Background: The production of D-lactic acid (D-LA) from non-detoxified corn stover hydrolysate is hindered by substrate-mediated inhibition and low cell utilization times. In this study, we developed a novel temperature-sensitive hydrogel, F127-IEA, for efficient D-LA production using a cell-recycle batch fermentation process. Results: F127-IEA exhibited a porous structure with an average pore size of approximately 1 μm, facilitating the formation of stable Lactobacillus bulgaricus clusters within the gel matrix. It also maintains excellent mechanical properties. It also maintains excellent mechanical properties. F127-IEA immobilized Lactobacillus bulgaricus T15 (F127-IEA-T15) can be used in cell-recycle fermentation for over 150 days from glucose and 50 days from corn stover hydrolysate, achieving high production rates of D-LA from glucose (2.71 ± 0.85 g/L h) and corn stover hydrolysate (1.29 ± 0.39 g/L h). F127-IEA-T15 enhanced D-LA production by adsorbing and blocking toxic substances present in corn stover hydrolysate that are detrimental to cellular activity. Conclusions: The newly developed hydrogels in this study provide a robust platform for large-scale extraction of D-LA from non-detoxified corn stover.

The probiotic bacterium, Lactobacillus reuteri, possesses a unique pathway for metabolizing glycerol into 3-hydroxypropionic acid (3-HP) and 1,3-propanediol (1,3-PD), which are two important platform chemicals. In this work, a potent L. reuteri strain, designated as FXZ014, was isolated and identified from healthy human feces for co-biosynthesis of 3-HP and 1,3-PD during whole cell biotransformation of glycerol. Initially, biotransformation conditions were optimized for this new strain using single factor optimization and response surface methodology (RSM). Under the optimum batch conditions, FXZ014 produced 8.74 ± 0.24 g/L of 3-HP and 9.94 ± 0.36 g/L of 1,3-PD with a corresponding net yield of 0.67 mol/mol. Finally, efficiency of the isolated strain in the co-production of these two metabolites was compared with the same of a reference strain, namely L. reuteri CICC 6118. Compared to the reference strain, the amounts of 3-HP and 1,3-PD produced by the isolated strain were significantly higher that corresponded to 38.4% and 21.14%, respectively. Therefore, FXZ014 could be a promising natural strain for co-biosynthesis of 3-HP and 1,3-PD from glycerol.

Chemical tethering NAD + to GfADH enhances enzyme-cofactor spatial proximity. • The application of conjugate and NOX reduced the NAD + demand by 13-fold. • The NAD + -GfADH conjugate and NOX were co-fixed via sequential recruitment. • Co-immobilization enhanced enzyme stability with improved cofactor regeneration. The green biomanufacturing of high-value compounds is limited by enzyme instability and cofactor dependence. Herein, we developed a tailored multi-module immobilization strategy that integrated cofactor tethering with sequential multi-enzyme co-immobilization to facilitate the efficient conversion of allitol into D-allulose catalyzed by the Gluconobacter frateurii -derived dehydrogenase (GfADH). In this strategy, nicotinamide adenine dinucleotide (NAD + ) was covalently tethered to the surface of GfADH via a flexible linker, creating an NAD + -GfADH conjugate. Coupling the conjugate with NADH oxidase (NOX) established an efficient cofactor self-regenerating system, reducing the external NAD + demand by 13-fold. Furthermore, this conjugate was encapsulated within hybrid nanoflowers, thereby improving its reusability. To facilitate cofactor transfer, NOX was directionally recruited and colocalized with GfADH using the SpyTag/SpyCatcher system. The co-immobilized enzyme exhibited a high D-allulose yield of 88.7% from 110 mM allitol and retained 85.0% of its initial activity after 6–8 cycles. This work provides a valuable integrated strategy for cofactor regeneration and enzyme stabilization in cofactor-dependent biosynthesis. To enhance the stability and intestinal delivery of the probiotic Lactobacillus sakei J15, a multilayer microencapsulation system was developed using layer-by-layer (LBL) technology, using milk protein-hyaluronic acid (HA) conjugates (casein–HA or β-lactoglobulin–HA) and chitosan (CHI). The microcapsules exhibited high encapsulation efficiency (up to 98.65%) and significantly improved probiotic viability under harsh conditions, including acid, bile salt, simulated gastrointestinal digestion, and heat. Electrostatic interaction was confirmed as the main force binding the amino groups in the conjugate to the carboxyl groups of CHI, and the surface charge of the samples reversed as a zigzag pattern whenever another outermost layer was added. The (CHI/CN–HA/CHI)3 and (CHI/β-Lg–HA/CHI)3 microcapsules exhibited the best adhesion to Caco-2 cells, reaching 24.47 and 26.71 CFU/cell, respectively. Microcapsules with two or four layers (milk protein-HA as the outer layer) showed superior resistance to environmental stresses, while three-layer capsules (chitosan as the outer layer) exhibited the strongest adhesion to Caco-2 cells.

