartap concentration, the IFE-decreased fluorescence was gradually recovered, realizing the fluorescence sensing of cartap. Under optimal conditions, the proposed method has a good linear relationship with cartap concentration in the range of 7–100 nM, and the detection limit is 3.34 nM. In addition, satisfactory results were obtained for cartap analysis using tap water and cabbage as real samples, which demonstrated that the method as-developed would have great practical application prospects.

The abnormal level of Cu2+ or GSH can cause variety of neurodegenerative diseases in humans. Thus, the selective and sensitive detection of Cu2+ and GSH has inspired intensive research efforts in biological sample analysis fields. Herein, an “on–off–on” fluorescent probe based on nitrogen and sulfur co-doped carbon dots (N,S-CDs) has been successfully prepared for the detection of Cu2+ and GSH. The “turn-off” process of fluorescence in the presence of Cu2+ ions was induced by forming a non-luminescent ground state complex due to the interaction between surface groups of the probe and Cu2+ ions. Moreover, the strong coordination between GSH and Cu2+ could destroy the structure of the complex and restore the fluorescence to “turn-on”. This fluorescent probe had excellent selectivity and high sensitivity toward Cu2+ and GSH with the limits of detection (LODs) of 38 nM and 41 nM. More importantly, the as-prepared N,S-CDs served as an efficient fluorescent probe for not only detecting Cu2+ ions in lake water and tap water, and GSH in BSA solution, but also sensing Cu2+ and GSH in living cells. Therefore, these N,S-CDs could be considered as a promising fluorescence probe candidate for environmental monitoring and biological imaging.

Nitrogen and phosphorus co-doped carbon dots (NPCDs) were synthesized by a microwave method, using citric acid monohydrate and diammonium hydrogen phosphate as raw materials. The morphology, structures and elemental compositions of NPCDs were characterized by transmission electron microscopy, X-ray diffractometry, and X-ray photoelectron spectroscopy. It was found that NPCDs show good selectivity to Hg2+ over other metal ions. And the fluorescence of the NPCDs-Hg2+ system can be recovered in the presence of glutathione (GSH). As such, a “turn-off-on” fluorescence probe can be constructed for the detection of Hg2+ and GSH. Under the optimum working conditions, the linear response of NPCDs to Hg2+ was in the range 0.10–30 μM with a detection limit of 9.9 nM. The linear range of GSH was 0.50–32 μM with a limit of detection of 11.2 nM. The proposed method has been successfully applied to the detection of Hg2+ and GSH in various biological samples. What is more, NPCDs have low cytotoxicity verified by the MTT assay. Using NPCDs as a probe, fluorescence images were obtained for HepG2 cells, which indicated that it could monitor the levels of Hg2+ and GSH in biological systems.

A dual-emission ratio-fluorescent sensing nanohybrid based on RadixHedysarigreen-synthesized carbon quantum dots (CDs) and glutathione-functionalized gold nanoclusters (GSH-AuNCs) had been developed for the determination of cefodizime sodium (CDZM). The designed fluorescence nanohybrid had two significant fluorescence emission peaks at 458 nm and 569 nm when excited at 360 nm, which was attributed to the CDs and GSH-AuNCs. With the addition of CDZM, the fluorescence at 458 nm was slightly weakened while the fluorescence at 569 nm was enhanced obviously. Based on the relationship between the I569/I458fluorescence intensity ratio and the concentration of CDZM, the designed nanohybrid exhibited a good linearity range of 1.0–1000.0 μM and the limit of detection (LOD) was 0.19 μM. The method was finally applied in the detection of CDZM in urine, showing the potential applications in complicated biological samples.