Gemcitabine (GEM) resistance remains a major challenge in the treatment of gallbladder cancer (GBC). This study investigated the synergistic effect of curcumin (CUR) combined with the probiotic Lactobacillus rhamnosus GG (LGG) in reversing chemoresistance through modulation of the gut microbiota. In GEM-resistant GBC-SD cells, the CUR–LGG combination significantly inhibited cell proliferation, suppressed migration and invasion, and induced apoptosis, as demonstrated by CCK-8, wound healing, Transwell, and flow cytometry assays. Western blot analysis revealed corresponding regulation of proliferation markers (Ki67, PCNA), apoptosis-related proteins (Bcl-2, Bax, cleaved Caspase-3), and epithelial–mesenchymal transition markers. In xenograft models, the combined treatment markedly suppressed tumor growth and altered gut microbial composition, increasing beneficial bacteria ( Lactobacillus , Bifidobacterium ) while reducing pathogenic taxa. LC–MS analysis further demonstrated restoration of bile acid homeostasis, characterized by elevated primary bile acids (GCA, CDCA) and decreased secondary bile acids (DCA, LCA). Mechanistically, the intervention significantly inhibited PI3K/AKT signaling, as confirmed by Western blot and immunohistochemistry. Bioinformatic analysis further identified PI3K/AKT as a central regulatory pathway. These findings indicate that probiotic-assisted CUR therapy reverses GEM resistance by remodeling the gut microbiota and its metabolic outputs, thereby suppressing oncogenic signaling pathways. This strategy provides a promising microbiota-based approach for improving therapeutic outcomes in GBC.

Pseudorabies virus (PRV) infection poses a significant health challenge to swine production, inducing oxidative stress, inflammatory responses, and reduced growth performance. Probiotic fermentation of medicinal plants is a promising strategy for generating novel probiotic-derived products for improving animal health. In this study, Phellodendron amurense (PA), a plant with both medicinal and dietary applications, was fermented with Lactobacillus plantarum R (LpR). We investigated the potential of fermented extract (FPAE) as a probiotic-derived feed additive for swine and its underlying mechanisms. Nontargeted UPLC‒MS/MS metabolomics revealed that fermentation significantly reshaped the metabolic profile of PAs, with marked enrichment of phenolic acids, flavonoids, and alkaloids. Specifically, the berberine content increased 2.5-fold, and the levels of antioxidant components such as ferulic acid and kaempferol were significantly increased. In vitro, FPAE inhibited PRV replication in PK-15 cells in a concentration-dependent manner (84.15% inhibition at 5 mg/mL). It also alleviated virus-induced oxidative stress, as evidenced by restored glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) activities and reduced malondialdehyde (MDA) levels. The supernatant of LpR showed no antiviral activity, confirming that the enhanced efficacy resulted from biotransformation of the plant matrix. In vivo, daily oral administration of FPAE (crude drug concentration of 125 mg/mL, 200 µL/mouse, equivalent to 892 mg/kg) significantly improved the survival rate (80% vs. 53.33% in the model group, and higher than the 73.33% in the unfermented PAE group); increased the average daily weight gain and feed conversion ratio; reduced the viral loads in the brain, lung, spleen and kidney; and, compared with PAE, more effectively reduced viral loads in the brain and alleviated pathological damage; and alleviated clinical symptoms and pathological damage to the brain. Furthermore, FPAE suppressed systemic inflammatory responses (decreased serum IL-6, P < 0.05) and enhanced antioxidant defense (increased GSH-Px activity, P < 0.05, P = 0.004). Through a multi-dimensional “constituent–cell–animal” approach, this study demonstrated that probiotic fermentation restructures the chemical composition of Phellodendron amurense and synergistically regulates host oxidative stress and inflammatory responses to confer protection against viral infection. These findings provide a theoretical basis for developing LpR-fermented Phellodendron amurense as a functional feed additive to control PRV infection in swine.

Passion fruit peel contained various phenolic compounds with α-glucosidase and α-amylase inhibitory activities in vitro . However, the bound state and glycosylated forms hindered their active expression. In this article, β-glucosidase-producing Lactobacilli were inoculated into fermented peels for targeted metabolism. The results showed that the release of bound polyphenols and the increase of free polyphenols were significantly correlated with β-glucosidase activity. The strain G1 with high enzyme activity degraded 48.20% of the bound polyphenols after 24 h. The ability of extracts to inhibit α-glucosidase and α-amylase activities was significantly enhanced and closely related to the content of free polyphenols (r = -0.96 and -0.88, P < 0.001). Computer- assisted screening identified 38 phenolic compounds as potential dual inhibitors of α-glucosidase and α-amylase. Among them, 22 α-glucosidase inhibitors and 24 α-amylase inhibitors were identified for the first time. Various inhibitors, including phenolic acids, flavones, and flavonols, increased in content after fermentation. The total content of 14 glycosylated polyphenols decreased to 28.38% at 48 h, which was opposite to the aglycone. Molecular docking indicated that glycosylated polyphenols bind to β-glucosidase through hydrogen bonds and van der Waals forces. Different glycosylations at the C-3 position of quercetin bound at same sites to β-glucosidase. This study contributes to the theory of enhancing the functional activity of natural products through targeted fermentation transformation.

Under the optimal conditions of immobilization and fermentation, the highest LA yield of 0.966 ± 0.006 g/g fructose and production rate of 2.426 ± 0.018 g/(L × h) with an error of -0.5% and -0.2% to the predicted results were obtained from batch fermentation by the CS film-coated SA-PVA immobilized L. pentosus cells. The LA yield and production rate of these immobilized cells were 2.7% and 10.1% higher than that of normal SA-PVA immobilized cells respectively, and they were 5.7% and 48.4% higher than that of free cells, respectively. The effect of temperature on different types of immobilized cells and free cells was significantly different, but the effect of pH on different types of cells was not much different. The kinetic models could effectively describe the different fermentation performances of three types of cells. The immobilized cells have excellent reusability to conduct 9 runs of repeated batch fermentation.

One-step fermentation, inoculated withLactobacillus fermentum(L. fermentum) in shrimp by-products, was carried out to obtain chitin and flavor protein hydrolysates at the same time. The fermentation conditions were optimized using response surface methodology, resulting in chitin with a demineralization rate of 89.48%, a deproteinization rate of 85.11%, and a chitin yield of 16.3%. The surface of chitin after fermentation was shown to be not dense, and there were a lot of pores. According to Fourier transform infrared spectroscopy and X-ray diffraction patterns, the fermented chitin belonged to α-chitin. More than 60 volatiles were identified from the fermentation broth after chitin extraction using gas chromatography–ion transfer spectrometry analysis.L. fermentumfermentation decreased the intensities of volatile compounds related to unsaturated fatty acid oxidation or amino acid deamination. By contrast, much more pleasant flavors related to fruity and roasted aroma were all enhanced in the fermentation broth. Our results suggest an efficient one-step fermentation technique to recover chitin and to increase aroma and flavor constituents from shrimp by-products.Keywords:shrimp by-products;L. fermuntum;one-step fermentation;chitin;flavor volatiles;gas chromatography–ion transfer spectrometry