The low targeting of immunity regulators & chemotherapy drugs and immune tolerance in the tumor microenvironment significantly hindered the chemo-immunotherapy effect. In this work, an MMP-2/GSH-responsive nanobomb loading Resiquimod (R848) and prodrug Cisplatin (Pt) with hierarchical drug delivery and tumor associated macrophages (TAMs) polarization traits was designed for improved chemo-immunotherapy. The Pt-carrying dendrimers penetrate the depth of tumor tissues under MMP-2 and activate the cytotoxicity of Pt prodrug in response to GSH, causing effective tumor damage and enhancing immunogenicity. Meanwhile, R848 is effectively uptake by TAMs, which were re-educated and then greatly relieved immune resistance. In vitro and in vivo results collectively revealed that the nanosystem could not only overcome the barrier of tumor penetration and the heterogeneity action sites of drugs but also re-educate TAMs and suppress immune evasion via activating extracellular signal-regulated kinase/mitogen-activated protein kinase (ERK/MAPK) signal pathway regulated by heparin binding EGF like growth factor ( Hbegf ) gene. The gene silencing or knockout in vitro and in vivo demonstrated that Hbegf could regulate the re-education of TAMs and the progression of tumors.

Considering the severe hazard to health posed by nitrophenol (NP) isomers, especially 4-NP, reliable and selective detection strategies are urgently challenged. In this study, a fluorescence platform based on glutathione-stabilized gold/silver nanoclusters (GSH-Au/Ag NCs) and zeolite imidazolate framework material (ZIF-8) was developed for 4-NP detection. The synergistic effect of gold and silver makes the bimetallic nanoclusters (GSH-Au/Ag NCs) much better in optical properties than gold nanoclusters. Subsequently, GSH-Au/Ag NCs with aggregation-induced emission (AIE) effect was encapsulated into ZIF-8 to obtain GSH-Au/Ag NCs@ZIF-8 nanocomposite. Compared with GSH-Au NCs, the fluorescence signal was efficiently further improved nearly 7 times through the confinement effect of ZIF-8, and the fluorescence lifetime was increased from 1.2 μs to 6.9 μs. The addition of 4-NP effectively quenched the fluorescence emission peak of GSH-Au/Ag NCs@ZIF-8 at 600 nm, and the assay exhibited highly sensitivity to 4-NP with the detection limit of 1.11 μM from 2.0 μM to 35 μM. Especially, results showed that 4-NP could be easily discriminated from nitrophenol isomers (3-NP and 2-NP). The mechanism research demonstrated that the absorption curve of GSH-Au/Ag NCs@ZIF-8 + 4-NP overlapped greatly with the fluorescence excitation spectrum of GSH-Au/Ag NCs@ZIF-8, and the fluorescence lifetime of GSH-Au/Ag NCs@ZIF-8 did not change when 4-NP was present or not. Therefore, the mechanism of GSH-Au/Ag NCs@ZIF-8 for 4-NP detection was mainly attributed to the inner filter effect (IFE). Furthermore, the developed detection assay was applied to water sample with satisfactory recovery results, demonstrating its high potential in monitoring 4-NP in environment.

Glutathione (GSH) plays crucial roles in various biological functions, the level alterations of which have been linked to varieties of diseases. Herein, we for the first time expanded the application of oxidase-like property of MnO2 nanosheet (MnO2 NS) to fluorescent substrates of peroxidase. Different from previously reported fluorescent quenching phenomena, we found that MnO2 NS could not only largely quench the fluorescence of highly fluorescent Scopoletin (SC) but also surprisingly enhance that of nonfluorescent Amplex Red (AR) via oxidation reaction. If MnO2 NS is premixed with GSH, it will be reduced to Mn2+ and lose the oxidase-like property, accompanied by subsequent increase in SC’s fluorescence and decrease in AR’s. On the basis of the above mechanism, we construct the first MnO2 NS-based ratiometric fluorescent sensor for ultrasensitive and selective detection of GSH. Notably, this ratiometric sensor is programmed by the cascade logic circuit (an INHIBIT gate cascade with a 1 to 2 decoder). And a linear relationship between ratiometric fluorescent intensities of the two substrates and logarithmic values of GSH’s concentrations is obtained. The detection limit of GSH is as low as 6.7 nM, which is much lower than previous ratiometric fluorescent sensors, and the lowest MnO2 NS-based fluorescent GSH sensor reported so far. Furthermore, this sensor is simple, label-free, and low-cost; it also presents excellent applicability in human serum samples.