We report for the first time the usage of Millettia speciosa Champ cellulose (MSCC) and carboxymethylcellulose (MSCCMC) for the fabrication of 3D-network hydrogel as delivery system for probiotics. The structural features, swelling behavior and pH-responsiveness of MSCC-MSCCMC hydrogels and their encapsulation and controlled-release behavior for Lactobacillus paracasei BY2 ( L. paracasei BY2) were mainly studied. Structural analyses demonstrated that MSCC-MSCCMC hydrogels with porous and network structures were successfully synthesized through the crosslinking of –OH groups between MSCC and MSCCMC molecules. An increasing concentration of MSCCMC significantly improved the pH-responsiveness and swelling ability of the MSCC-MSCCMC hydrogel toward neutral solvent. Besides, the encapsulation efficiency (50.38–88.91 %) and release (42.88–92.86 %) of L. paracasei BY2 were positively correlated with the concentration of MSCCMC. The higher the encapsulation efficiency was, the higher the release in the target intestine. However, due to the existence of bile salts , controlled-release behavior decreased the survivor rate and physiological state (degrading cholesterol) of encapsulating L. paracasei BY2. Even so, the number of viable cells encapsulated by hydrogels still reached the minimum effective concentration in the target intestine. This study provides an available reference for the practical application of hydrogels fabricated from the cellulose of the Millettia speciosa Champ plant for probiotic delivery.

The purpose of this study was to evaluate the inhibitory mechanism of cell-free supernatants of Lactobacillus plantarum (LCFS) on Proteus mirabilis and estimate the expression of histamine synthesis-related genes. The minimum inhibitory concentration (MIC) of the LCFS and antibacterial curve assay was determined by the tube double-dilution method. The antibacterial mechanism was explored by cell integrity, morphology, and structure of P. mirabilis . Further, the expression of the histidine decarboxylase ( hdcA ) gene and histidine/histamine antiporter ( hdcP ) gene of P. mirabilis and the intracellular and extracellular contents of histidine and histamine were determined. The MIC of the LCFS was 12.5 mg/mL, and the antibacterial curve assay showed an inhibited growth of P. mirabilis . The increase in the Alkaline Phosphatase (AKP) content, membrane permeability, and potential indicated the destruction of the cytoplasm, cell membrane, and cell wall of P. mirabilis . The results of SEM and TEM revealed that the LCFS disrupted the normal morphology and structure of the cell. Moreover, differences in the extracellular and intracellular histidine and histamine content between the MIC group and the control group was consistent with the decreased expression of hdcA and hdcP genes in the MIC group observed in the previous antimicrobial studies. Hence, the inhibitory mechanism of LCFS on histamine formation was clarified at cellular and molecular levels.

Scope Lactobacillus casei F0822-fermented milk has exhibited significant hypocholesterolemic activity in hamsters in the previous study. Under this premise, the objective of this study is to further explore whether bile salt hydrolase (BSH) and S-layer protein (SLP) of the strain have a significant influence on hypocholesterolemic activity of the fermented milk. Methods and results Independent and double interposon mutants of BSH and SLP genes are constructed from wild-type L. casei F0822 via chromosomal insertion of chloramphenicol or/and erythromycin resistance genes based on double-crossover homologous recombination. The mutants- and the wild-type strain-fermented milk is prepared (viable counts of approximately 8.0 × 10 8 colony-forming units mL –1 each) and intragastrically administered to high-cholesterol-fed hamsters once daily at a dose of 1.25 mL d –1 for 28 d, respectively. Both the BSH-deficient mutant- and the SLP-deficient mutant-fermented milk significantly ( p < 0.05) increase serum total and LDL-cholesterol levels in hamsters compared with the wild-type strain-fermented milk. However, only the BSH-deficient mutant-fermented milk could significantly ( p < 0.05) increase hepatic total and esterified cholesterol levels in hamsters. Conclusion Both BSH and SLP have a significant influence on the hypocholesterolemic activity of L. casei F0822-fermented milk in hamsters. Nevertheless, the BSH is greater than the SLP in this regard.
